Aufgabe der vorliegenden Erfindung ist die Schaffung neuer Arzneimittel. Diese Aufgabe warde gelöst durch das
Auffinden der neuen Verbindungen der Formel I, die aufgrund ihrer Eigenschaften zur Herstellung neuer Arzneimittel
verwendet v/erden können.
Eb wurde gefunden, daß die Prostansäur-ederivate der Formel
I wertvolle pharma,kologische Eigenschaften besitzen.
So treten beispielsweise blutdrucksenkende Wirkungen auf, die sich z.B. an der chloralosenarkotisierten Katze bei
Dauerinfusion zeigen. In diesem Test wird der arterielle
Blutdruck kymographisch registriert. Die Testsubstanzen werden über einen Zeitraum von 10 Minuten in wässeriger
Propylenglykol-Lösung infundiert.
Außerdom treten bei den Prostansäurederivaten der Formel I
vasodilatorische, antiphlogistisehe, diuxetische, bronchi
erientkranrpfende, die Magensaftsekretion, die Thrombozyten-Aggregation,
den Lipidabbau und die Noradrenalin-Freisetzung
hemmende, sov/ie Nasanschleimha/at-abschwellende
Eigenschaften auf, die ebenfalls nach hierfür geläufigen Methoden ermittelt werden können. Die Prostansäurederivate
der Formel I können auch die Funktion des corpus luteum, den Eitransport durch den Eileiter, die Nidation und die
Fertilität beeinflussen.
Die Verbindungen der Formel I und/oder ihre physiologisch unbedenklichen Salze können daher als Arzneimittel und
auch als Zwischenprodukte zur Herstellung anderer Arzneimittel verwendet werden.

Gegenstand der Erfindung sind Verbindungen der Formel I,

in denen A, B, R , R und R die oben angegebenen Bedeutungen
haben.
Die Verbindungen der Formel I enthalten mindestens zwei asymmetrische C-Atome am Fünfring. Wenn A -CHOH- bedeutet,
so sind im Ring drei AsymmetrieZentren vorhanden. Ein weiteres AsymmetrieZentrum wird durch das die OH-Gruppe
tragende C-Atom gebildet, welches sich in der ungesättigten Seitenkette befindet. Die Verbindungen der Formel I können
daher in einer Vielzahl stereoisomerer Formen auftreten; sie liegen in der Regel als racemische Gemische vor. °
Gegenstand der Erfindung sind neben den einzelnen Racematen
und racemischen Gemischen auch die optisch aktiven Isomeren der Formel I.
Gegenstand der Erfindung ist ferner ein Verfahren zur Herstellung einer Verbindung der Formel I sowie von deren
physiologisch unbedenklichen Salzen, dadurch gekennzeichnet, daß man eine Verbindung der allgemeinen Formel II

Formel I und/oder eines ihrer physiologisch unbedenklichen Salze, sowie ein Verfahren zur Herstellung von pharmazeutischen
Präparaten, dadurch gekennzeichnet, daß man eine Verbindung der Formel I zusammen mit mindestens einem
festen, flüssigen oder halbflüssigen Hilfs- oder Trägerstoff
und gegebenenfalls zusammen mit einem weiteren Wirkstoff in eine geeignete Dosierungsform bringt.
Wenn A eine -CHOH-G-ruppe bedeutet, so kann die OH-G-ruppe
cc- oder ß-ständig sein.
Die Doppelbindung in der Seitenkette einer Verbindung der Formel I ist vorzugsweise trans-substituiert.

Bei funktionell abgewandelten Hydroxygruppen handelt es sich vorzugsweise um z.B. mit einer gesättigten oder ungesättigten
aliphatischen, cycloaliphatischen oder aromatischen, substituierten oder unsubstituierten Carbonsäure
oder Sulfonsäure oder auch einer anorganischen Säure veresterten OH-G-ruppen. Bevorzugte Carbonsäuren sind Fettsäuren
mit 1 bis 18, vorzugsweise 1 bis 6 C-Atomen, wie Ameisen-, Essig-, Butter oder Isobuttersäure, oder auch
Pivalin-, Trichloressig-, Benzoe-, p-Nitrobenzoe-, Palmitin-,
Stearin- oder Ölsäure. Bevorzugte Sulfonsäuren sind solche, die sich von Alkylsulfonsäuren mit 1 bis

Es kann sich auch um substituierte Sulfonsäuren, wie 2-Hydroxyäthan- oder 4-Brombenzolsulfonsäure handeln.
Schwefelsäure und Orthophosphorsäure sind die als Esterkomponenten
bevorzugten anorganischen Säuren.
Punktioneil abgewandeltes OH kann weiterhin eine verätherte
OH-G-ruppe bedeuten, z.B. Aralkoxy mit vorzugsweise 7 bis 19 C-Atomen, wie Benzyloxy, p-Methylbenzyloxy.

