Description
- Bacillus thuringiensis (B.t.) produziert ein Insekten- Toxin, das als δ-Endotoxin bezeichnet wird. Es wird von der B.t.-Sporulationszelle produziert. Wenn das Toxin in kristalliner Form von dagegen empfindlichen Insektenlarven aufgenommen wird, wird es von den Darmsaft-Proteasen der Insekten in biologisch aktive Bestandteile umgewandelt. Das Hauptangriffsziel sind Insektenzellen des Darmepithels, die schnell zerstört werden.
- Wie berichtet wurde, umfaßt das Wirkungsspektrum von B.t. Insektenarten innerhalb der Ordnung der Lepidoptera von denen viele große Schäden in Land- und Forstwirtschaft anrichten. Das Wirkungsspektrum beinhaltet auch die Insektenordnung der Dioptera, zu der Stechmücken und Kriebelmücken gehören. Siehe Couch, T.L. (1980) "Mosquito Pathogenicity of Bacillus thuringiensis var. israelensis", Developments in Industrial Microbiology 22:61-76; Beegle, C.C., (1978) "Use of Entomogenous Bacteria in Agroecosystems", Developments in Industrual Microbiology 20:97-104. Krieg, et al., Z. ang. Ent. (1983) 96:500-508 beschreiben ein B.t.-Isolat, genannt Bacillus thuringiensis var. tenebrionis, von dem berichtet wird, daß er gegen zwei Käfer der Ordnung Coleoptera wirksam ist. Es handelt sich um den Kartoffelkäfer, Leptinotarsa decemlineata, und Agelastica alni.
- In der Europäischen Patentanmeldung 0 202 739 wird ein neues gegen Coleoptera wirksames B.t.-Isolat offenbart. Es ist als B. thuringiensis var. san diego (B.t.sd.) bekannt.
- Gegen Coleoptera wirksame Stämme, wie z.B. B.t.sd., können zur Bekämpfung blattfressender Käfer eingesetzt werden. Der Kartoffelkäfer (Leptinotarsa decemlineata) ist z.B. gegen das δ-Endotoxin des B.t.sd. empfindlich und seine Larven werden, wenn sie eine ausreichende Menge eines Sporen/Kristall-Präparats über damit behandelte Blätter aufnehmen, abgetötet.
- Viele Erntepflanzen werden von Erdflöhen befallen. Diese Käfer gehören zur Familie der Chrysomelidae, der decemlineata. Die ausgewachsenen Tiere können durch Blattfraß erhebliche Schäden verursachen.
- Die vorliegende Erfindung betrifft ein neues gegen Coleoptera-Schädlinge wirksames Bacillus thuringiensis (B.t.)-Isolat. Das neue B.t.-Isolat, das hier als Bacillus thuringiensis PS86B1 (B.t. PS86B1) bezeichnet wird, hat sich bisher z.B. als wirksam gegen den Kartoffelkäfer (Leptinotarsa decemlineata) erwiesen. Umfangreichere Untersuchungen hinsichtlich der Wirtstiere sind im Gange.
- Die vorliegende Erfindung umfaßt auch Mutanten von B.t. PS86B1, die im wesentlichen die gleichen pestiziden Eigenschaften wie B.t. PS86B1 besitzen. Verfahren zur Herstellung von Mutanten sind in der Mikrobiologie bekannt. Hierfür werden weithin ultraviolettes Licht und Nitrosoguanidin eingesetzt.
- Die Erfindung beinhaltet weiterhin die Behandlung von im wesentlichen intakten B.t. PS86B1-Zellen zur Verlängerung der pestiziden Wirkung, wenn die im wesentlichen intakten Zellen in der Umgebung eines Zielgruppen-Schädlings angewendet werden. Eine solche Behandlung kann auf chemischem oder physikalischem Weg oder durch eine Kombination chemischer oder physikalischer Wege erfolgen, solange das Verfahren weder die Eigenschaften des Pestizids negativ beeinflußt noch die Fähigkeit der Zellen, das Pestizid zu schützen, beeinträchtigt. Die behandelte B.t. PS86B1-Zelle wirkt als Schutzschicht für das pestizide Toxin. Das Toxin wird erst als solches wirksam, wenn es von einem Zielgruppen-Insekt aufgenommen wird.
- Das neue Bacillus thuringiensis-Isolat der vorliegenden Erfindung besitzt die folgenden Merkmale:
- Kolonien-Morphologie: große Kolonie, stumpfe Oberfläche, typisch für B.t.
- Vegetative Zellen-Morphologie: typisch für B.t.
- Züchtungsmethoden: typisch für B.t.
- Flagellarer Serotyp: PS86B1 gehört zu Serovar 9, Tolworthi.
- Einschlüsse: eine nicht-zerbrechliche, flache, an den Enden spitz zulaufende Ellipse und mehrere sehr kleine Einschlüsse.
