Det er velkendt, at man ved at forøge foderforbruget eller foderindtagelsen hos et dyr kan forøge dyrets vægt eller, når der er tale om malkekøer eller diegivende svin, forøge dyrets mælkeproduktion. F.eks. kan man ved 5 at forøge dyrets vækst på kort tid bringe dyrets vægt op på markedsvægten, hvilket er ensbetydende med betydelige besparelser for opdrætteren.
Selv om dyr altid har et vist niveau af væksthormon eller 10 dermed beslægtede substanser i blodet, vil en forøgelse af væksthormon-niveauet over det normale niveau bevirke, at foderindtagelsen eller foderforbruget hos visse dyr aftager. Derfor er det vigtigt at forøge foderindtagelsen hos sådanne dyr, eksempelvis hos svin og kvæg, eftersom 15 disse vil lide under en nedsat foderindtagelse.
Det har nu vist sig, at en indgivelse af thyroid-aktivt (dvs. skjoldbruskkirtelaktivt) ioderet casein vil forøge foderindtagelsen hos sådanne dyr, hvis niveauer af vækst-20 hormon i blodet er forhøjede eller over det normale.
Når malkekvæg har et niveau af væksthormon i blodet, som ligger over det normale, vil deres mælkeproduktion blive forøget, samtidigt med, at deres foderindtagelse hverken 25 forøges eller formindskes. Dette betyder, at en malkeko producerer mere mælk, uden at den behøver mere foder. En forøgelse af koens foderindtagelse vil således indebære en yderligere forøgelse af mælkeproduktionens kvalitet og kvantitet, hvilket er en fordel. Derfor er en tildeling 30 af ioderet casein til malkekvæg med henblik på at forøge dyrenes foderindtagelse også en del af opfindelsen.
Nærmere bestemt angår opfindelsen en fremgangsmåde til forøgelse af foderindtagelsen hos svin eller malkekvæg, 35 som er under behandling ved injektion eller implantering med væksthormon eller væksthormon-beslægtede forbindelser, idet fremgangsmåden ifølge opfindelsen er ejendomme-

2 lig ved, at man herefter eller samtidig indgiver svinene eller malkekvæget roderet casein.
Ved det "ioderede casein", som gives til svin eller mal-5 kekøer, skal ifølge The Third New International Dictionary forstås et iodholdigt, ud fra casein fremstillet præparat, hvis fysiologiske virkning ligner virkningen af thyroidhormon (se også Merck Index, 10. udgave, side 1349, nr. 9261).

Det har vist sig, at man hos svin (herunder lakterende søer) og malkekøer, som udviser et forhøjet niveau af væksthormon, hvilket bevirker en mindre foderoptagelse hos svin eller bortfald af en forhøjelse af foderoptagel-15 sen hos køer, kan forøge foderoptagelsen ved tildeling af ioderet casein. Dette er ikke nærliggende for fagmanden ud fra teknikkens stade.
Fra GB-PS 1 130 849 kendes ganske vist dyrefoder, som in-20 deholder et anabolisk middel og thyroxin eller et derivat heraf. Thyroxin er en thyroid-aktiv forbindelse. I det omtalte GB-PS antog man en vekselvirkning mellem disse to bestanddele (se side 1, 2. spalte linie 84 og videre).
Skønt væksthormon er en slags anabolisk middel, hører det 25 dog ikke til de anaboliske midler, som er omtalt som eksempler i afsnittet, der forbinder spalte 1 og 2 i nævnte GB-PS 1 130 849. Væksthormon er instabilt og kan ikke indgives peroralt. Genstanden for opfindelsen i GB-PS 1 130 849 er et dyrefodermiddel (se titlen), og det er 30 klart, at dette patent ikke er egnet til at beskrive anvendelsen af væksthormon, eller at denne anvendelse er nærliggende. Nævnte patentskrift omhandler derfor ikke anvendelsen af ioderet casein for at udligne formindskelsen af foderoptagelsen på grund af kunstigt eller exogent 35 væksthormon eller en dermed beslægtet forbindelse.

