Definitions
- the invention relates to methods and devices for cell examination, in particular cell assays or cell test arrangements, their production and methods for their use.
- the invention also relates to uses of cell traces on substrate surfaces.
- the object of the invention is to develop extremely low-stress methods and devices for cell examination which can also be used in the medical field and which use as many highly-specific detection techniques that have already been tried and tested, e.g. Immunofluorochromation of proteins, nucleotides and lipids, as well as destructive processes such as X-ray or electron beam microanalysis can be combined and allow the parallel examination of a large number of individual cells.
- the object of the invention is also to provide new applications of cell traces.
- the basic idea of the invention is, in particular, for the examination or cultivation of biological cells or their interactions with other cells or substances, first to generate cell traces of the cells to be examined on a substrate surface and then to subject these cell traces to the desired analysis or investigation.
- the generation of cell traces by adherent cells growing or moving on surfaces is known per se and is described for example by ED Hay et al. in "Exp. Biol. Med.”, Vol. 10, 1985, pp. 174 ff.
- the structures and properties of cell traces are explained below with reference to FIGS. 2 and 3.
- the cell traces are preferably generated using substrate surfaces which are at least partially microstructured and / or modified in a suitable manner.
- the microstructuring of the substrate surface is in particular intended to promote cell track production in certain substrate areas, for example along certain paths, and to hinder or exclude it in other substrate areas.
- the surface modification also means that not only can the material traces remaining on the substrate be examined naturally, but also artificially separated material residues (achieving larger amounts of traces).
- the substrate modification includes, in particular, the application of molecules that offer binding sites to which a specific, sought-after cell surface molecule can specifically couple.
- the examination methods used according to the invention include all techniques known per se for cell examination and cell treatment, it being possible to use both non-destructive and destructive techniques or, if appropriate, biochemical amplification techniques (e.g. PCR process).
- the invention also relates to the use of cell traces for manipulating the interaction of biological cells with solid substrates.
- cell traces By deliberately applying cell traces on synthetic or biological substrates in accordance with the principles explained in the present description, biocompatible carriers are provided for the biological cells to be manipulated.
- the manipulation consists in particular in the targeted cell cultivation (tissue building) on the substrate areas covered with cell traces.
- the substrate surfaces used according to the invention can consist of synthetic, inorganic or organic material or can also be formed by biological material (eg bone material).
- the invention has the following advantages. For the first time, a single cell-specific method for cell examination is specified, in which the cell being examined remains unaffected and unchanged by the examination process. This allows a significant expansion of the use of single cell examinations in pharmacology, toxicology, medical diagnostics and biochemistry.
- the cell examination can be performed highly parallel on a large number of cells by simultaneously generating many traces on a substrate. Since the microstructuring of the surfaces makes it possible to assign a cell trace to an investigated cell (donor cell), the highly parallel single cell examination also remains cell-specific.
- the microstructuring of the substrate surface can preferably be carried out using techniques which are known per se from semiconductor processing.
- the cells are not burdened by any staining or marking techniques except for the generation of traces. They are thus not contaminated or changed and can be subjected to medical use or cryopreservation or further cultivation. Instead of the cells in cells as before, the cell residues are subjected to a specific labeling or evaluation. This can also be destructive (e.g. step-by-step enzymatic degradation) or by immunofluorochromation in toxic concentration ranges.
- the cell cultivation according to the invention has the advantage that any substrate materials, such as are of interest for the implantation of bone materials, can be made biocompatible with cell traces. Novel substrates for in-vitro tissue construction are created, which have a considerably expanded range of applications.
- the cell track-based modification of substrate surfaces enables application-optimized cell cultures to be optimized Substrate materials are applied, which would not be compatible with each other without the cell traces.
- FIG. 2 shows a schematic illustration of the basic structures of cell traces in the form of filaments (A) and membrane spots (B),
- FIG. 5 shows an exemplary embodiment of the invention in which the basic principle illustrated in FIG. 1 is implemented with a large number of parallel tracks
- Fig. 6 shows another embodiment of the invention with a plurality of parallel cell lines
- Fig. 7 shows another embodiment of the invention with crossing cell lines.
