Description

Die vorliegende Erfindung betrifft ein System zur Inkubation von Probeflüssigkeiten, mit
  • einem Rack, bei dem Inkubationsgefäße in Bohrungen einer Halteplatte eingehängt sind,
  • einem Inkubationsblock mit Bohrungen zur Aufnahme von Inkubationsgefäßen,
  • einer Vorrichtung zur Temperierung des Inkubationsblockes, die thermisch mit dem Inkubationsblock verbunden ist,
wobei die Bohrungen in der Halteplatte und die Bohrungen im Inkubationsblock so aufeinander abgestimmt sind, daß sich die Inkubationsgefäße des Racks in die Bohrungen des Inkubationsblockes einpassen, wenn das Rack auf den Inkubationsblock gestellt wird, sowie ein Verfahren zur Inkubation von Probeflüssigkeiten.
Systeme zur Inkubation von Probeflüssigkeiten werden insbesondere im Bereich der klinischen Diagnostik eingesetzt. Zur Durchführung vieler diagnostischer Verfahren ist es erforderlich, Reaktionen bei einer vorgegebenen Temperatur ablaufen zu lassen, um die Reaktionsgeschwindigkeit steuern zu können. Eine Inkubation von Probeflüssigkeiten wird jedoch nicht ausschließlich zur Durchführung einer analytischen Reaktion eingesetzt, vielmehr werden Inkubatoren auch eingesetzt, um in Probeflüssigkeiten vorhandene Organismen wie Bakterien, Hefen, Pilze, Viren etc. zu vermehren, um nachfolgend die vermehrten Organismen bestimmen zu können. Inkubatoren werden beispielsweise auch zur Vermehrung von DNA oder RNA eingesetzt. Die entsprechenden Geräte werden als Thermocycler bezeichnet.
Im Stand der Technik sind Geräte zur Inkubation von Probeflüssigkeiten bereits seit langer Zeit bekannt. Die bekannten Geräte können prinzipiell in 2 Klassen eingeteilt werden. Zu der ersten Klasse gehören solche Inkubatoren, bei denen die Probegefäße durch eine fluide Phase beheizt werden. Vorrichtungen diesen Typs sind beispielsweise in EP-A-0 363 143 und EP-B-0 087 028 beschrieben. In den beiden genannten Dokumenten werden Vorrichtungen beschrieben, bei denen sich Probegefäße in einem Halter befinden und mit diesem Halter in eine Flüssigkeit eingetaucht werden. Eine Temperierung der Probengefäße bzw. Probeflüssigkeiten erfolgt durch die Flüssigkeit innerhalb des Inkubators. Aufgrund der fluiden Eigenschaften der flüssigen Phase paßt sich die Flüssigkeit der Form der Inkubationsgefäße perfekt an.
Die zweite Klasse der Inkubationsvorrichtungen umfaßt sogenannte Metallblockinkubatoren, bei denen sich innerhalb eines Inkubationsblockes aus wärmeleitendem Material Bohrungen befinden, die zur Aufnahme von Inkubationsgefäßen dienen. Derartige Inkubationsvorrichtungen sind beispielsweise aus EP-A-0 151 781 oder US-4,335,620 bekannt.
US-4,335,620 beschreibt einen Inkubator, der einen festen Block besitzt, der aus einem wärmeleitenden Material wie z. B. Aluminium besteht. Der Block dient sowohl zur Aufnahme von Probengefäßen als auch als Wärmesenke zur Temperaturkontrolle der Gefäße. Das Dokument bezieht sich auf eine spezielle Konstruktion zur thermischen Isolation der Vorrichtung gegenüber der Umgebung. Der Gesamtwärmeverlust der Apparatur wird minimiert.
Das Dokument US-4,727,034 betrifft eine Vorrichtung zur Thermostatisierung von Probeflüssigkeiten. Die Gefäße mit Probeflüssigkeiten werden dabei in ein Rack aus gut wärmeleitfähigem Material eingesetzt. Das Rack seinerseits wird in der Vorrichtung zwischen zwei Seitenwände eingeklemmt, von denen mindestens eine beheizt wird, so daß eine Temperierung des Racks und damit der Probeflüssigkeiten in den Gefäßen möglich ist.
