Claims (15)
1. Rekombinant værtscelle omfattende et heterologt polynukleotid, som koder for et polypeptid med carboanhydraseaktivitet, hvor værtscellen er i stand til at producere en større mængde C4-dicarboxylsyre sammenlignet med værtscellen uden det heterologe polynukleotid, når dyrkning sker under de samme betingelser; hvor den rekombinante værtscelle er en svampeværtscelle; og hvor det heterologe polynukleotid: (a) koder for en carboanhydrase med i det mindste 80% sekvensidentitet med SEQ ID NR: 55; (b) under middel-stringensbetingelser hybridiserer med (i) den fulde længe af den komplementære streng i SEQ ID NR: 54, eller (ii) den fulde længe af den komplementære streng af cDNA sekvensen i SEQ ID NR: 54; or (c) har i det mindste 80% sekvensidentitet med SEQ ID NR: 54 eller cDNA sekvensen i SEQ ID NR: 54.
2. Rekombinant værtscelle ifølge krav 1, hvor det heterologe polynukleotid koder for en carboanhydrase med i det mindste 90% sekvensidentitet med SEQ ID NR: 55.
3. Rekombinant værtscelle ifølge krav 1 eller 2, hvor det heterologe polynukleotid koder for en carboanhydrasevariant af SEQ ID NR: 55, hvor varianten omfatter en substitution, deletion, og/eller insertion af ikke flere end 10 positioner.
4. Rekombinant værtscelle ifølge ethvert af kravene 1-3, hvor det heterologe polynukleotid koder for en carboanhydrase, som omfatter eller består af SEQ ID NR: 55.
5. Rekombinant værtscelle ifølge ethvert af kravene 1-4, som endvidere omfatter et heterologt polynukleotid, der koder for en bicarbonattransporter.
6. Rekombinant værtscelle ifølge ethvert af kravene 1-5, som endvidere omfatter et heterologt polynukleotid, der koder for en C4-dicarboxylsyretransporter.
7. Rekombinant værtscelle ifølge ethvert af kravene 1-6, som endvidere omfatter et heterologt polynukleotid, der koder for en malatdehydrogenase.
8. Rekombinant værtscelle ifølge ethvert af kravene 1-7, som endvidere omfatter et heterologt polynukleotid, der koder for en pyruvatcarboxylase.
9. Rekombinant værtscelle ifølge ethvert af kravene 1-8, hvor værtscellen er en filamentøs svampeværtscelle.
10. Rekombinant værtscelle ifølge krav 9, hvor værtscellen er valgt fra gruppen bestående af en Acremonium, Aspergillus, Aureobasidium, Bjerkandera, Ceriporiopsis, Chrysosporium, Coprinus, Coriolus, Cryptococcus, Filibasidium, Fusarium, Humicola, Magnaporthe, Mucor, Myceliophthora, Neocallimastix, Neurospora, Paecilomyces, Penicillium, Phanerochaete, Phlebia, Piromyces, Pleurotus, Rhizopus, Schizophyllum, Talaromyces, Thermoascus, Thielavia, Tolypocladium, Trametes og Trichoderma.
11. Rekombinant værtscelle ifølge krav 10, hvor værtscellen er en Aspergillus værtscelle, som for eksempel en Aspergillus oryzae-værtscelle.
12. Rekombinant værtscelle ifølge ethvert af kravene 1-11, hvor C4-dicarboxylsyren er en æblesyre.
13. Rekombinant værtscelle ifølge ethvert af kravene 1-12, hvor værtscellen er i stand til at producere en større mængde C4-dicarboxylsyre med i det mindste 10%, fortrinsvis med i det mindste 50%, sammenlignet med værtscellen uden det heterologe polynukleotid, der koder for carboanhydrasen, når dyrkning sker under de samme betingelser.
14. Fremgangsmåde til fremstilling af en C4-dicarboxylsyre, omfattende: (a) dyrkning af den rekombinante værtscelle ifølge ethvert af kravene 1-13 i et medie under passende betingelser til at producere C4-dicarboxylsyren; og (b) indvinding af C4-dicarboxylsyren.
15. Fremgangsmåde ifølge krav 14, hvor C4-dicarboxylsyren er en æblesyre.
