Description
Process for the preparation of stable, neutral solutions of the blood sugar lowering agent Hormones of the pancreatic gland The solutions of the common for diabetes therapy Island hormones are known to show an acidic reaction, since they have a higher pH value the hormone flocculates or the solution over time causes a decrease in the effect value shows, whereby an application as a spray preparation is excluded. The restriction the hydrogen ion concentration to a low range has the major disadvantage that the preparations in sensitive patients more or less strong irritant effects cause.
It has now been found that it is possible to obtain stable, neutral aqueous islet hormone solutions to gain by making aqueous solutions of the hormone in the presence of polyvinyl alcohol .sets to the neutral point. For the neutralization you can use either acidic or also use alkaline hormone solutions. Neutralizes acidic solutions one with caustic soda or, more appropriately, with buffers, such as sec. Sodium phosphate while alkaline solutions preferably with hydrochloric acid or acidic buffers, such as primary sodium phosphate, be neutralized. The solutions obtained by the process are free of irritation tolerated and can be stored without decomposition. Another benefit of the The method consists in making the effects of the new solutions of the hormone last longer more lasting than that of the known acidic solutions. As is well known, shows an ordinary Islet hormone solution in starving rabbits has an effect which is evident after 21/2 hours has subsided while the effect of the solutions obtainable by the process Lasts 5 hours. The solutions can be applied gently to dry preparations to process. Example 42omg of crystallized islet hormone become dissolved in 10 ccm of normal hydrochloric acid and mixed with. 39o cc of a 5% polyvinyl alcohol solution diluted. This mixture will add so many cubic centimeters of an i o / o solution of secondary sodium phosphate added so that the pii = 7.2. The solution will be Filled to 50o ccm and contains 2o IU per cubic centimeter. It is slightly opalescent and can withstand one-hour heating to 60 ° C. This can be done with the same success sec. Replace sodium phosphate with caustic soda sodium acetate.