3 EP-A1-0 104 933 angår en formulering af exogent somatomedin. Det drejer sig her om en forbindelse, der stimuleres med væksthormon, og som sådan er det en med væksthormon beslægtet forbindelse. I nævnte EP offentliggørelses-5 skrift er der dog ikke nogen henvisning til, at et iode-ret casein skulle kunne anvendes sammen med somatomedin eller med en anden, med væksthormon beslægtet forbindelse. Yderligere er der ingen henvisning overhovedet, som ville være nærliggende, hvor det angives, at nedgangen i 10 foderoptagelse kunne udlignes ved anvendelse af ioderet casein.
Fremgangsmåden ifølge opfindelsen udføres fortrinsvis ved peroral administrering af en virksom mængde af ioderet 15 casein til dyret. Der kan imidlertid også anvendes andre administreringsmåder, f.eks. implanter eller intramusku-lær eller intravenøs injektion. Til peroral administrering blandes det ioderede casein fortrinsvis med egnede bærestoffer eller fortyndingsmidler, der er sædvanlige 20 til dyrehold inden for landbruget, f.eks. i forbindelse med foder. Typiske bære- og fortyndingsmidler, der anvendes i almindeligt foder, omfatter majsmel, sojabønnemel, lucernemel, risskaller, sojabønnemelprodukter, bomuldsfrøoliemel, benmel, formalet korn, majskolbemel, natrium-25 chlorid, urinstof og sukkerroemelasse. Disse bærestoffer begunstiger en ensartet fordeling af det ioderede casein i færdigfoderet, hvorved der opnås en gunstig fordeling af det ioderede casein i foderet. Yderligere kan også administreres forbindelser, der påvirker skjoldbruskkirt-30 len til dannelse af T^ (tetraiodthyroin, L-thyroxin eller levothyroxin) eller T^ (triiodthyroin, liothyroin, tert-oxin eller cytomel), og sådanne forbindelser kan omfatte TSH (thyroidstimulerende hormon, thyrotropin, thyrotro-pisk hormon, thytropar, Ambinon eller Dermathycin) og TRH 35 (thyrotropin-frigørende hormon).

Skønt den foretrukne metode til oral indgivelse af det ioderede casein består i at indgive dette via det daglige foder, kan det også inkorporeres i saltblokke og mineralsliksten, ligesom det kan sættes direkte til dyrenes 5 drikkevand med henblik på en bekvem oral indtagelse. Desuden kan forbindelsen formuleres sammen med polymorfe materialer eller voksarter, hvormed man kan opnå en lang-tidsvirkende kontrolleret frigivelse, idet forbindelsen enten kan indgives til dyret i form af en stor pille 10 eller indgives efter behov til opretholdelse af den ønskede daglige frigivelse af det ioderede casein.
Til parenteral indgivelse kan det ioderede casein blandes med konventionelle bærestoffer, såsom majsolie, sesam-15 olie, carbon-holdigt voks eller calciumstearat. Disse formuleringer kan indgives ved injektion, eller de kan indgives i form af en subkutan implantering med langsom frigivelse. Man kan også anvende andre typer af implantater til at afgive det ioderede casein, og disse implanta-20 ter kan anbringes subkutant. Disse implantater omfatter silastiske implanter, polymere implanter, såsom polyure-thaner og hydrogeler, mikrokapsler, mikrokugler og lipo-somer. Implantatet kan fjernes, når den ønskede administrationsperiode er afsluttet. Endvidere kan det ioderede 25 casein indgives ved hjælp af mekaniske pumpeanordninger eller med osmotiske og kemiske pumper.
"Den effektive mængde af ioderet casein", som skal tildeles et dyr, er den mængde, som er effektiv med hensyn til 30 at frembringe en stigning i foderforbruget eller foderindtagelsen. Den indgivne effektive mængde varierer afhængigt af den dyreart, som skal behandles, og formen af det indgivne ioderede casein. F.eks. er mængden af ioderet casein, som kan anvendes, på mellem 50 og 1000 35 mg/dag, og længden af administreringen er omkring 1 til 5 måneder for svin (længden af den tid, det tager at nå slutvægten) eller omkring 6 til 9 måneder for malkekvæg

