Abstract
Die Erfindung betrifft Nukleisäuresequenzen, die für ein Polypeptid mit Nitrilaseaktivität codieren, Nukleinsäurekonstrukte enthaltend die Nukleinsäuresequenzen sowie Vectoren enthaltend die Nukleinsäuresequenzen oder die Nukleinsäurekonstrukte. Die Erfindung betrifft weiterhin Aminosäuresequenzen, die durch die Nukleinsäuresequenzen codiert werden und Mikroorganismen enthaltend die Nukleinsäuresequenzen, die Nukleinsäurekonstrukte oder Vectoren enthaltend die Nukleinsäuresequenzen oder die Nukleinsäurekonstrukte. Außerdem betrifft die Erfindung ein enzymatisches Verfahren zur Herstellung von Carbonsäuren aus den entsprechenden Nitrilen.

Beschreibung
Die Erfindung betrifft Nukleinsauresequenzen, die für ein Poly- peptid mit Nitrilaseaktivität codieren, Nukleinsaurekonstrukte enthaltend die Nukleinsauresequenzen sowie Vectoren enthaltend die Nukleinsauresequenzen oder die Nukleinsaurekonstrukte. Die Erfindung betrifft weiterhin Aminosäuresequenzen, die durch die Nukleinsauresequenzen codiert werden und Mikroorganismen enthaltend die Nukleinsauresequenzen, die Nukleinsaurekonstrukte oder Vectoren enthaltend die Nukleinsauresequenzen oder die Nukleinsaurekonstrukte .
Außerdem betrifft die Erfindung ein enzymatisch.es Verfahren zur Herstellung von Carbonsäuren aus den entsprechenden Nitrilen.
Aliphatische, aromatische und heteroaromatische Carbonsäuren sind gesuchte Verbindungen für die organischen Synthesechemie. Sie sind Ausgangsprodukte für eine Vielzahl von pharmazeutischen Wirkstoffen oder Wirkstoffen für den Pflanzenschutz.
Aus der Literatur sind eine Reihe verschiedener enzymatischer Synthesezugänge zu achiralen oder chiralen Carbonsäuren bekannt . So werden beispielsweise optisch aktive Aminosäuren technisch über fermentative Verfahren gewonnen. Von Nachteil dabei ist, daß für jede Aminosäure ein eigenes Verfahren entwickelt werden muß. Um eine möglichst breite Palette verschiedener Verbindungen her- stellen zu können, werden deshalb chemische oder enzymatische
Verfahren verwendet. Nachteilig bei den chemischen Verfahren ist, daß das Stereozentrum in der Regel in mehrstufigen, nicht breit anwendbaren Synthese umständlich aufgebaut werden muß.
Die enzymatische Synthese chiraler Carbonsäuren sind einer Reihe von Patenten oder Patentanmeldungen zu entnehmen. W092/ 05275 beschreibt die Synthese enantiomerer α-Hydroxy-α-alkyl- oder α-Alkylcarbonsäuren in Gegenwart biologischen Materials. Weitere Synthesen zu optisch aktiven α-substituierten organischen Säuren mit Mikroorganismen werden in EP-B-0 348 901, EP-B-0 332 379,
EP-A-0 348 901 oder seinem US-Äquivalent US 5,283,193,
EP-A-0 449 648, EP-B-0 473 328, EP-B-0 527 553 oder seinem
US-Äquivalent US 5,296,373, EP-A-0 610 048, EP-A-0 610 049,
EP-A-0 666 320 oder WO 97/32030 beschrieben.
Die biotechnologische Synthese achiraler Carbonsäuren mit Mikro¬ organismen wird beispielsweise in EP-A-0 187 680, EP-A-0 229 042, WO 89/00193, JP 08173152, JP06153968, FR 2694571, EP-A0 502 476, EP-A-0 444 640 oder EP-A-0 319 344 beschrieben.
Von Nachteil bei diesen Prozessen ist, daß sie häufig nur zu Produkten mit einer geringen optischen Reinheit führen und/oder daß sie nur mit geringen Raum-Zeit-Ausbeuten ablaufen. Dies führt zu wirtschaftlich unattraktiven Prozessen. Von Nachteil ist außerdem, daß die Enzyme, die in den zur Synthese der achiralen oder chiralen Carbonsäuren verwendeten Mikroorganismen vorhanden sind, in der Regel nur ein eingeschränktes Substratspektrum haben, das heißt es werden immer nur bestimmte aliphatische, aromatische oder heteroaromatische Nitrile durch einen Mikro- Organismus umgesetzt. Speziell aromatische und heteroaromatische Nitrile wie beispielsweise Cyanothiophene oder Benzonitril werden schlecht oder gar nicht zu den entsprechenden Carbonsäuren umgesetzt .
Es bestand daher die Aufgabe, weitere Enzyme zur Herstellung von achiralen und/oder chiralen Carbonsäuren zu entwickeln, die in einem Verfahren zur Herstellung von achiralen und/oder chiralen Carbonsäuren verwendet werden können, das die oben genannten Nachteile nicht aufweist und speziell aromatische und/oder heteroaromatische Carbonsäuren aus den entsprechenden Nitrilen zugänglich macht.
Diese Aufgabe wurde durch die erfindungsgemäße isolierte Nuklein- säuresequenz gelöst, die für ein Polypeptid mit Nitrilase- aktivität codiert, ausgewählt aus der Gruppe:
a) einer Nukleinsäuresequenz mit der in SEQ ID NO: 1 dargestellten Sequenz,
b) Nukleinsauresequenzen, die sich als Ergebnis des degenerierten genetischen Codes von der in SEQ ID NO: 1 dargestellten Nukleinsäuresequenz ableiten,
c) Derivate der in SEQ ID NO : 1 dargestellten Nukleinsäure- Sequenz, die für Polypeptide mit der in SEQ ID NO: 2 dargestellten Aminosäuresequenzen codieren und mindestens 95 % Homologie auf Aminosäureebene aufweisen, ohne daß die enzymatische Wirkung der Polypeptide wesentlich reduziert ist .
Unter Homologe der erfindungsgemäßen Nukleinsäuresequenz mit der Sequenz SEQ ID NO : 1 sind beispielsweise Allelvarianten zu ver¬ stehen, die mindestens 95 % Homologie auf der abgeleiteten Amino¬ säureebene, vorteilhaft mindestens 97 % Homologie, bevorzugt min- destens 98 %, ganz besonders bevorzugt mindestens 99 % Homologie über den gesamten Sequenzbereich aufweisen. Über Teilbereiche der Sequenzen können die Homologien vorteilhaft höher liegen. Die von SEQ ID NO: 1 abgeleitete Aminosäuresequenz ist SEQ ID NO : 2 zu entnehmen. Allelvarianten umfassen insbesondere funktioneile Varianten, die durch Deletion, Insertion oder Substitution von Nukleotiden aus der in SEQ ID NO : 1 dargestellten Sequenz erhältlich sind, wobei die enzymatische Aktivität der abgeleiteten synthetisierten Proteine für das Einbringen eines oder mehrerer Gene in einen Organismus jedoch erhalten nicht wesentlich reduziert sein sollte. Unter nicht wesentlich reduzierter enzymatische Aktivität ist eine enzymatische Aktivität zu verstehen, die vorteilhaft mindestens 10 %, bevorzugt 30 %, besonders bevorzugt 50 %, ganz besonders bevorzugt 70 % der enzymatischen Aktivität des unter SEQ ID NO: 2 wiedergegebenen Enzyms aufweist. Die Erfindung betrifft damit auch Aminosäuresequenzen, die durch die oben dargestellte Gruppe von Nukleinsauresequenzen kodiert werden. Vorteilhaft betrifft die Erfindung Aminosäuresequenzen, die durch die Sequenz SEQ ID NO: 1 kodiert werden .
