Abstract
Die Erfindung betrifft ein Verfahren zur (2E)-2-(Hydroxyphenyl)-2-(alkoxyimino)-N-methylacetamiden der Formel (I) in welcher R für einfach bis dreifach durch Fluor subsituiertes oder unsubstituiertes C1-C6-Alkyl steht, durch Umsetzung von Benzofuran-2,3-dion-3-oxim mit Methylamin and anschliessender Alkylierung.
Description
Verfahren zur Herstellung von (2E)-2-(Hvdroxyphenyl)-2-(alkoxyimino)-N- methylacetamiden
Die Erfindung betrifft ein neues Verfahren zur Herstellung von (2E)-2-
(Hydroxyphenyl)-2-(alkoxyimino)-N-methylacetamiden.
Es ist bereits bekannt geworden, dass man 2-(2-(Hydroxyphenyl)-2-(methoxyimino)- N-methylacetamid (C) als ein Gemisch aus E- und Z-Isomeren erhält, wenn man 2- (2-Hydroxyphenyl)-2-(methoxyimino)-essigsäuremethylester (A) oder 1-Benzofuran-
2,3-dion-3-(O-methyloxim) (B), mit Methylamin umsetzt (vergleiche z.B. EP-A 398 692 oder WO 95/24396):

Die Nachteile dieser Verfahren liegen erstens in der schlechten Zugänglichkeit der Ausgangsstoffe, zweitens wird 2-(2-Hydroxyphenyl)-2-(methoxyimino)-N-methyl- acetamid (C) nach diesen Verfahren als ein Gemisch aus E- und Z-Isomeren erhalten.
Es wurde nun gefunden, dass man (2E)-2-(Hydroxyphenyl)-2-(alkoxyimino)-N- methylacetamide der allgemeinen Formel (I),
in welcher
R für einfach bis dreifach durch Fluor substituiertes oder unsubstituiertes Cι-C6-
Alkyl steht,
erhält, wenn man
a) die Verbindung der Formel (II)
mit Methylamin - oder einem Säureadditionskomplex davon -
gegebenenfalls in Gegenwart eines Verdünnungsmittels und gegebenenfalls in Gegenwart eines Säureakzeptors, umsetzt,
und
b) die dabei entstehende Verbindung der Formel (III)
gegebenenfalls ohne Aufarbeitung
mit einem Alkylierungsmittel der Formel (IV),

in welcher
R die oben angegebene Bedeutung hat, und
X für Chlor, Brom, Iod, -O-CO-O-R, -O-SO2-R, oder -O-SO2-O-R steht, wobei R die oben angegebene Bedeutung hat,
gegebenenfalls in Gegenwart eines Verdünnungsmittels und gegebenenfalls in Gegenwart eines Säureakzeptors, gegebenenfalls in Gegenwart eines Phasen- transferkatalysators, umsetzt.
In der Definition von R sind die gesättigten Cj-Cg-Alkyl-Kohlenwasserstoffketten jeweils geradkettig oder verzweigt. Insbesondere steht C Cö-Alkyl für Methyl, Ethyl, n- oder i-Propyl, n-, s- oder i- Butyl, n-Pentyl oder i-Pentyl.
R steht insbesondere für Trifluormethyl, Difluormethyl, Fluormethyl, Methyl, Ethyl oder n-Propyl.
R steht bevorzugt für Methyl, Ethyl oder n-Propyl.
R steht besonders bevorzugt für Methyl.
Die oben aufgeführten allgemein oder in Vorzugsbereichen angegebenen Restedefinitionen gelten sowohl für die Endprodukte der Formel (I) als auch entsprechend für die jeweils zur Herstellung benötigten Ausgangsstoffe bzw. Zwischenprodukte.
Die in den jeweiligen Kombinationen bzw. bevorzugten Kombinationen von Resten im einzelnen für diese Reste angegebenen Restedefinitionen werden unabhängig von der jeweilig angegebenen Kombination der Reste, beliebig auch durch Restedefini- tionen anderer Vorzugsbereiche ersetzt.
Es ist als ausgesprochen überraschend zu bezeichnen, dass beim erfindungsgemäßen Verfahren in Verfahrensschritt a) die Ringöffnung der Verbindung der Formel (II) selektiv unter Bildung der E-Isomere der 2-(2-Hydroxyphenyl)-2-(alkoxyimino)-N- methylacetamide erfolgt, da bei der Ringöffnung des l-Benzofuran-2,3-dion-3-(O- methyloxim) (B) mit Methylamin ein Gemisch aus E- und Z-Isomeren erhalten wird (vergleiche z.B. EP-A 398 692 oder WO 95/24396). Es ist auch überraschend dass bei der Alkylierung der Verbindung der Formel (III) die Alkylierung selektiv am Oximsauerstoff erfolgt.