Bei den nachstehend "beschriebenen chemischen Verfahren
zur Herstellung der Ausgangsverbindungen, aber auch der Verbindungen der Formel I handelt es sich um Analogieverfahren,
welche unter an sich bekannten Reaktionsbedingungen durchgeführt werden, wie sie in der Literatur
(z.B. in Standardwerken der präparativen organischen Chemie, wie HOUBEN-WEYL, Methoden der organischen Chemie,
Georg-Thieme-Verlag, Stuttgart) für analoge Reaktionen
angegeben sind.
Die Ausgangsstoffe zur Herstellung der Verbindungen der Formel I sind teils neu, teils bekannt und können gewünschtenfalls
auch in situ gebildet werden, derart, daß man sie aus dem Reaktionsgemisch nicht isoliert, sondern
direkt weiter zu den Verbindungen der Formel I umsetzt
(gegebenenfalls nach vorheriger Abtrennung unerwünschter Nebenprodukte).
So sind die Verbindungen der Formel II bekannt oder können z.B. aus 7-(2-Hydroxy-5-formyl-cyclopentyl)-heptansäuremethylester
durch Umsetzen mit einem gegebenenfalls substituierten Dimethyl-2-ketophosphonat der Formel

Die Dimethyl-2-ketophosphonate (CH2O)2P(O)-CH2COR2 und
(CH3O)2P(O)-CH2COBR^ sind nach an sich bekannten Verfahren
erhältlich durch Umsetzen von (CH^O)0P(O)-CH-* mit
einem Ester einer Säure R COOH oder R^BCOOH, vorzugsweise den Methyl- oder Äthylestern dieser Säuren in
Gegenwart einer starken Base,vorzugsweise einer Alkyllithiumverbindung
wie Butyllithium in einem inerten Lösungsmittel, beispielsweise einem Äther wie Diäthyläther,
Dimethoxyäthan oder Tetrahydrofuran unter Feuchtigkeitsausschluß und bei Temperaturen etwa zwischen -100
und -30°, insbesondere etwa zwischen -80° und -60 .
Die Umsetzung einer Verbindung der Formel II mit einer
Organometallverbindung der Formel III erfolgt in den für solche Reaktionen üblichen Lösungsmitteln, vorzugsweise
Äthern, wie Diäthyläther, Diisopropyläther, 1,2-Dimethoxyäthan,
Tetrahydrofuran (= THF), Dioxan, Anisol, oder Kohlenwasserstoffen, wie Benzol, Toluol, Xylol oder auch
in Gemischen dieser Lösungsmittel. Man kann entweder die Verbindung der Formel II oder eine Lösung dieser Verbindung
zu einer Lösung der Organometallverbindung der Formel III zugeben oder auch umgekehrt eine Lösung der Organometallverbindung
III zu einer Lösung der Verbindung II hinzufügen. Die Reaktionstemperatur ist nicht kritisch
und liegt etwa zwischen -25° und Raumtemperatur.
Die Verbindungen der Formel IV sind bekannt oder können beispielsweise wie bei den Verbindungen der Formel II
angegeben, hergestellt werden.
Als reduzierende Mittel, mit welchen man eine Verbindung der Formel IV oder eines ihrer Salze in eine Verbindung

z.B. Natriumborhydrid, gegebenenfalls in Gegenwart von Aluminiumchlorid oder von Lithiumbromid; ferner Lithiumborhydrid,