- FIG. 1 shows the basic structure of a cell trace-based system according to the invention.
- the surface of a substrate 11 in the ⁇ m and mm range is structured as follows or its surface properties are changed.
- a preferred path is formed by areas 12 of the surface on which cells can adhere only poorly and areas 13, 15, 17 (web surface areas) where cells can adhere well, on which a cell 16 can actively move.
- the field 15 in the surface area of the web has been modified (chemically, mechanically etc.) in such a way that the cells lose parts of their membrane and internal components 14a, 14b which adhere to the substrate.
- filaments 14a and membrane patches (or membrane patches) 14b which are explained in detail below with reference to FIGS. 2A and 2B.
- the cell continues to move in the direction of arrow 18.
- the cell track can now be analyzed non-destructively or destructively.
- the material left behind characterizes the donor cell with regard to the membrane composition (receptors, carriers, lipids, etc.), but also with regard to internal components of the cytoplasm, from which medical, toxicological, pharmacological and other applications can be derived. Either one or more cells can move on a path and create tracks.
- the substrate 11 consists, for example, of glass, mica, inorganic crystal material or semiconductor material.
- the substrate surface is structured or modified to form the preferred path on which the cell moves preferentially and leaves cell traces.
- the microstructuring and / or modification of the substrate surface comprises a locally selective influencing of the preferred path between the regions 12 of the substrate surface.
- the segmentation of the preferred path, for example in the areas 13, 15 and 17, is provided depending on In the design of the respective area, the cell traces are particularly numerous or particularly small or are left behind in relation to a specific composition. This can also be seen from the examples explained below.
- microstructuring or modification of the preferred path includes, for example:
- the layer thickness can be selected from the thickness of a molecular layer down to the ⁇ m range.
- the molecular monolayers are preferably applied using the Langmuir blodget technique. In general, thick-film techniques and / or plasma treatments can also be used to apply the layer.
- Nano or micro structuring of surfaces i.e. Application of patterns in nm or ⁇ m dimensions, to which membrane parts, but especially natural contact molecules of the cell, such as those of the integrin and catherine family, can adhere (e.g. structuring via photo or electron beam lithography.
- the substrate loading takes place, for example, by rinsing from a suspension, for example through a channel of the microsystem, with a manipulator (capillary, separate microsystem or optical tweezers) or also by active growth.
- a manipulator capillary, separate microsystem or optical tweezers
- filament or membrane spot When the cells migrate over substrate surfaces (eg over a clean glass surface), the cells leave behind physiological conditions filamentous or spot-like traces, which are referred to below as filament or membrane spot and are explained with reference to FIGS. 2A and 2B.
- the traces are usually structures that are membrane-covered and filled with cell contents. Typical sizes of these structures in terms of width and height are in the ⁇ m and sub- ⁇ m range. While the length of a membrane spot generally corresponds essentially to its width, the length of a filament is variable. The filament length can be up to a few millimeters.
- the components of the cells of interest that can also be found in the cell traces are membrane proteins 210, surface proteins and receptors 211, 212, cytoplasm components 213, 214 and the lipids 215 in the membrane (see FIG. 2A).
- these constituents which include membrane proteins 220 and lipid composition 225 in FIG. 2B, vesicles 221, organelles 222 and genetic material 223 also occur in the membrane spots.
- Cytoplasm 224 is also present.
- the cell traces contain material capable of analysis, which includes the constituents mentioned, in sufficient quantity. This means that the known analysis or investigation methods can advantageously be implemented without separate enrichment steps.