EP-A-0 488 769 befasst sich ebenfalls mit einer Vorrichtung zur Inkubation von Flüssigkeiten.
Jede der zwei genannten Klassen von Inkubatoren besitzt ihre eigenen Vor- und Nachteile. Bei der Verwendung von Fluiden zur Wärmeübertragung können Anordnungen von Inkubationsgefäßen verwendet werden, die zusammenhängend sind. Dies macht es möglich, eine Vielzahl von Gefäßen gleichzeitig in das Wärmebad zu tauchen. Ein Nachteil dieser Inkubationstypen liegt jedoch darin, daß die Fluide an den Außenwandungen der Inkubationsgefäße anhaften. Bei einem Herausnehmen der Inkubationsgefäße z. B. für nachfolgende Analyseschritte tropft diese Flüssigkeit ab und führt zu einer Verschmutzung des Gerätes. Wesentlich nachteilhafter ist es noch, wenn an den Gefäßen anhaftende Flüssigkeit oder Flüssigkeit aus dem Inkubator verspritzt und zu einer Störung der Analyse führt. Insgesamt ist die Handhabung von feuchten Reagenzgefäßen nachteilig und sollte demnach vermieden werden.
Die genannten Nachteile besitzen Metallblockinkubatoren nicht, jedoch besitzt dieser Inkubatortyp seinerseits andere Nachteile. Zur Gewährleistung eines guten Wärmeübertrages zwischen Probeflüssigkeit und Inkubator muß gewährleistet sein, daß die Inkubationsgefäße mit dem Inkubator auf möglichst großer Fläche in direkten Kontakt kommen, um einen guten Wärmeübertrag zu gewährleisten. Bei den in US-4,335,620 und US-4,727,032 beschriebenen Inkubatoren wird diese Forderung erreicht, in dem die Vorrichtung innerhalb des Inkubators Bohrungen aufweist, die in ihrer Form den Inkubationsgefäßen entsprechen. Die Reagenzgefäße und die Bohrungen im Inkubator müssen demnach genau aufeinander abgestimmt sein. Aufgrund der mechanischen Gegebenheiten war es bisher nicht möglich, eine Anordnung bestehend aus mehreren Gefäßen in einen Metallblockinkubator einzusetzen, ohne daß es zu einem Verklemmen kommt.
Die Erfindung hatte die Aufgabe, diesen Nachteil von Metallblockinkubatoren zu beseitigen und eine Inkubationsvorrichtung vorzuschlagen, die die Vorteile von Metallblockinkubatoren und Fluidinkubatoren miteinander verbindet. Insbesondere war es Aufgabe der Erfindung ein System sowie ein Verfahren zur Inkubation vorzuschlagen, das ohne eine Verwendung von Fluiden zur Wärmeübertragung auskommt, das eine einfache, schnelle und zuverlässige Bestückung von Inkubatoren mit einer Vielzahl von Inkubationsgefäßen ermöglicht, und das ein Verklemmen der Inkubationsgefäße verhindert .
Ausgehend von EP-A-0488769 wird diese Aufgabe gelöst durch ein System bzw. durch ein Verfahren zur Inkubation von Probeflüssigkeiten, wie es in Anspruch 1 bzw. in Anspruch 7 definiert ist.
Unter Probeflüssigkeiten für die Inkubation sollen Blutproben, Serumproben, Urin, Lebensmittelproben, Wasserproben, Reaktionsansätze und dergleichen verstanden werden. Insbesondere sind auch solche Flüssigkeiten mit umfaßt, die aus Probematerialien durch Versetzen mit Reagenzien gewonnen werden. Beispielsweise sollen auch DNA-enthaltende Proben umfaßt sein, die zur Amplifikation der DNA mit Reagenzien versetzt werden.
Ein erfindungsgemäßes System kann insbesondere zur chemischen und klinischchemischen Analyse verwendet werden, da in diesem Bereich die Einhaltung bestimmter Temperaturen bzw. die Vorbehandlung von Proben bei bestimmten Temperaturen von entscheidender Bedeutung für die Verläßlichkeit der Analyseergebnisse ist. Ein erfindungsgemäßes System zur Inkubation kann beispielsweise auch als sogenannter Thermocycler eingesetzt werden, der bei der Amplifikation von DNA Verwendung findet.
