Abstract
Die Erfindung betrifft die Verwendung von 2-[4-({[(2R)-8-Isopropoxy-chroman-2-yl]methyl}amino)butyl]-1,2-benzisothiazol-3(2H)-on 1,1-dioxid, dessen physiologisch unbedenklichen Salzen, Hydraten und/oder Solvaten, insbesondere dessen Hydrochlorid, zur Herstellung eines Arzneimittels zur Prophylaxe und/oder Behandlung der Parkinsonschen Krankheit.
Description
Verwendung von Chromanen
Die Erfindung betrifft die Verwendung von 2-[4-({[(2R)-8-Isopropoxy-chroman-2- yl]methyl}amino)butyl]-l,2-benzisothiazol-3(2H)-on 1,1-dioxid, dessen physiologisch unbedenklichen Salzen, Hydraten und/oder Solvaten, insbesondere dessen Hy- drochlorid, zur Herstellung eines Arzneimittels zur Prophylaxe und/oder Behandlung der Parkinsonschen Krankheit.
Die Parkinsonschen Krankheit ist eine chronische, progressive Erkrankung des zentralen Nervensystems. Sie wird verursacht durch die Degeneration dopaminerger Neurone in der Substantia nigra, welche den Neurotransmitter Dopamin produzieren und freisetzen. Die daraus resultierende Verringerung der dopaminergen Neuro- transmission führt zu massiven Dysfunktionen des extrapyramidalen Systems der Bewegungskontrolle. Diese Störungen betreffen nicht nur die Basalganglien sondern auch andere eng verknüpfte Gehirnareale.
Die Ätiologie des idiopathischen Parkinsonismus ist immer noch weitgehend unbekannt. Zunehmende Evidenzen deuten jedoch darauf hin, dass der Zelltod dopaminerger Neurone der Substantia nigra durch Apoptose in Folge mitochondrialer
Fehlfunktionen zustande kommt. Neben möglichen genetischen Störungen, werden auch erhöhte Glutamatspiegel und/oder eine defiziente Versorgung mit neurotrophen Faktoren als Ursache für die mitochondrialen Fehlfunktionen diskutiert.
Davon ausgehend sollte ein weiter fortschreitender neuronaler Zelltod durch eine neuroprotektive pharmakologische Beeinflussung der neurodegenerativen Prozesse zu verhindern sein, womit die Progression der Erkrankung aufgehalten werden könnte ohne notwendigerweise mit den kausalen pathophysiologischen Mechanismen zu interagieren.
Es ist gezeigt worden, dass eine Stimulation neuronaler 5-HTiA-Rezeptoren in verschiedenen in vitro- und in vivo- Systemen sowohl neuroprotektive, als auch anti- apoptotische und neurotrophe Effekte hat. Eine Stimulation von 5-HTIA- Rezeptoren könnte demnach auch die weitere Degeneration dopaminerger Neurone in der Parkinsonschen Krankheit verhindern und damit letztendlich die Progression der
Erkrankung verzögern.
Die derzeit klinisch verwendeten Therapeutika für Parkinsonsche Krankheit verfolgen in der Mehrzahl einen rein symptomatischen Ansatz. Ziel dieser Therapien ist, entweder direkte Substitution des fehlenden Dopamins durch ein Dopaminvor- läufermolekül (L-DOPA), das im Körper zu Dopamin metabolisiert wird, oder aber Stimulation defizitärer dopaminerger Neurotransmissionsprozesse mittels Agonisten an Dopaminrezeptoren oder durch Verminderung des Dopaminabbaus (MAO- Inhibitoren, COMT-Inhibitoren). Alle derzeitigen Therapien sind jedoch durch starke Nebenwirkungen (z.B. Dyskinesien, Psychosen, Schlafstörungen) oder langfristigen
Wirkungsverlust gekennzeichnet.
