Description
- VERFAHREN ZUR HERSTELLUNG VON α-HYDROXYKETONEN
- Die Erfindung betrifft ein Verfahren zur Herstellung von α-Hydroxyketonen, die in der Natur weit verbreitet sind und wegen ihrer physiologischen Wirkungen von großer Wichtigkeit sind. So sind zum Beispiel Adriamycin und Daumycin als Anti- Krebsmittel bekannt (siehe J. Am. Chem. Soc., Vol. 85, S.5334 (1964)); Betamethason und Cortison sind als Steroidhormone bekannt (siehe J. Am. Chem. Soc., Vol. 80, S.6687 (1958)); und Cyproteron ist als antiandrogenes Mittel bekannt (siehe Ger Offen, Nr. 1189, S.91 (1965)).
- Bekannte Verfahren zur Synthese dieser α-Hydroxyketone schließen zum Beispiel ein: Die Oxidation von Diolen (siehe J. Organomet. Chem., Vol 5, S.263 (1966)) die oxidative Spaltung von Epoxiden (siehe J. Org.Chem, Vol 26, S.1981 (1961)), und die Acyloin-Kondensation (siehe J. Org. Chem, Vol. 405. 393 (1975)) und zusätzlich, von Olefinen ausgehende Synthesen unter Verwendung einer stöchiometrischen Menge an Kaiumpermanganat.
- Andererseits ist von Rutheniumverbindungen bekannt, daß sie die Oxidationsreaktion einer bestimmten Art organischer Verbindungen katalysieren, wie in Kagaku, Vol. 45, Nr.6, S.426-427, Kagaku Dojin K.K. (1990) berichtet. In Anbetracht der Spezifität ihrer katalytischen Aktivität war es gänzlich unerwartet daß Rutheniumverbindungen ebenfalls eine katalytische Wirkung bei der Olefin-Oxidation zur Gewinnung von α-Hydroxyketonen entfalten.
- Ziel der vorliegenden Erfindung ist die Bereitstellung eines Verfahrens zur Herstellung von α-Hydroxyketonen durch Oxidation von Olefinen mit guter Selektivität und in hoher Ausbeute.
- Als Ergebnis ausgedehnter Studien wurde von den Erfindern gefunden daß obiges Ziel der vorliegenden Erfindung bei Verwendung einer Rutheniumverbindung als Katalysator erreicht werden kann.
- Die vorliegende Erfindung bezieht sich auf ein Verfahren zur Herstellung eines α-Hydroxyketones dargestellt durch die Formel (I):
- worin R¹ eine substituierte oder unsubstituierte Alkylgruppe, eine substituierte oder unsubstituierte Aralkylgruppe oder eine Alkoxycarbonylgruppe darstellt; und R² und R³ jeweils ein Wasserstoffatom, eine substituierte oder unsubstituierte Alkylgruppe oder eine substituierte oder unsubstituierte Aralkylgruppe darstellen, unter der Voraussetzung, daß sie nicht zugleich ein Wasserstoffatom darstellen; oder ein Paar R¹ und R², ein Paar R¹ und R³ oder ein Paar R² und R³ zusammengenommen einen Ring darstellen; oder das Paar R¹ und R² und das Paar R² und R³ zugleich einen Ring bilden,
- welches die Reaktion einer Verbindung der Formel (II):
- worin R¹, R² und R³ wie oben definiert sind,
- mit einem Oxidationsmittel in Gegenwart einer Rutheniumverbindung, von Wasser und einem organischen Lösungsmittel, ausgewählt aus Methylenchlorid, Dichlorethan und Chlorbenzol oder deren Mischungen, bei einer Temperatur von -10 ºC bis +30 ºC, umfaßt.
- In den Formeln (I) und (II) schließt die substituierte Alkylgruppe Acetoxyalkyl-, 2-Methyl-4-acetoxypentyl-, Benzyloxymethyl- und Benzoyloxyethylgruppen ein. Die substituierte Aralkylgruppe schließt eine Acetoxybenzylgruppe ein. Die Alkoxycarbonylgruppe schließt Methoxycarbonyl- und Ethoxycarbonylgruppen ein.
- Spezifische Beispiele der durch Formel (II) dargestellten Olefine, die bei der vorliegenden Erfindung als Ausgangsmaterial verwendet werden können, sind Cydopenten, Cyclohexen, Cycloocten, trans-2-Buten, cis-2-Buten, 4-Octen, β-Methylstyrol, Cyclohexadien, Cyclooctadien, Inden, 1-Methylcyclopenten, 1-Methylcydohexen, Ethylidencyclohexan, 2-Methyl-4,4-dimethylpenten, Crotonsäuremethylester, Orotonsäureethylester, Zimtsäuremethylester, 3-Methylcrotonsäureethylester, Pentylidencyclohexan und Ethylidencyclopentan.
