Claims (11)

  1. IKKE-SIMIANCELLER TIL DYRKNING AF PORCINT REPRODUKTIONS- OG RESPIRATIONSSYNDROM- (PRRS) VIRUS
    1. Isoleret føtal porcin alveolær makro fagcelle, der er i stand til at reproducere PRRS-virus, hvor cellen er ZMAC-1, en deponering hos American Type Culture Collection betegnet ATCC-patentdeponering nr. PTA-8764, eller afledt deraf, eller en immortaliseret cellevariant eller et derivat af cellen, hvor cellen, der er afledt deraf, varianten eller derivatet er i stand til at reproducere PRRS-virus.
  2. 2. Fremgangsmåde til generering af afkom af et PRRS-virus, hvilken fremgangsmåde omfatter: a) tilvejebringelse af en isoleret eller oprenset føtal porcin alveolær makrofagcelle, der er i stand til at replikere PRRS-virusset, hvor cellen dyrkes i mindst 5 passager, og hvor cellen eventuelt er opnået fra et individuelt dyr; b) eksponering af cellen for PRRS-virusset og c) tilladelse til, at PRRS-virusset replikeres i cellen; hvorved der genereres afkom af PRRS-virusset.
  3. 3. Fremgangsmåde til fremstilling af en PRRS-vaccine, hvilken fremgangsmåde omfatter tilvejebringelse af en modificeret levende virus- (MLV) stamme af PRRSV og dyrkning af MLV-stammen i en isoleret eller oprenset føtal porcin alveolær makrofagcelle, der er i stand til at replikere PRRSV MLV-stammen, hvor cellen er en primærcelle, en cellepopulation, en cellelinje eller en variant eller et derivat deraf, og hvor cellen eller varianten eller derivatet er i stand til at reproducere PRRS-virus.
  4. 4. Fremgangsmåde ifølge krav 2 eller 3, hvor cellen er repræsenteret af cellen ifølge krav 1.
  5. 5. Fremgangsmåde til dyrkning af en PRRS-virus, hvilken fremgangsmåde omfatter: (a) isolering af en makrofagcelle fra en porcin føtal lunge, hvor isoleringen foregår i trin, der omfatter tilvejebringelse af et porcint føtalt individ; opnåelse af en celle indeholdende bronchoalveolær lavageprøve fra individet og separation af en cellulær bestanddel fra prøven; (b) dyrkning af cellen og (c) etablering af kontakt mellem cellen og virusset for således at tillade virusreplikation; hvorved vimsset dyrkes.
  6. 6. Fremgangsmåde ifølge krav 5, hvor dyrkningen omfatter passage af cellen i mindst 5 passager og/eller dyrkning af cellen i mindst 10 dage med kontinuerlig dyrkning.
  7. 7. Sammensætning, der omfatter et PRRS-virus dyrket i en isoleret føtal porcin alveolær makrofagcelle, der er i stand til at reproducere PRRS-virus, hvor cellen er opnået fra et individuelt dyr og dyrket i mindst 5, 10, 20, eller 50 passager.
  8. 8. Fremgangsmåde ifølge et hvilket som helst af kravene 2 til 6, hvilken fremgangsmåde endvidere omfatter etablering af kontakt mellem cellen og en vækstfaktorsammensætning.
  9. 9. Fremgangsmåde ifølge krav 8, hvor vækstfaktorsammensætningen omfatter makrofagkoloni-stimulerende faktor (MCSF) eller granulocyt-makrofagkoloni-stimulerende faktor (GMCSF).
  10. 10. Vaccine fremstillet ved dyrkning af et virus, der er et PRRS-virus, en PRRS-virusvaccinestamme, eller et PRRS-modificeret levende virus i en celle med ATCC-patentdeponering nr. PTA-8764 eller en celle, der er afledt deraf, hvor cellen eller cellen, der er afledt deraf, er i stand til at reproducere PRRS-virus.
  11. 11. Sammensætning ifølge krav 7, hvor PRRS-virusset dyrkes i cellen ifølge krav 1.
