Verfahren zur Darstellung von 17-Chlor-A4-androstenon-3. Es wurde gefunden, dass man 17-Chlor- A 4-androstenon-3 erhält, ein Produkt von gro sser physiologischer<B>,</B> Wirksamkeit, das für pharmazeutische Zwecke oder als Zwischen produkt zur Herstellung von Arzneimitteln dient, wenn man 17-Chlor-A.--androstenol-3 unter intermediärem Schutz der Ringdoppel- bindung der Einwirkung eines Oxydations mittels unterwirft, welches eine sekundäre Alkoholgruppe in eine Ketogruppe überzu führen vermag.
Als Oxydationsmittel kommen, geeignete Verbindungen des 6wertigen Chroms, wie Chromsäureanhydrid und dergleichen, in Be tracht, es können aber auch andere Oxyda- fionsmittel Verwendung finden, wie sie zum B eispiel in Houben <B>-</B> Weyl, "Methoden der organischen Chemie", 11). Band,<B>3.</B> Auflage, Leipzig 1925, Seite 47<B>ff .</B> angeführt sind.
Zum Schutz der Ringdoppelbindung <B>wäh-</B> rend der axydativen Behandlung kann man an das 17-Chlor-A,-androstenol-3 Mittel an lagern, die sieh nach erfolgter Oxydation wieder abspalten lassen, wie z. B. Halogen. Die Entferuneg des Halogens kann in an sich bekannter Weise, z. B. durch Zinkstaub, er folgen. Nähere Einzelheiten hierüber sind aus Houben-Weyl, "Methoden der organi schen Chemie", 2. Die Isolierung der gebildeten neuen Keto- verbindung kann mit Hilfe von Ketonreagen- zien, wie z. B. Semicarbazid, oder mit Hilfe der fraktionierten Kristallisation oder der gleichen erfolgen.
Aus Aceton kristallisiert sie in kleinen farblosen Blättchen vom Schmelzpunkt 148'<B>C.</B> <I>Beispiel:</I> Eine Lösung von<B>0,8 g</B> 17-Chlor-A,-andro- stenol-3 in<B>60</B> cm' Eisessio, wird unter Schüt- e teln mit einer Lösung von 0,414<B>g</B> Brom in 20 cm# Eisessig vermischt. Hierzu gibt man <B>259</B> mg -Chromsäureanhydrid in 20 cm# Eis essig gelöst.
Das Reaktionsgemisch wird während 48 Stunden stehen gelassen und das Reaktionsprodukt unter Wasserkühlung durch Behandlung mit 2<B>g</B> Zinkstaub während <B>8</B> Stunden entbromt. Die Lösung wird dann vom Zinkstaub abfiltriert, in Wasser gegos sen und mit Äther extrahiert. Die ätherische Lösung wird mit Alkalilauge und Wasser gewaschen und zur Trockne verdampft. Der Rückstand der Ätherlösung enthält<B>0,7 g</B> 17- Chlor-,4.-androstenon-3.
Process for the preparation of 17-chloro-A4-androstenone-3. It has been found that 17-chloro-A 4-androstenone-3 is obtained, a product of great physiological activity, which is used for pharmaceutical purposes or as an intermediate product for the production of medicaments if one 17-chloro-A .-- androstenol-3, with intermediate protection of the ring double bond, is subjected to the action of an oxidizing agent which is able to convert a secondary alcohol group into a keto group.

To protect the ring double bond <B> during </B> rend the axydative treatment, the 17-chloro-A, -androstenol-3 agent can be added to, which can be split off again after oxidation, such as B. Halogen. The new keto compound formed can be isolated with the aid of ketone reagents, such as. B. semicarbazide, or with the help of fractional crystallization or the like.
It crystallizes from acetone in small, colorless flakes with a melting point of 148 '<B> C. </B> <I> Example: </I> A solution of <B> 0.8 g </B> 17-chlorine-A, -andro- stenol-3 in <B> 60 </B> cm 'Eisessio, is mixed with a solution of 0.414 <B> g </B> bromine in 20 cm # of glacial acetic acid while shaking. To this end, <B> 259 </B> mg of chromic anhydride dissolved in 20 cm # of glacial acetic acid are added.
The reaction mixture is left to stand for 48 hours and the reaction product is defbromated by treatment with 2 g zinc dust for 8 hours while cooling with water. The solution is then filtered off from the zinc dust, poured into water and extracted with ether. The essential solution is washed with alkali and water and evaporated to dryness. The residue of the ether solution contains <B> 0.7 g </B> 17-chloro-, 4-androstenone-3.