Weiterhin sind unter Homologe der SEQ ID NO: 1 beispielsweise pilzliche oder bakterielle Homologe, verkürzte Sequenzen, Einzel- strang-DNA oder RNA der codierenden und nichtcodierenden DNA- Sequenz zu verstehen. Homologe der SEQ ID NO: 1 besitzen auf DNA-Ebene eine Homologie von mindestens 60 %, bevorzugt von mindestens 70 %, besonders bevorzugt von mindestens 80 % , ganz besonders bevorzugt von mindestens 90 % über den gesamten in SEQ ID NO: 1 angegebenen DNA-Bereich.
Außerdem sind unter Homologe der SEQ ID NO : 1 Derivate wie beispielsweise Promotorvarianten zu verstehen. Die Promotoren, die den angegebenen Nukleotidsequenzen vorgeschalten sind, können durch ein oder mehrere Nukleotidaustausche, durch Insertion (en) und/oder Deletion (en) verändert sein, ohne daß aber die Funktionalität bzw. Wirksamkeit der Promotoren beeinträchtigt sind. Des weiteren können die Promotoren durch Veränderung ihrer Sequenz in ihrer Wirksamkeit erhöht oder komplett durch wirksamere Promotoren auch artfremder Organismen ausgetauscht werden.
Unter Derivaten sind auch Varianten zu verstehen, deren Nukleotidsequenz im Bereich von -1 bis -200 vor dem Startkodon oder 0 bis 1000 Basenpaare nach dem Stopkodon so verändert
wurden, daß die Genexpression und/oder die Proteinexpression verändert, bevorzugt erhöht wird.
Vorteilhaft läßt sich die SEQ ID NO: 1 oder seine Homologen aus Bakterien, vorteilhaft aus gram-positiven Bakterien, bevorzugt aus Bakterien der Gattungen Nocardia, Rhodococcus, Streptomyces , Mycobacterium, Corynebacterium, Micrococcus, Proactinomyces oder Bacillus , besonders bevorzugt aus Bakterien der Gattung Rhodo¬ coccus, Mycobacterium oder Nocardia, ganz besonders bevorzugt aus der Gattung und Art Rhodococcus sp., Rhodococcus rhodochrous , Nocardia rhodochrous oder Mycobacterium rhodochrous über dem Fachmann bekannte Methoden isolieren.
SEQ ID No: 1 oder seine Homologen oder Teile dieser Sequenzen lassen sich beispielsweise mit üblichen Hybridisierungsverfahren oder der PCR-Technik aus anderen Pilzen oder Bakterien isolieren. Diese DNA-Sequenzen hybridisieren unter Standardbedingungen mit den erfindungsgemäßen Sequenzen. Zur Hybrisierung werden vorteilhaft kurze Oligonukleotide der konservierten Bereiche beispiels- weise aus dem aktiven Zentrum, die über Vergleiche mit anderen Nitrilasen oder Nitrilhydratasen in dem Fachmann bekannter Weise ermittelt werden können, verwendet. Es können aber auch längere Fragmente der erfindungsgemäßen Nukleinsäuren oder die vollständigen Sequenzen für die Hybridisierung verwendet werden. Je nach der verwendeten Nukleinsäure Oligonukleotid, längeres Fragment oder vollständige Sequenz oder je nachdem welche Nukleinsäureart DNA oder RNA für die Hybridisierung verwendet werden, variieren diese Standardbedingungen. So liegen beispielsweise die Schmelztemperaturen für DNA: DNA-Hybride ca 10°C niedriger als die von DNA:RNA-Hybriden gleicher Länge.
Unter Standardbedingungen sind beispielsweise je nach Nukleinsäure Temperaturen zwischen 42 und 58°C in einer wäßrigen Pufferlösung mit einer Konzentration zwischen 0,1 bis 5 x SSC (1 X SSC = 0,15 M NaCl, 15 mM Natriumeitrat, pH 7,2) oder zusätzlich in Gegenwart von 50 % Formamid wie beispielsweise 42°C in 5 x SSC, 50 % Formamid zu verstehen. Vorteilhafterweise liegen die Hybridisierungsbedingungen für DNA: DNA-Hybride bei 0,1 x SSC und Temperaturen zwischen etwa 20°C bis 45°C, bevorzugt zwischen etwa 30°C bis 45°C. Für DNA:RNA-Hybride liegen die Hybridisierungsbedingungen vorteilhaft bei 0 , 1 x SSC und Temperaturen zwischen etwa 30°C bis 55°C, bevorzugt zwischen etwa 45°C bis 55°C. Diese angegebenen Temperaturen für die Hybridisierung sind beispielhaft kalkulierte Schmelztemperaturwerte für eine Nukleinsäure mit einer Länge von ca. 100 Nukleotiden und einem G + C-Gehalt von 50 % in Abwesenheit von Formamid. Die experimentellen Bedingungen für die DNA-Hybridisierung sind in einschlägigen Lehrbüchern der Genetik wie beispielsweise Sambrook et al . , "Molecular Cloning" ,

den gram-positiven Promotoren amy und SP02 , in den Hefe- oder Pilzpromotoren ADC1, MFα, AC , P-60, CYC1, GAPDH, TEF, rp28, ADH. In diesem Zusammenhang sind auch die Promotoren der Pyruvat- decarboxylase und der Methanoloxidase aus beispielsweise Hansenula vorteilhaft. Es können auch künstliche Promotoren für die Regulation verwendet werden.
Das Nukleinsäurekonstrukt wird zur Expression in einem Wirtsorganismus vorteilhafterweise in einen Vektor wie beispielsweise einem Plasmid, einem Phagen oder sonstiger DNA inseriert, das eine optimale Expression der Gene im Wirt ermöglicht. Diese Vektoren stellen eine weitere Ausgestaltung der Erfindung dar. Geeignete Plasmide sind beispielsweise in E. coli pLG338, pA- CYC184, pBR322, pUC18, pUC19, pKC30, pRep4 , pHSl, pHS2, pPLc236, pMBL24, pLG200, pUR290, pIN-III113-Bl, λgtll oder pBdCI, in Streptomyces pIJlOl, pIJ364, pIJ702 oder pIJ361, in Bacillus pUBHO, pC194 oder pBD214, in Corynebacterium pSA77 oder pAJ667, in Pilzen pALSl, pIL2 oder pBB116, in Hefen 2μM, pAG-1, YEp6, YEpl3 oder pEMBLYe23 oder in Pflanzen pLGV23, pGHlac+, pBIN19, pAK2004 oder pDH51. Die genannten Plasmide stellen eine kleine Auswahl der möglichen Plasmide dar. Weitere Plasmide sind dem Fachmann wohl bekannt und können beispielsweise aus dem Buch Cloning Vectors (Eds. Pouwels P. H. et al . Elsevier, Amsterdam- New York-Oxford, 1985, ISBN 0 444 904018) entnommen werden.
Vorteilhafterweise enthält das Nukleinsäurekonstrukt zur Expression der weiteren enthaltenen Gene zusätzlich noch 3' und/oder 5' Terminale regulatorische Sequenzen zur Steigerung der Expression, die je nach ausgewähltem Wirtorganismus und Gen oder Gene für eine optimale Expression ausgewählt werden.
Diese regulatorischen Sequenzen sollen die gezielte Expression der Gene und der Proteinexpression ermöglichen. Dies kann beispielsweise je nach Wirtsorganismus bedeuten, daß das Gen erst nach Induktion exprimiert oder überexprimiert wird, oder daß es sofort exprimiert und/oder überexprimiert wird.
Die regulatorischen Sequenzen bzw. Faktoren können dabei vorzugsweise die Genexpression der eingeführten Gene positiv beeinflussen und dadurch erhöhen. So kann eine Verstärkung der regulatorischen Elemente vorteilhafterweise auf der Transkriptionsebene erfolgen, indem starke Transkriptionssignale wie Promotoren und/oder "Enhancer" verwendet werden. Daneben ist aber auch eine Verstärkung der Translation möglich, indem beispielsweise die Stabilität der mRNA verbessert wird.