- the surface proteins and receptors 211, 212 comprise, for example, a trace protein 211 in the membrane and a coupled receptor 212 with a chromophoric group. With suitable light excitation, the receptor 212 emits fluorescent light which indicates the presence of the trace protein 211. Since the receptor coupling is protein-specific, the protein complex present in the trace can be detected with the fluorescent light. In an analogous manner, other detection techniques can also be implemented, such as those provided for in the ELISA and RIAS assays.
- the specific detection of certain components of the donor cell is thus carried out on the cell traces.
- the components can be arranged on the surface or inside the cell traces. In the latter case, it is provided to dissolve the membrane of the cell track with suitable solvents or to permeate it mechanically or electrically.
- the destructive measurement on the cell tracks without changing the donor cell to detect molecular or microscopic components in the cell tracks represents a particular advantage of the invention.
- the cell trace-based analyzes are particularly suitable for combination with highly sensitive measurement techniques. These include, for example, fluorescence correlation analysis for the detection of single molecules and for the determination of binding constants, mass spectrometry for elemental analysis and confocal laser scanning microscopy. Genetic material in the traces can e.g. are amplified via a PCR process, which provides a novel technique of genetic analysis which does not influence the donor cell in its physiological cells.
- the amount of cell residues is recorded as a quantitative measure of the strength of the donor cell adhering to the substrate surface and thus of the amount of certain binding complexes in its membrane.
- the trace structure is recorded as a measure, for example the ratio The proportion of filaments, the proportion of branches, the proportion of patches, etc. (comparison of cell trace base elements in terms of their quantity).
- the material composition of the tracks is recorded as a measure, e.g. Lipid / protein content, specific appearance of certain receptors (including the immunoglobulin families), specific appearance of lipids, nucleotides etc.
- Optical parameters of the tracks are recorded as a measure, such as absorption, transmission, non-linear properties etc.
- the change in a cell track by a subsequent cell of the same or a different type is used as a measure detected .
- the track characterization takes place after fixation or contrasting, e.g. using high-resolution microscope methods (scanning electron microscopy, AFM, SNOM etc.).
- a negative or other duplicate or a multiplication of trace parts as in the PCR technique is used as a measure for the comparison.
- the dimensions mentioned here are recorded as sizes specific for the donor cells, which are used for given comparison values to characterize the donor cell or to compare the dimensions with the corresponding results in other cells to characterize the different behavior of the cells.
- Embodiments of devices according to the invention are explained below with reference to FIGS. 3 to 7. Details of the substrate surfaces used according to the invention are discussed. Details of an overall apparatus for cell lane examination are not shown, since these are known per se with regard to the handling of assays or substrates loaded with samples and the adaptation to the respectively desired examination methods.
- the figures 3 and 4 are schematic sectional views of substrates 31, 41, each of which is equipped with a modification layer 32 or 42 in the web surface area 35 or 45 for the preferred adhesion of cells and leaving cell traces 34 and 44, respectively.
- the modification layer 32 (or 42) offers binding sites at which a specific, sought-after cell surface olekül 33 (or 43) can couple.
- a cell which has such a sufficient number of such molecules will accordingly be more firmly bound and leave more trace material 34 (or 44) as it migrates across the substrate than other cell types.
- the detection of the amount of the material left behind provides information about the content of the predetermined cell surface molecule in the examined cell. The cell itself is not burdened by the measurement.
- the cell trace measurement can be modified according to FIG. 4. After generation of the cell traces 44, these are treated with the solution of a fluorescent marker, which as the marker molecule 46 e.g. is nonspecifically incorporated into the lipid parts of cell track 44. With suitable light excitation and fluorescence measurement, the number of marker molecules 46 and thus the quantitative amount of cell trace material 44 can be inferred from the intensity of the fluorescent light.
- a fluorescent marker which as the marker molecule 46 e.g. is nonspecifically incorporated into the lipid parts of cell track 44.
- the preferred path according to FIG. 1 or the modified path surface area 35 (or 45) according to FIG. 3 (or 4) can be formed one or more times with the most varied geometries on the substrate. Preferred lanes are described below. However, with a suitable substrate structuring, curved (e.g. circular) preferred paths are also possible.
