Description
- Gegenstand der vorliegenden Erfindung sind spezifische Lipidester von Nucleotiden der allgemeinen Formel I: worin
R1 ein geradkettiger oder verzweigter, gesättigter oder ungesättigter Alkylrest mit 1 bis 20 C-Atomen ist, gegebenenfalls einfach oder mehrfach substituiert durch Halogen, C1-C6-Alkoxy-, C1-C6-Alkylmerkapto-, C1-C6-Alkoxycarbonyl-, C1-C6-Alkylsulfinyl- oder C1-C6-Aklysulfonylgruppen
R2 Wasserstoff, ein geradkettiger oder verzweigter, gesättigter oder ungesättigter Alkylrest mit 1 bis 20 C-Atomen ist, gegebenenfalls einfach oder mehrfach substituiert durch Halogen, C1-C6-Alkoxy-, C1-C6-Alkylmerkapto-, C1-C6-Alkoxycarbonyl- oder C1-C6-Aklysulfonylgruppen,
R3 Amino ist
X für Schwefel, eine Sulfinyl- oder Sulfonylgruppe steht und
Y ein Sauerstoffatom ist,
deren Tautomere und physiologisch verträgliche Salze anorganischer und organischer Säuren und Basen; sowie Verfahren für ihre Herstellung, und Arzneimittel, welche diese Verbindungen als Wirkstoff enthalten. - Die Aminogruppe im Adeninrest der allgemeinen Formel I kann auch durch an sich gut bekannte Aminoschutzgruppen geschützt sein.
- Da die Verbindungen der allgemeinen Formel I asymmetrische Kohlenstoffatome enthalten sind sämtliche optisch aktiven Formen und racemischen Gemische dieser Verbindungen ebenso Gegenstand der vorliegenden Erfindung.
- J. Biol. Chem. 265, 6112 (1990) und EP-A-0,350,287 beschreiben die Herstellung und Verwendung von Liponucleotiden als antivirale Medikamente. Darin werden jedoch nur Dimyristylphosphatidyl- und Dipalmylphosphatidylreste mit allgemein bekannten Nucleosiden wie AZT und DDC offenbart, einschließlich deren Fettsäureesterstruktur.
- J. Med. Chem. 33, 1380, (1990) beschreibt Nucleosidkonjugate von Thioetherlipiden mit Cytidindiphosphat, die eine antitumorale Wirkung haben und in der Onkologie Verwendung finden können.
- Chem. Pharm. Bull. 36, 209 (1988) beschreibt 5'-(3-sn-phosphatidyl)nucleoside mit antileukämischer Wirkung sowie deren enzymatische Synthese aus den entsprechenden Nucleosiden und Phosphocholinen in Gegenwart von Phospholipase D mit Transferase-Aktivität.
- Die Patentanmeldung WO 92/03462 beschreibt Thioetherlipidkonjugate mit antiviraler Wirkung, insbesondere für die Behandlung von HIV-Infektionen.
- WO 96/15234 beschreibt Phospholipidnucleosidkonjugate, welche im Vergleich zu den nicht konjugierten Nucleosiden eine deutlich verbesserter Wirkstärke zeigen.
- Die Herstellung von 2-Chlor-9-(2'-desoxy-2-fluor-β-D-arabinofuranosyl)-adenin ist in J. Org. Chem. 34, 2632–2636 (1969), in der Patentanmeldung WO 01/60383 und in US Patent 6,680,382 beschrieben.
- Die pharmakologische Wirkung von 2-Chlor-9-(2'-desoxy-2'-fluor-β-D-arabinofuranosyl)-adenin als Hemmstoff der DNA-Replikation im Vergleich zu anderen Nucleosiden wird auch in Hematology 463 (1999) beschrieben.
- Andere halogenierte Arabinoadenosine mit antitumoraler Wirkung sind in den Patentanmeldungen
US 5,384,310 und WO 92/20347 erwähnt. - Die antivirale Wirkung von Purin-Derivaten ist in
EP 0 314 011 gezeigt. - 2-Chlor-9-(2'-desoxy-2'-fluor-β-D-arabinofuranosyl)-adenin (Clofarabin) ist ein sehr bekanntes Entwicklungsprodukt in klinischen Studien (J. Med. Chem., 35, 397, (1992) und Bioorg. & Med. Chem. Letters, 5 (24), 2999, (1995)).
- Die Verbindungen der allgemeinen Formel I gemäß der vorliegenden Erfindung, welche die chemische Struktur 2-Chlor-9-(2'-desoxy-2'-fluor-β-D-arabinofuranosyl)-adenin beinhalten, besitzen eine biologische Wirksamkeit, die sie von dem Stammnucleosid unterscheiden. Insbesondere zeigen die Verbindungen gemäß der vorliegenden Erfindung eine antitumorale Wirkung und sind brauchbar da, in pharmakologisch relevanten Dosierungen, ein oder mehrere toxische Nebenwirkungen der Stammverbindung abgeschwächt wird/werden, und/oder die kovalent gebundenen Lipideinheit die Bioverfügbarkeit der konjugierten Wirksubstanz verbessert und damit zur gesteigerten Selektivität und Wirksamkeit der Verbindungen beizutragen scheint.
- Die Verbindungen gemäß der vorliegenden Erfindung haben wertvoll pharmakologische Eigenschaften. Insbesondere sind sie zur Therapie und Prophylaxe von bösartigen Tumoren, einschließlich Karzinomen, Sarkomen und Leukämien, geeignet.
