Claims (20)
1. Fremgangsmåde til rensning af daptomycin omfattende trinene a) påfyldning af en opløsning indeholdende delvis renset daptomycin på en an-i o n bytte rkro matog raf i sø j I e; b) påfyldning af opløsningen fra trin a) på en omvendt fase-kromatografisøjle; c) eventuelt påfyldning af opløsningen fra trin b) på endnu en omvendt fase-kro-matografisøjle mindst én gang; hvor elueringsbufferen i a) er en monovalent saltopløsning, og elueringsbufferen i b) er vandig alkohol.
2. Fremgangsmåde ifølge krav 1, ved hvilken der udføres ét eller flere klaringstrin forud for anionbytterkromatografitrinet a).
3. Fremgangsmåde ifølge krav 1 - 2, ved hvilken produktet fra omvendt fase-kromatografitrinet b) renses yderligere ved ét eller flere filtreringstrin.
4. Fremgangsmåde ifølge krav 3, ved hvilken produktet fra den omvendte fase yderligere følges af et frysetørringstrin.
5. Fremgangsmåde ifølge krav 1, ved hvilken elueringsopløsningen i a) er NaCI.
6. Fremgangsmåde ifølge krav 2, ved hvilken elueringsbufferen er 0,1-1,5 M NaCI.
7. Fremgangsmåde ifølge krav 1, ved hvilken elueringsbufferen i b) er vandig ethanol.
8. Fremgangsmåde ifølge krav 7, ved hvilken elueringsbufferen er 5-80% ethanol.
9. Fremgangsmåde ifølge krav 1, ved hvilken anionbytterkromatografitrinet udføres under anvendelse af en stærkt tværbundet agaroseharpiks med dextran-overfladeextender.
10. Fremgangsmåde ifølge krav 1, ved hvilken omvendt fase-kromatografitrinet udføres under anvendelse af en styren-baseret harpikssøjle.
11. Fremgangsmåde ifølge krav 1, ved hvilken trin b) gentages én gang.
12. Fremgangsmåde ifølge krav 11, ved hvilken den første omvendt fase-kro-matografisøjle elueres ved en pFI-værdi på 6,5-8,5.
13. Fremgangsmåde ifølge krav 11, ved hvilken den anden omvendt fase-kro-matografisøjle elueres ved en pFI-værdi på 2,5-3,5.
14. Fremgangsmåde til rensning af daptomycin omfattende trinene a) udsættelse af en fermenteringsvæske indeholdende daptomycin for ét eller flere klaringstrin; b) påfyldning af opløsningen fra trin a) på en anionbytterkromatografisøjle; c) påfyldning af opløsningen fra trin b) på en omvendt fase-kromatografisøjle; d) eventuelt påfyldning af opløsningen fra trin c) på endnu en omvendt fase-kromatografisøjle mindst én gang; e) udsættelse af den resulterende opløsning fra trin d) for ét eller flere filtreringstrin; f) udsættelse af filtratet fra d) for frysetørring; resulterende i et renset pulver af daptomycin; hvor elueringsbufferen i trin c) og eventuelt i trin d) er vandig alkohol.
15. Fremgangsmåde ifølge krav 14, ved hvilken klaringstrinene i a) er én af eller en kombination af de følgende metoder: omvendt osmose, centrifugering, filtrering, ultrafiltrering, nanofiltrering, anionbytterkromatografi.
16. Fremgangsmåde ifølge krav 14, ved hvilken elueringsopløsningen i b) er NaCI.
17. Fremgangsmåde ifølge krav 16, ved hvilken elueringsbufferen er 0,1-1,5 M NaCI.
18. Fremgangsmåde ifølge krav 14, ved hvilken elueringsbufferen i c) er vandig ethanol.
19. Fremgangsmåde ifølge krav 18, ved hvilken elueringsbufferen er 5-80% ethanol.
20. Fremgangsmåde ifølge krav 14, ved hvilken trin c) gentages én gang.