Chroman-Derivate und speziell 2-[4-({[(2R)-8-Isopropoxy-chroman-2-yl]methyl}- amino)butyl]-l,2-benzisothiazol-3(2H)-on 1,1-dioxid Hydrochlorid und deren agonistische Wirkung am 5-HTIA- Rezeptor sind als Mittel zur Behandlung von
Erkrankungen des Zentralen Nervensystems aus der EP-A-0 352 613 bzw. der EP-A- 0 749 970 bekannt.
Die WO 99/26621 beschreibt Chroman-Derivate, insbesondere 2-[4-({[(2R)- Chroman-2-yl]methyl}amino)butyl]-l,2-benzisothiazol-3(2H)-on 1,1-dioxid Hydrochlorid (genetischer Name: Repinotan Hydrochlorid), als Mittel zur Förderung der Neuroregeneration bei neurologischen Erkrankungen wie beispielsweise der Parkinsonschen Krankheit.
Überraschenderweise wurde nun mit 2-[4-({[(2R)-8-Isoproρoxy-chroman-2-yi]- methyl}amino)butyl]-l,2-benzisothiazol-3(2H)-on 1,1-dioxid ein 5-HT1A- Rezeptor-
Agonist gefunden, der nicht nur neuroprotektiv wirkt, sondern zusätzlich auch eine symptomatische Wirksamkeit aufweist und so auf doppelte Weise den Verlauf der Parkinsonschen Erkrankung positiv beeinflusst.
Gegenstand der Erfindung ist daher die Verwendung von 2-[4-({[(2R)-8-Isoproρoxy- chroman-2-yl]methyl}amino)butyl]-l,2-benzisothiazol-3(2H)-on 1,1-dioxid, dessen physiologisch unbedenklichen Salzen, Hydraten und/oder Solvaten insbesondere 2- [4-({[(2R)-8-Isopropoxy-chroman-2-yl]methyl}amino)butyl]-l,2-benzisothiazol- 3(2H)-on 1,1-dioxid Hydrochlorid, zur Herstellung eines Arzneimittels zur Prophylaxe und/oder Bekämpfung der Parkinsonschen Krankheit sind.
2-[4-({[(2R)-8-Isopropoxy-chroman-2-yl]methyl}amino)butyl]-l,2-benzisothiazol- 3(2H)-on 1,1-dioxid hat folgende Struktur:
Physiologisch unbedenkliche Salze der erfindungsgemäß verwendeten Verbindungen können Salze der Verbindungen mit Mineralsäuren, Carbonsäuren oder Sulfonsäuren sein. Besonders bevorzugt sind z.B. Salze mit Chlorwasserstoffsäure, Bromwasser- stoffsäure, Schwefelsäure, Phosphorsäure, Methansulfonsäure, Ethansulfonsäure,
Toluolsulfonsäure, Benzolsulfonsäure, Naphthalindisulfonsäure, Essigsäure, Propion- säure, Milchsäure, Weinsäure, Oxalsäure, Zitronensäure, Fumarsäure, Maleinsäure oder Benzoesäure.
Hydrate im Sinne der Erfindung sind stöchiometrische Zusammensetzungen von 2-[4-
( {[(2R)-8-Isopropoxy-chroman-2-yl]methyl} amino)butyl]-l ,2-benzisothiazol-3(2H)- on 1,1-dioxid oder seinen Salzen mit Wasser.
Solvate im Sinne der Erfindung sind stöchiometrische Zusammensetzungen von 2-[4- ({[(2R)-8-Isopropoxy-chroman-2-yl]methyl}amino)butyl]-l,2-benzisothiazol-3(2H)- on 1,1-dioxid oder seinen Salzen mit Lösungsmittel.
Die erfindungsgemäß verwendeten Verbindungen können nach den in der EP-A- 0 749 970 angegebenen Verfahren hergestellt werden. Beispielsweise entspricht 2-[4- ( { [(2R)-8-Isopropoxy-chroman-2-yl]methyl} amino)butyl]- 1 ,2-benzisothiazol-3 (2H)- on 1,1-dioxid Hydrochlorid (im folgenden Beispiel 11 genannt) in der EP-A- 0 749 970 dem Beispiel 7.