Unter Inkubation soll die Temperierung von Probeflüssigkeiten über einen bestimmten Zeitraum mit einem vorgegebenen Temperaturprofil verstanden werden. Bei den am häufigsten eingesetzten Inkubatoren besitzt der Inkubationsblock eine genau definierte Temperatur, die über einen langen Zeitraum konstant gehalten werden kann. In diesem Falle wird die Inkubation durch ein Einstellen des Inkubationsgefäßes in den Inkubationsblock begonnen und durch ein Herausnehmen des Inkubationsgefäßes aus dem Block beendet. Es ist jedoch erfindungsgemäß auch vorgesehen, daß die Temperatur des Inkubationsblockes zeitabhängig gesteuert wird. Auf diese Weise kann eine Probeflüssigkeit während der Anwesenheit des Inkubationsgefäßes im Inkubationsblock wechselnden Temperaturen ausgesetzt werden. Zeitlich wechselnde Temperaturen werden beispielsweise in sogenannten Thermocyclern zur Amplifikation von DNA mit der "Polymerase Chain Reaction" eingesetzt.
Ein System zur Inkubation von Probeflüssigkeiten im Sinne der Erfindung kann ein sogenanntes "Stand alone-Modul" oder eine Teileinheit innerhalb eines Analysegerätes darstellen. Als Inkubationsblock wird der Teil des Inkubationssystemes bezeichnet, der Bohrungen zur Aufnahme von Inkubationsgefäßen besitzt. Erfindungsgemäße Inkubatoren gehören demnach zur Klasse der Metallblockinkubatoren. Der Inkubationsblock ist bevorzugt aus einem Stück gefertigt, beispielsweise aus einem Zylinder, in den Bohrungen eingebracht werden oder durch Gießen eines Materials in eine Form. die Vertiefungen für Inkubationsgefäße vorsieht. Geeignete Materialien für den Inkubationsblock sind Metalle, insbesondere Aluminium und auch Legierungen, wie beispielsweise Messing.
Die Bohrungen innerhalb des Inkubationsblockes besitzen in der Regel eine zylindrische Form bzw. sind Kegelstümpfe. Durch die Bezeichnung "Zylinder" sollen sowohl Zylinder mit einem runden als auch eckigen Querschnitt umfaßt sein. Die Bohrungen haben in der Regel eine Tiefe von wenigen Zentimetern und laufen bevorzugt zum Inneren des Inkubationsblockes hin konisch zu.
Der Inkubationsblock kann weiterhin Führungselemente besitzen, die die Positionierung eines erfindungsgemäßen Racks erleichtern. Im Inneren des Inkubationsblockes oder an seiner Außenseite können sich Sensoren (z. B. Lichtschranken) befinden, welche die Anwesenheit eines Racks detektieren.
Der Inkubationsblock steht weiterhin in thermischen Kontakt mit einer Vorrichtung zur Temperierung. Ist der Inkubationsblock ausschließlich zur Erwärmung von Inkubationsgefäßen oberhalb Raumtemperatur vorgesehen, so kann diese Temperierungsvorrichtung eine einfache elektrische Heizung sein. Es ist jedoch von Vorteil, neben einem heizenden Element auch eine Möglichkeit zur Kühlung des Inkubationsblockes vorzusehen. Ein Kühlung kann beispielsweise durch einen Teil des Inkubationsblockes realisiert werden, der von kaltem Wasser durchflossen wird. Bei größeren Inkubationssystemen kann ebenfalls zur Kühlung eine Wärmepumpe analog einem Kühlschrank eingesetzt werden. Vorrichtungen, mit denen sowohl Erwärmung als auch Kühlung möglich ist, sind Peltier-Elemente, die mittels elektrischer Energie Wärme transportieren.
Die genannte Vorrichtung zur Temperierung des Inkubationsblockes kann entweder thermisch an den Inkubationsblock angekoppelt werden, wie dies beispielsweise in US-4,335,620 beschrieben ist, oder die Mittel zur Temperierung können sich innerhalb des Inkubationsblockes selbst befinden.