- Die bei der vorliegenden Erfindung als Katalysator verwendbare Rutheniumverbindung schließt die nachfolgen aufgelisteten Verbindungen ein, die in Comprehensive Organometallic Chemistry, Vol. 4, S.651-930, Pergamon Press (1982) beschrieben sind:
- 1) Rutheniumverbindungen, dargestellt durch Formel (III):
- RuX&sub3;;
- worin X ein Halogenatom, eine Acyloxygruppe der Formel R&sup4;COO (worin R&sup4; eine niedere Alkylgruppe bedeutet) oder eine Acetylacetonatogruppe darstellt.
- Spezifische Beispiele solcher Rutheniumverbindungen sind Rutheniumtrichlorid, Rutheniumtribromid und Rutheniumtriiodid sowie deren Hydrate, Trisacetylacetonatoruthenium und Rutheniumacetat.
- 2) Rutheniumkomplexe
- (i) Ruthenium-Phosphinkomplexe, z.B.,
- HRuCl(PPH&sub3;)&sub3;,
- RuCl&sub2;(PPH&sub3;)&sub3;,
- H&sub2;Ru(PPH&sub3;)&sub4;,
- Ru&sub2;Cl&sub4;(BINAP)&sub2;(NEt&sub3;),
- Ru(OAc)&sub2;(BINAP),
- Ru&sub2;Cl&sub4;(1,4-diphos)&sub2;,
- HRuCl(BINAP)&sub2;
- [Ru(bpy)&sub2;(O)(PPh&sub3;)](ClO&sub4;)&sub2;,
- [Ru(bpy)&sub2;(O)(PEt&sub3;)](ClO&sub4;)&sub2; und

- worin Et eine Ethylgruppe darstellt; Ph eine Phenylgruppe darstellt; Ac eine Acetylgruppe darstellt; BINAP bedeutet eine 2,2'-Bis(diphenylphosphino)-1,1'-binaphthylgruppe; 1,4-diphos bedeutet 1,4-Bis(diphenylphosphino)butan und bpy eine Bipyridylgruppe; nachfolgend ebenso.
- (ii) Ruthenium-Nitrosylkomplexe, z.B.,
- Ru(NO)Cl&sub3; x H&sub2;O
- (iii) Ruthenium-Carbonylkomplexe, z.B.,
- Ru&sub3;(CO)&sub1;&sub2;,
- [RuCl&sub2;(CO)&sub3;]&sub2; und
- RuCl&sub2;(CO)&sub2;(PPH&sub3;)&sub2;
- (iv) Ruthenium-Oxokomplexe, z.B.,
- RuO&sub2;
- (v) Ruthenium-Olefinkomplexe, z.B.,
- RuCl&sub2;(COD)
- worin COD 1,5-Cyclooctadien darstellt; nachfolgend ebenso.
- 3) Metallische Rutheniumpulver eingeschlossen:
- Ruthenium-auf-Kohle, Ruthenium-auf-Graphit, Ruthenium-auf-Aluminiumoxid, Ruthenium-auf-Siliziumdioxid, Ruthenium-auf-Zeolith und Ruthenium-auf- Zirkon
- Als Katalysator wird die Rutheniumverbindung gewöhnlich in einer Menge von 0,01 bis 0,2 mol und vorzugsweise von 0,03 bis 0,1 mol pro mol Ausgangsolefin verwendet.
- Das Oxidationsmittelv welches bei der vorliegenden Erfindung verwendet werden kann, unterliegt keinen besonderen Einschränkungen. Geeignete Oxidationsmittel schließen verschiedene Persäuren ein, z.B. Peressigsäure, Perpropionsäure und m-Chlorperbenzoesäure. Jedes im Handel erhältliche Oxidationsmittel kann verwendet werden oder aus einer Carbonsäure und Wasserstoffperoxid vor der Reaktion hergestellt werden.
- Das Oxidationsmittel wird üblicherweise in einer Menge von 1 bis 8 mol und vorzugsweise von 2 bis 3 mol pro mol Ausgangsolefin verwendet.