Die Salze des 2-[4-({[(2R)-8-Isopropoxy-chroman-2-yl]methyl}amino)butyl]-l,2- benzisothiazol-3(2H)-on 1,1-dioxids können erhalten werden, indem man die freie Base in einem geeignete Lösemittel mit stöchiometrischen oder überstöchiome- trischen Mengen der dem Salz zugrundeliegenden Säure in einem Temperaturbereich von 0°C bis zum Siedepunkt des Lösemittels umsetzt. Geeignete Lösemittel sind beispielsweise Wasser, aliphatische Alkohole wie Methanol, Ethanol, oder 2- Propanol, aliphatische offenkettige oder cyclische Ether wie Diethylether, tert.Butyl- methylether, Dioxan, Tetrahydrofuran oder aliphatische Ketone wie 2-Propanon, 2- Butanon, sowie deren Gemische. Die Salze werden direkt aus dieser Mischung, gegebenenfalls nach teilweisem oder vollständigem Abdestillieren des Lösemittels, als Feststoff gewonnen; sie können durch Umkristallisation oder Umfällung in zum Beispiel oben aufgeführten Lösemitteln oder deren Gemischen gereinigt werden.
Der Wirkstoff kann systemisch und/oder lokal wirken. Zu diesem Zweck kann er auf geeignete Weise appliziert werden, wie z.B. oral, parenteral, pulmonal, nasal, sublingual, lingual, buccal, rectal, transdermal, conjunctival, otisch oder als Implantat. Vorzugsweise erfolgt die Applikation oral.
Für diese Applikationswege kann der Wirkstoff in geeigneten Applikationsformen verabreicht werden.
Für die orale Applikation eignen sich bekannte, den Wirkstoff schnell und/oder modifiziert abgebende Applikationsformen, wie z.B. Tabletten (nichtüberzogene sowie überzogene Tabletten, z.B. magensaftresistente Überzüge), Kapseln, Dragees, Granulate, Pellets, Pulver, Emulsionen, Suspensionen und Lösungen.
Die parenterale Applikation kann unter Umgehung eines Resorptionsschrittes geschehen (intravenös, intraarteriell, intrakardial, intraspinal oder intralumbal) oder unter Einschaltung einer Resorption (intramuskulär, subcutan, intracutan, percutan, oder intraperitoneal). Für die parenterale Applikation eignen sich als Applikations- formen u.a. Injektions- und Infusionszubereitungen in Form von Lösungen, Suspensionen, Emulsionen, Lyophilisaten und sterilen Pulvern.
Für die sonstigen Applikationswege eignen sich z.B. Inhalationsarzneiformen (u.a. Pulverinhalatoren, Nebulizer), NasentropferJ-lösungen, Sprays; lingual, sublingual oder buccal zu applizierende Tabletten oder Kapseln, Suppositorien, Ohren- und
Augenpräparationen, Vaginalkapseln, wässrige Suspensionen (Lotionen, Schüttelmixturen), lipophile Suspensionen, Salben, Cremes, Milch, Pasten, Streupuder oder Implantate.
Die Wirkstoffe können in an sich bekannter Weise in die angeführten Applikationsformen überführt werden. Dies geschieht unter Verwendung inerter nichttoxischer, pharmazeutisch geeigneter Hilfsstoffe. Hierzu zählen u.a. Trägerstoffe (z.B. mikrokristalline Cellulose), Lösungsmittel (z.B. flüssige Polyethylenglycole), Emul- gatoren (z.B. Natriumdodecylsulfat), Dispergiermittel (z.B. Polyvinylpyrrolidon), synthetische und natürliche Biopolymere (z.B. Albumin), Stabilisatoren (z.B.
Antioxidantien wie Ascorbinsäure), Farbstoffe (z.B. anorganische Pigmente wie Eisenoxide) oder Geschmacks- und / oder Geruchskorrigentien.
