Verfahren zur Darstellung von Estern von 2-Phenytchino ! inearbonsäuren- (4).
Die harnsäureausscheidende Wirkung der 2-Phenylchinolincarbonsäuren- (4) und ihrer Derivate ist bekannt und führte zu deren Verwendung als Heilmittel. Auch die Ester dieser Säuren (s. z. B. deutsche Patentschriften Nr. 275963,281136, 287959, 520155) sind hiezu mit wechselndem Erfolg herangezogen worden, wobei man gleichzeitig auf Erreichung guter Resorbierbarkeit, möglichst weitgehender Ungiftigkeit und einer Wirkung hinsichtlich der Steigerung der Harnsäureausscheidung bemüht war.
Es wurde nun gefunden, dass man durch Veresterung der 2-Phenylchinolincarbonsäure- (4) oder deren Derivaten mit Glycerin oder mit Glycerinhalogenhydrinen zu bisher nicht beschriebenen Verbindungen gelangt, welche unmittelbar therapeutisch anwendbar sind, aber auch als Zwischenprodukte zur Darstellung anderer, gleichfalls therapeutisch brauchbarer Stoffe Anwendung finden können.
Weiters wurde gefunden, dass sich die durch Umsetzung von 2-Phenylchinolincarbonsäure- (4) bzw. deren Derivaten mit Glycerinhalogenhydrinen erhältlichen halogenhaltigen Ester mit Alkalisalzen von 2-Phenylchinolincarbonsäuren- (4) zu mehrere Säurereste enthaltenden halogenfreien Glycerinestern umsetzen lassen, die für die gleichen Zwecke verwendbar sind.
Die nach vorliegendem Verfahren erhältlichen Verbindungen unterscheiden sich von bereits bekannten Estern der 2-Phenylchinolincarbonsäure-(4) teils durch bessere Wirksamkeit hinsichtlich der Beeinflussung der Harnsäureausscheidung aus dem tierischen und menschlichen Organismus, teils durch bessere Resorbierbarkeit und grössere Ungiftigkeit.
Beispiel 1 : 5 Gewichtsteile 2-Phenylchinolincarbonsäure- (4), 70 Gewichtsteile Glycerin und 16'6 Gewichtsteile Schwefelsäure (D = 1'84) werden einige Stunden auf dem Wasserbade erwärmt.
Dann wird alkalisch gemacht, der abgeschiedene Niederschlag abgesaugt, gewaschen und getrocknet.
Der so erhaltenen α-Mono-[2-phenylchinolincarbonsäure-(4)]-glycerinester kann durch Überführung in das Hydrochlorid gereinigt werden ; aus Benzol umkristallisiert bildet er weisse Nadeln vom F = 99 , die in den gebräuchlichen Lösungsmitteln, Alkohol, Äther und Benzol löslich sind.
Zu dem gleichen Ester gelangt man durch Umsetzung des Chlorids der 2-Phenylchinolincarbon-

Beispiel 2 : 30 Gewichtsteile Kaliumsalz der 2-phenylchinolincarbonsäure-(4) und 6'7 Gewichtsteile 1. 3-Dichlorpropanol-(2) [symm. Glycerindichlorhydrin] werden 10-20 Stunden auf 160-170 erhitzt. Dann wird mit Natriumcarbonat verrieben, mit Wasser gewaschen, mit Äther extrahiert und der Extraktionsrückstand im Vakuum getrocknet. Zwecks weiterer Reinigung wird in Olivenöl gelöst und mit Petroläther gefällt. Der so erhaltene a. α'-Bis-[2-phenylchinolincarbonsäure-(4)]-glycerinester stellt ein in Methyl-oder Äthylalkohol leicht lösliches weisses Pulver dar, das bei 580 schmilzt.
Zu dem gleichen Ester gelangt man auch durch Umsetzung des im Beispiel 4 beschriebenen 1-[2'-Phenylchinolincarbonsäure-(4')]-3-chlorpropandiol-(1.2)-esters mit dem Kaliumsalz der 2-Phenyl- chinolincarbonsäure- (4) oder durch Erhitzen von 2 Mol des Säurechlorids der letztgenannten Säure