Das erfindungsgemäße Verfahren a) weist eine Reihe von Vorteilen auf. So lassen sich selektiv die E-Isomeren der 2-(2-Hydroxyphenyl)-2-(alkoxyimino)-N-methyl- acetamide in hohen Ausbeuten und Reinheiten aus leicht zugänglichen Ausgangsstoffen unter technisch völlig problemlosen Bedingungen erhalten.
Die Verbindung der Formel (II) ist bereits bekannt und lässt sich nach bekannten Methoden herstellen (vergleiche z.B. DE-A-2162717, EP-A-711 751).
Das weiterhin zur Durchführung des erfindungsgemäßen Verfahrens im Verfahrens- schritt a) als Ausgangsstoff benötigte Methylamin - oder dessen Säureadditionskomplexe - ist eine kommerziell erhältliche Synthesechemikalie. Es kann sowohl als
Gas, als auch als alkoholische oder wässrige Lösung eingesetzt werden. Bevorzugte Säureadditionskomplexe sind die Hydrochloride, Hydrogensulfate oder Sulfate.
Als weiterhin zur Durchführung des erfindungsgemäßen Verfahrens im Verfahrens- schritt b) als Ausgangsstoff benötigte Alkylierungsmittel der Formel (IV) kommen alle chemischen Verbindungen infrage, die Alkylgruppen übertragen können. Bevorzugt sind Chlormethan, Brommethan, Iodmethan, Dimethylsulfat und Dimethyl- carbonat. Besonders bevorzugt sind Dimethylsulfat und Chlormethan.
Als Verdünnungsmittel zur Durchführung des erfindungsgemäßen Verfahrens a) kommen alle inerten organischen Lösungsmittel in Betracht. Hierzu gehören beispielhaft und vorzugsweise aliphatische, alicyclische oder aromatische Kohlenwasserstoffe, wie beispielsweise Petrolether, Hexan, Heptan, Cyclohexan, Methylcyclo- hexan, Benzol, Toluol, Xylol oder Decalin; halogenierte Kohlenwasserstoffe, wie beispielsweise Chlorbenzol, Dichlorbenzol, Dichlormethan, Chloroform, Tetrachlormethan, Dichlorethan oder Trichlorethan; Ether, wie beispielsweise Diethyl- ether, Diisopropylether, Methyl-t-butylether, Methyl-t-Amylether, Dioxan, Tetra- hydrofuran, 1,2- Dimethoxyethan, 1,2-Diethoxyethan oder Anisol; Nitrile, wie beispielsweise Acetonitril, Propionitril, n- oder i-Butyronitril oder Benzonitril; Ami- de, wie N,N-Dimethylforrnamid, N,N-Dimethylacetamid, N-Methylformanilid, N-
Methylpyrrolidon oder Hexamethylphosphorsäuretriamid; Sulfone, wie Sulfolan; Alkohole, wie Methanol, Ethanol, n- oder i-Propanol, n-, i-, sek- oder tert-Butanol, Ethandiol, Propan-l,2-diol, Ethoxyethanol, Methoxyethanol, Diethylenglykolmono- methylether, Diethylenglykolmonoethylether, deren Gemische mit Wasser oder reines Wasser.
Verfalirensschritt a) des erfindungsgemäßen Verfahrens wird gegebenenfalls in Gegenwart eines geeigneten Säureakzeptors durchgeführt. Als solche kommen alle üblichen anorganischen oder organischen Basen infrage. Hierzu gehören beispielhaft und vorzugsweise Erdalkalimetall- oder Alkalimetallhydroxide, -alkoholate, -acetate,
-carbonate oder -hydrogencarbonate, wie beispielsweise Natriummethanolat, Na-
triumethanolat, Kalium-tert.-butanolat, Natriumhydroxid, Kaliumhydroxid, Natrium- acetat, Kaliumacetat, Natriumcarbonat, Kaliumcarbonat, Kaliumhydrogencarbonat oder Natriumhydrogencarbonat, sowie tertiäre Amine, wie beispielsweise Trimethyl- amin, Triethylamin, Tributylamin, N,N-Dimethylanilin, N,N-Dimethyl-benzylamin, Pyridin, N-Methylpiperidin, N-Methylmorpholin, N,N-Dimethylaminopyridin,
Diazabicyclooctan (DABCO), Diazabicyclononen (DBN) oder Diazabicycloundecen (DBU). Bevorzugt werden in Verfahren a) Kaliumcarbonat, Natriumhydroxid oder Kaliumhydroxid verwendet.