Im allgemeinen hat es sich als vorteilhaft erwiesen, bei parenteraler Applikation Mengen von etwa 0,001 bis 30 mg/kg, vorzugsweise etwa 0,01 bis 10 mg/kg
Köφergewicht zur Erzielung wirksamer Ergebnisse zu verabreichen. Bei oraler
Applikation beträgt die Menge etwa 0,01 bis 100 mg/kg, vorzugsweise etwa 0,1 bis 30 mg/kg Köφergewicht.
Trotzdem kann es gegebenenfalls erforderlich sein, von den genannten Mengen abzuweichen, und zwar in Abhängigkeit von Köφergewicht, Applikationsweg, individuellem Verhalten gegenüber dem Wirkstoff, Art der Zubereitung und Zeitpunkt bzw. Intervall, zu welchem die Applikation erfolgt.
Bestimmung der agonistisehen Wirkung an humanen rekombinanten 5-HTIA- Rezeptoren
An humanen rekombinanten 5-HTιA-Rezeptoren wurden Signaltransduktionsstudien durchgeführt unter Verwendung der Guanosin-5'-O-(3-[S-35]thio)-triphosphat (GTP gamma[S-35]) Bindungs-Technik (modifiziert nach Elliott und Reynolds Europ J. Pharmacol 1999, 386, 313-315 und Sim et al. J Neurosci 1996, 16, 8057-8066).
Repinotan Hydrochlorid und Beispiel 11 erzielten in diesem Test EC50 -Werte von 0,51 nM bzw. 0,19 nM, das heisst, beide Substanzen sind 5-HT1A- Agonisten, wobei Beispiel 11 etwa zweifach potenter als Repinotan Hydrochlorid wirkt.
MPTP-Affen- Modell
Die in vivo- Wirkung von Repinotan Hydrochlorid und von Beispiel 11 wurde in einem Affenmodell für die Parkinsonsche Krankheit, dem sogenannten chronischen MPTP-Modell (Bezard et al. Brain Res. 1997, 766, 107-112.), getestet. MPTP (=1- methyl-4-phenyl-l,2,3,6-tetrahydropyridine) ist ein Neurotoxin, das in Menschen und Tieren die für die Parkinsonsche Krankheit typische Degeneration dopaminerger Neurone in der Substantia nigra verursacht. Darüber hinaus erzeugt MPTP im Menschen und im Affen die für Parkinsonsche Krankheit typischen Motorsymptome. Diese Symptome werden auf einer Parkinsonskala für Affen bewertet.
Für die Experimente wurden Rhesusaffen (Macaca fascicularis) täglich mit MPTP (0,2mg/kg i.v.) behandelt, bis sie auf der Parkinsonskala einen Score von 8 erreicht hatten. Die ersten Parkinson-Symptome treten nach 5-10 Tagen MPTP-Behandlung auf. Aufgrund der Langzeitwirkung des Neurotoxins entwickeln sich die klinischen Symptome der Tiere weiter bis zum vollen Parkinsonismus (score>15). Fünf Gruppen von Tieren wurden untersucht: die erste erhielt nur MPTP, die zweite erhielt MPTP plus Repinotan Hydrochlorid (2mg/kg p.o. bid), die dritte erhielt Beispiel 11 (lmg/kg p.o. bid). Die Behandlung mit Repinotan Hydrochlorid und Beispiel 11 begann jeweils ab dem Tag, an dem die Tiere zum ersten Mal klinische Symptome zeigten. Sowohl Repinotan Hydrochlorid als auch Beispiel 11 waren nach oraler Gabe neuroprotektiv wirksam, das heißt, beide Substanzen verlangsamten die Entwicklung der Parkinsonismus-Symptome in diesem Affenmodell. Völlig überraschend war aber die Beobachtung, dass Beispiel 11 darüber hinaus auch den Schweregrad der Symptome verringerte, also eine symptomatische Wirkung hatte (22% Reduktion gegenüber Kontrolle). Eine solche symptomatische Wirkung wurde dagegen mit Repinotan Hydrochlorid nicht beobachtet (vgl. Tabelle 1).