Innerhalb des Inkubationsblockes, bevorzugt in Nähe der Bohrungen für die Inkubationsgefäße, können sich Temperaturdetektoren befinden, mit denen die Einstellung der gewünschten Temperatur kontrolliert werden kann. Die Regelung der Vorrichtungen zur Temperierung ist im Stand der Technik hinlänglich bekannt (EP-B-0 273 969).
Der Inkubationsblock besitzt eine Zahl von nach oben offenen Bohrungen, die in der Regel periodisch angeordnet sind. Die Bestückung dieser Bohrungen mit Inkubationsgefäßen erfolgt im Stand der Technik entweder manuell oder mit Hilfe eines Roboterarmes. Während eine manuelle Bestückung zeit- und personalintensiv ist, macht eine Vorrichtung zur automatischen Bestückung ein Gerät relativ aufwendig und damit teuer. Selbst bei einer automatischen Bestückung müssen dem Gerät Inkubationsgefäße von außen in einer Form zugeführt werden, die für eine Robotik geeignet ist.
Bei der vorliegenden Erfindung erfolgt die Bestückung des Inkubationblockes mit Hilfe eines Racks, in dem sich Inkubationsgefäße befinden. Erfindungsgemäß hängen die Inkubationsgefäße in Bohrungen einer Halteplatte. Die Bohrungen innerhalb der Halteplatte sind entsprechend den Bohrungen innerhalb des Inkubationsblockes angeordnet. Zur Bestückung des Inkubationsblockes mit Inkubationsgefäßen wird das Rack über den Inkubationsblock bewegt und dort abgesenkt, so daß sich die Inkubationsgefäße in die Bohrungen des Inkubationsblockes bewegen.
Eine Halteplatte besitzt die Form einer dünnen Scheibe, in der Bohrungen angebracht sind, die zur Aufnahme von Inkubationsgefäßen dienen. Die Halteplatte kann aus einer Vielzahl von formstabilen Materialien gefertigt sein. In der Anwendung kommen Metalle und insbesondere Kunststoffe in Frage. Die Anordnung der Bohrungen in der Halteplatte entspricht der Anordnung der Bohrungen an der Oberseite des jeweils verwendeten Inkubationsblockes. Eine Halteplatte kann beispielsweise ein Vollkreis sein oder bevorzugt ein Kreissegment bzw. ein Ausschnitt aus einer Kreisscheibe. Die Dicke der Halteplatte muß so bemessen sein, daß sie eine mechanische Stabilität gewährleistet, wenn Inkubationsgefäße in der Halteplatte hängen. Ist die Halteplatte aus Kunststoff gefertigt, so wird die Materialstärke in der Regel ein bis mehrere Millimeter betragen. Selbstverständlich ist eine Reduktion der Materialstärke möglich, wenn Verstrebungen eingebaut sind, die die mechanische Stabilität erhöhen.
Die Bohrungen innerhalb der Halteplatte können mit Werkzeugen in eine Halteplatte aus Vollmaterial eingebracht werden oder sie sind bereits in der Form zur Herstellung der Halteplatte vorgesehen. Erfindungsgemäß ist der Querschnitt dieser Bohrungen von einiger Bedeutung, so daß die Toleranzen für den Bohrungsquerschnitt unter 0,05 mm liegen sollten.
Bei der Auswahl der Materialien für die Halteplatte sind schlecht-wärmeleitende Materialien bevorzugt, da bei einer bevorzugten Ausführungsform die Halteplatte direkt auf dem Inkubationsblock zu liegen kommt. Schlecht-wärmeleitende Halteplatten verringern daher den Wärmeverlust des Inkubationsblockes an die Umgebung.
An der Unterseite der Halteplatte können sich bevorzugt Standelemente befinden, die gewährleisten, daß die Inkubationsgefäße, die in der Halteplatte hängen, nicht auf der Unterlage aufstoßen. Die Standelemente machen die Halteplatte zu einem Gestell, in dem die Inkubationsgefäße auch außerhalb eines Inkubationsgerätes aufbewahrt werden können. Dies ist insbesondere wichtig, wenn die Halteplatte bereits werksmäßig mit Inkubationsgefäßen bestückt werden soll. In diesem Falle ermöglichen die Standelemente eine einfachere Verpackung der Halteplatte mit den Inkubationsgefäßen und weiterhin ein leichteres Handling für den Benutzer.