- Im Vergleich zu den unkonjugierten Nucleosidderivaten, die bislang in der Therapie bösartiger Tumoren Verwendung finden, haben die Verbindungen gemäß der vorliegenden Erfindung eine gesteigerte Potenz/Wirksamkeit für spezielle Indikationen oder eine geringere Toxizität und damit ein höhere therapeutische Breite. In einigen Ausführungsformen der vorliegenden Erfindung kann die Verabreichung der pharmazeutischen Zusammensetzungen umfassend diese Verbindungen kontinuierlich über einen längeren Zeitraum erfolgen. Zeiten des Absetzens der Zubereitung oder einer intermittierenden Verabreichung, die aufgrund der unerwünschten Nebenwirkungen häufig zur Routine bei chemotherapeutischen Wirkstoffen gehören, können mit den Verbindungen der vorliegenden Erfindung im Vergleich zu den Stammverbindungen reduziert werden. Weiterhin können durch die abgeschwächten toxischen Nebenwirkungen aufgrund der gesteigerten Selektivität der Tumortoxizität höhere Dosierungen angewandt werden.
- Die Lipidesterverbindungen der vorliegenden Erfindung sind auch zur Behandlung von Autoimmunerkrankungen, einschließlich Multiple Sklerose, Rheumatoide Arthritis, Lupus, systemische Vaskulitis, entzündliche Darmerkrankungen, Skleroderm und Sjorgens Syndrom, geeignet.
- Die Lecithin-artige Struktur der Lipideinheit ist für die beanspruchten Verbesserungen der Verbindungen gemäß der allgemeinen Formel I wünschenswert. Der Durchtritt durch Membranen und Resorptionsbarrieren wird erleichtert und die Konjugate gemäß Formel I zeigen einen Depoteffekt in verschiedenen Geweben.
- Die Bildung der Lipidkonjugate kann aufgrund einer verbesserten Diffusion oder aktiver Transportprozesse auch den Durchgang durch die Blut-Hirnschranke erleichtern.
- Analog können die Verbindungen der vorliegenden Erfindung und ihre pharmazeutischen Formulierungen in freier oder fixer Kombination mit anderen Arzneimitteln zur Therapie oder Prophylaxe der zuvor genanten Erkrankungen angewendet werden.
- Beispiele für diese Arzneimittel schließen Wirkstoffe wie z.B. Mitoseinhibitoren, wie Colchicin, Vinblastin; alkylierende Cytostatika wie Cyclophosphamid, Melphalan, Myleran oder cis-Platin; Antimetaboliten wie Folsäureantagonisten (Methotrexat) und Antagonisten von Purin- und Pyrimidinbasen (Mercaptopurin, 5-Fluoruridin, Cytarabin); zytostatisch wirksame Antibiotika wie die Anthracycline (z.B. Doxorubicin, Daunorubicin); Hormone wie Fosfesterol, Tamoxifen; Taxane, z.B. Taxol, und andere cytostatisch/cytotoxisch wirksame chemotherapeutische und biologische Wirkstoffe ein.
- Ausführungsformen der Erfindung umfassen auch die Verbindungen der allgemeinen Formel I, einschließlich Alkali-, Erdalkali- und Ammoniumsalze der Phosphatgruppe. Beispiele für Alkalisalze schließen Lithium-, Natrium- und Kaliumsalze ein.
- Erdalkalisalze umfassen Magnesium und Calcium, und unter Ammoniumsalzen sind solche, die das Ammoniumion, welches bis zu viermal mit Alkylresten mit 1–4 Kohlenstoffatomen und/oder Arylresten wie dem Benzylrest substituiert sein kann, zu verstehen. Hierbei können die Substituenten gleich oder verschieden sein.
- Die Verbindungen der allgemeinen Formel I können basische Gruppen, insbesondere Aminogruppen enthalten, welche in Säureadditionssalze überführt sein können durch geeignete anorganische oder organische Säuren. In dieser Hinsicht mögliche Säure sind insbesondere: Salzsäure, Bromwasserstoffsäure, Schwefelsäure, Phosphorsäure, Fumarsäure, Bernsteinsäure, Weinsäure, Zitronensäure, Milchsäure, Maleinsäure oder Methansulfonsäure.
- In der allgemeinen Formel I steht R1 vorzugsweise für einen geradkettigen C8-C16-Alkylrest, der zusätzlich durch eine C12-C6-Alkoxy- oder eine C1-C6-Alkylmercaptogruppe substituiert sein kann. Besonders bevorzugt steht R1 für einen Nonyl-, Decyl-, Undecyl-, Dodecyl-, Tridecyl-, Tetradecyl- oder Pentadecylrest. Vorzugsweise sind Methoxy-, Ethoxy-, Butoxy- und Hexyloxygruppen die möglichen Substituenten des R1 Restes. Wenn R1 durch einen C1-C6-Alkylmercaptorest substituiert ist, ist darunter insbesondere der Methylmercapto-, Ethylmercapto-, Propylmercapto, Butylmercapto- und Hexylmercaptorest zu verstehen.
- Vorzugsweise steht R2 für eine C8-C15-Alkylgruppe die zusätzlich durch eine C1-C6-Alkoxy- oder eine C1-C6-Alkylmercaptogeruppe substituiert sein kann. Ganz besonders bevorzugt stellt R2 eine Octyl-, Nonyl-, Decyl-, Undecyl-, Dodecyl-, Tridecyl- oder Tetradecylgruppe dar. Vorzugsweise sind Methoxy-, Ethoxy-, Propoxy-, Butoxy- und Hexyloxygruppen bevorzugt als C1-C6-Alkoxysubstituenten von R2. Wenn R2 durch einen C1-C6-Alkylmercaptorest substituiert ist, ist darunter insbesondere der Methylmercapto-, Ethylmercapto-, Propylmercapto, Butylmercapto-, Pentylmercapto- und Hexylmercaptorest zu verstehen.