Herstellungsbeispiele
Beispiel 1
2-Hy droxy-3 -methoxy-benzonitril
Eine Suspension von 6375 g (41,94 mol) o-Vanillin, 3823 g (55 mol) Hydroxylamin- Hydrochlorid und 6375 g (93,75 mol) Natriumformiat in 14 1 Ameisensäure wurde unter Rühren auf ca. 90 bis 95°C erhitzt. In diesem Temperaturbereich setzten verstärkte Gasentwicklung und exotherme Reaktion ein (Heizung wurde abgestellt). Die exotherme Reaktion hielt ca. 10 bis 15 Minuten an (Temperaturanstieg auf ca. 115°C). Danach wurde noch 45 Minuten unter Rückfluß gerührt. Nach Beendigung der Reaktion wurde das Gemisch auf ca. +6°C abgekühlt und in ein Gemisch aus
6 kg Eis und 25 1 Wasser eingerührt. Nach 1 h wurde abgesaugt und mit ca. 12 1 Wasser gewaschen. Danach wurde 24 h bei Raumtemperatur im Frischluft- Trockenschrank und 120 h über P2O5 im Vakuum-Trockenschrank (Raumtemperatur) getrocknet.
Ausbeute: 4816 g (77 %) Kristalle, Schmp. 54°C, Rf: 0,34 (Toluol-Essigester 3:1)
Beispiel 2
2-Hydroxy-3-methoxy-acetophenon
In einem trockenen mit Stickstoff gespülten Reaktionsgefäß wurden 750 g (30,8 mol) Magnesium-Späne und 3 g Jod vorgelegt. Dazu wurden 10 1 Methoxybenzol gegeben und man erwärmte unter langsamen Rühren auf 40°C. Das Rühren wurde unter- brachen und man gab 25 ml Methyljodid und einen Startansatz1) direkt in die
Magnesiumspäne. Nach dem Anspringen der Reaktion wurde das Rührwerk wieder angestellt, und man gab unter mäßiger Kühlung eine Lösung von 1916 ml (30,8 mol) Methyljodid in 2,5 1 Methoxybenzol so zu, dass eine Temperatur von 40 bis 43°C gehalten werden konnte (1,5 h). Anschließend wurde noch 5 h bei 40°C und 15 h bei Raumtemperatur gerührt. Man kühlte auf +5°C, ließ innerhalb von 1,5 h eine Lösung von 1840 g (12,3 mol) 2-Hydroxy-3-methoxy-benzonitril in 6,5 1 Methoxybenzol zulaufen und rührte 1,5 h bei 40°C nach. Nach Beendigung der Umsetzung (DC- Kontrolle / Toluol-Essigester 3:1) wurde das Reaktionsgemisch auf +10°C gekühlt und in ein Gemisch aus 24 kg Eis und 8 1 Wasser eingerührt. Anschließend säuerte man durch Zugabe von 12 1 6N Salzsäure an, wobei 0 bis 5°C nicht überschritten wurden. Die organische Phase wurde abgetrennt und mit 2,5 1 6N Salzsäure gewaschen. Die vereinigten wässrigen Phasen wurden 3x mit je 41 Toluol extrahiert. Dann wurde die wässrige Phase 1,5 h bei einer Innentemperatur von 98°C gerührt. Man stellte die Heizung ab und gab 6 kg Natriumchlorid zu und rührte über Nacht bei Raumtemperatur nach.
Nach längerem Kühlen auf +5°C erhielt man einen Niederschlag von ockerfarbenen Kristallen. Diese wurde abgesaugt, mit 4 1 Eiswasser nachgewaschen (2x) und 5 Tage (120 h) im Vakuumtrockenschrank über P2O5 und NaOH-Plätzchen getrocknet.