An der Oberseite der Halteplatte können Laschen befestigt sein, die ein Benutzer ergreifen kann, wenn er das Rack transportieren will. Die Laschen sind insbesondere hilfreich beim Einsetzen des Racks in den Inkubationsblock, da hier ein Ergreifen der Halteplatte selbst unbequem wäre.
In die Bohrungen der Halteplatte werden bevorzugt werksseitig Inkubationsgefäße eingesetzt. Eine Bestückung der Halteplatte mit Inkubationsgefäßen ist jedoch auch manuell durch den Benutzer möglich. Inkubationsgefäße sind im Stand der Technik bereits vielfach bekannt. Mehrfach verwendbare Inkubationsgefäße sind in der Regel aus Glas gefertigt, während Gefäße zur Einmalverwendung in der Regel aus Kunststoffen hergestellt werden. Geeignete Kunststoffe sind beispielsweise Polyethylen, Polypropylen. Polystyrol und Polymethylmetacrylat. Die Form der Inkubationsgefäße entspricht in der Regel der von Röhren mit einem offenen und einem geschlossenen Ende. Wie bereits weiter oben beschrieben, ist es für die Verwendung von Inkubationsgefäßen in Inkubationsblöcken mit Bohrungen wichtig, daß die Außenseite der Inkubationsgefäße an der Innenwandung der Bohrungen anliegt, um einen möglichst guten Wärmeübertrag zu gewährleisten. Für Inkubationsgefäße sind Röhren bevorzugt, die zu ihrem geschlossenen Ende hin, d. h. zur Unterseite, konisch verjüngt sind. Es hat sich herausgestellt, daß aus Herstellgründen eine Verjüngung mit einer Steigung von 0,05 bis 0,5 besonders günstig ist. Erfindungsgemäß besitzen die Inkubationsgefäße weiterhin Haltevorrichtungen, die ein Hindurchrutschen der Inkubationsgefäße durch die Bohrungen in der Halteplatte verhindern. Die Inkubationsgefäße hängen so in der Halteplatte, daß ihr geschlossenes Ende nach unten weist. Eine Halterung der Inkubationsgefäße ist möglich, wenn die Inkubationsgefäße konisch verjüngt sind und das offene Ende einen Querschnitt aufweist, der größer ist als der Querschnitt einer Bohrung der Halteplatte. Wird ein solches Inkubationsgefäß in eine Bohrung eingebracht, so rutscht es hindurch, bis es sich an einer Stelle befindet, an der der Außenquerschnitt dem Bohrungsdurchmesser entspricht. An dieser Stelle findet ein Verklemmen von Inkubationsgefäß und Halteplatte statt.
Erfindungsgemäß ist es jedoch bevorzugt, wenn das Inkubationsgefäß Halteelemente aufweist, die eine besser definierte Halteposition des Inkubationsgefäßes innerhalb der Halteplatte vorgeben. Diese Halteelemente können beispielsweise Stege sein, die auf der Außenumfangsfläche des Inkubationsgefäßes angebracht sind, und die in einer bestimmten Höhe des Inkubationsgefäßes eine plötzliche Verbreiterung des effektiven Querschnittes bewirken. Besonders bevorzugt ist es, wenn das Inkubationsgefäß so aufgebaut ist, daß ein Plateau existiert, welches im wesentlichen senkrecht zur Längsachse des Inkubationsgefäßes ist, und mit dem die Inkubationsgefäße auf der Halteplatte aufliegen. Ein solches Plateau kann beispielsweise durch einen Kreisring erzeugt werden, der um das Inkubationsgefäß verläuft oder das Inkubationsgefäß kann einen unteren Teil besitzen, dessen Querschnitt kleiner ist als der Querschnitt der Bohrungen und sich auf diesem Teil ein zweiter Teil befindet, dessen Querschnitt größer ist als der Bohrungsquerschnitt. Bevorzugt sind die beiden Teile durch ein Materialstück bevorzugt senkrecht zur Längsachse des Inkubationsgefäßes miteinander verbunden. Dieses Verbindungsstück bildet das Plateau, das auf der Halteplatte aufliegt.
