0 I US patentskrift nr. 3.547.935 beskrives fremstillingen af herbicidt virksomme 3-nitro-pyridiner blandt andet ved omsætning af substituerede 4-chlor-3--nitropyridiner med diethyl- eller di-n-propylamin.
5 Lignende forbindelser med yderligere alkylsubstituenter i 2- og/eller 6-stillingen beskrives imidlertid ikke id©fc nævnte patentskrift.
Det har nu overraskende vist sig, at der ved indføring af egnede primære og sekundære amingrupper i 10 4-stillingen i 2,6-dialkyl-3-nitro- (eller cyano)-py- ridiner fås forbindelser med fremragende farmakologiske egenskaber, hvor den bronchospasmolytiske virkning står i forgrunden. De udgør en ny strukturtype blandt de hidtil kendte bronchospasmolytika.

Den foreliggende opfindelse angår følgelig en analogifremgangsmåde til fremstilling af substituerede 4-aminopyridiner med formlen R1 R2 20 X y

Foretrukne forbindelser med formlen I og deres salte er sådanne, hvor R3 og R4 hver især betyder alkyl med op til 4 carbonatomer, Y betyder en nitro- X 2 15 eller cyanogruppe, og enten R og R^ sammen med det 4-stillede nitrogenatom danner en morpholin- eller thiomorpholinring, eller R^· betyder hydrogen eller o alkyl med op til 4 carbonatomer, R betyder med halogen, alkylthio med op til 3 carbonatomer eller en ΡΠ eventuelt substitueret phenylthiogruppe substitueret alkyl med op til 4 carbonatomer, eller R3, betyder hydrogen, og R^ betyder benzyl, hvis CE^-gruppe kan være substitueret med methyl, og hvis phenylring kan være substitueret med alkoxy med op til 2 carbonatomer, halogen, en sulfamoylgruppe eller en methylendioxygruppe.
Forbindelserne med formlen I er hidtil ukendte og har værdifulde farmakologiske, især bronchospasmo-lytiske egenskaber.

Til fremstilling af syreadditionssalte af forbindelserne med formlen I kan der eksempelvis anvendes hydrogenhalogenidsyrer, især saltsyre, desuden 30 svovl-, phosphor-, eddike-, mælke-, malein-, fumar-, oxal-, vin-, citron-, glucon-/ p-toluensulfon-, methan-sulfon- og cyclohexylamidosulfonsyre.
De fleste af udgangsforbindelserne med formlerne II-IV er kendte fra litteraturen, eller de kan let 35 fremstilles ved fremgangsmåder, som er kendte fra litteraturen .

Som eksempler på egnede forbindelser med formlen II kan nævnes de symmetriske 2,6-dialkyl-4--halogen-3-nitropyridiner såsom 4-chlor-2,6-dimethyl-3--nitropyridin (P. Nantka-Namirski, Acta Polon. Pharm.
5 18, 449 (1961)) og 4-chlor-2,6-dipropyl-3-nitropyri- din, som kan fremstilles ud fra acyleddikesyreestere via 6-alkyl-3-acyl-2,3-dihydro-2,4-dioxy-pyraner ved omsætning med ammoniak efterfulgt af nitrering i 3- stillingen og halogenering i 4-stillingen, 2,6-dial- 10 kyl-3-*cyano-4-halogenpyridiner såsom 4-chlor-3-cyano- -2,6-dimethylpyridin (T. Kato et al., Yakugaku Zasshi 91, 740 (1871)) samt de asymmetriske 2,6-dialkyl-4--halogen-3-nitropyridiner såsom 2-hexyl-6-methyl- eller 2-methyl-6-hexyl-4-chlor-3-nitropyridin, som eksempel-15 vis kan fremstilles ved omsætning af tilsvarende 3-alkyl- -3-aminoacrylsyreestere med diketen (T. Kato et al.,
Yakugaku Zasshi 9_1, 740 (1971)) eller vilkårlige acyleddikesyreestere og efterfølgende forsæbning, decarboxylering, nitrering, isomeradskillelse og 20 halogenering.

Omsætningen af forbindelser med formlen II med aminerne med formlen III gennemføres hensigtsmæssigt i et opløsnings- eller fordelingsmiddel. Hertil kan der anvendes alko-5 holer såsom methanol, ethanol, isopropanol, n-propanol, de forskellige butanoler samt blån-dinger deraf eller blandinger deraf med vand, ethere såsom tetrahydrofuran og dioxan, aprotiske opløsningsmidler såsom pyridin, dimethylformic) amid, dimethylacetamid, dimethylsulfoxid og hexa-methylphosphorsyretriamid eller carbonhydrider såsom benzen, toluen og xylen.
Ved kondensationsreaktionen er det fordelagtigt at anvende den mindst dobbelte molære 15 mængde af den valgte anvendte amin. Det er også muligt at anvende ækvimolære mængder af de to reaktanter, men så anbefales tilsætningen af et syrebindende middel, f.eks. et alkalimetal-, eller jordalkalimetalhydroxid eller -carbonat 20 eller en organisk base såsom triethylamin, i en mindst støkiometrisk mængde. Sædvanligvis gennemføres reaktionen i det første trin ved temperaturer på mellem 0°C og kogepunktet for det pågældende opløsningsmiddel, fortrinsvis 25 ved temperaturer på 2Q-100°C, og reaktionstiden kan være på nogle minutter til flere timer.
Til halogenering, især chlorering, af 2 forbindelser med formlen I, som i R bærer en 30 til alkyl bundet hydroxylgruppe, anvendes egnede halogenerings-, især chloreringsmidler såsom phosphoroxychlorid, phosphorpentachlorid eller blandinger deraf, en kombination af tri-phenylphosphin med carbontetrachlorid, thionyl-35 chlorid eller -bromid. Som opløsningsmidler hertil kan der foruden halogeneringsmidlerne selv fortrinsvis anvendes sådanne, som under reaktionsbetingelserne er indifferente over