Ausbeute: 1587 g (78 %), Schmp. 53°C, Rf: 0.33 (ToluokEssigester 9:1)
als "Startansatz" diente dieselbe Reaktion im 1 mol-Maßstab
Beispiel 3
8-Methoxy-chromon-2-carbonsäure-ethylester
Zu einer gekühlten Lösung von 1024 g (15,04 mol) Natriumethylat in 20 1 Ethanol gab man bei 50°C eine weitgehend gelöste Mischung aus 1140 g (6,86 mol) 2- Hydroxy-3-methoxy-acetophenon, 2 1 (14,86 mol) Diethyloxalat und 7 1 Ethanol rasch zu. Man erwärmte für 3 h zum Rückfluß. Man kühlte auf 50°C, gab 2 1 konz.
Salzsäure zu und erhitzte 30 min zum Rückfluß. Danach kühlte man auf 50°C, saugte ab, wusch den Filterrückstand mit Ethanol und engte das Filtrat im Rotationsverdampfer ein.
Der gelbe kristalline Rückstand wurde in 15 1 Dichlormethan gelöst und mit 14 1
10 %iger NaHCθ3-Lösung 30 Minuten gut durchgerührt. Die organische Phase wurde abgetrennt, mit 2 1 10 %iger NaHC03 -Lösung gewaschen und über einem Gemisch aus 2 kg Na2S04 und 1 kg Tonsil getrocknet. Danach wurde über Kieselgur abgesaugt, mit Dichlormethan nachgewaschen und das Filtrat im Rotationsver- dampfer eingeengt. Der Rückstand wurde im Rotationsverdampfer mit 2,5 1 Petrol- ether versetzt und 15 min. bei Raumtemperatur verrührt, auf +5°C gekühlt und abgesaugt. Die blaßgelben Kristalle wurden 2 h im Frischluft-Trockenschrank und 24 h im Vakuumtrockenschrank über P2O5 getrocknet.
Ausbeute: 1304 g (76 %) Kristalle, Schmp. 129-130°C, Rf.: 0,53 (ToluohEssig- ester = 85:15)
Beispiel 4
8-Methoxy-chroman-2-carbonsäure-ethylester
Ein Gemisch von 5,30 kg (22,45 mol) 8-Methoxy-chromon-2-carbonsäureethylester in 100 1 Ethylacetat und 50 1 Eisessig wurden in Gegenwart von 500 g 10 %ig Pd/C 24 h bei 50°C unter einem Druck von 3 bar Wasserstoff hydriert. Zur Aufarbeitung wurde die Reaktionslösung über Kieselgur abgesaugt und das Filtrat im Rotationsverdampfer eingeengt. Um Reste vom Eisessig azeotrop zu entfernen, wurde der Kolbeninhalt 2x mit 6 1 Toluol versetzt und eingeengt. Nach Trocknen des Rückstandes im Dampfstrahlvakuum (8 h/8 mm) erhielt man das Produkt als dunkles Öl.
Ausbeute: 5,002 kg (94 %) Öl, Sdp. 110-114°C/0,04 mm, Rf: 0,42 (Toluol: Ethylacetat 3:1)
Beispiel 5
(R)-2-Hydroxymethyl-8-methoxy-chroman
Zu 540 g 65 % RedAl in Toluol wurden 235 g (R)-8-Methoxy-chroman-2-carbon- säureethylester (erhalten nachdem in der DE-A 4430089 angegebenen Verfahren aus (R,S)-8-Methoxy-chroman-2-carbonsäureethylester) in Toluol langsam zugetropft; insgesamt wurden 1,5 1 Toluol verwendet. Nach 18 h Rühren bei Raumtemperatur wurde zunächst für 1 h auf 50°C, anschließend für eine weitere Stunde auf 80°C erhitzt. Nach Abkühlen auf Raumtemperatur wurden unter externer Kühlung 100 g Eis zugesetzt, gefolgt von 600 ml 15 % Kalium-Natrium-Tartratlösung. Die Mischung wurde mit 500 ml Toluol und 500 ml Essigester verdünnt. Die organische Phase wurde abgetrennt, über Magnesiumsulfat getrocknet und durch Zusatz von Tonsil geklärt. Nach Aufkonzentrieren auf ein Volumen von ca. 500 ml wurde 1 1 Cyclohexan zugesetzt und 30 min bei Raumtemperatur gerührt. Der ausgefallene Feststoff wurde abgesaugt, nachgewaschen und getrocknet. Man erhielt so 135,5 g Zielverbindung.