Erfindungsgemäß ist das Verhältnis des Bohrungsquerschnittes und des Außenumfanges des Inkubationsgefäßes, der sich auf Höhe der Halteplatte befindet, wenn das Inkubationsgefäß in der Halteplatte eingehängt ist, von Bedeutung. Einerseits muß das Inkubationsgefäß durch die Halteplatte ausreichend genau innerhalb der Ebene der Halteplatte gehaltert werden, so daß ein gezieltes Einführen der Inkubationsgefäße in die Bohrungen des Inkubationsblockes möglich ist. Andererseits muß das Inkubationsgefäß innerhalb der Bohrung ein gewisses Spiel in der Ebene der Halteplatte besitzen, damit geometrische Abweichungen der Bohrungen in der Halteplatte und der Bohrungen im Inkubationsblock nicht zu einem Verklemmen der Inkubationsgefäße führen. Ein mechanisches Spiel zwischen Inkubationsgefäßen und Halteplatte ist bei Inkubatoren von besonderer Wichtigkeit, da sich durch die Temperaturunterschiede Wärmedehnungen einstellen, die bei einer starren Anordnung aus Inkubationsgefäßen und Halteplatte zu einem Verklemmen führen, sofern mehr als ein Inkubationsgefäß vorhanden ist. Bereits ein geringfügiges Verklemmen führt dazu, daß ein dichtes Anliegen der Inkubationsgefäße an den Innenwandungen der Bohrungen des Inkubationsblockes nicht mehr gewährleistet ist.
Ein geeignetes mechanisches Spiel kann erhalten werden, wenn der Außenquerschnitt der Inkubationsgefäße in dem Bereich, der von der Halteplatte umschlossen ist, um 0,2 bis 1 mm kleiner ist als der Innenquerschnitt der Bohrungen der Halteplatte.
Zur Gewährleistung eines engen Anliegen eines Inkubationsgefäßes innerhalb einer Bohrung ist der Teil der Inkubationsgefäße, der sich unterhalb der Halteplatte befindet, länger als die Bohrungen des Inkubationsblockes tief sind. Wird die Halteplatte mit den Inkubationsgefäßen auf den Inkubationsblock gestellt, so stoßen die Inkubationsgefäße auf dem Boden der Bohrungen auf und die Vorrichtungen werden von der Halteplatte abgehoben. Bei der Verwendung von sich konisch verjüngenden Inkubationsgefäßen führt dies dazu, daß sich die Haltevorrichtungen der Inkubationsgefäße von der Halteplatte abheben, und das Spiel zwischen Inkubationsgefäß und Bohrung in der Halteplatte vergrößert wird, so daß ein Verklemmen vermieden werden kann. Auf diese Weise wird ein enges Anliegen der Inkubationsgefäße an den Innenwandungen der Bohrungen auch dann erreicht, wenn die Inkubationsgefäße produktionstechnisch Längenschwankungen aufweisen.
Bei einer besonders bevorzugten Ausführungsform wird die Halteplatte bestückt mit Inkubationsgefäßen angeliefert, so daß sie vom Benutzer lediglich in den Inkubationsblock eingesetzt werden muß.
Die Erfindung bietet demnach den Vorteil, daß mit dem erfindungsgemäßen Rack eine einfache Bestückung von Inkubatoren erfolgen kann. Die Vorteile von Fluidinkubatoren und Metallblockinkubatoren können durch Verwendung eines erfindungsgemäßen Racks demnach kombiniert werden. Es ist eine einfache Bestückung eines Inkubators mit einer Vielzahl von Inkubationsgefäßen möglich, ohne daß eine Vielzahl von manuellen Arbeitsschritten nötig ist, eine Robotik benötigt wird, oder daß die mit Fluiden verbundenen Nachteile auftreten.