- Die am meisten bevorzugten Verbindungen sind [2-Chlor-9-(2'-desoxy-2'-fluor-β-D-arabinofuranosyl)-adenin]-5'-phosphorsäure-(3-dodecylmercapto-2-decyloxy)propylester, [2-Chlor-9-(2'-desoxy-2'-fluor-β-D-arabinofuranosyl)-adenin]-5'-phosphorsäure-(3-dodecylsulfinyl-2-decyloxy)propylester, [2-Chlor-9-(2'-desoxy-2'-fluor-β-D-arabinofuranosyl)-adenin]-5'-phosphorsäure-(3-dodecylsulfonyl-2-decyloxy)propylester sowie [2-Chlor-9-(2'-desoxy-2'-fluor-β-D-arabinofuranosyl)-6-methoxy-9N-purin]-5'-phosphorsäure-(3-dodecylmercapto-2-decyloxy)propylester, [2-Chlor-9-(2'-desoxy-2'-fluor-β-D-arabinofuranosyl)-6-methoxy-9H-purin]-5'-phosphorsäure-(3-dodecylsulfinyl-2-decyloxy)propylester und [2-Chlor-9-(2'-desoxy-2'-fluor-β-D-arabinofuranosyl)-6-methoxy-9H-purin]-5'-phosphorsäure-(3-dodecylsulfonyl-2-decyloxy)propylester.
- Die Verbindungen der allgemeinen Formel I können hergestellt werden durch
- 1. Umsetzung einer Verbindung der allgemeinen Formel II, oder eines Salzes davon worin R1, R2, X und Y die angegebenen Bedeutungen haben, mit einer Verbindung der allgemeinen Formel III worin R3 Amino oder OR4 ist, wobei R4 C1-C8-Alkyl ist, und die 3'-Hydroxygruppe gegebenenfalls durch eine dem Fachmann bekannte Sauerstoffschutzgruppe geschützt sein kann und die Verbindung der Formel II in Gegenwart eines geeigneten Säurecfilorides, wie 2,4,6-Triisopropylbenzolsulfonsäurechlorid, und einer tertiären Stickstoffbase, z.B. Pyridin oder Lutidin, in einem inerten Lösungsmittel, wie Toluol, oder direkt in wasserfreiem Pyridin aktiviert sein kann und gegebenenfalls, nach der anschließenden Hydrolyse die Sauerstoffschutzgruppen mit in der Nucleosidchemie üblichen Verfahren entfernt werden und Umsetzung der OR4-Gruppe in 6'-Stellung des Purin in eine Aminogruppe oder Umsetzung eines Lipidalkohols (entsprechend Formel II) mit einem Nucleosid-5'-monophosphat (entsprechend der Formel III, worin R3 Amino ist) in gleicher Weise wie oben erwähnt, oder
- 2. Umsetzung einer Verbindung der allgemeinen Formel IV worin R1, R2, X und Y die vorgenannten Bedeutungen haben, mit einer Verbindung der allgemeinen Formel III, worin R3 Amino oder OR4 ist, wobei R4 C1-C8-Alkyl ist, und die 3'-Hydroxygruppe gegebenenfalls durch eine dem Fachmann bekannte Sauerstoffschutzgruppe geschützt sein kann, in Gegenwart von Phospholipase D aus Streptomyces in einem inerten Lösungsmittel, wie Chloroform, in Gegenwart eines geeigneten Puffers, und gegebenenfalls im Anschluss an die Reaktion Entfernung der Sauerstoffschutzgruppe mit in der Nucleosidchemie üblichen Verfahren und Umsetzung der OR4-Gruppe in 6'-Stellung des Purin in eine Aminogruppe.
- Die Herstellung der Verbindungen der allgemeinen Formeln II und IV erfolgt analog zu Lipids 22, 947 (1987) und J. Med. Chem. 34, 1377 (1991). Verbindungen der Formel III werden analog zu J. Org. Chem. 34, 2632–2636 (1996), J. Med. Chem. 35, 397–401 (1992) oder WO 01/60383 hergestellt, wenn R3 eine Aminogruppe ist oder in zwei Schritten, wenn R3 = OR4. Der erste Schritt umfasst die Herstellung von und 2,6-Dichlor-9-(3',5'-O-dibenzoyl-2'-desoxy-2'-fluor-β-D-arabinofuranosyl)-9H-purin durch Umsetzung von 2,6-Dichlorpurin mit einem geschützten 2-Desoxy-2-fluor-α-D-arabinofuranosylhalogenid in einem geeigneten Lösungsmittel in Gegenwart einer gehinderten Kaliumbase, vorzugsweise Kalium-tert.-Butyloxid oder Kalium-tert.-Amylat. Geeignete Schutzgruppen umfassen Benzoyl und Acetyl. Geeignete Halogenide umfassen Brom und Chlor. Geeignete inerte Lösungsmittel umfassen tert.-Butylalkohol, Acetonitril, Dichlormethan, Dichlorethan, tert.-Amylalkohol, Tetrahydrofuran, oder Mischungen davon, sind jedoch nicht auf diese beschränkt. Ein bevorzugtes Lösungsmittel umfasst eine Mischung aus Acetonitril, tert.-Butanol und 1,2-Dichlorethan. Calciumhydrid kann gegebenenfalls der Reaktionsmischung zugefügt werden. Der zweite Schritt umfasst eine Behandlung des 2,6-Dichlorpurinnucleosidderivtes mit Bedingungen, die eine Schutzgruppenentfernung und eine nucleophile, aromatische Substitutionsreaktion bewirken, z.B. Natriumhydroxid und ein C1-C8-Alkohol oder Natrium-C1-C8-Alkoholat in dem entsprechenden C1-C8-Alkohol (z.B. Methanol mit Natriummethanolat, Ethanol mit Natriumethanolat etc.) oder einem anderen geeigneten, nicht-alkoholischen Lösungsmittel, um die gewünschte C1-C8-6-Alkoxy-Purin-Nucleosid-Verbindung der Formel III bereitzustellen.