In einer weiteren Ausgestaltungsform des Vektors kann der das erfindungsgemäße Nukleinsäurekonstrukt oder die erfindungsgemäße Nukleinsäure enthaltende Vektor auch vorteilhafterweise in Form einer linearen DNA in die Mikroorganismen eingeführt werden und über heterologe oder homologe Rekombination in das Genom des WirtsOrganismus integriert werden. Diese lineare DNA kann aus einem linearisierten Vektor wie einem Plasmid oder nur aus dem Nukleinsäurekonstrukt oder der Nukleinsäure bestehen.
Für eine optimale Expression heterologer Gene in Organismen ist es vorteilhaft die Nukleinsauresequenzen entsprechend des im Organismus verwendeten spezifischen "codon usage" zu verändern. Der "codon usage" läßt sich anhand von Computerauswertungen anderer, bekannter Gene des betreffenden Organismus leicht ermitteln.
Als Wirtsorganismen für die erfindungsgemäße Nukleinsäure oder dem Nukleinsäurekonstrukt kommen prinzipiell alle prokaryonti- schen oder eukaryontisehen Organismen in Frage. Vorteilhafter- weise werden als Wirtsorganismen Mikroorganismen wie Bakterien, Pilze oder Hefen verwendet. Vorteilhaft werden gram-positive oder gram-negative Bakterien, bevorzugt Bakterien der Familie Enterobacteriaceae, Pseudomonadaceae, Streptomycetaceae, Myco- bacteriaceae oder Nocardiaceae, besonders bevorzugt Bakterien der Gattungen Escherichia, Pseudomonas, Nocardia, Mycobacterium, Streptomyces oder Rhodococcus verwendet . Ganz besonders bevorzugt sind die Gattung und Art Escherichia coli, Rhodococcus rhodochrous, Nocardia rhodochrous, Mycobacterium rhodochrous oder Streptomyces lividans.
Der Wirtsorganismus gemäß der Erfindung enthält dabei vorzugsweise mindestens ein proteinisches Agenz zur Faltung der von ihm synthetisierten Polypeptide und insbesondere der in dieser Erfindung beschriebenen Nukleinsauresequenzen mit Nitrilase- aktivität und/oder die dieses Agenz codierenden Gene, wobei dieses Agenz in einer Menge vorliegt, die größer ist als die, die der Grundmenge des betrachteten Mikroorganismus entspricht . Die für dieses Agenz codierenden Gene sind im Chromosom oder in extrachromosomalen Elementen wie zum Beispiel Plasmiden enthalten.
Ein weiterer Gegenstand der Erfindung ist ein Verfahren zur Herstellung von chiralen oder achiralen Carbonsäuren, dadurch gekennzeichnet, daß man Nitrile in Gegenwart einer durch die erfindungsgemäßen Nukleinsäuren kodierten Aminosäuresequenz oder einem wachsenden, ruhenden oder aufgeschlossenen oben genannten Mikroorganismus (= Wirtsorganismus) , der entweder eine
erfindungsgemäße Nukleinsäuresequenz, ein erfindungsgemäßes Nukleinsäurekonstrukt, das eine erfindungsgemäße Nukleinsäure mit einem oder mehreren Regulationssignalen verknüpft enthält, oder einen erfindungsgemäßen Vector enthält, zu den chiralen oder achiralen Carbonsäuren umsetzt.
Ein vorteilhafte Ausführungsform des Verfahrens ist die Umsetzung chiraler oder achiraler aliphatischer Nitrile zu den entsprechenden Carbonsäuren.
Eine weitere bevorzugte Ausführungsform des Verfahrens ist ein Verfahren zur Herstellung von chiralen oder achiralen Carbonsäuren, dadurch gekennzeichnet, daß man Nitrile der allgemeinen Formel I
in Gegenwart einer durch die erfindungsgemäßen Nukleinsäuren kodierten Aminosäuresequenz oder einem wachsenden, ruhenden oder aufgeschlossenen oben genannten Mikroorganismus, der entweder eine erfindungsgemäße Nukleinsäuresequenz, ein erfindungsgemäßes Nukleinsäurekonstrukt, das eine erfindungsgemäße Nukleinsäure mit einem oder mehreren Regulationssignalen verknüpft enthält, oder einen erfindungsgemäßen Vector enthält, zu Carbonsäuren der allgemeinen Formel II

Als Hetaryl- seien substituierte oder unsubstituierte, einfache oder kondensierte aromatische Ringsysteme mit einem oder mehreren heteroaromatischen 3- bis 7gliedrigen Ringen, die ein oder mehrere Heteroatome wie N, 0 oder S enthalten können und gegebenenfalls über eine Cι-Cιo-Alkyl- , C3-Cg-Alkenyl- oder C3-C8~Cycloalkylkette an das Grundgerüst gebunden sein können, genannt. Beispiele für derartige Hetarylreste sind Pyrazol, Imidazol, Oxazol, Isooxazol, Thiazol, Triazol, Pyridin, Chinolin, Isochinolin, Acridin, Pyrimidin, Pyridazin, Pyrazin, Phenazin, Purin oder Pteridin. Die Hetarylreste können über die Heteroatome oder über die verschiedenen Kohlenstoffatome im Ring oder Ringsystem oder über die Substituenten an das Grundgerüst gebunden sein. Auch aromatische oder aliphatische gesätttigte oder ungesättigte Dinitrile lassen sich im erfindungsgemäßen Verfahren vorteilhaft umsetzen.
Das erfindungsgemäße Verfahren wird vorteilhaft bei einem pH-Wert von 4 bis 11, bevorzugt von 4 bis 9 durchgeführt.
Weiterhin werden im Verfahren vorteilhaft von 0,01 bis 10 Gew.-%, bevorzugt 0,1 bis 10 Gew.-%, besonders bevorzugt 0,5 bis 5 Gew.-% Nitril verwendet. Je nach Nitril können unterschiedliche Mengen an Nitril in der Reaktion verwendet werden. Die geringsten Mengen
an Nitril werden vorteilhaft bei Nitrilen (Cyanhydrine) verwendet (= Mengen zwischen 0,01 bis 5 Gew.-%), die im Gleichgewicht mit den entsprechenden Aldehyden und Blausäure stehen. Da der Aldehyd für die Mikroorganismen oder Enzyme in der Regel toxisch ist. Nitrile, die leicht flüchtig sind, werden ebenfalls vorteilhaft in Mengen zwischen 0,01 bis 5 Gew.-% eingesetzt. Bei höheren Cyanhydrin- bzw. Nitrilmengen läuft die Reaktion verzögert ab. Bei Nitrilen, die nur geringe oder nahezu keine Lösungsmitteleigenschaften haben oder Nitrilen, die sich nur in sehr geringen Mengen in wäßrigen Medium lösen, können vorteilhaft auch größere als die oben angegebenen Mengen eingesetzt werden. Zur Erhöhung des Umsatzes und der Ausbeute wird die Reaktion vorteilhafterweise unter kontinuierlicher Zugabe des Nitrils durchgeführt. Das Produkt kann nach Ende der Reaktion isoliert werden oder aber in einem Bypass kontinuierlich entfernt werden.
Das erfindungsgemäße Verfahren wird vorteilhaft bei einer Temperatur zwischen 0°C bis 80°C, bevorzugt zwischen 10°C bis 60°C, besonders bevorzugt zwischen 15°C bis 50°C durchgeführt
Vorteilhaft werden im erfindungsgemäßen Verfahren aromatische oder heteroaromatische Nitrile wie 2-Phenyl-propionitril , 2-Hydroxy-phenylessigsäurenitril, 2-Amino-2-phenyl-acetonitril , Benzonitril, Phenylacetonitril , trans-Zimtsäurenitril , 3-Cyan- thiophen oder 3-Cyanomethyl-thiophen genannt.