- Plasmidpräparate: bei der Agarosegel-Elektrophorese der Plasmidpräparate wird B.t. PS86B1 von B.t.sd. und anderen B.t.-Isolaten unterschieden.
- Alkali-lösliche Proteine: SDS-Polyacrylamidgele weisen 61000, 68000 und 75000 Dalton Proteine auf.
- Coleoptera-Toxin: der Bioassay zeigt Wirksamkeit gegen Kartoffelkäfer bei einer LC&sub5;&sub0; von 8,4 ug/ml.
- Ein Vergleich der Merkmale der bekannten B.t.-Stämme B. thuringiensis var. kurstaki (HD-1) und B. thuringiensis var. san diego (B.t.sd.) mit B. thuringiensis PS86B1 (B.t. PS86B1) ist in Tabelle 1 gezeigt. - Ein Teil der oben angegebenen Morphologie des B.t. PS86B1 ist in den Zeichnungen, die die am 23.02.88 eingereichte US Stammpatentanmeldung Nr. 159144 begleiten, zu sehen.
- Eine Kultur von Bacillus thuringiensis PS86B1 wurde am 03.02.88 in der Agricultural Research Service Patent Culture Collection (NRRL), Northern Regional Research Center, 1815 North University Street, Peoria, Illinois, 61604, USA hinterlegt. Die Hinterlegungsnummer ist NRRL B-18299.
- B.t. PS86B1, NRRL B-18299, kann mit Hilfe von Standard- Medien und Fermentationstechniken gezüchtet werden. Nach Beendigung des Fermentationszyklus können die Bakterien geerntet werden, indem man zunächst die B.t.-Sporen und - Kristalle auf dem Fachmann bekannte Weise von der Fermentationsbrühe trennt. Die gewonnenen B.t.-Sporen und - Kristalle können zur leichteren Handhabung und Anwendung für bestimmte Zielgruppen-Schädlinge durch Zugabe von oberflächenaktiven Stoffen, Dispergiermitteln, inerten Trägersubstanzen und anderen Bestandteilen zu Spritzpulver, flüssigem Konzentrat, Granulat oder anderen Formulierungen formuliert werden. Diese Formulierungs- und Anwendungsverfahren sind dem Fachmann alle bekannt.
- Formulierte Produkte können zur Bekämpfung pflanzenfressender Käfer oder Raupen auf die Blätter gesprüht oder aufgetragen werden.
- Ein weiterer möglicher Ansatz ist, daß man die Sporen und Kristalle des B.t. PS86B1 in Köder-Granulat, das einen Lockstoff enthält, gibt und dieses Granulat zur Bekämpfung am Boden lebender Coleoptera auf den Boden aufbringt. Formulierter B.t. PS86B1 kann auch als Saatgut-Überzug, zur Behandlung der Wurzeln oder zur Behandlung der ganzen Pflanze aufgetragen werden.
- Die B.t. PS86B1-Zellen können vor der Formulierung zur Verlängerung der pestiziden Wirkung behandelt werden, wenn die Zellen in der Umgebung eines Zielgruppen-Schädlings angewendet werden. Eine solche Behandlung kann auf chemischem oder physikalischem Wege, oder durch eine Kombination chemischer und/oder physikalischer Wege erfolgen, solange das Verfahren weder die Eigenschaften des Pestizids negativ beeinflußt noch die Fähigkeit der Zellen, das Pestizid zu schützen, beeinträchtigt. Beispiele für chemische Reagenzien sind Halogeniermittel, insbesondere Halogene der Ordnungszahlen 17 bis 80. Insbesondere Jod kann unter schonenden Bedingungen und über einen ausreichend langen Zeitraum hinweg verwendet werden, um die gewünschten Ergebnisse zu erzielen. Andere geeignete Verfahren beinhalten die Behandlung mit Aldehyden, wie z.B. Formaldehyd und Glutaraldehyd; Desinfektionsmitteln, wie z.B. Zephiranchlorid; Alkoholen, wie z.B. Isopropanol und Ethanol; verschiedenen histologischen Fixierungsmitteln, wie z.B. Bouins-Fixativ und Hellys-Fixativ (siehe: Humason, Gretchen. L., Animal Tissue Techniques, W.H. Freeman and Company, 1967); oder einer Kombination von physikalischen (Wärme) und chemischen Mitteln, die die Wirkung des in der Zelle produzierten Toxins verlängern, wenn die Zelle in der Umgebung des bzw. der Zielgruppen-Schädlinge eingesetzt wird. Beispiele für physikalische Mittel sind kurzwellige Strahlen, wie z.B. Gamma-Strahlen und Röntgenstrahlen, Gefrieren, UV-Bestrahlung, Gefriertrocknung u.ä.
- Es folgen Beispiele, die Verfahren, einschließlich der besten Verfahrensweise, zur Durchführung der Erfindung erläutern. Diese Beispiele sollen nicht als beschränkend verstanden werden. Alle Prozentangaben sind Gewichtsprozent und alle Lösungsmittel-Mischungsverhältnisse beziehen sich, soweit nicht anders angegeben, auf das Volumen.