- Die Verbindungen der Formel I, in denen X = Sulfinyl oder Sulfonyl ist, können durch Oxidation der entsprechenden Verbindungen gemäß Formel I, in denen X = Schwefel ist, hergestellt werden, z.B. mit H2O2/Essigsäure oder durch Verwendung geeigneter Ausgangsverbindungen der Formel II oder IV.
- Salze der Verbindungen gemäß der allgemeinen Formel I werden hergestellt, indem die freie Säure mit Alkali- oder Erdalkalihydroxiden, -alkholaten oder -acetaten umgesetzt wird.
- Die „Enantiomere" der Lipidteile der Verbindungen gemäß Formel I können durch Trennung der diastereomeren Salze oder durch enantioselektive Synthese der Lipidreste ausgehend von optisch aktiven C3-Vorläufern der Formel II hergestellt werden.
- Die Arzneimittel enthaltend Verbindungen der Formel I zur Behandlung von Krebs können in flüssiger oder fester Form auf oralem oder parenteralem Weg verabreicht werden. Gebräuchliche Darreichungsformen sind möglich, wie Tabletten, Kapseln, beschichtete Tabletten, Sirupe, Lösungen oder Suspensionen.
- Vorzugsweise wird Wasser als Injektionsmedium verwendet, enthaltend Additive wie Stabilisatoren, Lösungsvermittler und Puffer die in Injektionslösungen üblich sind. Solche Additive sind z.B. Weinsäure und Zitronensäurepuffer, Ethanol, Komplexbildner wie Ethylendiamintetraessigsäure und dessen nicht-toxische Salze, hochmolekulare Polymere wie flüssiges Polyethylenoxid zur Viskositätseinstellung. Flüssige Träger für Injektionslösungen müssen steril sein und werden vorzugsweise in Ampullen gefüllt.
- Feste Träger sind zum Beispiel Stärke, Laktose, Mannitol, Methylzellulose, Talkum, hochdisperse Kieselsäure, hochmolekulare Fettsäuren wie Stearinsäure, Gelatine, Agar-Agar, Calciumphosphat, Magnesiumstearat, tierische und pflanzliche Fette, feste hochmolekulare Polymere wie Polyethylenglykol etc. Gewünschtenfalls können Formulierungen für die orale Applikation Geschmacksstoffe oder Süßungsmittel enthalten.
- Die Dosierung kann von vielen Faktoren abhängen, wie Darreichungsform, Art, Alter oder individueller Konstitution.
- Die Verbindungen gemäß der Erfindung können geeigneterweise oral oder intravenös (i.v.) in Mengen im Bereich von 0,1–100 mg, vorzugsweise im Bereich von 0,2–80 mg pro kg Körpergewicht und Tag verabreicht werden. In einigen Dosierungsbereichen wird die Tagesdosis in 2–5 Anwendungen aufgeteilt, wobei Tabletten mit einem Wirkstoffgehalt im Bereich von 0,5–500 mg bei jeder Anwendung verabreicht werden.
- Analog können die Tabletten eine verzögerte Freisetzung aufweisen, wodurch die Anzahl der Anwendungen auf z.B. 1–3 pro Tage reduziert wird. Der Wirkstoffgehalt von Retard-Tabletten kann im Bereich von 2–1000 mg liegen. Der Wirkstoff kann auch durch i.v. Bolusinjektion oder eine kontinuierliche Infusion verabreicht werden, wobei Mengen im bereich von 5–1000 mg pro Tag normalerweise ausreichen.

- Herstellung von 2-Chlor-6-methoxy-9-(2'-desoxy-2'-fluor-β-D-arabinofuranosyl)-9H-purin

- In einen 1000 ml Kolben wurden 2,6-Dichlorpurin (12,65 g, 66,9 mmol), Calciumhydrid (2,43 g, 57,7 mmol) und Acetonitril (150 ml) gegeben und mit Rühren begonnen. Eine Lösung von Kalium-tert.-Butanolat (60,6 ml, 60,6 mmol, 1,0 M in tert.-Butanol) wurde innerhalb von 5 min. zugefügt, wodurch eine viskose aber rührbare Suspension erhalten wurde. Eine Lösung von 3,5-O-Dibenzoyl-2-desoxy-2-fluor-α-D-arabinofuranosylbromid (26,88 g, 63,5 mmol) in 1,2-Dichlorethan (200 ml) wurde innerhalb 45 min. bei Umgebungstemperatur zugefügt. Nachdem die Zugabe vollständig erfolgt war, wurde die Mischung 16 Stunden bei Umgebungstemperatur gerührt. Die Mischung wurde durch Celite gefiltert und Kolben sowie Feststoffe mit Acetonitril (100 ml) gewaschen. Die flüchtigen Bestandteile wurden mittels Rotationsverdampfer entfernt wobei eine gelbe gummiartige Masse (38,1 g) erhalten wurde. Essigester (100 ml) wurde zugefügt, der pH kontrolliert und mit 8 festgestellt. Essigsäure (0,5 ml) wurde zugefügt und der pH erneut kontrolliert und mit 4 festgestellt. Die trübe Lösung wurde durch Whatman 541 Filterpapier filtriert. Kolben und Filter wurden mit Essigester (100 ml) gewaschen. Es konnte keine Klärung der Lösung beobachtet werden. Die organische Phase wurde mit Wasser (100 ml) und Salzlösung (100 ml) gewaschen. Die organische Phase wurde getrocknet (MgSO4) und mit einem Rotationsverdampfer und anschließend einer Hochvakuumpumpe eingeengt, wobei ein weißer Schaum (34,0 g) erhalten wurde. Das Rohmaterial wurde mittels Säulenchromatographie (Silicagel 60, 230–400 mesh, 14 cm Durchmesser, 14,5 cm Höhe, 2232 ml) gereinigt. Eine Gradienten-Elution mit Hexan/Essigester wurde durchgeführt und die Fraktionen, welche das reinste Produkt enthielten, mittels Rotationsverdampfer eingeengt, zweimal in Methanol aufgeschlämmt, filtriert und mit Methanol gewaschen, wobei ein weißer Feststoff (13,4 g, 92,6% AUC) erhalten wurde. Die weniger reinen Fraktionen wurden vereinigt, mittels Rotationsverdampfer eingeengt und mittels Säulenchromatographie weitergereinigt, wobei ein weißer Feststoff (3,85 g, 94,7% AUC) erhalten wurde. Die Gesamtausbeute betrug 17,3 g (56%). Ein Teil wurde zur Charakterisierung in Methanol resuspendiert (97,9% AUC). - Ein 500 ml Kolben wurde mit geschütztem 2,6-Dichlor-9-α-D-(3',5'-O-dibenzoyl-2'-desoxy-2'-fluor-β-D-arabinofuranosyl)-9H-purin (13,33 g, 25,1 mmol) und Methanol (300 ml) befüllt. Der pH wurde mit einer NaOH-Lösung (2 ml, 1,0 N in H2O) auf 9,5 eingestellt. Die Suspension wurde bei Umgebungstemperatur 16,5 Stunden gerührt. Der pH wurde geprüft und betrug 5,5. Weitere NaOH-Lösung (11,3 ml) wurde zugefügt (pH = 11) und die Mischung bei Umgebungstemperatur 1,5 Stunden gerührt. Der pH wurde geprüft und betrug 6. Ein Dünnschichtchromatogramm (10% EtOH/90% CH2Cl2, UV254) zeigte 3 Flecken bei Rf 0,28, 0,72 und 0,88. Weitere NaOH-Lösung (13,3 ml) wurde zugefügt (pH = 11). Nach weiteren 5 min. Rühren bei Umgebungstemperatur wurde die Reaktionsmischung eine klare farblose Lösung und nach einem weiteren Rühren für 2,5 Stunden wurde die Reaktion gemäß Dünnschichtchromatogramm als vollständig angesehen. Essigsäure (0,8 ml) wurde zur Neutralisierung der Base zugefügt (pH = 5). Rotationsverdampfung ergab einen zweiphasigen Rückstand. Isopropylalkohol (100 ml) wurde zugefügt, wobei eine weiße Suspension erhalten wurde. Wasser wurde mittels azeotroper Rotationsverdampfung entfernt. Dieses Verfahren wurde zweimal mit weiterem Isopropylalkohol (100 ml) wiederholt. Die Rotationsverdampfung wurde gestoppt als etwa 50 ml im Kolben verblieben und die Suspension wurde filtriert und Kolben sowie Filterkuchen mit dem Filtrat und dann mit Isopropylalkohol (10 ml) gewaschen. Der Feststoff wurde getrocknet (50°C, 27 Torr, 16,5 h). Das Gewicht an Feststoff betrug 5,58 g (92,4% AUC). Das Filtrat wurde mittels Rotationsverdampfer und Hochvakuumpumpe eingeengt. Das Gewicht des Rückstandes betrug 6,79 g (70,9% AUC). Beide Feststoffe wurden separat durch Säulenchromatographie gereinigt (Silicagel 60, 230–400 mesh, 10% Ethanol, 90% Dichlormethan). Das Gewicht des gereinigten Materials des rohen Feststoffs betrug 4,62 g (95,5% AUC). Das Gewicht des gereinigten Materials des Rückstandes war 1,69 g (98,1% AUC). Die Gesamt ausbeute betrug 6,31 g (79%). - Herstellung von [2-Chlor-9-(2'-desoxy-2'-fluor-β-D-arabinofuranosyl)adenin]-5'-phosphorsäure-(3-dodecylmercapto-2-decyloxy)propylester
-
- 3,12 g Phosphorsäure-(3-dodecylmercapto-2-decyloxy)propylester wurden zweimal mit 60 ml wasserfreiem Pyridin behandelt und durch Abdamfen konzentriert. Der Rückstand wurde bei Raumtemperatur in 60 ml wasserfreiem Pyridin aufgenommen, mit 3,80 g 2,4,6-Triisopropylbenzolsulfonylchlorid (Trisylchorid) in Stickstoffatmosphäre behandelt und bei 20°C 2 Stunden gerührt. Dann wurden 2,00 g 2-Chlor-9-(2'-desoxy-2'-fluor-β-D-arabinofuranosyl)-6-methoxy-9H-purin auf einmal zugefügt und der Ansatz 16 Stunden in Stickstoffatmosphäre gerührt. Die Hydrolyse wurde durch Zugabe von 10 ml Wasser durchgeführt, die Mischung weitere 0,5 Stunden bei Raumtemperatur gerührt, im Vakuum vom Lösungsmittel befreit, und zweimal mit 20 ml Toluol gestrippt. Der Rückstand wurde in t-Butylmethylether (160 ml) bei 40°C 0,5 Stunden gerührt. Nach Abkühlen auf Raumtemperatur wurde das Pyridinsulfonsäuresalz abfiltriert. Das Filtrat wurde zweimal mit 40 ml 2N Salzsäure gewaschen und zur Trockne eingedampft. Die verbleibenden 7,38 g sirupöses Material werden ohne weitere Reinigung im nächsten Schritt eingesetzt.