Unter chiralen Nitrilen im erfindungsgemäßen Verfahren sind Nitrile zu verstehen, die aus einem 50:50 Gemisch der beiden Enantiomere oder aus einem beliebigen anderen Gemisch mit einer Anreicherung eines der beiden Enantiomere im Gemisch bestehen. Beispielhaft seien für derartige Nitrile 2-Phenyl-propionitril , 2-Hydroxy-phenylessigsäurenitril , 2-Amino-2-phenyl-acetonitril , 2-Chlor-propionitril oder 2-Hydroxy-propionitril genannt.
Unter chiralen Carbonsäuren sind im erfindungsgemäßen Verfahren zu verstehen, die eine Enantiomerenanreicherung zeigen. Bevorzugt werden im Verfahren Enantiomerenreinheiten von mindestens 90 %ee, bevorzugt von min. 95 %ee, besonders bevorzugt von min. 98 %ee, ganz besonders bevorzugt min. 99 %ee erreicht.
Das erfindungsgemäße Verfahren ermöglicht die Umsetzung einer großen Vielzahl von chiralen oder achiralen Nitrilen zu den entsprechenden chiralen oder achiralen Carbonsäuren. Im Verfahren lassen sich mindestens 25 mmol Nitril/h x mg Protein oder min- destens 25 mmol Nitril/h x g Trockengewicht der Mikroorganismen umsetzen, bevorzugt mindestens 30 mmol Nitril/h x mg Protein oder mindestens 30 mmol Nitril/h x g Trockengewicht, besonders bevor-
zugt mindestens 40 mmol Nitril/h x mg Protein oder mindestens 40 mmol Nitril/h x g Trockengewicht, ganz besonders bevorzugt mindestens 50 mmol Nitril/h x mg Protein oder mindestens 50 mmol Nitril/h x g Trockengewicht.
Für das erfindungsgemäße Verfahren können wachsende Zellen ver¬ wendet werden, die die erfindungsgemäßen Nukleinsäuren, Nuklein¬ saurekonstrukte oder Vektoren enthalten. Auch ruhende oder aufgeschlossene Zellen können verwendet werden. Unter auf- geschlossenen Zellen sind beispielsweise Zellen zu verstehen, die über eine Behandlung mit beispielsweise Lösungsmitteln durchlässig gemacht worden sind, oder Zellen die über eine Enzymbehandlung, über eine mechanische Behandlung (z.B. French Press oder Ultraschall) oder über eine sonstige Methode auf- gebrochen wurden. Die so erhaltenen Rohextrakte sind für das erfindungsgemäße Verfahren vorteilhaft geeignet . Auch gereinigte oder angereinigte Enzyme können für das Verfahren verwendet werden. Ebenfalls geeignet sind immobilisierte Mikroorganismen oder Enzyme, die vorteilhaft in der Reaktion Anwendung finden können.
Die im erfindungsgemäßen Verfahren hergestellten chiralen oder achiralen Carbonsäuren lassen sich vorteilhaft aus der wäßrigen Reaktionslösung über Extraktion oder Kristallisation oder über Extraktion und Kristallisation gewinnen. Hierzu wird die wäßrige Reaktionslösung mit einer Säure wie einer Mineralsäure (z.B. HC1 oder H S0 ) oder einer organischen Säure angesäuert vorteilhaft auf pH-Werte unter 2 und anschließend mit einem organischen Lösungsmittel extrahiert . Die Extraktion kann zur Erhöhung der Ausbeute mehrfach wiederholt werden. Als organische Lösungsmittel können prinzipiell alle Lösungsmittel verwendet werden, die mit Wasser gegebenenfalls nach Zugabe von Salzen eine Phasengrenze zeigen. Vorteilhafte Lösungsmittel sind Lösungsmittel wie Toluol, Benzol, Hexan, Methyltertiärbutylether oder Essigester. Auch eine Reinigung der Produkte kann auch vorteilhaft über Bindung an einen Ionentauscher und anschließendes eluieren mit einer Mineralsäure oder Carbonsäure wie HCL, HS0 , Ameisensäure oder Essigsäure erfolgen.
Nach Einengen der wässrigen oder organischen Phase können die Produkte in der Regel in guten chemischen Reinheiten, das heißt größer 90 % chemische Reinheit, gewonnen werden. Nach Extraktion kann die organische Phase mit dem Produkt aber auch nur zum Teil eingeengt werden und das Produkt auskristallisiert werden. Dazu wird die Lösung vorteilhaft auf eine Temperatur von 0°C bis 10°C abgekühlt. Die Kristallisation kann auch direkt aus der organischen Lösung erfolgen. Das auskristallisierte Produkt kann
nochmals in im gleichen oder in einem anderen Lösungsmittel zur erneuten Kristallisation aufgenommen werden und nochmals kristallisiert werden. Durch die anschließende mindestens einmalige Kristallisation kann die Enantiomerenreinheit des Produktes je nach Lage des Eutektikum weiter gesteigert werden.
Die chiralen oder achiralen Carbonsäuren können jedoch auch direkt nach Ansäuern mit einer Säure auf einen pH-Wert vorteilhaft unter 2 aus der wäßrigen Reaktionslösung auskristallisiert werden. Vorteilhaft wird dazu die wäßrige Lösung unter Erwärmen eingeengt und in ihrem Volumen um 10 bis 90 %, bevorzugt 20 bis 80 %, besonders bevorzugt 30 bis 70 % reduziert. Vorzugsweise wird die Kristallisation unter Kühlung durchgeführt. Temperaturen zwischen 0°C bis 10°C sind für die Kristallisation bevorzugt. Aus kostengründen ist die direkte Kristallisation aus der wäßrigen Lösung bevorzugt. Ebenfalls bevorzugt wird eine Aufarbeitung der chiralen Carbonsäuren über ein Extraktion und gegebenenfalls anschließender Kristallisation.
Bei diesen bevorzugten Aufarbeitungsarten läßt sich das Produkt des erfindungsgemäßen Verfahrens in Ausbeuten von 60 bis 100 %, bevorzugt von 80 bis 100 %, besonders bevorzugt von 90 bis 100 % bezogen auf das für die Reaktion eingesetzte Nitril isolieren. Das isoliert Produkt zeichnet sich durch eine hohe chemische Reinheit von > 90 %, bevorzugt > 95 % besonders bevorzugt von > 98 % aus. Weiterhin zeichnen sich die Produkt bei chiralen Nitrilen bzw. chiralen Carbonsäuren durch eine hohe Enantiomerenreinheit, die durch die Kristallisation weiter gesteigert werden kann.
Die so gewonnenen Produkte eignen sich als Ausgangsmaterial für organischen Synthesen zur Herstellung von Pharmaka oder Agro- chemikalien oder zur Racematspaltung.

Isolierung und heterologe Expression des nitA-Gens aus Rhodococcus rhodochrous NCIMB 11216
Beispiel 1: Isolierung des niCA-Gens aus Rhodococcus rhodochrous NCIMB 11216
Zur Isolierung des nitA-Gens aus Rhodococcus rhodochrous NCIMB 11216 wurde zelleigene DNA isoliert, eine Phagen-Genbank angelegt und diese mit einer Oligo-Nukleotid-Sonde durchsucht.