5 (længden af laktationsperioden).
Man kan også indgive ioderet casein i en mængde på mellem ca. 1 og ca. 1000 ppm af den totale daglige foderindta-5 gelse. Denne mængde vil give en dosering på mellem ca.
0,05 og ca. 50 mg/kg/dag. I en foretrukken udførelsesform anvender man omkring 1 til 500 ppm, især omkring 5 til 250 ppm. En typisk mængde af den aktive bestanddel, som indgives til svin, vil være fra omkring 25 til omkring 10 150 ppm. Når man gennemfører fremgangsmåden ifølge opfin delsen i praksis, kan man f.eks. sætte ioderet casein til dyrets daglige foderration i en mængde på omkring 100 ppm af foderet.
15 Dyrene, som skal behandles med ioderet casein, har et niveau af væksthormon eller somatomedin i blodet på over det normale niveau. Disse blod-niveauer af væksthormon eller somatomedin på over det normale niveau fremkaldes ved kunstig eller exogen indgivelse af væksthormon (soma-20 totropin) eller en væksthormon-beslægtet substans. Indgivelsen af en væksthormon-frigivende faktor, såsom somatomedin eller en hvilken som helst anden substans, bevirker, at produktionen af endogent væksthormon stiger, og som følge deraf stiger også produktionen af somatomedin. Sub-25 stanser, som stimulerer det endogene væksthormon, omfatter enkephaliner eller enkephalin-lignende forbindelser, prostaglandiner, α-adrenerge forbindelser, benzodiazepi-ner, barbiturater, opiat-antagonister, r-aminosmørsyre (GABA), GABAnerge forbindelser og lignende. Andre sub-30 stanser, som kan indgives, omfatter vilkårlige analoge af væksthormon eller den væksthormon-frigivende faktor, såsom methionyl-oksevæksthormon, den 29 aminosyrer store væksthormon-frigivende faktor og lignende. 1
De typer af væksthormon eller væksthormon-beslægtede substanser, som kan indgives, kan stamme fra naturlige eller rekombinante kilder, eller de kan fremstilles ved syn-

6 teseteknikker i fast fase. Desuden behøver væksthormonet eller den væksthormon-beslægtede substans ikke at udvise artsspecificitet. Eksempelvis kan man anvende svine-, okse- eller humant væksthormon ved indgivelse af vækst-5 hormon til svin.
Mængden af væksthormon eller den med væksthormon beslægtede substans, som er nødvendig for at opretholde et niveau af væksthormon i dyrets blod på over det normale 10 niveau, er på mellem ca. 5 ug/kg/dag og ca. 300 ug/kg/dag. Hos svin vil en indgivelse på mellem ca. 10 og ca. 300 ug/kg væksthormon bevirke, at der opnås et niveau i blodet, som er over det normale. Den foretrukne indgivelse til svin er på mellem ca. 20 og ca. 100 μg/kg/dag.

Da væksthormonet eller den væksthormon-beslægtede substans er et protein, er det ikke ønskeligt at indgive hormonet oralt. Man kan imidlertid anvende intramuskulæ-re, intravenøse eller subkutane injektioner. Desuden kan 20 man indgive væksthormonet eller den væksthormon-beslægtede substans ved implantering. Typisk kan dyret modtage væksthormonet eller den væksthormon-beslægtede substans ad parenteral vej, mens det ioderede casein indgives enten oralt eller parenteralt. Væksthormonet eller den 25 væksthormon-beslægtede substans og det ioderede casein kan imidlertid indgives parenteralt via en afgivelsesanordning til et enkelt medikament, hvorved man opnår den ønskede virkning. Disse anordninger omfatter implanterede anordninger, mikrokapsler, pumper og lignende.

Virkningen af en tildeling af ioderet casein på foderindtagelsen hos svin er vist i følgende forsøg.
Forsøg 1 35 72 krydsnings frembragte (Yorkshire X Hampshire) orner og gylter, som vejede omkring 65 kg, blev anbragt i båse med

7 fire svin i hver bås. Dyrene blev fodret ad libitum med majs/soja svinefoder indeholdende omkring 16% råprotein.