- Eine Probe des obigen Rohmaterials wurde mittels Säulenchromatographie an Lichrospher 60 RPSelect B mit Methanol/wässrigem 40 mM Natriumacetat 90:10 als Elutionsmittel gereinigt. Die Produkt-haltigen Fraktionen wurden eingedampft und der Rückstand zwischen 50 ml tert.-Butylmethylether und 10 ml 2N Salzsäure verteilt. Die organische Phase wurde eingedampft und der Rückstand in einer Mischung aus 5 ml Toluol und 5 ml Methanol aufgenommen. Der pH wurde durch Zugabe von Natriummethanolat auf pH 7 eingestellt. Das Lösungsmittel wurde abgestrippt und der Rückstand im Vakuum getrocknet. Das Natriumsalz von [2-Chlor-9-(2'-fluor-β-D-arabinofuranosyl)-6-methoxy-9H-purin]-5'-phosphorsäure-(3-dodecylmercapto-2-decyloxy)propylester wurde als amorpher Feststoff erhalten, der bei 65–75°C schmilzt und eine spezifische Drehung von [α]20 Hg436 = + 31,9 (c = 1,0 in Methanol) zeigt.

- Der Aminolyse-Schritt wurde in einem Edelstahlreaktor bei 80°C durchgeführt.
- Das oben genannte Rohmaterial (7,38 g) wurde in 30 ml 7M NH3 in Ethanol (gesättigt bei –5°C) gelöst. Nach 20 Stunden erhitzen konnte kein Edukt [2-Chlor-9-(2'-fluor-β-D-arabinofuranosyl)-6-methoxy-9H-purin]-5'-phosphorsäure-(3-dodecylmercapto-2-decyloxy)propylester-Methoxyderivat nachgewiesen werden. Das Produkt wurde mittels Säulenchromatographie an Lichrospher 60 RPSelect B mit Methanol/wässrigem 40 mM Natriumacetat 85:15 als Elutionsmittel gereinigt. Die Produkt-haltigen Fraktionen werden eingedampft. Der Rückstand wird zwischen 100 ml tert.-Butylmethylether und 50 ml 2N Salzsäure verteilt. Die organische Phase wird eingedampft, der Rückstand in einer Mischung aus 30 ml Methanol aufgenommen und der pH durch Zugabe von Natriummethanolat (30% in Methanol) auf pH 7 eingestellt. Das Lösungsmittel wird abgestrippt und das zurückbleibende Natriumsalz im Vakuum getrocknet. Das Produkt (2,90 g) wird mit einer Gesamtausbeute von 57% bezogen auf die Umsetzung des 2-Chlor-9-(2'-desoxy-2'-fluor-β-D-arabinofuranosyl)-6-methoxy-9H-purin erhalten. Die mittels HPLC ermittelte Reinheit betrug 93,6 Flächen-%. Schmelzpunkt: 130–131°C. MS (FAB–): m/z = 780 [M – Na+], UV (Methanol) λmax 263,4 nm.

- Herstellung von [2-Chlor-9-(2'-desoxy-2'-fluor-β-D-arabinofuranosyl)adenin]-5'-phosphorsäure-(3-dodecylmercapto-2-decyloxy)propylester aus 2-Chlor-9-(2'-desoxy-2'-fluorarabinofuranosyl)adenin
- 0,91 g Phosphorsäure-(3-dodecylmercapto-2-decyloxy)propylester werden zweimal mit 20 ml wasserfreiem Pyridin behandelt und durch Abdampfen konzentriert. Der Rückstand wird bei Raumtemperatur in 20 ml wasserfreiem Pyridin aufgenommen, mit 1,07 g 2,4,6-Triisopropylbenzolsulfonsäurechlorid unter Stickstoff behandelt und bei 25°C 0,5 Stunden gerührt. Dann werden 0,5 g 2-Chlor-9-(2'-desoxy-2'-fluorarabinofuranosyl)adenin auf einmal zugefügt, und der Ansatz unter Stickstoff 20 Stunden stehen gelassen. Die Hydrolyse wird durch Zugabe von 5 ml Wasser durchgeführt, die Mischung für weitere 0,5 Stunden bei Raumtempeartur gerührt, unter Vakuum vom Lösungsmittel befreit und zweimal unter Verwendung von 50 ml Toluol gestrippt. Der Rückstand wird mittels Säulenchromatographie an Lichrospher 60 RPSelect B mit Methanol/wässrigem 40 mM Natriumacetat 88:12 als Elutionsmittel gereinigt. Die Produkt-haltigen Fraktionen werden eingedampft. Der Rückstand wird zwischen 50 ml tert.-Butylmethylether und 10 ml 2N Salzsäure verteilt. Die organische Phase wird eingedampft. Der Rückstand wird in einer Mischung aus 5 ml Toluol und 5 ml Methanol aufgenommen. Der pH wird durch Zugabe von Natriummethanolat auf pH 7 eingestellt. Das Lösungsmittel wird abgestrippt und der Rückstand im Vakuum getrocknet.
Die Ausbeute sind 0,82 g (62%) weißes Pulver. - Der Phosphorsäure-(3-dodecylmercapto-2-decyloxy)propylester wird wie in WO 92/03462 beschrieben hergestellt.