- Bei der Ausführung des erfindungsgemäßen Verfahrens werden eine Verbindung (II), Wasser und eine Rutheniumverbindung in einem geeigneten Lösungsmittel wie Acetonitril, Methylenchlorid, Dichlorethan, Chlorbenzol und deren Mischungen gelöst oder suspendiert und zu dieser Lösung oder Suspension wird bei einer Temperatur von -10 ºC bis +30 ºC die Lösung einer Persäure in Essigsäureethylester oder Essigsäure unter Rühren zugegeben. Während die Reihenfolge oder die Art und Weise der Zugabe des Ausgangsmaterials, des Katalysators usw. keinen besonderen Einschränkungen unterliegen, wird das Oxidationsmittel vorzugsweise zuletzt zugegeben.
- Die Isolierung des Reaktionsproduktes aus der Reaktionsmischung kann durch bekannte Trennverfahren, wie Destillation oder Säulenchromatographie erfolgen.
- Die vorliegende Erfindung wird nachfolgend unter Bezugnahme auf die Beispiele detaillierter erläutert, es versteht sich jedoch, daß damit keine Beschränkung der vorliegenden Erfindung darauf beabsichtigt ist. Alle Prozentangaben sind auf Gewicht bezogen, soweit nichts anderes angegeben ist.
- In einen mit einem Thermometer, einem Tropftrichter und einem Rührer ausgerüsteten 50 ml Kolben wurden 10 ml Methylenchlorid, 10 ml Acetonitril, 10 ml Wasser, 0,82 g (10 mmol) Cyclohexen und 78 mg (0,3 mmol) RuCl&sub3; x 3H&sub2;0 eingebracht. Die Mischung wurde unter Rühren mit einem Magnetrührer auf 18 ºC abgekühlt und eine 30 %ige Essigsäureethylesterlösung von 7,6 g (30 mmol) Peressigsäure über 2 Stunden tropfenweise zugegeben. Nach 2-stündigem Rühren bei Raumtemperatur wurde die Reaktionsmischung dreimal mit 50 ml Portionen Methylenchlorid extrahiert. Die vereinigten Extrakte wurden mit 50 ml einer gesättigten wäßrigen Natriumchlorid lösung gewaschen und über wasserfreien Magnesiumsulfat getrocknet. Das Lösungsmittel wurde durch Destillation entfernt unter Erhalt von 0,843 g (prozentuale Ausbeute: 74 %) 2-Hydroxycyclohexanon (α-Hydroxycyclohexanon).

- In einen mit einem Thermometer, einem Tropftrichter und einem Rührer ausgerüsteten 50 ml Kolben wurden 10 ml Methylenchlorid, 10 ml Acetonitril, 10 ml Wasser, 1,12 g (10 mmol) 4-Octen und 78 mg (0,3 mmol) Rutheniumtrichlorid eingebracht und eine 30 %ige Essigsäureethylesterlösung von 7,6 g (30 mmol) Peressigsäure bei 15 ºC während 1,5 Stunden unter Rühren mit einem Magnetrührer tropfenweise der Mischung zugesetzt. Nach der Zugabe wurde 2 Stunden bei Raumtemperatur weiter gerührt. Die Reaktionsmischung wurde dreimal mit 30 ml Portionen Methylenchlorid extrahiert. Die vereinigten Extrakte wurden mit 30 ml einer wäßrigen gesättigten Natriumchloridlösung gewaschen und über wasserfreien Magnesiumsulfat getrocknet. Das Lösungsmittel wurde durch Destillation entfernt unter Erhalt eines rohen 5-Hydroxy4-octanon welches dann mittels Silicagel-Säulenchromatographie unter Verwendung einer 10:1 (Volumenverhältnis) Mischung von n- Hexan und Essigsäureethylester als Eluens gereinigt wurde, wobei 0,87 g (prozentuale Ausbeute: 59 %) des 5-Hydroxy-4-octanons erhalten wurden.

- In einen mit einem Thermometer und einem Tropftrichter ausgerüsteten 50 ml Kolben wurden unter Stickstoffatmosphäre 10 ml Methylenchlorid, 10 ml Acetonitril, 5 ml Wasser, 0,82 g (10 mmol) Cyclohexen und 277 mg (0,3 mmol) HRuCl(PPH&sub3;)&sub3; eingebracht. Zu dieser Mischung wurden eine 30 %ige Essigsäureethylesterlösung von 7,6 g (30 mmol) Peressigsäure bei 20 ºC während 2 Stunden unter Rühren tropfenweise zugesetzt. Nach der Zugabe wurde 1 Stunde bei Raumtemperatur weiter gerührt und die Mischung in der gleichen Weise wie im Beispiel 1 aufgearbeitet, wobei 0,68 g (prozentuale Ausbeute. 60 %) 2-Hydroxycyclohexanon (α-Hydroxycyclohexanon) erhalten wurden.
