Abstract

La présente invention concerne un procédé simple, peu onéreux et précis permettant d'introduire une substance étrangère dans une population prédéterminée de cellules cibles. En un premier temps, le procédé consiste à disposer (a) des microsondes biologiquement inertes positionnées sur un support, (b) une masse solide ou quasi-solide de cellules cibles définissant une interface avec les microsondes, et (c) une substance biologique à l'interface. En un second temps, le procédé consiste à faire entrer les cellules en contact avec les microsondes, de sorte que les microsondes perforent de façon non létale les parois ou membranes cellulaires des cellules. Les microsondes, qui font de préférence corps avec le support, sont préparées par attaque chimique d'une plaquette unique d'un cristal telle que le silicium. La forme des microsondes est de préférence pyramidale. Le contact entre cellules cibles et microsondes peut se faire soit in vitro, soit in situ. Ce procédé convient avantageusement à presque tous les types de cellules et de substances biologiques susceptibles d'être introduites dans les cellules décrites ici.