- Beispiel 4
- Herstellung von [2-Chlor-9-(2'-desoxy-2'-fluor-β-D-arabinofuranosyl)adenin]-5'-diphosphorsäure, (3-dodecylmercapto-2-decyloxy)propylester
-
- Ein Kolben wird mit 2-Chlor-6-methoxy-9-(2'-desoxy-2'-fluor-β-D-arabinofuranosyl)purin und Triethylphosphat (z.B. 2,3 ml/mmol Nucleosid) unter Stickstoff befällt. Die resultierende Mischung wird gekühlt (z.B. –25°C) und dann POCl3 (z.B. 3 eq.) zugefügt. Nach Erwärmung auf Umgebungstemperatur wird die Mischung gerührt (z.B. 3 h). Eis (z.B. 1,4 g/mmol Nucleosid) und Wasser (z.B. 8,7 ml/mol Nucleosid) werden dann unter Rühren zugegeben und die Mischung in einen Scheidetrichter überführt. MTBE (z.B. 4,4 ml/mol Nucleosid) werden zugegeben und die Phasen nach dem Schütteln getrennt. Die organische Phase wird zweimal mit Wasser (z.B. 8,7 ml/mmol Nucleosid) gewaschen. Die vereinigten wässrigen Extrakte werden mit NaOH (z.B. 50% aq.) auf etwa pH 2 angesäuert und dann eine ausreichende Zeit (z.B. 2 h) mit Aktivkohle (z.B. 5,7 g/mmol Nucleosid) gerührt. Die Mischung wird filtriert und das Filtrat verworfen. Die Aktivkohle wird mit einer Mischung aus MeOH (z.B. 4,4 ml/mmol Nucleosid), Ammoniumhydroxid (konz.) (z.B. 44 ml/mmol Nucleosid) und Wasser (z.B. 3,9 ml/mmol Nucleosid) für eine ausreichende Zeit (z.B. 30 min) gerührt und filtriert. Der Vorgang wird wiederholt (z.B. 5 mal) und die Filtrate werden vereinigt. Eindampfen der vereinigten Filtrate ergibt rohes 2-Chlor-6-methoxy-9-(2'-desoxy-2'-fluor-5'-O-phosphat-β-D-arabinofuranosyl)-9H-purin. Dieses wird in Wasser gelöst (z.B. 8,7 ml/mmol Nucleosid) und eine ausreichende Zeit (z.B. 30 min) mit Dowex 50WX8-100 kationischem Harz (z.B. 4 g/mmol Nucleosid) unter Rühren behandelt. Die Mischung wird filtriert und das Harz mit Wasser gerührt (z.B. 9 ml/mmol Nucleosid) und filtriert. Das Harz wird mit Wasser extrahiert (z.B. viermal) und die vereinigten Wasser-Filtrate werden eingedampft, wobei 2-Chlor-6-methoxy-9-(2'-desoxy-2'-fluor-β-D-arabinofuranosyl)-9H-purin (z.B. in 30–100% Ausbeute) erhalten wird.

- 2-Chlor-6-methoxy-9-(2'-desoxy-2'-fluor-β-D-arabinofuranosyl)purin wird in wasserfreiem Ethanol in einem Druckbehälter gelöst und unter Stickstoffamosphäre gekühlt (z.B. –5°C). Ammoniak wird in die Lösung eingeleitet bis eine gesättigte Lösung erreicht ist. Das System wird dann eine ausreichende Zeit (z.B. >20 h) erhitzt (z.B. auf 80°C). Der Fortschritt der Reaktion wird durch Probennahme und HPLC Analyse verfolgt. Sobald die Reaktion vollständig ist, wird das Lösungsmittel abgedampft, wobei rohes 2-Chlor-9-(2'-desoxy-2'-fluor-5'-O-phosphat-β-D-arabinofuranosyl)adenin erhalten wird.
- Der dritte Schritt ist die Herstellung von (2-Chlor-9-(2'-desoxy-2'-fluor-β-D-arabinofuranosyl)adenin-5'-diphosphorsäure-(3-dodecylmercapto-2-decyloxy)propylester durch Umsetzung des Morpholinsalzes von Phosphorsäure-mono-(3-dodecylmercapto-2-decyloxy)-1-propylester mit 2-Chlor-9-(2'-desoxy-2'-fluor-5'-O-phosphat-β-D-arabinofuranosyl)adenin:
- Das Morpholinsalz von Phosphorsäure-mono(3-dodecylmercapto-2-decyloxy)-1-propylester wird in Analogie zu Bioorg. Med. Chem., 7, 1195–1200, (1999) hergestellt, wobei Phosphorsäure-mono(3-dodecylmercapto-2-decyloxy)-1-propylester und Morphilin in einer Mischung aus Wasser und tert-Butanol (z.B. 1:1 bezogen auf das Volumen) gelöst werden. Dicylohexylcarbodiimide (DCC) in tert-Butanol wird der Lösung zugefügt (z.B. etwa 4 molarer Überschuss an DCC bezogen auf Phosphorsäure-mono(3-dodecylmercapto-2-decyloxy)-1-propylester) und die Reaktion am Rückfluss gehalten (z.B. 3,5 h). Das Volumen wird durch Abdampfen reduziert und die Mischung gekühlt wodurch das Phosphomorpholinsalz ausgefällt wird.
- Das Phosphomorpholinsalz (z.B. 1,13 mol pro mol Adenosinderivat) wird als ein wasserfreies Pyridin (z.B. 23 ml/mmol Adenosinderivat) hergestellt und 2-Chlor-9-(2'-desoxy-2'-fluor-5'-O-phosphat-β-D-arabinofuranosyl)adenin unter Rühren zugefügt, alles unter Stickstoff. Die Mischung wird gerührt (z.B. bei 40°C für mindestens 16 h) und dann Wasser (z.B. 4,5 ml/mmol Adenosinderivat) zugefügt und weiter gerührt (z.B. 1 h). Das Lösungsmittel wird abgedampft und das Produkt chromatographiert (z.B. Silicagel, Elution mit einer Mischung aus CHCl3, MeOH und NH4OH (aq)) wobei (2-Chlor-9-(2'-desoxy-2'-fluor-β-D-arabinofuranosyl)adenin-5'-diphosphorsäure-[3-dodecylmercapto-2-decyloxy)propylester als weißer Feststoff (z.B. mit 20–100% Ausbeute) erhalten wird.