- FIG. 5 shows an example of a parallel embodiment of the basic principle explained in FIG. 1 with a large number of straight, preferred webs running in parallel.
- Materials 52 that prevent cell adhesion are applied to a substrate 51, which can be made of glass, silicon or plastic, for example.
- the tracks 57 range from input depots 53 via surface fields 55 to output depots 58.
- the surface fields 55 are treated in such a way that cell traces are preferably generated. If the cells reach the starting depots 58, which have also been prepared for adhesion, they are held or removed there in order to be subjected to cultivation, cryopreservation or another procedure. The trace can be analyzed using all common micro detection methods (fluorescence, isotope labeling, element analysis etc.).
- the surface fields 55 with the cell tracks (not shown) are arranged as segments of the tracks 57 as a row, each with the same distance from the respective input depot 53. This facilitates the parallel, simultaneous evaluation of the cell traces.
- FIG. 6 shows a substrate surface in the form of a microsystem, which was compartmentalized into different surface areas in a similar manner as is the case in FIG. 5.
- the substrate here consists either of two parts 61a, 61b or a part with a predetermined breaking point 61c.
- the cells leave traces on the fields 65. If they have arrived at the output depots 68 on their hike over the substrate, the part 61b is removed or broken off. As shown in the left part of FIG. 6, the traces and cells are then on separate substrates 31 and 32, so that they can be further processed in different ways.
- FIG. 7 shows a substrate surface which is structured in such a way that two preferential paths which are set up for the migration of the donor cells intersect.
- the webs 77a and 77b run essentially perpendicular to one another on the substrate 71.
- a surface modification or structuring for requesting the adherence of cell traces is applied in the intersection area 75, as was explained above.
- the substrate 71 is structured such that there is no cell migration and no cell traces adhere.
- interactions between different cells can preferably be examined.
- the cell 76a first migrates over the area 75 and leaves cell traces there.
- the cell 76b then moves over the same area 75 with the cell traces present.
- An optical-microscopic method or another examination method is then used to determine whether the traces of the first cell 76a have been changed, superimposed or removed by the second cell 76b. It can also be detected whether geometric correlations occur between the cell tracks, i.e. whether the successor cells follow the traces of the predecessor cells or just avoid them. This in turn can be used to develop cell-based assays for medicine, biotechnology and pharmacy with high specificity.
- a substrate with crossed paths can in turn be formed on a common support in order to achieve parallel processing.
- a substrate according to the invention is preferably produced in such a way that a carrier material is first provided with a coating which is suitable for cell migration and -adhesion is unfavorable (eg strongly negatively charged molecules).
- This coating is then structured in accordance with the desired course of the preferred webs by ablation, so that the carrier material is exposed in accordance with certain geometric shapes, which then form the preferred webs.
- the preferred web is segmented, ie the structuring and / or modification of the carrier material is applied for increased cell adhesion.
- the web widths are preferably adapted to the characteristic extent of an adherent cell and amount to approx. 50 ⁇ m.
- the web lengths can also be selected depending on the application. They are e.g. 3 to 4 characteristic cell diameters (i.e. around 150 to 200 micrometers) up to longer lengths in the millimeter range.
- cell traces are applied to substrates for cell cultivation.
- Solid or flat solid materials of synthetic or biological origin are used as substrates.
- glass, ceramic or plastic materials or polished bone disks are used as the substrate.
- tissue-producing cells such as, for example, chondrocytes, osteoplasts or epithelial cells are placed on the substrate surfaces in accordance with the principles explained above, in order to migrate there, leaving cell traces.
- the substrate surface can be structured or otherwise modified to apply the largest possible cell traces (see above). After the formation of a closed cell trace surface, tissue builds up on the modified substrate. Tissue-producing cells, preferably of the type with which the cell tracks were generated, are cultivated on the modified substrate. On Depending on the application, substrate with cultivated tissue cells is then used as an implant in the human body.