Figur 1a zeigt einen Inkubationsblock (1) in Aufsicht sowie in Figur 1b einen Detail-Querschnitt. Die Oberseite des Inkubationsblockes hat eine im wesentlichen kreisförmige Gestalt. Die Bohrungen (2) im Inkubationsblock sowie die Bohrungen (12) in der Halteplatte sind in vier konzentrischen Kreisen angeordnet. Die Halteplatte (3) besitzt im wesentlichen die Form eines Kreissegmentes und hat an ihrer Unterseite mehrere Standelemente (4a, 4b), die außerdem zur Positionierung der Halteplatte auf dem Inkubationsblock dienen. Die Standelemente (4a) besitzen einen winkligen Querschnitt, der in Zusammenwirkung mit den Aussparungen (5) im Inkubationsblock (1) zur Positionierung dient. Weiterhin besitzt die Halteplatte ein Standelement (4b), das in eine kreisförmige Ausnehmung (6) innerhalb des Inkubationsblockes (1) eingreift. Figur 1b zeigt, daß die Form des Inkubationsgefäßes (20) und die Bohrung (2) im Inkubationsblock (1) so aufeinander abgestimmt sind, daß die Wandung des Inkubationsgefäßes eng an der Innenseite der Bohrung (2) anliegt, um einen guten Wärmeübergang zu gewährleisten.
In Figur 2 ist ein Inkubationsgefäß (20) vergrößert dargestellt. Das dargestellte Inkubationsgefäß (20) ist einteilig, kann jedoch in zwei Abschnitte aufgeteilt werden. Der Abschnitt (21) läuft konisch nach unten zu und ist an der Unterseite verschlossen. Der Abschnitt (22), der sich oberhalb der Halteplatte (3) befindet, wenn das Gefäß in die Halteplatte eingehängt ist, besitzt eine zylindrische Gestalt und ist nach oben hin offen. Von besonderer Bedeutung ist der Übergang zwischen den beiden Abschnitten des Inkubationsgefäßes. Aufgrund der unterschiedlichen Außendurchmesser der genannten Abschnitte bildet sich zwischen ihnen ein Plateau (23), mit dem das Inkubationsgefäß auf der Halteplatte (3) aufliegt. Das Plateau (23) besitzt eine Breite von etwa 0,3 bis 0,6 mm.
Figur 3 zeigt ein Rack (10) in Seitenansicht mit einem eingehängten Inkubationsgefäß (20) und freien Bohrungen (12). Oberhalb der Halteplatte (3) befinden sich die zwei Grifflaschen (11). Unterhalb der Halteplatte (3) sind sowohl die Standelemente (4a) als auch das Standelement (4b) zu erkennen.
Figur 4a zeigt ein System zur Inkubation in perspektivischer Ansicht. Ein Rack (10) wird so auf den Inkubationsblock (1) gestellt, daß die Inkubationsgefäße (20) in die Bohrungen (2) des Inkubationsblockes eintauchen. Die Standelemente (4a) schieben sich über den Rand der Ausnehmung (5) und das Standelement (4b) tritt in die Ausnehmung (6) ein. In Figur 4a ist ebenfalls eine Lichtschrankte (24) zu erkennen, die sich innerhalb des Inkubationsblockes befindet. Die Lichtschranke detektiert die Anwesenheit eines Racks dadurch, daß ein Standelement (4b) den Lichtweg am unteren Ende der Ausnehmung (6) unterbricht. Zur Verdeutlichung ist ein Ausschnitt aus dieser Figur in Seitenansicht in Figur 4b dargestellt. Es ist in diesen Figuren zu erkennen, daß das Inkubationsgefäß (20) zunächst in das Rack (10) eingehängt ist. Wird das Inkubationsgefäß mit Hilfe des Racks in eine Bohrung (2) des Inkubationsblockes (1) eingebracht, so stößt das Inkubationsgefäß auf den Boden der Bohrung des Inkubationsblockes auf. Die Länge des Inkubationsgefäßes (20) ist so bemessen, daß zwischen dem Plateau (23) und der Oberseite der Halteplatte (3) ein Spalt entsteht, wenn die Halteplatte auf dem Inkubationsblock aufliegt. In Figur 4b ist ebenfalls zu erkennen, daß sich der Zwischenraum zwischen dem Bohrungsrand und dem Inkubationsgefäß vergrößert, wenn das Inkubationsgefäß nach oben aus der Halteplatte herausgeschoben wird.