- Tabletten Formulierung

- 1,50 kg [2-Chlor-9-(2'-desoxy-2'-fluor-β-D-arabinofuranosyl)adenin]-5'-phosphorsäure-(3-dodecylmercapto-2-decyloxy)propylester-Natriumsalz,
- 1,42 kg mikrokristalline Cellulose,
- 1,84 kg Laktose,
- 0,04 kg Polyvinylpyrrolidon und
- 0,20 kg Magnesiumstearat wurden in trockener Form gemischt, mit Wasser angefeuchtet und granuliert. Nach der Trocknung wurde das Material zu Tabletten mit 500 mg Gewicht verpresst.
- Beispiel 6
- Formulierung zur Injektion
- 10,0 g kg [2-Chlor-9-(2'-desoxy-2'-fluor-β-D-arabinofuranosyl)adenin]-5'-phosphorsäure-(3-dodecylmercapto-2-decyloxy)propylester-Natriumsalz wurden in 500 ml physiologischer Kochsalzlösung gelöst, jeweils 5 ml in Ampullen gefüllt und sterilisiert. Die Lösung kann durch intravenöse Injektion appliziert werden.
- Beispiel 7
- Antitumorale Wirksamkeit von [2-Chlor-9-(2'desoxy-2'-fluor-β-D-arabinofuranosyl)adenin]-5'-phosphorsäure-(3-dodecylmercapto-2-decyloxy)propylester ("Nukcleotidkonjugat") und 2-Chlor-9-(2'-desoxy-2'-fluor-β-D-arabinofuranosyl)adenin ("Nucleosid") in einem humanen Darmkarzinom Xenograft-Modell (HCT-15) in vivo
- Die Antitumorwirkung des Nucleotidkonjugats und des entsprechenden Nucleosids wurde in dem humanen Darmkarzinom Xenograft Modell HCT-15 an Nacktmäusen verglichen.
- Mäuse mit Tumoren wurden an Tag 7 nach der Inokulation mit HCT-15-Tumorzellen randomisiert und in Behandlungsgruppen mit 9 Tieren pro Gruppe aufgeteilt. Die Behandlung wurde an Tag 8 begonnen. Die Tiere wurden intraperitoneal (ip) einmal am Tag an 5 aufeinanderfolgenden Tagen mit dem Nucleotidkonjugat oder dem Nucleosid behandelt. Die Dosierung beinhaltete 50% und 25% der maximal tolerierten Dosis (MTD). Zur Kontrolle wurde Tieren das entsprechende Lösungsmittel (Träger 1 oder 2) injiziert. Am 28. Tag wurden die Primärtumoren entnommen und die Tumorgewichte bestimmt. Der Median der Tumorgewichte ist in Tabelle 1 gezeigt. Tabelle 1

- Die Antitumorwirkung des Nucleotidkonjugats war bei beiden Dosierungen signifikant (p < 0,01) stärker als die des entsprechenden Nucleosids.
Claims (8)
- Nucleotid-derivate der Formel I wobei R1 ausgewählt ist unter geradkettigen oder verzweigten, gesättigten oder ungesättigten Alkylketten mit 1–20 Kohlenstoffatomen, die unsubstituiert sind oder zumindest einfach substituiert sind mit einem Halogenatom, einer C1-C6 Alkoxy-, C1-C6 Alkylmercapto-, C1-C6 Alkoxycarbonyl-, C1-C6 Alkylsulfinyl- oder C1-C6 Alkylsulfonylgruppe; R2 ausgewählt ist unter Wasserstoff, einer geradkettigen oder verzweigten, gesättigten oder ungesättigten Alkylkette mit 1–20 Kohlenstoffatomen, die unsubstituiert ist oder zumindest einfach substituiert ist mit einem Halogenatom, einer C1-C6 Alkoxy-, C1-C6 Alkylmercapto-, C1-C6 Alkoxycarbonyl- oder C1-C6 Alkylsulfonylgruppe; R3 Amino ist; X ausgewählt ist unter einem Schwefelatom, einer Sulfinylgruppe und einer Sulfonylgruppe; Y Sauerstoff ist; wobei, wenn R3 Amino ist, die Aminogruppe unsubstituiert ist oder durch eine bekannte Aminoschutzgruppe substituiert sein kann, ihre Tautomere, optisch aktiven Formen und racemischen Mischungen und ihre physiologisch verträglichen Salze mit anorganischen und organischen Säuren oder Basen.
- Nucleotid-derivate gemäß Anspruch 1, wobei R1 eine geradkettige C8-C15 Alkylgruppe ist, die unsubstituiert ist oder durch eine C1-C6 Alkoxy- oder eine C1-C6 Alkylmercaptogruppe substituiert ist.
- Nucleotid-derivate gemäß Anspruch 1, wobei R2 eine geradkettige C8-C15 Alkylgruppe darstellt, die unsubstituiert ist oder durch eine C1-C6 Alkoxy- oder eine C1-C6 Alkylmercaptogruppe substituiert ist.
- Pharmazeutische Zusammensetzung umfassend mindestens eine Verbindung gemäß einem der Ansprüche 1 bis 4 in Kombination mit einem pharmazeutisch verträglichen Zusatz oder Träger.
- Verwendung einer Verbindung gemäß einem der Ansprüche 1 bis 4 zur Herstellung von Arzneimitteln zur Behandlung von bösartigen Tumoren, wobei die Verbindung in einer Menge, die zur Behandlung des Tumors wirksam ist, an einen Patienten, der eine solche Behandlung benötigt, verabreicht werden soll.
- Verwendung gemäß Anspruch 6, wobei der Tumor ausgewählt ist unter Karzinomen, Sarkomen und Leukämie.
- Verwendung gemäß Anspruch 6 oder 7 in fester oder freier Kombination mit anderen Antitumormitteln.