1.1 Isolierung von DNA aus R . rhodochrous NCIMB 11216
Zur Präparation genomischer DNA aus Rhodococcus rhodochrous NCIMB 11216 nach Sa brook et al . , 1989, wurden 2 x 100 ml Über- nacht-Kultur (in dYT-Medium, Sambrook, J., Fritsch, E. F. and Maniatis, T. , 1989, Molecular cloning: a laboratory manual, 2nd edition, Cold Spring Harbor Laboratory Press. Cold Spring Harbor, New York.) abzentrifugiert und die Pellets in 8 ml 25 mM Tris/ HC1, 25 mM EDTA, 10 % Saccharose (w/v) , pH 8,0 resuspendiert. Nach Lysozymbehandlung der vereinigten Kulturen für 15 min bei
37°C (Zugabe von 2 ml Lysozym, 100 mg/ml in 10 mM Tris/HCl, 0 , 1 mM EDTA, pH 8,0) wurden 2 ml 10 % (w/v) Na-Lauroylsarcosinat zuge¬ geben und bei mehrmaliger Durchmischung 15 min bei 65°C inkubiert. Anschließend wurde CsCl in einer Endkonzentration von 1 g/ml hin- zugegeben, bei 65°C gelöst und nach .Zugabe von Ethidiumbromid in einer Endkonzentration von 0,4 mg/ml eine Ultrazentrifugation im Festwinkelrotor (Sorvall T1270, 83500 g, 48 h, 17°C) durchgeführt. Die chromosomale DNA-Bande wurde unter UV-Licht abgezogen, 2 h gegen TE 10.1 (10 mM Tris/HCl, 1 mM EDTA, pH 8,0) dialysiert und 3 mal mit Phenollösung (gesättigt mit 10 mM Tris/HCl, pH 8) extrahiert. Schließlich wurde die DNA noch 3 mal gegen TE 10.01 (10 mM Tris/HCl, 0,1 mM EDTA, pH 8,0), dialysiert und bei 4°C gelagert. So wurden etwa 1,5 ml DNA-Lösung mit einer Konzentration von etwa 500 μg/ml gewonnen.
1.2 Herstellung einer Phagen-Genbank aus der DNA von R . rhodochrous NCIMB 11216
Als Vektor für die Genbank wurde der Phage λ±RESIII verwendet: Dieser Substitutionsvektor enthält das lux-Operon als Replace- ment-Fragment , was eine visuelle Erfassung des Hintergrundes durch Bioluminiszenz ermöglicht, sowie integrierte res ("reso- lution") -sites aus Tnl722 und die Replikationsfunktionen von pTW601-l, so daß der Vektor in einem Stamm mit entsprechender Transposase in ein autonom replizierendes Plasmid umgewandelt werden kann (Altenbuchner , 1993, A new λ RES vector with a built- in Tn2721-encoded, excision System, Gene 123, 63-68).
1.2.1 Isolierung von λ±RESIII-DNA (nach Sambrook et al . , 1989)
Aus einer Übernachtkultur von E. coli TAP 90 (LBo, Sambrook et al . , 1989, mit 10 mM MgS04, 0,2 % Maltose (w/v)) wurden 1010 Zellen abzentrifugiert und das Pellet in 3 ml SM-Phagenpuffer (50 mM Tris/HCl, 100 mMNaCl, 8 M MgS04, 0,01 % (w/v) Gelatine) resuspendiert. Nach Infektion mit 1,5 x 108 - 1,5 x 109 plaque forming units (pfu) λ RESIII-Phagenlysat für 20 min bei 37°C wurde der Ansatz im 2 1 Erlenmeyer-Kolben zu 500 ml LBQ , 10 mM MgS0 ,
0,2 % Maltose gegeben. Insgesamt 4 solcher Ansätze wurden 9 bis 12 h bei 37°C gerührt, bis die Zellyse erkennbar wurde. Zur vollständigen Lyse wurde jeder Kolben mit 10 ml Chloroform versetzt, weitere 30 min bei 37°C gerührt. Zelluläre Nukleinsäuren 5 wurden durch Zugabe von DNase und RNase (je 1 μg/ml) 30 min bei Raumtemperatur unter Rühren verdaut. Dann wurden zu jedem Ansatz 29,2 g NaCl zugegeben, gelöst, 10 min bei 8300 g zentrifugiert und die Überstande mit 10 % PEG 6000 versetzt. Zur anschließenden Phagenpräzipitation wurden die Ansätze über Nacht bei 4°C ge-
10 rührt und anschließend 15 min bei 14000 g abzentrifugiert. Nach Trocknen der Pellets wurden diese in jeweils 5 ml SM-Puffer aufgenommen, mit 5 ml Chloroform versetzt und 15 min bei 3000 g zentrifugiert. Die wässrigen Phasen mit den Phagen wurden vereinigt, mit 0,75 g/ml CsCl versetzt und nach vollständiger Lösung
15 24 h zentrifugiert (Festwinkelrotor Sorvall T1270, 98400 g, 48 h, 17°C) . Die sichtbare Phagenbande wurde abgezogen und 2 x gegen 50 mM Tris/HCl, 10 mM NaCl, 10 mM MgCl2, pH 8 , 0 dialysiert. Nach Zugabe von 20 mM EDTA, 50 μg/ml Proteinase K und 0,5 % SDS erfolgte eine Inkubation für 1 h bei 65°C. Anschließend wurde je 1 x
20 mit Phenol (gesättigt mit 10 mM Tris/HCl, pH 8 ) , l x mit Phenol (gesättigt mit 10 mM Tris/HCl, pH 8) / Chloroform (50/50 v/v) und 1 x mit Chloroform extrahiert. Schließlich wurde die DNA 3 x gegen TE 10.1 und 1 x gegen TE 10.01 dialysiert, der Titer auf E. coli TAP 90 bestimmt (siehe 1.2.3) und die λ±RESIII-DNA bei 4°C
25 gelagert.
1.2.2 Klonierung genomischer DNA in λ±RESIII-Vektoren
Für die Klonierung genomischer R . rhodochrous NCIMB 11216 DNA- 30 Fragmente wurden zuerst die λ±RESIII-Armfragmente präpariert, indem λ±RESIII-DNA in einem Volumen von 100 μl 2 μg mit 20 U BamHI 5 h bei 37°C verdaut wurden. Nach Extraktion mit Phenol (gesättigt mit 10 mM Tris/HCl, pH 8) / Chloroform (50/50 v/v), Isopropanol- Fällung und Waschen mit 70 % bzw. 100 % Äthanol (vorgekühlt auf 5 -20°C) wurde die DNA in TE 10.01 gelöst und mit 20 U Sall nachbehandelt (5 h bei 37°C) . Erneut erfolgten Phenol / Chloroform- Extraktion, Isopropanol-Fällung, Waschen und Lösung in TE 10.01.
Zur Herstellung der genomischen DNA-Fragmente wurden - nach Auf- 0 nähme einer Zeitkinetik für die verwendete Enzymcharge - 10 μg genomische DNA in einem 100 μl-Ansatz für 5 min mit 0,5 U Sau3AI partial verdaut. Nach elektrophoretischer Auftrennung über ein 0,8 %iges Low-melting-point-Agarosegel wurde der Fragmentbereich von 8 bis 14 kb isoliert und nach Parker & Seed (1980) aus dem 5 Gel eluiert . Die genomischen DNA-Fragmente wurden über Nacht bei 16°C mit den λ+RESIII-Armen ligiert.
Die Ligationsansätze wurden schließlich in vi tro mit Phagenex- trakten verpackt, .die zuvor aus dem "packaging extract donor" E. coli BHB 2688 ("freeze thaw lysate", FTL, Sambrook et al . , 1989) und dem "prehead donor" E. coli BHB 2690 ("sonicated 5 extract", SE, Sambrook et al., 1989) präpariert worden waren. Für die Verpackung wurden 5 μl Ligationsansatz , 7 μl Puffer A (20 mM Tris/HCl, 3 mM MgCl2, 1 mM EDTA, 0,05 % ß-Mercaptoäthanol, pH 8,0), 7 μl Puffer Ml ( 6 , 7 mM Tris/HCl, 33 mM Spermidin, 100 mM Putrescin, 17,8 mM ATP, 0,2 % ß-Mercaptoäthanol, 20 mM MgCl2, pH 10 8) , 15 μl SE und 10 μl FTL gemischt und 1 h bei Raumtemperatur inkubiert. Anschließend wurden 500 μl SM-Puffer und 1 Tropfen Chloroform zugegeben, gemischt, die Ansätze abzentrifugiert und bei 4°C gelagert.