Weiterhin sind Aluminiumalkoholate, z.B. Triisopropyl- oder Triisobuty!aluminiumalkoholate
in Gegenwart des alkoholatbildenden Alkohols als Reduktionsmittel geeignet.
Die Reduktionen einer -CO-Gruppe zu einer -CHOH-Gruppe werden in einem inerten Lösungsmittel, beispielsweise
einem Alkohol, vorzugsweise einem niederen aliphatischen Alkohol, wie Methanol, Äthanol oder Isopropy!alkohol,
einem Äther wie Diäthyläther, Tetrahydrofuran oder Dioxan, oder auch in Wasser, bzw. in Gemischen dieser Lösungsmittel
bei Temperaturen zwischen -20° und 40°, vorzugsweise bei Raumtemperatur ausgeführt. Die Reaktionszeiten liegen
meist zwischen 15 Minuten und 6 Stunden.
Solvolysierende Mittel sind Wasser oder organische Lösungsmittel, meist in Gegenwart eines sauren oder basischen
Katalysators. Als organische Lösungsmittel kommen z.B. in Frage Alkohole mit vorzugsweise bis zu 7 C-Atomen,
insbesondere aliphatische Alkohole wie Methanol, Äthanol,

zu 6 C-Atomen, wie Aceton oder Butanon, Amine, vorzugsweise mit bis zu 12 C-Atomen, insbesondere aliphatische
Amine wie Triethylamin, Äthanolamin, Triäthanolamin oder
Diisopropylamin, aber auch alicyclische oder heterocyclische
Amine, wie Cyclohexylamin, Pyrrolidin oder Pyridin, aprotisch dipolare Lösungsmittel, vorzugsweise Amide wie Dimethylformamid
(= DMF) oder Hexamethylphosphorsäuretriamid (= HMPT), aber auch z.B. Nitrile wie Acetonitril,
oder Schwefelverbindungen wie Dimethylsulfoxid (= DMSO)
oder Tetrahydrothiophen-S,S-dioxid, sowie Gemische dieser
Lösungsmittel, vorzugsweise die Wasser enthaltenden Gemische.
Als saure Katalysatoren eignen sich für eine Solvolyse anorganische Säuren, beispielsweise Protonensäuren, wie
Salz-, Schwefel-, Phosphor- oder Bromwasserstoffsäure
oder Lewis-Säuren, wie BF,, aber auch organische Säuren, z.B. Carbonsäuren wie Chloressigsäure, Trichloressigsäure
oder Trifluoressigsäure oder Sulfonsäuren wie Methan-, Äthan-, Benzol- oder p-Toluolsulfonsäure. Als basische
Katalysatoren verwendet man bei der Solvolyse zweckmäßig Alkalimetall- oder Erdalkalimetallhydroxide, wie Natrium-,
Kalium- oder Calciumhydroxid, oder basische Salze, wie Natrium- oder Kaliumcarbonat. Wenn das Lösungsmittel(gemisch)
bereits basisch oder sauer ist, kann auch auf den Zusatz eines basischen oder eines sauren Katalysators
verzichtet werden.

Verbindungen, die sonst der allgemeinen Formel I entsprechen,
in denen aber vorhandene COOR - und/oder Hydroxy- und/oder Carbonylgruppen in funktionell abgewandelter
Form vorliegen, können durch Umsetzen mit solvolysierenden Mitteln in die Verbindungen der Formel I
umgewandelt werden. Die Solvolysen führt man z.B. bei
Temperaturen zwischen etwa -20° und 100° aus. In der Regel arbeitet man in Gegenwart eines sauren oder eines
basischen Katalysators unter Verwendung eines geeigneten Lösungsmittels. Die Solvolysezeiten liegen ungefähr zwischen
einer Stunde und 48 Stunden.
Als oxidierende Mittel, mit welchen es gelingt, eine Gruppe A = -CHOH- in eine andere Gruppe A= -CO- umzuwandeln,
kommen vor allem solche in Frage, die im basischen, neutralen oder auch schwach sauren Milieu oxidierend
wirken. Man verwendet beispielsweise Ketone, vorzugsweise aliphatische oder cycloaliphatische wie Aceton
oder Cyclohexanon, aber auch Chinone wie 1,4-Benzochinon in Gegenwart eines Aluminiumalkoxids, vorzugsweise Aluminium-
tri-tert.-butoxid oder Aluminium-tris-isopropoxid (Oppenauer-Oxidation) , Sulfoxide, vorzugsweise Dimethylsulfoxid,
gegebenenfalls in Gegenwart eines weiteren Hilfsstoffes, beispielsweise eines Carbodiimids, wie Dicyclohexylcarbodiimid,

