Description
Process for the preparation of o-aminocarboxylic acids of the benzene series. A large number of o-aminocarboxylic acids of the benzene series is technically difficult accessible.
It has now been found that by oxidizing isatin, one's Substitution products or homologues by means of oxidizing agents in alkaline solution; z. B. with persalts, sodium peroxide, especially with hydrogen peroxide, smooth the corresponding o-aminocarboxylic acids are obtained. It arises z. B. from 7-methylisatin quantitative 1-methyl-2-aminobenzene-3-carboxylic acid.
It is true that one already has anthranilic acid through the oxidation of isatin with chromic acid obtained in glacial acetic acid solution; but isatoic acid is formed first, which then must be converted into anthranilic acid by splitting off carbonic acid. In contrast this is obtained directly by oxidation with alkaline oxidizing agents and quantitatively the o-aminocarboxylic acids. The process is particularly valuable where the corresponding isatins are easily accessible.
Beispiel 1.Example 1.
16,1 Gewichtsteile 7-Methylisatin werden in 15o Gewichtsteilen Wasser und 48 Gewichtsteilen Natronlauge von 40'B6 gelöst.16.1 parts by weight of 7-methylisatin are dissolved in 150 parts by weight of water and 48 parts by weight of 40'B6 sodium hydroxide solution.
In this solution are allowed at 1o to 15'7o parts by weight of hydrogen peroxide, which contains 5.48 parts by weight of hydrogen peroxide in Zoo parts by weight. The temperature is kept at 10 to 15 ° by throwing in ice. After three hours Stirring is filtered from a slight cloudiness and acidified with acetic acid. After standing overnight, 90 percent of theory is r-methyl-2-aminobenzene-3-carboxylic acid deposited. The rest can be obtained by shaking out with ether.
Beispiel 2.Example 2.
Der Rest kann noch durch Ausäthern der Mutterlauge gewonnen werden.16.1 parts by weight of 7-methylisatin are dissolved in 50 parts by weight of water and 34 parts by weight of 40 ° Be sodium hydroxide solution and then 15 parts by weight Sodium perborate, from the zoo parts by weight 3 parts by weight of hydrogen peroxide correspond, dissolved in zoo parts by weight of water, added. After four hours Stirring is acidified with 60 parts by volume hydrochloric acid and the weak mineral acid Reaction removed again by adding a little sodium acetate. It divorced 12.8 parts by weight of 1-methyl-2-amino-3-carboxylic acid. The rest can still get through Ethereal the mother liquor can be obtained.
Beispiel 3.Example 3.
Es scheiden sich 12,8 Gewichtsteile i-Methyl-q.-aminobenzo1-3-carbonsäure ab, die durch Absaugen gewonnen werden können.14.7 parts by weight isatin in 15o parts by weight of water and 48 parts by weight of 40 ° Be sodium hydroxide solution dissolved. 75 parts by weight are then left Flow in hydrogen peroxide of 5.48 percent, whereby one goes through cooling keeps the temperature between 10 and 15 °. After five hours of stirring, it is 65 parts by volume Acidified 5% acetic acid. It divorces; Parts by weight of anthranilic acid away. A large part of the acid still remains in solution. It can be obtained through etherification. Example 16.1 parts by weight of 5-methylisatin in 15o parts by weight of water and 48 parts by weight of sodium hydroxide solution of -.o ° Be dissolved and with cooling at 10 to 15 ° 70 parts by weight of hydrogen peroxide of 5.4.8 percent were added. After three hours Stirring is acidified with 65 parts by volume of 5% acetic acid and left overnight stand. 12.8 parts by weight of i-methyl-q.-aminobenzo1-3-carboxylic acid separate which can be obtained by suction.
Verwendet man statt 16,1 Gewichtsteilen 5-Methylisatin 24 Gewichtsteile 5-Bron1-7-Methylisatin und arbeitet sonst in der gleichen Weise, wie in Beispiel .a. angegeben, so erhält man 22,6 Gewichtsteile i-MethyI-5-brOm-2-aminobenzo1-3-carbonsäure vom F.229°.If, instead of 16.1 parts by weight of 5-methylisatin, 24 parts by weight of 5-bron1-7-methylisatin are used and the procedure is otherwise the same as in Example a. indicated, 22.6 parts by weight of i-methyl-5-brOm-2-aminobenzo1-3-carboxylic acid with a temperature of 229 ° are obtained.
