Description

Procedure. for the preparation of.solutions or suspensions of the blood sugar lowering Pancreatic hormone In patent 725,439 is a method of manufacture of durable neutral solutions of the blood sugar-reducing hormone of the pancreatic gland described, which is characterized in that one aqueous solutions of the hormone adjusts to neutral point in the presence of polyvinyl alcohol. It is on it it has been pointed out that neutral aqueous solutions of the islet hormone are not stable but flocculate within a relatively short time. This disadvantage can, as explained, by using polyvinyl alcohol in the solution avoid, also show the solutions made with the help of polyvinyl alcohol after the injection a longer lasting effect than the previously known acidic ones Solutions.

In a further embodiment of this invention it has now been found that you can get to hormone preparations with a prolonged effect even if you aqueous solutions of the island hormone, which contain an addition of polyvinyl alcohol, set to a pg value between 4.5 and the neutral point. If it in this case in contrast to the method described in patent 725,439 when falling below a certain pH limit no longer succeeds in keeping the hormone in solution, the addition of polyvinyl alcohol leads to apart from the aforementioned protracted effect, a precipitation which is essential is more finely divided than a precipitate obtained without the addition of the substance mentioned. This has a particularly advantageous effect during the injection, since it is natural it is much easier and more pleasant to inject a finely dispersed suspension as a coarsely dispersed.

Example 420 mg of crystallized islet hormone are dissolved in 10 cc of normal hydrochloric acid and diluted with 390 cc of a 5% solution of polyvinyl alcohol. In this mixture, which is heated to 40 ° C., and into which a glass electrode is immersed in the usual arrangement for measuring the hydrogen ion concentration, a 0.2% solution of sodium carbonate is allowed to flow in a thin stream with thorough stirring until the pH = 5.5. It is then cooled to room temperature with constant stirring. The product is now made up to 500 cc with water.

Instead of the sodium carbonate solution, caustic soda or sodium phosphate solution can be used use.