15 Zur TiterbeStimmung der hergestellten Phagengenbank wurde der Stamm E. coli TAP 90 (Patterson & Dean, 1987) infiziert. Hierzu wurden logarithmisch wachsende Zellen (Anzucht in LBo, 10 mM MgCl , 0,5 % Maltose) mit 100 μl verschiedener Verdünnungen des Verpackungs- oder Phagenlysats in SM-Puffer für 30 min bei
20 37°C inkubiert. Die Ansätze wurden dann mit je 3 ml auf 42°C temperierten Top-Agar (0,8 % Bacto-Agar, 10 mM MgCl , 0,5 % Maltose) versetzt, kurz gemischt und auf LBrj-Agarplatten mit 10 mM MgCl (auf 37°C vorgewärmt) aufgeschichtet. Nach 12-16 h Inkubation bei 37°C wurden die Plaques zur Titerbestimmung aus-
25 gezählt. Der Titer der hergestellten Genbank betrug etwa 4 x 105 pfu/ml .
1.2.3 Umwandlung der rekombinanten λ±RESIII-Phagen in ein Plasmid

Die erhaltenen rekombinanten λ±RESIII-Phagen wurden in dem Stamm E. coli HB 101 F' [ : : Tnl 7391ac] , der das Transposon Tnl735 mit dem Resolvasegen unter Kontrolle des tac-Promotors trägt (Altenbuch- ner, 1993, siehe oben), in ein autonom replizierendes Plasmid
35 umgewandelt. Dieser Stamm wurde zur Infektion in 5 ml LBo mit 10 mM MgCl2 und 0,5 % Maltose bis zu einer OD6oo von 0,6 bis 0,8 angezogen und 100 μl davon mit einer geeigneten Menge an Phagen- lysat für 30 min bei Raumtemperatur infiziert. Der Ansatz wurde in 5 ml vorgewärmtem dYT, 1 mM Isopropyl-ß-thiogalactopyranosid 0 (IPTG) 1 h bei 37°C gerollert, abzentrifugiert, im Rücklauf resuspendiert, und die Zellen auf dYT Agarplatten mit 100 μg/ml Kanamycin plattiert und über Nacht bei 37°C inkubiert.
Zur Visualisierung von Zellen, deren umgewandeltes λ±RESIII- 5 Molekül noch das ursprüngliche Replacement-Fragment mit dem lux-Operon und somit kein genomisches Insert enthält (Genbankhintergrund) , wurden die Platten 3 h bei 30°C induziert und die
bioluminiszierenden Zellen im Dunkeln ausgezählt. Der Genbankhintergrund betrug dem Anteil leuchtender Zellen zufolge 13 %.
1.3. Screening des Nitrilasegens ni tA aus R . rhodochrous 5 NCIMB 11216
Rekombinante λ±RESIII-Phagen, die chromosomale DNA-Fragmente mit dem Nitrilasegen aus R . rhodochrous NCIMB 11216 enthalten, wurden durch Hybridisierung der Phagenplaques mit der Oligo-Nukleotid- 10 Sonde

identifiziert. Die Sequenz des Oligo-Nukleotids wurde aus einem konservierten Aminosäuresequenzbereich mit dem mutmaßlichen
25 katalytischen Cystein-Rest (Kobayashi et al . , J. Biol . Chem. 267, 1992, 20746-20751 und Proc . Natl . Acad. Sei. USA, 90, 1993, 247-251) . Dieses Motif konnte auch in den bereits bekannten DNA- Sequenzen der Nitrilasegene aus den Stämmen Rhodococcus rhodochrous Jl (GenBank Acc. # D11425) und R . rhodochrous K22 (GenBank
30 Acc. # D12583) gefunden werden.
1.3.1 Transfer von DNA und Hybridisierung
Auf 5 Agarplatten mit insgesamt 2500 Plaques , die wie für die 35 Titerbestimmung in 1.2.3 beschrieben hergestellt wurden, wurden Nylon-Rundmembranen für 1 min aufgelegt . Die Membranen wurden 2 x 5 min mit der Plaque-Seite nach oben auf Filterpapier mit Denaturierungslösung (1,5 M NaCl, 0,5 M NaOH) und dann 2 x 5 min auf Filterpapier mit Neutralisierungslösung (0,5 M Tris/HCl, 40 1,5 M NaCl, pH 7,5) gelegt. Anschließend wurden sie kurz in 50 M NaCl gewaschen, getrocknet und die DNA über 30 min bei 120°C fixiert .
Zur Hybridisierung wurden die Membranen mit 50 ml Hybridisie- rungspuffer für 2 h bei 37°C präinkubiert und dann über Nacht in 45 12 ml Hybridisierungspuffer bei 37°C mit 10 pmol 32P-markiertem Oligo-Nukleotid hybridisiert. Das Oligo-Nukleotid wurde in einem 30 μl-Ansatz mit 80 μCi (γ-32P)-ATP durch 10 U T4-Polynukleotid-
kinase markiert und über eine Tropfsäulen-Gelfiltration mit Sephadex G-25 von überschüssigem (γ-32P)-ATP getrennt.
Nach der Hybridisierung wurden die Nylonmembranen 1 5 min bei Raumtemperatur mit 0,5 g/1 NaCl, 8,8 g/1 Na-Citrat (2 x SSC), 0,1 % SDS und 2 x 15 min bei 32°C mit 0,125 g/1 NaCl, 2,2 g/1 Na-Citrat (0,5 x SSC), 0,1 % SDS gewaschen und für 5 Tage in einer Filmkassette mit Verstärkerfolie mit einem Röntgenfilm exponiert .
1.3.2 Identifizierung und Sequenzierung des nitA-Gens
Es wurden insgesamt 3 positive Klone identifiziert, von denen zwei ein unvollständiges nitA-Genfragment und einer das voll- ständige nitA-Gen trug. Die positiven Plaques wurden ausgestochen, 2 h bei Raumtemperatur in je 0,5 ml SM-Puffer inkubiert und nach Zugabe von 2 Tropfen Chloroform bei 4°C gelagert. Das nach Umwandlung des rekombinanten λ±RESIIIPhagen mit dem vollständigen nitA-Gen resultierende Plasmid (siehe 1.2.3) wurde mit pDHE 6 bezeichnet (Fig. 1 gibt pDHE 6 mit 12 kb genomischen
Genbankfragment von Rhodococcus rhodochrous NCIMB 11216 wieder) und die Umgebung des nitA-Gens mit Hilfe von Southern-Hybridi- sierungen mit der Oligo-Nukleotid-Sonde "nitllower" restriktions- kartiert. Ein 1,5 kb Pvul-Fragment mit dem gesamten nitA-Gen wurde mit Klenow-Fragment behandelt und in EcoRV behandelten pBluescriptSK+ subkloniert (pDHE 7 mit dem 1,5 kb Pvul-Fragment aus dem genomischen 12 kb Genbankfragment von Rhodococcus rhodochrous NCIMB 11216 in pDHE 6, Fig. 2) . Nach weiterer Subklonierung der überlappenden pDHE 7-Fragmente Hindlll (Vektor) /EcoRI, Kpnl /Xhol , EcoRV/BamHI und Apal /EcόRI (Vektor) in jeweils entsprechend verdauten pBluescriptSK+ wurde das Pvul-Fragment nach der Methode von Sanger et al . (Proc. Natl . Acad. Sei. USA 74, 1977, 5463-5467) mit Hilfe eines automatischen Sequenziergerätes doppelsträngig sequenziert. Die Sequenzierreaktion wurde mit einem kommerziell erhältlichen Sequenzierkit mit den ebenfalls kommerziell erhältlichen "universal"- und "reverse"-Primern (Vieira & Messing, Gene, 19, 1982: 259 - 268) durchgeführt. Die für das 1,5 kb Pvul-Fragment ermittelte DNA-Sequenz ist in SEQ ID NO: 1 wiedergegeben. Die abgeleitete Aminosäuresequenz ist SEQ ID NO: 2 zu entnehmen.