Oxidationen der Gruppe A = -CHOH- zu -CO- sollen unter
relativ milden Reaktionsbedingungen ausgeführt werden. Oxidiert man daher z.B. mit Chromsäureanhydrid, so verwendet
man eine organische Base, vorzugsweise Pyridin als Lösungsmittel. Man kann aber auch, vor allem "bei
Verwendung anderer Oxidationsmittel in anderen inerten Lösungsmitteln, "beispielsweise Ketonen wie Aceton, oder

Verbindungen der Formel I (R = H) können nach an sich
bekannten Methoden mit einem veresternden Mittel verestert werden. Man arbeitet in einem geeigneten inerten,
vorzugsweise wasserfreien Lösungsmittel, beispielsweise einem Äther wie Diethylether oder THF, einem Alkohol, vorzugsweise
einem niederen verzweigten oder unverzweigten Alkanol, wie Methanol, Äthanol, Propanol, Isopropylalkohol
oder Butanol, oder auch in einem Kohlenwasserstoff, wie Petroläther, Hexan, Benzol oder Toluol, oder Gemischen
dieser Lösungsmittel bei Temperaturen zwischen etwa -10° und 40°, vorzugsweise bei Raumtemperatur. Die Reaktionszeiten
liegen in der Regel zwischen 30 Minuten und 24 Stunden.
Umesterungen v/erden vorzugsweise in den wasserfreien
Alkoholen R OH (R ungleich H) durchgeführt, deren Alkylrest
im gewünschten Ester erscheinen soll. Man arbeitet bei Temperaturen zwischen etwa 0° und 40°, vorzugsweise
bei Raumtemperatur.
Man kann die freien Carbonsäuren der Formel I (R = H)
durch Umsetzung mit einer Base in eines ihrer physiologisch unbedenklichen Metall- bzw. Ammoniumsalze überführen.
Als Salze kommen insbesondere die Natrium-, Kalium-, Magnesium-, Calcium- und Ammoniumsalze in Betracht, ferner
substituierte Ammoniumsalze, wie z.B. die Dimethyl- und Diäthylammonium-, Monoäthanol-, Diäthanoi- und Triäthanolammonium-,
Cyclohexylammonium-, Dicyclohexylammonium-
und Dibenzyläthylendiammonium-Salze. Umgekehrt können
Säuren der Formel I aus ihren Metall- und Ammoniumsalzen durch Behandlung mit Säuren, vor allem Mineralsäuren, wie
Salz- oder Schwefelsäure, in Freiheit gesetzt.werden.
Die Verbindungen der Formel I haben, wie bereits erläutert, im allgemeinen mehrere AsymmetrieZentren, stets aber wenig-

stens drei. Sie werden daher meist als Gemische ver.i'.•hiedener
stereoisomerer Formen erhalten, d.h. als Racenate oder in der Regel als Gemische von Racematen. Da verschiedene
Racemate zueinander diastereomer sind, können sie aufgrund ihrer unterschiedlichen physikalischen Eigenschaften
aus ihren Gemischen isoliert und rein erhalten werden, beispielsweise durch Umkristallisieren aus geeigneten
Lösungsmitteln (wobei anstelle der Verbindungen selbst auch gut kristallisierende Derivate eingesetzt werden
können), durch destillative Trennung, insbesondere aber mit Hilfe chromatographischer Methoden, wobei sowohl
adsorptionschromatographische oder verteilungschromatographische Methoden als auch Kischformen in Frage kommen.
Die Racemate können nach einer Vielzahl bekannter Methoden, wie sie in der Literatur angegeben sind, in ihre optischen
Antipoden getrennt werden. Die Methode der chemischen Trennung ist zu bevorzugen. Danach werden aus dem
racemischen Gemisch durch Umsetzung mit einem optisch aktiven Hilfsmittel Diastereomere gebildet.
So kann man gegebenenfalls eine optisch aktive Base mit der Carboxylgruppe einer Verbindung der Formel I umsetzen.
Zum Beispiel kann man diastereomere Salze der Verbindungen der Formel I (R1 = H) mit optisch aktiven Aminen, wie
Chinin, Brucin, 1-Phenyläthylamin, 1-Naphthyläthylamin
oder basischen Aminosäuren, wie Lysin, Arginin, bilden. In ähnlicher Weise lassen sich Ester-Diastereomere durch
Veresterung von Verbindungen der Formel I (R = H) mit optisch aktiven Alkoholen, wie Borneol, Menthol oder
Octancl-2, herstellen. Der Unterschied in der Löslichkeit
der anfallenden diastereomeren Salze bzw. Ester erlaubt die selektive Kristallisation der einen Form und die
Regeneration der jeweiligen optisch aktiven Verbindungen aus dem Gemisch.