Die Reaktionstemperaturen können bei der Durchführung des Verfahrensschritts a) des erfindungsgemäßen Verfahrens in einem größeren Bereich variiert werden. Im Allgemeinen arbeitet man bei Temperaturen von 15°C bis Rückflusstemperatur, vorzugsweise bei Temperaturen von 20°C bis 40°C, insbesondere bei Temperaturen von 20°C bis 25°C.
Verfahrensschritt a) des erfindungsgemäßen Verfahrens wird im Allgemeinen unter Normaldruck durchgeführt. Es ist jedoch auch möglich, unter erhöhtem oder vermindertem Druck - im Allgemeinen zwischen 0,1 bar und 10 bar - zu arbeiten.
Zur Durchführung des Verfahrensschritts a) des erfindungsgemäßen Verfahrens zur
Herstellung der Verbindungen der Formel (I) setzt man pro Mol der Verbindung der Formel (II) im Allgemeinen 1 bis 4 Mol, vorzugsweise 1 bis 2 Mol Methylamin ein.
Zur Durchführung des Verfahrensschritts a) des erfindungsgemäßen Verfahrens zur Herstellung der Verbindungen der Formel (I) geht man im Allgemeinen wie folgt vor. Methylamin wird zu der Verbindung der Formel (II), vorzugsweise in Gegenwart eines Verdünnungsmittels gegeben. Man rührt wird in einem Verdünnungsmittel gegebenenfalls bei erhöhter Temperatur bis die Umsetzung vollständig ist. Nach beendeter Reaktion wird in üblicher Weise aufgearbeitet.
Als Verdünnungsmittel zur Durchführung des erfindungsgemäßen Verfahrens kommen in Verfahrensschritt b) alle inerten organischen Lösungsmittel in Betracht. Hierzu gehören beispielhaft und vorzugsweise aliphatische, alicyclische oder aromatische Kohlenwasserstoffe, wie beispielsweise Petrolether, Hexan, Heptan, Cyclohexan, Methylcyclohexan, Benzol, Toluol, Xylol oder Decalin; halogenierte Kohlenwasserstoffe, wie beispielsweise Chlorbenzol, Dichlorbenzol, Dichlormethan, Chloroform, Tetrachlormethan, Dichlorethan oder Trichlorethan; Ether, wie beispielsweise Di- ethylether, Diisopropylether, Methyl-t-butylether, Methyl-t-Amylether, Dioxan, Tetrahydrofuran, 1,2- Dimethoxyethan, 1 ,2-Diethoxyethan oder Anisol; Ketone, wie beispielsweise Aceton, Butanon, Methyl-isobutylketon oder Cyclohexanon; Nitrile, wie beispielsweise Acetonitril, Propionitril, n- oder i-Butyronitril oder Benzonitril; Amide, wie N,N-Dimethylformamid, N,N-Dimethylacetamid, N-Methylformanilid, N-Methylpyrrolidon oder Hexamethylphosphorsäuretriamid; Ester wie Essigsäuremethylester oder Essigsäureethylester; Sulfoxide, wie Dimethylsulfoxid; Sulfone, wie Sulfolan; Alkohole, wie Methanol, Ethanol, n- oder i-Propanol, n-, i-, sek- oder tert-Butanol, Ethandiol, Propan-l,2-diol, Ethoxyethanol, Methoxyethanol, Diethylen- glykolmonomethylether, Diethylenglykolmonoethylether, deren Gemische mit Wasser oder reines Wasser.
Der Verfahrensschritt b) des erfindungsgemäßen Verfahrens wird gegebenenfalls in
Gegenwart eines geeigneten Säureakzeptors durchgeführt. Als solche kommen alle üblichen anorganischen oder organischen Basen infrage. Hierzu gehören beispielhaft und vorzugsweise Erdalkalimetall- oder Alkalimetallhydroxide, -alkoholate, -acetate, -carbpnate oder -hydrogencarbonate, wie beispielsweise Natriummethanolat, Na- triumethanolat, Kalium-tert.-butanolat, Natriumhydroxid, Kaliumhydroxid, Natrium- acetat, Kaliumacetat, Natriumcarbonat, Kaliumcarbonat, Kaliumhydrogencarbonat oder Natriumhydrogencarbonat, sowie tertiäre Amine, wie beispielsweise Trimethyl- amin, Triethylamin, Tributylamin, N,N-Dimethylanilin, N,N-Dimethyl-benzylamin, Pyridin, N-Methylpiperidin, N-Methylmorpholin, N,N-Dimethylaminopyridin, Diazabicyclooctan (DABCO), Diazabicyclononen (DBN) oder Diazabicycloundecen
(DBU). Bevorzugt werden in Verfahrensschritt b) Kaliumcarbonat, Natriumhydroxid oder Kaliumhydroxid verwendet.