2 Heterologe Expression des nitA-Gens aus R . rhodochrous NCIMB 11216 in E. coli und Reinigung des rekombinanten Nitrilase-Proteins
5 Zur Klonierung in einen Expressionsvektor wurde das nitA-Gen aus R . rhodochrous NCIMB 11216 vom Translations-Start- bis zum Translations-Stop-Codon amplifiziert . Hierzu wurden die Primer "nit Ndel " (upper) mit der Sequenz:

verwendet, in denen an das 5'-nitA-Ende eine mit dem Translationsstart überlappende Wdel-Schnittstelle und an das 3'-nitA-
20 Ende eine mit dem Stopcodon überlappende Hindlll-Schnittstelle angefügt ist. Mit diesem Primerpaar wurde das nitA-Gen aus pDHE 7 mit der Pwo-Polymerase in 40 μl Reaktionsvolumen mit je 8 pmol Primer, 100 pg pDHE 7-Template und 2,5 Units Pwo in 10 mM Tris- HC1, pH 8.85, 25 M KC1, 5 mM (NH4)S04, 2 mM MgS0 , 0,2 mM dATP,
25 0,2 mM dTTP, 0 , 2 mM dGTP und 0 , 2 mM dCTP unter den folgenden Bedingungen amplifiziert :
Denaturierung für 3' bei 94°C;
30 25 Zyklen mit Denaturierung für l'bei 93°C, Primeranlagerung für 1' 30'' bei 48°C und Polymerisation für 1' 30'' bei 72°;
Abschluß-Polymerisation für 5' bei 72°C.
35 Das erhaltene ni -PCR-Fragment wurde gereinigt, mit Ndel/HindiII verdaut, in analog verdaute Vektormoleküle pJOE 2702 (Volff et al., Mol. Microbiol . , 21, 1996: 1037 - 1047) integriert und das resultierende Plasmid mit pDHE 17 (Fig. 2: pDHE 17 mit nitA in dem L-Rhamnose-induzierbaren Expressionsvektor pJOE 2702)
40 bezeichnet. Durch die Integration über Ndel/Hindlll steht das nitA-Gen in dem Plasmid pDHE 17 unter Transskriptionskontrolle des in pJOE 2702 enthaltenen Promotors rhap, der aus dem L-Rhamnose-Operon rhaBAD in E. coli (Egan & Schleif, Mol. Biol . 243, 1994: 821 - 829) stammt. Die Transkriptions-Termination 5 des nitA-Gens und die Translations-Initiation der Transkripte erfolgen ebenfalls über Vektorsequenzen (Volff et al . , 1996) . Nach Transformation von pDHE 17 in E. coli JM 109 ist das nitA-Gen
aus R. rhodochrous NCIMB 11216 durch Zugabe von L-Rhamnose induzierbar .
Zur Reinigung des rekombinanten Nitrilase-Proteins über Imidazol- Affinitätschromatographie wurde das nitA-Gen außerdem mit einer 3 ' -Sequenz für ein C-terminales His6~Motiv fusioniert, indem für die Amplifikation des nitA-Gens, die unter den oben genannten Bedingungen erfolgte, neben dem 5 ' -Primer "nitiVdel" (upper) ein modifizierter 3 ' -Primer ohne Stop-Codon mit der Sequenz 5 ' -CGAGGGTGGCTGTCGCCCG-3 ' verwendet wurde und das erhaltene
PCR-Fragment in einen modifizierten pJOE 2702-Vektor integriert wurde, der hinter der BamHI-Schnittstelle die Sequenz [CATJδTGA enthielt. Nach BamHI-Verdau, Klenow-Behandlung und IVdel-Verdau des Vektors wurde das iVdel-nachgeschnittene ni A-Pwo-Amplifikat so durch Ligation am 3 ' -Ende durch "blunt ends" im Leserahmen mit der His6-Motiv-Sequenz fusioniert und das erhaltene Plasmid mit pDHE 18 bezeichnet.
Zur heterologen Expression im Labormaßstab wurde JM 109 (pDHE 17) aus einer 37°C Übernachtkultur 1:200 in 50 ml dYT-Vollmedium (Sambrook et al . , 1989) mit 0,2 % L-Rhamnose angeimpft und die Kultur für 8 h bei 30°C im Schüttelwasserbad unter Induktion kultiviert. Anschließend wurden die Zellen einmal in 50 mM Tris/ HCl, pH 7,5 gewaschen, entsprechend einer ODgoo von 10 in dem- selben Puffer resuspendiert und durch Ultraschall-Behandlung aufgeschlossen. Mit JM 109 (pDHE 18) wurde in analoger Weise verfahren. Das Proteinmuster der durch Ultraschallbehandlung erhaltenen, über Zentrifugation geklärten Rohextrakte im Vergleich zur nicht-induzierten Kontrolle wurde durch SDS-Polyacrylamid- Gelelektrophorese ermittelt; nach den genannten Induktionsbedingungen betrug der Nitrilase-Anteil am Gesamtprotein für JM 109 (pDHE 17) und JM 109 (pDHE 18) je etwa 30 %.
Zur Reinigung der Nitrilase mit Hisg-Motiv aus JM 109 (pDHE 18) wurden die Zellen in 50 mM Tris/HCl, pH 7 , 5 gewaschen, entsprechend etwa 50 ODgoo l resuspendiert und Extrakte mit einer French-Press (2 x bei 20000 psi) hergestellt. Nach Klärung der Extrakte durch Zentrifugation für 30 min bei 15000 g erfolgte die Reinigung über QIAexpress-Ni2+-NTA (QIAGEN) . Pro ml Rohextrakt wurde 1 ml Matrix verwendet, die mit 20 mM Tris/HCl, pH 7 , 5 äquilibriert wurde. Nach dem Beladen der Säule wurde mit 5 Säulenvolumina 20 mM Tris/HCl, 300 M NaCl, 40 mM Imidazol, pH 7,0 gewaschen und mit 20 mM Tris/HCl, 300 mM NaCl, 300 mM Imidazol, pH 7 , 5 eluiert. Die gelelektrophoretische Reinheit des so gewonnenen Nitrilaseproteins war > 90 %. Nach zweimaliger
Dialyse gegen 50 mM Tris/HCl, 0 , 1 mM DTT, 0,5 M (NH4)2S04, pH 7 , 5 konnte die gereinigte Nitrilase bei -20°C gelagert werden.
Für die Rohextrakte wurden für den Umsatz von 2-Benzonitril zu Benzoesäure jeweils um 2 U/mg und für die über QIAexpress-Ni2+-NTA gereinigte Nitrilase mit His6-Motiv bei einer Enzymkonzentration von 50 μg/ml um 11 U/mg ermittelt. 1 Unit entspricht dabei der Produktion von 1 μmol Benzoesäure bei einer anfänglichen Benzo- nitrilkonzentration von 10 mM, 30°C und pH 7,5. Die Umsätze von 2-Benzonitril zu Benzoesäure durch den Nitrilase-Rohextrakt erfolgten in 50 mM Tris/HCl, pH 7,5, die Umsätze mit gereinigter Nitrilase in 50 mM Tris/HCl, pH 7 , 5 , 0 , 1 mM DTT. Die Benzoesäure- Bildung wurde mittels HPLC (Säule RP18, 250 x 4, Laufmittel 47 % Methanol, 0,3 % H3P04,) bestimmt.
