Abstract

Способ энзиматического синтеза пептидов, включающий энзиматическое получение эфира или тиоэфира из (i) защищенной с N-конца аминокислоты, С-концевого эфира защищенной с N-конца аминокислоты, защищенного с N-конца пептида или С-концевого эфира защищенного с N-конца пептида и (ii) спирта, представленного формулой HO-CX-Z, или же тиола, представленного формулой HS-CX-Z, где каждый Х независимо означает атом галогена или атом водорода; a Z выбирается из группы sp-гибридизованных атомов углерода, содержащих по меньшей мере два заместителя, содержащих гетероатом, присоединенный непосредственно к sp-гибридизованному углероду, и sp-гибридизованных атомов углерода, содержащих один или два заместителя, содержащих гетероатом, присоединенный непосредственно к sp-гибридизованному углероду, причем получение эфира или тиоэфира осуществляется в реакционной среде, содержащей 2 мас.% или меньше воды, исходя из массы жидкости в реакционной среде; и энзиматическое присоединение полученного эфира или тиоэфира к необязательно защищенной с С-конца аминокислоте или необязательно защищенному с С-конца пептиду, при этом пептид синтезируется в реакционной среде, содержащей 2 мас.% или меньше воды, исходя из общей массы реакционной среды.