Der Verfahrensschritt b) des erfindungsgemäßen Verfahrens wird gegebenenfalls in Gegenwart eines geeigneten Phasentransferkatalysators durchgeführt. Hierzu gehören beispielhaft und vorzugsweise quartäre Ammoniumsalze, wie beispielsweise Tetra- butylammoniumbromid, -chlorid, -hydrogensulfat oder -sulfat, Methyl-trioctyl- ammoniumbromid oder -chlorid, -hydrogensulfat oder -sulfat oder 4-Dimethyl- amino-N-(2-ethyl-hexyl)pyridiniumbromid oder -chlorid, -hydrogensulfat oder -sul- fat, quartäre Phosphoniumsalze, wie beispielsweise Tributyl-tetradecyl-phospho- niumbromid oder -chlorid, Tetraphenylphosphoniumbromid oder -chlorid, Kronenether, wie beispielsweise Dibenzo-18-krone-6, Guanidiniumsalze, wie beispielsweise Hexaalkylguanidiniumchlorid, sowie Polyethylenglycolderivate. Bevorzugt wird in Verfahrensschritt b) Tetrabutylammoniumbromid oder Tetrabutylammoniumchlorid verwendet.
Die Reaktionstemperaturen können bei der Durchführung des Verfahrensschritts b) des erfindungsgemäßen Verfahrens in einem größeren Bereich variiert werden. Im Allgemeinen arbeitet man bei Temperaturen von -20°C bis 80°C, vorzugsweise bei Temperaturen von 0°C bis 60°C.
Der Verfahrensschritt b) des erfindungsgemäßen Verfahrens wird im Allgemeinen unter Normaldruck durchgeführt. Es ist jedoch auch möglich, unter erhöhtem oder vermindertem Druck - im Allgemeinen zwischen 0,1 bar und 10 bar - zu arbeiten.
Zur Durchführung des Verfahrensschritts b) des erfindungsgemäßen Verfahrens zur Herstellung der Verbindungen der Formel (I) setzt man pro Mol der Verbindung der Formel (III) im Allgemeinen 1 bis 15 Mol, vorzugsweise 1 bis 4 Mol Alkylierungsmittel der Formel (IV) ein.
Zur Durchführung des Verfahrensschritts b) des erfindungsgemäßen Verfahrens geht man im Allgemeinen wie folgt vor. Die Verbindung der Formel (III) wird vorzugsweise in Gegenwart eines Verdünnungsmittels mit einer Base und gegebenenfalls einem Phasentransferkatalysators versetzt. Hierzu gibt man das Alkylierungsmittel der Formel (IV) und rührt, gegebenenfalls bei erhöhter Temperatur bis die Umsetzung vollständig ist. Nach beendeter Reaktion wird in üblicher Weise aufgearbeitet.
Das erfindungsgemäße Verfahren wird insbesondere zur Herstellung von (2E)-2- (Hydroxyphenyl)-2-(methoxyimino)-N-methylethanamid verwendet, das ein wichtiges Zwischenprodukt für die Herstellung von Schädlingsbekämpfungsmitteln ist (EP-A-937 050). Nach dem erfindungsgemäßen Verfahren wird von (2E)-2-(Hy- droxyphenyl)-2-(methoxyimino)-N-methylethanamid in konstant hohen Reinheiten und guten Ausbeuten erhalten. Das neue Verfahren erleichtert daher die Herstellung von bekannten Schädlingsbekämpfungsmitteln.
Die nachfolgenden Beispiele dienen zur Erläuterung der Erfindung. Die Erfindung ist jedoch nicht auf die Beispiele limitiert.