- Eine Subkultur von B.t. PS86B1, NRRL B-18299 kann zur Beimpfung des folgenden Mediums, eines Pepton-, Glucose-, Salzmediums, verwendet werden.
- Bactopepton 7,5 g/l
- Glucose 1,0 g/l
- KH&sub2;PO&sub4; 3,4 g/l
- KP&sub2;HPO&sub4; 4,35 g/l
- Salzlösung 5,0 ml/l
- CaCl&sub2;-Lösung 5,0 ml/l
- Salzlösung (100 ml)
- MgSO&sub4;.7H&sub2;O 2,46 g
- MnSO&sub4;.H&sub2;O 0,04 g
- ZnSO&sub4;.7H&sub2;O 0,28 g
- FeSO&sub4;.7H&sub2;O 0,40 g
- CaCl&sub2;-Lösung (100 ml)
- CaCl&sub2;.2H&sub2;O 3,66 g
- pH 7,2
- Die Salzlösung und die CaCl&sub2;-Lösung werden filtersterilisiert und bei der Beimpfung der unter Druck gekochten Brühe zugegeben. Kolben werden 64 Stunden lang bei 200 Upm auf einer Rotationsschüttelmaschine bei 30ºC inkubiert.
- Das oben beschriebene Verfahren läßt sich durch dem Fachmann bekannte Verfahren leicht auf große Fermentatoren übertragen.
- Die bei der oben beschriebenen Fermentation erhaltenen B.t.-Sporen und -Kristalle können durch dem Fachmann bekannte Verfahren isoliert werden. In einem häufig verwendeten Verfahren wird die erhaltene Fermentationsbrühe Trennverfahren, z.B. einer Zentrifugation, unterzogen.
- B.t. PS86B1, NRRL B-18299-Sporen und -Kristalle wurden gegen den Kartoffelkäfer getestet. B.t. PS86B1 hat in dem Kartoffelkäfer-Test eine LC&sub5;&sub0; von 8,4 ug Protein/ml. Die Prüfung für den Kartoffelkäfer wurde wie folgt durchgeführt.
- Kartoffelkäfer-Bioassay - Leptinotarsa decemlineata-Larven der frühen zweiten Erscheinungsform werden auf Kartoffelblätter gesetzt, die in Suspensionen, die Bacillus thuringiensis-Präparate enthalten, getaucht worden waren. Die Larven werden 4 Tage lang bei 25ºC gehalten und die Larven-Sterblichkeitsrate wird aufgezeichnet und durch Probit-Analyse analysiert.
Claims (10)
1. Bacillus thuringiensis PS86B1 mit den
kennzeichnenden Merkmalen von NRRL B-18299 oder
eine Mutante davon, die gegen einen Insekten-
Schädling aus der Ordnung Coleoptera wirksam ist.
2. Gegen einen Coleoptera-Schädling wirksames Toxin,
produziert von B. thuringiensis PS86B1, der die
kennzeichnenden Merkmale von NRRL B-18299 besitzt.
3. Zusammensetzung, umfassend einen Mikroorganismus
nach Anspruch 1, z.B. als Sporen oder Kristalle, in
Verbindung mit einer Insektizid-Trägersubstanz oder
mit Formulierungsbestandteilen, die als Saatüberzug
aufgetragen werden.
4. Zusammensetzung nach Anspruch 3, worin die
Trägersubstanz Käfer-Phagostimulantien oder
Lockstoffe umfaßt.
5. Verfahren zur Bekämpfung von Coleoptera-Insekten-
Schädlingen, umfassend das Kontaktieren der
Schädlinge oder ihrer Umgebung mit einem
Mikroorganismus nach Anspruch 1.
6. Verfahren nach Anspruch 5, umfassend das Plazieren
eines Köder-Granulats, das den Mikroorganismus,
z.B. als Sporen oder Kristalle, enthält, auf oder
im Boden beim Einpflanzen des Samens einer Pflanze,
von der sich der Schädling bekanntermaßen ernährt.
7. Verfahren nach Anspruch 6, worin das Köder-Granulat
gleichzeitig mit dem Einbringen von Getreidesaat in
den Boden gegeben wird.
8. Verfahren nach Anspruch 5, worin der Schädling auf
gelagerten Produkten vorhanden ist.
9. Verfahren nach irgendeinem der Ansprüche 5 bis 8,
worin der Schädling der Kartoffelkäfer ist.
10. Verfahren nach irgendeinem der Ansprüche 5 bis 9,
worin im wesentlichen intakte Zellen des
Mikroorganismus behandelt werden, um die pestizide
Wirkung zu verlängern, wenn im wesentlichen intakte
Zellen in die Umgebung des Schädlings ausgebracht
werden.