Svinene blev inddelt i tre grupper med henblik på den videre behandling, således at hver gruppe indeholdt tre bå-20 se med orner og tre båse med gylter. Den første gruppe blev benyttet som kontrolgruppe, mens de øvrige to grupper blev behandlet med enten 1,5 eller 3,0 mg/dag pGH (svinevæksthormon) i de første 28 dage af forsøget. Fra dag nr. 29 til forsøgets afslutning blev doseringen for-25 øget til enten 2,0 eller 4,0 mg/dag.
Hormonet blev bragt i opløsning under anvendelse af en steriliseret 0,2 M kaliumphosphatpuffer (pH 7,8). Denne puffer var fremstillet med pyrogen-frit vand. Den endeli-30 ge koncentration af pGH i pufferen var 1,0 mg/ml.

Koncentrationen af pGH-opløsningen var 1,0 mg/ml bæremateriale. [Opløsningens koncentration og injektionsvo-15 lumenerne valgtes således, at der blev afgivet ca. 50 μg/kg/dag].
Svinene blev vejet individuelt ved forsøgets begyndelse, og derefter blev de vejet hver 14 dag indtil forsøgets 20 afslutning. Svinene udgik af forsøget, når de nåede en omtrentlig legemsvægt på 104 kg (i købet af omkring 55 dage).
Man registrerede endvidere foderindtagelsen for hvert 25 svin. Den gennemsnitlige daglige foderoptagelse og den gennemsnitlige daglige tilvækst i kilo blev beregnet og registreret.
Den nedenstående tabel 1 viser den dosis-afhængige reduk-30 tion i foderoptagelsen, som skyldes tildelingen af væksthormonet alene.

Femten ved krydsning frembragte orner, der hver vejede omkring 90 kg, blev holdt i individuelle båse af rustfrit stål og fodret med et svinefoder på basis af majs og 20 soja. Alle svinene fik en konstant mængde foder, nærmere bestemt omkring 2 kg pr. dag, som blev opdelt i to fodringer på hvert et kilo. På fodringstidspunktet blandedes vand i foderet.
25 Svinene blev opdelt i fem grupper, som hver bestod af tre svin. Disse grupper var følgende: 1 - kontrol (4,5 mg bærestof/dag) 2 - pGH (9 mg/dag) 30 3 - pGH (9 mg/dag) + IC ( 250 mg/dag) 4 - pGH (9 mg/dag) + IC ( 500 mg/dag) 5 - pGH (9 mg/dag) + IC (1000 mg/dag)
Der benyttedes IC (ioderet casein) i grupperne 3-5. Halv-35 delen af den daglige dosis til hvert svin blev blandet i foderet ved hver af de to daglige fodringer.

Man fremstillede pGH-opløsningen på samme måde som beskrevet under forsøg 1 med den undtagelse, at koncentrationen af pGH i pufferen var 2,0 mg/ml. Indgivelsen af pGH foregik dagligt ved subkutan injektion i siden af dy-5 rets krop. De daglige injektioner med opløsningen var følgende: pGH-krav og formuleringer 10 Daglig Daglig

Foderindtagelsen for hvert svin blev registreret dagligt i en periode på 12 dage. De opnåede resultater med hensyn til foderindtagelsen er vist i den nedenstående tabel 2.

Dyrene blev vejet ved forsøgets begyndelse og efter 12 dages forløb. Vægtforøgelsen efter de 12 dage i hver gruppe blev beregnet og registreret i kilo, og de fundne resultater er vist i tabel 3.

Claims (3)
1. Fremgangsmåde til forøgelse af foderindtagelsen hos 5 svin eller malkekvæg, som er under behandling med væksthormon eller en væksthormon-beslægtet substans ved injektion eller implantering, kendetegnet ved, at man tildeler svinene eller malkekvæget roderet casein samtidig hermed eller efter behandlingen. 10
2. Fremgangsmåde ifølge krav 1, kendetegnet ved, at det ioderede casein indgives i en mængde på mellem 1 og 1000 ppm beregnet på den daglige foderindtagelse.
3. Fremgangsmåde ifølge krav 2, kendetegnet ved, at det ioderede casein tildeles i en mængde på mellen 5 og 250 ppm beregnet på den daglige foderindtagelse. 20 25 1 35