Beispiel 6
(R)-8-Hydroxy-2-hydroxymethylchroman
135,5 g (0.7 Mol) (-)-2-Hydroxymethyl-8-methoxychroman werden 20 Stunden in 0,7 1 48 %ige wäßriger HBr-Lösung erhitzt. Nach Abkühlen und Verdünnen mit 1,2 1 Eiswasser wird 30 Minuten gerührt, und der ausgefallenen Niederschlag abgesaugt. Waschen mit Eiswasser und Trocknen über Phosphoφentoxid liefern 109,5 g (87 %) der Titelverbindung, Schmp. 131-132°C. α20 289 = -133,8 (c = 0,7 Methanol)
Beispiel 7
(R)-2-Hydroxymethyl-8-isopropoxy-chroman
4,5 g (25 mMol) (R)-8-Hydroxy-2-hydroxymethyl-chroman, 4,6 g (27 mMol) 2-Iod- propan und 5,2 g (37,5 mMol) gepulvertes Kaliumcarbonat in 50 ml Dimethylformamid werden 40 Stunden auf 60°C erhitzt. Nach Zugabe weiterer 0,9 g lodpropan wird 24 Stunden auf 80°C und anschließend weitere 24 Stunden auf 95°C erhitzt. Nach Ab-
kühlen wird zwischen Toluol/Wasser verteilt und über Celite® filtriert. Die orgamsche Phase wird getrocknet (Magnesiumsulfat) und eingeengt. Nach Flashchromatographie (Kieselgel; Elution mit Toluol/Essigester-Gradienten 3:1 - 2:1) erhält man 7 g Rohprodukt, das durch Chromatographie auf Kieselgel (Gradient Toluol/Essigester 1 :0 - 8:1) gereinigt wird. Ausbeute: 2,9 g (52%) Öl. RF (Kieselgel, Toluol/Essigester 1:1): 0,4

Beispiel 8
(R)-8-Isopropoxy-2-mesyloxymethyl-chroman
Zu 114 g (0,51 Mol) der Verbindung aus Beispiel 7 in 95 g Pyridin werden bei Raumtemperatur 68 g (0,6 Mol) Methansulfonsäurechlorid getropft. Nach 18 Stunden Rühren bei Raumtemperatur wird mit 700 ml Wasser verdünnt und mit Dichlormethan extrahiert. Filtration über Kieselgel und Einengen ergeben 150 g Rohprodukt, das durch Kristallisation aus 1,5 1 Cyclohexan/Toluol-Gemisch 3:1 gereinigt wird. Die Mutterlauge wird nach Einengen aus Cyclohexan umkristallisiert. Man erhält so insgesamt 112g Titelverbindung als farblosen Feststoff, Schmp. 77- 78°C. α289 20 = -56.2 [c=0.9, CH3OH]
Beispiel 9
(R)-2-Benzylaminomethyl-8-isopropoxy-chroman
112 g (0.37 Mol) der Verbindung aus Beispiel 8, 200 g (1.87 Mol) Benzylamin und 3.0 g (0.02 Mol) Natriumiodid werden 5 Stunden auf 100°C erhitzt. Nach Abkühlen wird von Feststoff abgetrennt und die organische Phase 2x mit je 2.5 1 Wasser gewaschen. Das verbleibende Öl wird mit 1 1 Essigester aufgenommen. Waschen der Essigesteφhase mit Wasser und gesättigter Kochsalzlösung sowie anschließendes Trocknen und Einenengen ergeben 114.5 g (quant.) der Titelverbindung (HPLC- Reinheit: 93 %) als Öl, die in die nächste Stufe eingesetzt wird. α289 20 = -104 [c=0.5, CH3OH]
Beispiel 10
(R)-2-(N-Benzyl-N-(4-(l , 1 -dioxido-3-oxo-2,3-dihydro-benzisothiazol-2-yl)-2- butinyl)- aminomethyl)-8-isopropoxy-chroman Hydrochlorid