Herstellungsbeispiele:
Beispiel 1
(2E)-2-(Hydroxyimino)-2-(2-hydroxyphenyl)-N-methylethanamid:
Verfahrensschritt a)
5.0 g Benzofuran-2,3-dion-3-oxim werden in 20 ml Methanol gelöst und bei Raum- temperatur mit 3.4 g Methylamin (40 %ig in Methanol) innerhalb von 15 Minuten versetzt. Anschließend rührt man 2 Stunden bei Raumtemperatur und engt den Ansatz bis zur Trockene ein. Man erhält (2E)-2-(Hydroxyimino)-2-(2-hydroxy- phenyl)-N-methylethanamid (GC: 98.8 %) in quantitativer Ausbeute.
(2E)-2-(Hydroxyphenyl)-2-(methoxyimino)-N-methylethanamid:
Verfahrensschritt b)
5.0 g (2E)-2-(Hydroxyimino)-2-(2-hydroxyphenyl)-N-methylethanamid und 7.1 g
Pottasche werden in 45 ml Aceton vorgelegt und bei Raumtemperatur innerhalb von 4 Stunden mit einer Lösung aus 3.6 g Dimethylsulfat und 25 ml Aceton versetzt. Man rührt 1.5 Stunden bei Raumtemperatur nach, destilliert das Lösungsmittel ab,
versetzt mit 150 ml Wasser, stellt mit Salzsäure pH 6-7 ein und extrahiert dreimal mit Methylenchlorid. Die vereinigten organischen Extrakte werden mit Wasser gewaschen, mit Natriumsulfat getrocknet und anschließend bis zur Trockene eingeengt. Man erhält 4.7 g (2E)-2-(Hydroxyphenyl)-2-(methoxyimino)-N- methylethanamid (HPLC: 83.6 %; Ausbeute: 73.3 % d. Th.).
Claims
Verfahren zur Herstellung von Verbindungen der allgemeinen Formel (I),
in welcher
R für einfach bis dreifach durch Fluor substituiertes oder unsubstituiertes
Cι-C6- Alkyl steht,
dadurch gekennzeichnet, dass man
a) die Verbindung der Formel (II)
mit Methylamin - oder einem Säureadditionskomplex davon -
gegebenenfalls in Gegenwart eines Verdünnungsmittels und gegebenenfalls in Gegenwart eines Säureakzeptors, umsetzt,
und
b) die dabei entstehende Verbindung der Formel (III)
gegebenenfalls ohne Aufarbeitung
mit einem Alkylierungsmittel der Formel (IV),
R-X (IV)
in welcher
R die oben angegebene Bedeutung hat, und
X für Chlor, Brom, Iod, -O-CO-O-R, -O-SO2-R, oder -O-SO2- O-R steht, wobei R die oben angegebene Bedeutung hat,
gegebenenfalls in Gegenwart eines Verdünnungsmittels und gegebenenfalls in Gegenwart eines Säureakzeptors, gegebenenfalls in Gegenwart eines Phasentransferkatalysators, umsetzt.
2. Verfahren gemäß Anspruch 1, dadurch gekennzeichnet, dass die Verfahrensschritte a) und b) als Eintopfverfahren durchgeführt werden.
3. Verfahren gemäß mindestens einem der Ansprüche 1 bis 2, dadurch gekenn- zeichnet, dass im Verfahrensschritt b) als Alkylierungsmittel der Formel (IV)
Dimethylsulfat oder Methylchlorid verwendet wird.
4. Verfahren gemäß mindestens einem der Ansprüche 1 bis 3, dadurch gekennzeichnet, dass der Verfahrensschritt a) bei Temperaturen von 20°C bis Rück- flusstemperatur durchgeführt wird.
5. Verfahren gemäß mindestens einem der Ansprüche 1 bis 4, dadurch gekennzeichnet, dass der Verfahrensschritt b) bei Temperaturen von -20°C bis 80°C durchgeführt wird.
6. Verfahren gemäß mindestens einem der Ansprüche 1 bis 5, dadurch gekenn- zeichnet, dass zur Durchführung des Verfahrensschritts a) zur Herstellung der
Verbindungen der Formel (I) pro Mol der Verbindung der Formel (II) 1 bis 4 Mol Methylamin eingesetzt werden.
7. Verfahren gemäß mindestens einem der Ansprüche 1 bis 6, dadurch ge- kennzeichnet, dass zur Durchführung des Verfahrensschritts b) zur Herstellung der Verbindungen der Formel (I) setzt pro Mol der Verbindung der Formel (III) 1 bis 15 Mol Alkylierungsmittel der Formel (IV) eingesetzt werden.
8. Verfahren gemäß mindestens einem der Ansprüche 1 bis 7, dadurch gekennzeichnet, dass R für Methyl steht.