Analog dem oben beschriebenen Beispiel wurden eine Reihe von Nitrilen umgesetzt und die erzielten Umsätze bestimmt.
Mit den E. coli-Stämmen JM 109 (pDHE 17 bzw. pDHE 18) wurden verschiedene Nitrile umgesetzt. Die Zellen wurden dazu in 250 ml LB/Amp-Medium + 2 g/1 Rhamnose bei 30°C und 200 upm für 9 Stunden (= h) angezogen. Die Zellernte erfolgt durch Zentrigfugation (20 min, 4°C, 5000 rpm) . Die Zellen wurden anschließend in 10 mM Phosphatpuffer, pH 7,2 resuspendiert, so daß die Konzentration an Biotrockenmasse (BTM) bei 2 g BTM/1 lag. Je 150 μl der Zellsuspension wurden pro well in eine Mikrotiterplatte pipettiert. Die Platte wurde anschließend zentrifugiert. Der Überstand wurde abgesaugt und die Zellpellets zweimal mit Na2HP04 (1,42 g/1 in Finnaqua, pH 7,2) gewaschen. Nach erneutem Zentrifugations- schritt, werden die Zellpellets in der jeweiligen Substratlösung (150 μl) resuspendiert. Je eine 12-er Reihe der Mikrotiterplatte wurde mit einem Substrat versetzt. Als Kontrolle wurde eine Reihe mit der Substratlösung ohne Zellen genommen (Leerwert-Blank) . Die Mikrotiterplatten wurden bei 30°C und 200 rpm für 1 h im Schüttel- inkubator inkubiert. Danach wurden die Zellen abzentrifugiert und im Überstand die entstandene Menge an NH4-Ionen mit Hilfe des Biomek-Geräts bestimmt Die Messung erfolgte bei 620 nm gegen eine Eichkurve, die mit verschiedenen NH4OH-Lösungen erstellt worden war. Als Substrate wurden in Experiment 1 (siehe Figur 3, Tabelle 1) folgende Substrate eingesetzt: Benzonitril (=1), 3-Hydroxypropionitril (=2), 2-Methylglutaronitril (=3), 4-Chlor-3-hydroxybutyronitril (=4), Malonodinitril (=5), Crotono- nitril (=6), Geranonitril (=7), Octandisäuredinitril (=8), Pivalonitril (=9), Aminocapronitril (=10), 3 , 4-Dihydroxybenzo- nitril (=11), 3 , 5-Dibrom-4-hydroxybenzonitril ( =12), 3-Cyan- pyridin (=13), 4-Brombenzylcyanid (=14), 4-Chlorbenzylcyanid (=15), 2-Phenylbutyronitril (=16), 2-Chlorbenzylcyanid (=17),
2-Pyridylacetonitril (=18) , 4-Fluorbenzylcyanid (=19) , 4-Methyl- benzonitril (=20) , Benzylcyanid (=21) . In Experimet 2 (siehe Figur 4, Tabelle 2) , das analog zu Experimet 1 durchgeführt wurde, wurden folgende Substrate verwendet: 2-Phenylpropionitril (=1), Mandelonitril (=2), 2-Amino-2-phenylacetonitril (=3), 2-Hydroxypropionitril (=4), 3 , 3-Dimethoxypropionitril (=5), 3-Cyanthiophen (=6), 3-Cyanmethylthiophen (=7), Benzonitril (=8), Propionitril (=9) , trans-Zimtsäurenitril (=10) , 2-Hydroxy-4- phenylbutyronitril (=11), 3-Phenylglutarsäuredinitril (=12), Fumaronitril (=13), Glutaronitril (=14) Valeronitril (=15).

Claims
1. Isolierte Nukleinsäuresequenz, die für ein Polypeptid mit Nitrilaseaktivität codiert, ausgewählt aus der Gruppe:
a) einer Nukleinsäuresequenz mit der in SEQ ID NO: 1 dargestellten Sequenz,
b) Nukleinsauresequenzen, die sich als Ergebnis des degenerierten genetischen Codes von der in SEQ ID NO: 1 dargestellten Nukleinsäuresequenz ableiten,
c) Derivate der in SEQ ID NO: 1 dargestellten Nukleinsäure- sequenz, die für Polypeptide mit der in SEQ ID NO: 2 dargestellten Aminosäuresequenzen codieren und mindestens 95 % Homologie auf Aminosäureebene aufweisen, ohne daß die enzymatische Wirkung der Polypeptide wesentlich reduziert ist.
2. Aminosäuresequenz codiert durch eine Nukleinsäuresequenz gemäß Anspruch 1.
3. Aminosäuresequenz nach Anspruch 2, codiert durch die in SEQ ID NO: 1 dargestellte Sequenz.
4. Nukleinsäurekonstrukt enthaltend eine Nukleinsäuresequenz gemäß Anspruch 1, wobei die Nukleinsäuresequenz mit einem oder mehreren Regulationssignalen verknüpft ist.
5. Vector enthaltend eine Nukleinsäuresequenz gemäß Anspruch 1 oder ein Nukleinsäurekonstrukt gemäß Anspruch 4.
6. Rekombinanten Mikroorganismus enthaltend eine Nukleinsäure- sequenz gemäß Anspruch 1, ein Nukleinsäurekonstrukt gemäß
Anspruch 4 oder einen Vector gemäß Anspruch 5.
7. Rekombinanten Mikroorganismus nach Anspruch 6, wobei es sich bei dem Mikroorganismus um ein Bakterium der Gattungen Escherichia, Rhodococcus, Nocardia, Streptomyces oder Mycobacterium handelt.
8. Verfahren zur Herstellung von chiralen oder achiralen Carbonsäuren, dadurch gekennzeichnet, daß man Nitrile in Gegenwart einer Aminosäuresequenz gemäß Anspruch 2 oder 3 oder einem wachsenden, ruhenden oder aufgeschlossenen Mikroorganismus gemäß Anspruch 6 oder 7 zu den chiralen oder achiralen Carbonsäuren umsetzt.
9. Verfahren zur Herstellung von chiralen oder achiralen Carbonsäuren nach Anspruch 8, dadurch gekennzeichnet, daß man Nitrile der allgemeinen Formel I
in Gegenwart einer Aminosäuresequenz gemäß Anspruch 2 oder 3 oder einem wachsenden, ruhenden oder aufgeschlossenen Mikroorganismus gemäß Anspruch 6 oder 7 zu Carbonsäuren der allgemeinen Formel II
umsetzt,
wobei die Substituenten und Variablen in den Formeln I und II folgende Bedeutung haben:
n = 0 oder 1
m = 0, 1, 2 oder 3, wobei bei m > 2 zwischen zwei benach- barten Kohlenstoffatomen eine oder keine Doppelbindung vorhanden ist,
p = 0 oder 1
A, B, D und E unabhängig voneinander CH, N oder CR3
H = 0, S, NR4, CH oder CR3 , wenn n = 0, oder CH, N oder CR3 , wenn n = 1 , wobei zwei benachbarte Variablen A, B, D, E oder H zusammen einen weiteren substituierten oder unsubstituierten aromatischen, gesättigten oder teilweise gesättigten Ring mit 5 bis 8 Atomen im Ring bilden können, der ein oder mehrere Hetero- atome wie 0, N oder S enthalten kann und wobei nicht mehr als drei der Variablen A, B, D, E oder H ein Heteroatom sind,

12. Verfahren nach den Ansprüchen 8 bis 11, dadurch gekennzeichnet, daß das Verfahren bei einer Temperatur zwischen
35 0°C bis 80°C durchgeführt wird.
13. Verfahren nach den Ansprüchen 8 bis 12 , dadurch gekennzeichnet, daß die achirale oder chirale Carbonsäure über Extraktion oder Kristallisation oder Extraktion und 40 Kristallisation in Ausbeuten von 60 bis 100 % aus der Reaktionslösung gewonnen wird.
