Verfahren zur Darstellung von Estern in der allphatischen Seitenkette
jodierter aromatischer Alkyloxy- und Alkylaminocarbonsäuren. Es wurde gefunden,
daß die bis-her unbekannten in der aliphatischen Seitenkette, jodierten Ester aromatischer
Alkyloxy- oder Alkylaminocarbonsäuren der allgemeinen Formeln
(R = Alkylrest, R, = Alkyl) eine hervorragende therapeutische Wirkung
bei Kreislaufstörungen entfalten. Man gelangt zu den neuen Verbindungen, g i n dem
man aromatische j,odalkyloxy- oder jodalkylaminocarbonsäuren verestert. Man kann
auch von den in der Seitenkette hydroxylierten, Estern, d.h. der Oxalkyloxy- und
Oxalkylaminosubstitutionsprodukte aromatischer Carbonsäurealkylester, ausgehen und
diese Oxy-,ester entweder unmittelbar in die entsprechenden jodester überführen,
oder man kann aus ihnen zunächst die entsprechenden Chlor-oder Bromverbindungen
herstellen und diese ihrerseits mit jodsalzen umsetzen. Gegenüber den entsprechenden,
teilweise bereits bekannten Bromverbindungen zeichnen sich die neuen Verbindungen
durch ganz wesentlich erhöhte Wirksamkeit aus, die sehr überraschend ist, insbesondere
auch mit Rücksicht darauf, daß die den jodhaltigen Estern zugrunde liegenden jodalkylierten
Säuren keine analoge physIologische Wirkung entfalten. B ei sp i e 1 e. i.
p-j#odäthoxybenzoesäurem,ethylestier: In eine Suspension von io Teilen _p-Jodäthoxybenzoesäure
in 4o Teilen. Methylalkohol wird in der Kälte Salzsäuregas bis zur Sättigung eingeleitet
und 12 Stunden stehen gelassen. Nach dem Abdestillieren des größten Teiles des Methylalkohols
wird in überschüssige Sodalösung gegossen, wobei der jodester ausfällt. Die neue
Verbindung kristallisiert aus Methylalkohol in weißen Nadeln vom Schmelzpunkt 7o'.

182', liefert. Diese gibt mit Phosphorpentachlorid das p-Chloräthoxybenzoylchlorid,
Siedepunkt
i8o' unter io min Druck, Schmelzpunkt 4--0, das beim Verseifen die p-Chlorätho.xybenzoesäure,
Schmelzpunkt 256c', liefert. Durch Ersatz des Chloratorns durch Jod mittels jogäÜenl-'erhält

dem man etwas Bisulfit zugesetzt hat. Hierbei fällt die neuc Vei#bindung aus, die,
aus Alkohol umgelöst, weiße Nadeln vom Schmelzpunkt 77 bis, 78"
bildet.
Den p-Chloräthoxybemo,esäurehenzylester, Schmelzpunkt 62#, kann man durch Einwirkung
von Benzylalk-ohol auf das in Beispiel i beschriebene p-Chloräthoxyb=oylchlorid

das p-i3-Chlorpropyloxybenzoylchl#orid, das durch Einwirkung von Methylalkohol in