114 g (0.37 Mol) der Verbindung aus Beispiel 9 und 13.5 g (0.45 Mol) Para- formaldehyd in 1 1 Dioxan werden mit 4 g Kupfer(II)acetat versetzt und auf 50°C erwärmt. Bei dieser Temperatur werden 81 g (0.37 Mol) Propargylsaccharin zugesetzt. Nach 2 Stunden Rühren bei 80°C wird eingeengt und der Rückstand zwischen Toluol/Wasser unter Zusatz von Tonsil verteilt. Nach Filtration des Gemisches über Celite® wird die organische Phase abgetrennt und durch Flashchromatographie über Kieselgel (Toluol/Essigester 10:1) gereinigt. Fällung des Hydrochlorids aus Ether mit etherischer Salzsäure ergibt 226 g Rohprodukt. Dieses Produkt wird nach Freisetzung der freien Base mit Natriumhydrogencarbonat durch Chromatographie auf Kieselgel (Elution mit Toluol/Essigester 20:1) gereinigt. Die Produktfraktionen werden mit etherischer Salzsäure behandelt. Man erhält so 139 g (65 %) Titelverbindung als Feststoff, Schmp. 106-109°C. α289 20 = - 64.1 [c=0.8, CH3OH]
Beispiel 11
2-[4-({[(2R)-8-Isopropoxy-chroman-2-yl]methyl}amino)butyl]-l,2-benzisothiazol-
3(2H)-on 1,1-dioxid hat folgende Struktur:
120 g (0.21 mMol) der Verbindung aus Beispiel 10 in 1.4 1 Methanol werden mit 400 ml konz. Salzsäure und 20 g 10 % Palladium auf Aktivkohle versetzt. Nach 4 Stunden Hydrieren bei Normaldruck und 20°C wird vom Katalysator abfiltriert und eingeengt. Der Rückstand wird 2x mit Toluol eingeengt und mit 400 ml Essigester gelöst. Zusatz von 800 ml Diethylether und 18 h Rühren bei Raumtemperatur ergeben nach Absaugen und Trocknen im Vakkum 90.5 g Feststoff. Umkristallisation aus 1 1 Acetonitril und Waschen der Kristalle mit Dieethylether ergeben 70.8 g (69 %) Titelverbindung als farblose Kristalle, Schmp. 153-154°C.

Claims
1. Verwendung von 2-[4-({[(2R)-8-Isopropoxy-chroman-2-yl]methyl}amino)- butyl]-l,2-benzisothiazol-3(2H)-on 1,1-dioxid, dessen physiologisch uribe- denklichen Salze, Hydrate und/oder Solvate zur Herstellung eines Arzneimittels zur Prophylaxe und/oder Behandlung der Parkinsonschen Krankheit.
2. Verwendung nach Anspruch 1, wobei der Wirkstoff 2-[4-({[(2R)-8- Isopropoxy-chroman-2-yl]methyl}amino)butyl]-l,2-benzisothiazol-3(2H)-on 1,1 -dioxid Hydrochlorid ist.
3. 2-[4-({[(2R)-8-Isopropoxy-chroman-2-yl]methyl}amino)butyl]-l,2-benziso- thiazol-3(2H)-on 1,1 -dioxid, dessen physiologisch unbedenkliche Salze, Hydrate und/oder Solvate zur Prophylaxe und/oder Behandlung der Parkinsonschen Krankheit.
4. 2-[4-({[(2R)-8-Isopropoxy-chroman-2-yl]methyl}amino)butyl]-l,2-benziso- thiazol-3(2H)-on 1,1 -dioxid Hydrochlorid zur Prophylaxe und/oder Behandlung der Parkinsonschen Krankheit.