Beispiel 3 ; 3'6 Gewichtsteile des im Beispiel 4 beschriebenen 2-[2'-PhenylchinoIinearb & n- säure- (4')]-1. 3-dichlorpropanol- (2)-esters und 5'8 Gewichtsteile Kaliumsalz der 2-Phenylchinolin- carbonsäure- (4) werden 10-20 Stunden auf 160-1700 erhitzt. Dann wird mit Natriumcarbonat verrieben, mit Wasser gewaschen, mit Äther extrahiert und der Extraktionsrückstand im Vakuum getrocknet. Zwecks Reinigung wird aus Amylalkohol und aus Benzol umkristallisiert. Der so erhaltene α.ss.#-Tri-[2-phenylchinolincarbonsäure-(4)]-glycerinester schmilzt bei 1350.
Beispiel4 : 9#5 Gewichtsteile des Chlorids der 2-Phenylchinolincarbonsäure-(4) und 3#5 Gewichts- teile 3-Chlorpropandiol- (1. 2) werden in ungefähr 70 Gewichtsteilen Benzol aufgeschlemmt und bis zur Beendigung der Salzsäureentwicklung auf dem Wasserbade erhitzt. Dann wird das Lösungsmittel abdestilliert, der Rückstand mit Natriumcarbonat verrieben, mit Wasser gewaschen, in Äther gelöst und nach Filtration der Äther entfernt. Der Rückstand liefert aus Alkohol umkristallisiert den 1-[2'-Phenylchinolincarbonsäure-(4')]-3-chlorpropandiol-(1.2)-ester in farblosen Nadeln vom F =109 .
Zu dem gleichen chlorhaltigen Ester kann man auch gelangen, wenn man 2-Phenylchinolincarbon- säure- (4) und Glycerin-α-monochlorhydrin bei Gegenwart von Schwefelsäure in bekannter Weise verestert oder wenn man das Kaliumsalz der 2-Phenylchino% carbonsäure- (4) und symm. Glycerindichlorhydrin in monomolekularem Verhältnis miteinander zur Reaktion bringt.
Die Verwendung von 1. 3-Dichlorpropanol- (2) führt zum 2-[2'-Phenylchinolincarbonsäure-(4')]- 1. 3-dichlorpropanol- (2)-ester, der aus Alkohol umkristallisiert in farblosen, monoklinen Kristallen erhalten wird, die bei 960 schmelzen.
Die Einwirkung von 1 Mol 3-Chlorpropandiol- (1. 2) auf 2 Mol des Säurechlorids ergibt den

1. Verfahren zur Darstellung von Estern von 2-Phenylchinolincarbonsäuren-(4) durch Veresterung dieser Säuren mit aliphatischen Alkoholen nach üblichen Methoden, dadurch gekennzeichnet, dass man 2-Phenylehinolincarbonsäure- (4) oder deren Derivate mit Glycerin oder Glycerinhalogenhydrinen verestert.

The uric acid-releasing effect of 2-phenylquinolinecarboxylic acids- (4) and their derivatives is known and has led to their use as medicinal products. The esters of these acids (see e.g. German Patent Nos. 275963, 281136, 287959, 520155) have been used for this with varying degrees of success, while efforts have been made to achieve good absorbability, as far as possible non-toxicity and an effect in increasing uric acid excretion .
It has now been found that esterification of 2-phenylquinolinecarboxylic acid- (4) or its derivatives with glycerol or with glycerol halohydrins leads to compounds not previously described which are directly therapeutically applicable, but also as intermediates for the preparation of other, likewise therapeutically useful substances Can find application.
Furthermore, it has been found that the halogen-containing esters obtainable by reacting 2-phenylquinolinecarboxylic acid (4) or their derivatives with glycerol halohydrins can be converted with alkali salts of 2-phenylquinolinecarboxylic acids (4) to form several acid radicals containing halogen-free glycerol esters which are used for the same purposes are usable.
The compounds obtainable by the present process differ from the already known esters of 2-phenylquinolinecarboxylic acid- (4) partly by better effectiveness with regard to influencing uric acid excretion from the animal and human organism, partly by better absorbability and greater non-toxicity.
Example 1: 5 parts by weight of 2-phenylquinoline carboxylic acid (4), 70 parts by weight of glycerol and 16,6 parts by weight of sulfuric acid (D = 1'84) are heated for a few hours on the water bath.

The same ester is also obtained by reacting the 1- [2'-phenylquinolinecarboxylic acid (4 ')] -3-chloropropanediol (1.2) ester described in Example 4 with the potassium salt of the 2-phenylquinolinecarboxylic acid (4) or by heating 2 moles of the acid chloride of the latter acid

The residue gives the 1- [2'-phenylquinolinecarboxylic acid (4 ')] -3-chloropropanediol (1.2) ester recrystallized from alcohol in colorless needles with an F = 109.
The same chlorine-containing ester can also be obtained if 2-phenylquinolinecarboxylic acid- (4) and glycerol-α-monochlorohydrin are esterified in the presence of sulfuric acid in a known manner or if the potassium salt of the 2-phenylquinolinecarboxylic acid- (4 ) and symm. Glycerol dichlorohydrin reacts with one another in monomolecular ratio.
The use of 1. 3-dichloropropanol (2) leads to the 2- [2'-phenylquinoline carboxylic acid (4 ')] - 1. 3-dichloropropanol (2) ester, which is recrystallized from alcohol and obtained in colorless, monoclinic crystals that will melt at 960.
