Claims (15)
1. Rekombinant Escherichia co/z-værtscelle til fremstilling af et Escherichia coli L-asparaginase Il-enzym, der omfatter et Escherichia co/z'-kromosom og mindst én kopi af en rekombinant ekstrakromosomal vektor, hvor den rekombinante ekstrakromosomale vektor koder for en underenhed af L-asparaginase Il-enzymet, hvor værtscellekromosomet ligeledes koder for den samme underenhed af L-asparaginase Il-enzymet, hvor værtkromosomet ikke koder for nogen anden isoform af L-asparaginase II, hvor den kodede L-asparaginase II-underenhed omfatter SEQ ID NO: 1; og den rekombinante ekstrakromosomale vektor omfatter et DNA-molekyle, der omfatter SEQ ID NO: 2.
2. Rekombinant Escherichia co/z-værtscelle ifølge krav 1, hvor den ekstrakromosomale vektor er et plasmid.
3. Rekombinant Escherichia co/z-værtscelle ifølge krav 1, hvor den rekombinante ekstrakromosomale vektor omfatter et DNA-molekyle, der koder for underenheden af L-Asparaginase Il-enzymet, der er operativt bundet til en egnet promotor.
4. Rekombinant Escherichia cø/z-værtscelle ifølge krav 3, hvor promotoren er udvalgt fra gruppen bestående af T7, araB, λΡκ/λΡΓ , phoA, tre og trp-promotorer.
5. Rekombinant Escherichia co/z-værtscelle ifølge krav 3, hvor den rekombinante ekstrakromosomale vektor endvidere omfatter en operatør, et ribosombindingssted, en signalsekvens, en transskriptionsterminator, en antibiotisk selektionsmarkør, et replikationsorigin og en reguleret kopi af repressoren.
6. Rekombinant Escherichia co/z-værtscelle ifølge krav 3, hvor kromosomet omfatter et DNA-molekyle ifølge SEQ ID NO: 3.
7. Isoleret nukleinsyremolekyle, der koder for en L-asparaginase II-enzymunderenhed af SEQ ID NO: 1, hvor det isolerede nukleinsyremolekyle består af en nukleinsyre udvalgt fra gruppen bestående af SEQ ID NO: 2 og SEQ ID NO: 3.
8. Rekombinant ekstrakromosomal vektor, hvor den rekombinante ekstrakromosomale vektor omfatter SEQ ID NO: 2 og koder for en underenhed af L-asparaginase Il-enzymet.
9. Ekstrakromosomal vektor ifølge krav 8, hvor vektoren er et plasmid.
10. Ekstrakromosomal vektor ifølge krav 9, hvor vektoren er plasmid pEN537.
11. Escherichia co/z'-værtscelle, der omfatter plasmidet ifølge krav 2, hvor plasmidet er pEN537, betegnet som EN538 og deponeret under ATCC-nr. PTA 7490.
12. Fremgangsmåde til fremstilling af en rekombinant L-asparaginase Il-enzym i alt væsentligt fri for andre L-asparaginase II-isomerer, hvilken fremgangsmåde omfatter dyrkning af værtscellen ifølge krav 11 og isolering af det fremstillede L-asparaginase Il-enzym.
13. Isoleret DNA-molekyle, der koder for en underenhed af L-asparaginase Il-enzym, der omfatter SEQ ID NO: 2.
14. Rekombinant ekstrakromosomal vektor ifølge krav 8, hvor SEQ ID NO: 2 er operativt bundet til en egnet promotor.
15. Rekombinant ekstrakromosomal vektor ifølge krav 14, hvor promotoren er udvalgt fra gruppen bestående af T7, araB, λΡ^/λΡ^ phoA, tre og trp-promotorer.