- ¹H-NMR: Dasselbe wie in Beispiel 1.
- Die in der nachfolgenden Tabelle 1 gezeigten Verbindungen wurden auf die gleiche Weise wie in Beispiel 1 hergestellt. Die NMR-Spektren dieser Verbindungen sind ebenfalls gezeigt. - In einen mit einem Thermometer, einem Tropftrichter und einem Rührer ausgerüsteten 50 ml Kolben wurden 7 ml Methylenchlorid, 7 ml Wasser, 2 mmol 1-Methyl-1-cyclohexen und 0,1 mmol jeweils eines der in der nachfolgenden Tabelle 2 gezeigten Ruthenium-Katalysatoren eingebracht. Zu der Mischung wurden eine 30 %ige Essigsäureethylesterlösung von 1,52 g (6 mmol) Peressigsäure bei Raumtemperatur während 2 Stunden unter Rühren mit einem Magnetrührer tropfenweise zugesetzt. Nach der Zugabe wurde das Rühren 2 Stunden bei Raumtemperatur fortgesetzt. Die Reaktionsmischung wurde dreimal mit 20 ml Portionen Methylenchlorid extrahiert. Der Extrakt wurde mit 30 ml einer gesättigten wäßrigen Natriumchloridlösung gewaschen und über wasserfreien Magnesiumsulfat getrocknet. Das Lösungsmittel wurde durch Destillation entfernt und das zurückbleibende Rohprodukt mittels Silicagel-Säulenchromatographie unter Verwendung einer 5:1 (Volumenverhältnis) Mischung von n-Hexan und Essigsäureethylester als Eluens gereinigt, wobei 2-Methyl-2-hydroxy-1-cyclohexanon erhalten wurde. die prozentuale Ausbeute des Produktes ist in Tabelle 2 gezeigt. - Die in der nachfolgenden Tabelle 3 gezeigten Verbindungen wurden auf die gleiche Weise wie in Beispiel 1 hergestellt. Tabelle 3 Beispiel Nr. - Die vorliegende Erfindung stellt so ein industriell vorteilhaftes Verfahren zur Herstellung von α-Hydroxyketonen zur Verfügung, die als physiologisch aktive Stoffe oder als Zwischenprodukte bei guter Reaktionsselektivität und hoher Ausbeute für ihre einfache Herstellung verwendbar sind.
Claims (4)
1. Verfahren zur Herstellung eines α-Hydroxyketons dargestellt durch Formel (I):
worin R¹ eine substituierte oder unsubstituierte Alkylgruppe, eine substituierte
oder unsubstituierte Aralkylgruppe oder eine Alkoxycarbonylgruppe darstellt
und R² und R³ jeweils ein Wasserstoffatom, eine substituierte oder
unsubstituierte Alkylgruppe oder eine substitu erte oder unsubstituierte
Aralkylgruppe darstellen, unter der Voraussetzung, daß sie nicht zugleich ein
Wasserstoffatom darstellen; ein Paar R¹ und R², ein Paar R¹ und R³ oder ein
Paar R² und R³ zusammengenommen einen Ring darstellen; oder das Paar R¹
und R² und das Paar R² und R³ zugleich eine Ringstruktur bilden,
welches die Reaktion einer Verbindung der Formel (II):
worin R¹, R² und R³ wie oben definiert sind,
mit einem Oxidationsmittel in Gegenwart einer Rutheniumverbindung, von
Wasser und einem organischen Lösungsmittel, ausgewählt aus Methylenchlorid,
Dichlorethan und Chlorbenzol oder deren Mischungen, bei einer Temperatur
von -10 ºC bis +30 ºC, umfaßt.
2. Verfahren gemäß Anspruch 1, bei dem die genannte Rutheniumverbindung eine
durch die Formel (III)
RuX&sub3;:
dargestellte Verbindung ist, worin X ein Halogenatom, eine Acyloxygruppe der
Formel R&sup4;COO, worin R&sup4; eine niedere Alkylgruppe darstellt, oder eine
Acetylacetonatgruppe darstellt.
3. Verfahren gemäß Anspruch 1, bei dem die genannte Rutheniumverbindung ein
Rutheniumkomplex, ausgewählt aus einem Ruthenium-Phosphinkomplex, einem
Ruthenium-Nitrosylkomplex, einem Ruthenium-Carbonylkomplex, einem
Ruthenium-Oxokomplex und einem Ruthenium-Olefinkomplex, ist.
4. Verfahren gemäß Anspruch 1 bei dem die genannte Rutheniumverbindung
metallisches Ruthenium oder Ruthenium auf einem Träger ist.