for reaktanterne, f.eks. carbonhydrider såsom hexan, benzen og toluen eller halogenerede carbonhydrider såsom dichlormethan, chloroform, carbon-tetrachlorid og chlorbenzen.
5 Halogeneringsreaktionerne gennemføres sæd vanligvis ved temperaturer på 10-10Q°C, fortrinsvis 20-70°C, idet reaktionstiden også her kan være på nogle minutter til flere timer.
Til den videre omsætning af forbindelser 2 10 med formlen I, som i R indeholder et halogenatom, med mercaptanerne med formlen IV anvendes overvejende vand, alkoholer såsom methanol, ethanol, propanol, isopropanol eller blandinger deraf med vand, dimethylformamid, dimethylacetamid, dime-15 thylsulfoxid, hexamethylphosphorsyretriamid eller pyridin som opløsningsmiddel. Denne omsætning med mercaptaner gennemføres fordelagtigt i nærværelse af et syrebindende middel såsom et alkalimetal- eller jordalkalimetalhydroxid, 20 hensigtsmæssigt ved temperaturer på 0-150°C, fortrinsvis 50-100°C.
Til oxidation af forbindelser med form- 1 2 len I med en svovlholdig NR R -gruppe til dannelse af sulfoxider eller sulfoner kan der 25 f.eks. anvendes oxidationsmidler såsom-natri- ummetaperiodat, salpetersyre eller undertiden også frit chlor, idet oxidationsmidlet selv eller vand kan tjene som opløsningsmiddel, og reaktionstemperaturerne sædvanligvis er 30 på 20-100°, fortrinsvis 0-70°C.

Den nødvendige dosering ved behandlingen af en voksen patient, som lider af bronchialobstruk-tioner, er ved oralt indgift sædvanligvis på 30-150 5 mg pr. dag, fortrinsvis 40-100 mg pr. dag. Ved intravenøs indgift er mængden sædvanligvis på 10-50 mg pr. dag, fortrinsvis 15-30 mg pr. dag.
De omhandlede forbindelser med formlen I samt deres salte kan kombineres med andre egnede virk-10 somme stoffer som f.eks. antiallergika, anti- tussiva, sekretolytika, sedativa, perifere va-soterapeutika, antihistaminer samt andre bron-chospasmolytika såsom 32“sympathomimetika eller parasympatholytika.

Efter endt omsætning fjernes opløsningsmidlet under formindsket tryk. Remanensen optages i chloroform og udrystes tre gange med vand. Chloroformfasen tørres og kon- 30 centreres og omdannes til hydrochloridet ved opløsning i ethanol og tilsætning af ethanolisk saltsyre. Bundfaldet frasuges og omkrystalliseres fra methanol. Udbytte:44,8 g (77,3% af det teoretiske). Smp. 240°C (under sønderdeling).

Pharmak. 195 (1940) 71) i sammenligning med det kendte bronchospasmolytikum theophyllin-10 -ethylendiamin, ved hvilken hæmningen af eksperi mentelle bronchialspasmer - fremkaldt ved intravenøs indgift af spasmogene aminer såsom acetyl-cholin, histamin og serotonin - undersøges hos marsvin af begge køn i urethan-narkose (1/25 15 g/kg i.p.).
Forsøgsforbindelserne indgives i en vandig opløsning enten intravenøst (i.v.) eller intra-duodenalt (i.d.). Vanduopløselige forbindelser indgives i carboxymethylcellulose-suspensioner 20 i.d. ED^q-Værdierne/ som betegner den dosis i mg/kg, ved hvilken den eksperimentelt fremkaldte spasme reduceres til halvdelen i forhold til spasmen hos ubehandlede dyr, bestemmes grafisk ud fra dosis-virkningskurverne.
25 2. Akut toksicitet
Bestemmelsen af LD^-værdierne eller Lovområderne sker standardmæssigt over den i løbet af 7 dage hos NMRI-mus efter én intraperitoneal (i.p.) 30 indgift forekomne dødelighed.
Resultaterne af disse undersøgelser, som illustrerer overlegenheden af de her omhandlede forbindelser med formlen I over for standardpræparatet theophyllin-ethylendiamin (især også i betragtning 35 af det gunstigere forhold mellem LD^ og ED^q) er anført i den nedenstående tabel II.
