Abstract

Новый способ основан на использовании мРНК-кодируемой транспозазы (например, транспозазы Sleeping Beauty) в комбинации с кодируемым миникольцевой ДНК перемещающимся элементом. Новый способ делает возможными более высокие коэффициенты переноса генов и одновременно менее токсичен, чем стандартный подход, который состоит в использовании кодируемой плазмидной ДНК транспозазы в комбинации с кодируемым плазмидной ДНК перемещающимся элементом. Применения изобретения включают, но не ограничиваясь этим, стабильное встраивание трансгена, кодирующего иммунный рецептор (например, T-клеточный рецептор или синтетический химерный антигенный рецептор), в T-лимфоциты человека, где иммунный рецептор обеспечивает специфичность к молекуле, экспрессируемой опухолевой клеткой. мРНК транспозазы и транспозонную миникольцевую ДНК можно вводить в лимфоциты с помощью способов, включающих в качестве неограничивающих примеров электроперенос, такой как электропорация и нуклеофекция.