Claims (9)

  1. System zur Inkubation von Probeflüssigkeiten, das die folgenden Elemente besitzt
    einen Inkubationsblock (1) mit Bohrungen (2) zur Aufnahme von Inkubationsgefäßen,
    ein Rack (10), bei dem konisch verjüngte Inkubationsgefäße (20) mit Haltevorrichtungen in Bohrungen (12) einer Halteplatte (3) eingehängt sind,
    eine Vorrichtung zur Temperierung des Inkubationsblockes, die thermisch mit dem Inkubationsblock verbunden ist,
    wobei die Bohrungen in der Halteplatte (12) und die Bohrungen im Inkubationsblock (2) in ihrer geometrischen Anordnung so aufeinander abgestimmt sind, daß sich die Inkubationsgefäße des Racks in die Bohrungen des Inkubationsblockes einpassen, wenn das Rack auf den Inkubationsblock gestellt wird, dadurch gekennzeichnet, daß der Teil der Inkubationsgefäße, der sich unterhalb der Halteplatte befindet, länger ist als die Bohrungen des Inkubationsblockes tief sind, so daß, wenn das Rack auf den Inkubationsblock gestellt wird, sich die Haltevorrichtungen der Inkubationsgefäße von der Halteplatte abheben und sich aufgrund der konischen Verjüngung der Inkubationsgefäße das Spiel in der Ebene der Halteplatte vergrößert, so daß ein Verklemmen verhindert wird.
  2. System gemäß Anspruch 1, bei dem die Bohrungen im Inkubationsblock und die Außenwandungen der Inkubationsgefäße so aufeinander abgestimmt sind, daß die Außenwandung des Inkubationsgefäßes an der Innenwandung der Bohrung des Inkubationsblockes eng anliegt.
  3. System gemäß Anspruch 1, wobei an der Unterseite der Halteplatte Standelemente (4a, 4b) angebracht sind und sich im Inkubationsblock Ausnehmungen zur Aufnahme dieser Standelemente befinden.
  4. System gemäß Anspruch 1, bei dem sich auf der Oberseite der Halteplatte ein oder mehrere Grifflaschen (11) befinden.
  5. System gemäß Anspruch 1 oder 3 mit einem Sensor innerhalb des Inkubationsblockes, welcher die Anwesenheit eines Racks detektiert.
  6. System gemäß Anspruch 1, bei dem die Halteplatte die Form eines Kreissegmentes oder eines Teiles eines Kreisringes besitzt.
  7. Verfahren zur Inkubation von Probeflüssigkeiten mit
    einem Inkubationsblock (1) mit Bohrungen zur Aufnahme von Inkubationsgefäßen,
    einem Rack (10), bei dem konisch verjüngte Inkubationsgefäße (20) mit Haltevorrichtungen in Bohrungen (12) einer Halteplatte (3) eingehängt sind und der Teil der Inkubationsgefäße, der sich unterhalb der Halteplatte befindet länger ist als die Bohrungen des Inkubationsblockes tief sind, sowie
    einer Vorrichtung zur Temperierung des Inkubationsblockes, die thermisch mit dem Inkubationsblock verbunden ist,
    wobei die Bohrungen in der Halteplatte (12) und die Bohrungen im Inkubationsblock (2) in ihrer geometrischen Anordnung so aufeinander abgestimmt sind, daß sich die Inkubationsgefäße des Racks in die Bohrungen des Inkubationsblockes einpassen, wenn das Rack auf den Inkubationsblock gestellt wird, mit den Schritten
    Einstellen des Racks in den Inkubationsblock, wobei sich die Haltevorrichtungen der Inkubationsgefäße von der Halteplatte abheben und sich aufgrund der konischen Verjüngung der Inkubationsgefäße das Spiel in der Ebene der Halteplatte vergrößert,
    Temperierung der Inkubationsgefäße.
  8. Verfahren gemäß Anspruch 7, bei dem das Rack (10) vom Benutzer manuell mit Inkubationsgefäßen (20) bestückt wird.
  9. Verfahren gemäß Anspruch 7, bei dem die Bestückung des Racks (10) mit Inkubationsgefäßen (20) automatisch erfolgt und das bestückte Rack dem Benutzer zur Verfügung gestellt wird.