Aber auch die anderen in den Verbindungen der Formel I
vorhandenen funktioneilen Gruppen können zur Bildung von Diastereomeren herangezogen werden. So kann man z.B. OH-G-ruppen
mit optisch aktiven Säuren wie (+)- und (-)-Weinsäure oder Camphersäure verestern und Ketogruppen mit
optisch aktiven Hydrazinen, wie Menthylhydrazin umsetzen
und aus diesen Derivaten die reinen Enantiomeren gewinnen.
Weiterhin ist es natürlich möglich, optisch aktive Verbindungen nach den beschriebenen Methoden zu erhalten, indem
man Ausgangsstoffe verwendet, die bereits optisch aktiv sind.
Bevorzugt sind solche Verbindungen der Formel I, in denen die beiden Seitenketten trans-ständig sind und bei denen
die C=C-Doppe!bindung trans-substituiert ist.
Die neuen Verbindungen können im Gemisch mit festen, flüssigen und/oder halbflüssigen Arzneimittelträgern als Arzneimittel
in der Human- oder Veterinärmedizin verwendet werden. Als Trägersubstanzen kommen solche organischen oder
anorganischen Stoffe in Frage, die für die parenterale, enterale oder topikale Applikation geeignet sind und die
mit den neuen Verbindungen nicht in Reaktion treten, wie beispielsweise Wasser, pflanzliche Öle, Benzylalkohol,
Polyathylenglykole, G-elatine, Lactose, Stärke, Magnesiumstearat,
Talk, Vaseline, -Cholesterin. Zur parenteralen Applikation dienen insbesondere Lösungen, vorzugsweise
ölige oder wässerige Lösungen, sowie Suspensionen, Emulsionen oder Implantate. Für die enterale Applikation
eignen sich Tabletten, Dragees, Sirupe, Säfte oder Suppositorien, für die topikale Anwendung Salben, Cremes oder

Puder. Die angegebenen Zubereitungen können gegebenenfalls sterilisiert oder mit Hilfsstoffen, wie Gleit-,
Konservierungs-, Stabilisierungs- oder Netzmitteln, Emulgatoren, Salzen zur Beeinflussung des osmotischen Druckes,
Puffersubstanzen, Parb-, Geschmacks- und/oder Aromastoffen versetzt werden.
Die Substanzen v/erden vorzugsweise in einer Dosierung von 0,001 bis 10mg pro Dosierungseinheit verabreicht;

und Rühren eine G-rignardlösung, hergestellt aus 3,39 g Methyljodid und 520,5 mg Magnesiumspänen in 60 ml
trockenem Diäthyläther und rührt weitere 60 Minuten bei Raumtemperatur. Man hydrolysiert durch Zutropfen einer
gesättigten, wässerigen Ammoniumchloridlösung unter Eiskühlung, trennt die organische Phase ab, wäscht mit Wasser, trocknet über Na2SO., destilliert das Lösungsmittel ab und erhält nach chromatographischer Reinigung des Rückstandes (Kieselgel/Dichlormethan:Aceton = 95:5) 2-(6-Carboäthoxyhexyl )-3-Z3~kyäroxy-3-niethyl-4- (4-f luorphenoxy )-1 -

Man tropft zu 3,12 g 2-(6-Carboäthoxyhexyl)-3-Z3-oxo-4-(4-chlorphenoxy)-1-butenyl7-cyclopentanol,
gelöst in 100 ml trockenem THP unter Stickstoff, Feuchtigkeitsausschluß
und Rühren eine G-rignardlösung, hergestellt aus 3,4 g Methyljodid und 521 mg Magnesiumspänen in 60 ml
trockenem THF und rührt weitere 90 Minuten bei Raumtemperatur. Man hydrolysiert durch Zutropfen einer gesättigten,
wässerigen Ammoniumchloridlösung unter Eiskühlung, trennt die organische Phase ab, wäscht mit Wasser, trocknet
über Natriumsulfat, destilliert das Lösungsmittel ab und erhält nach chromatographischer Reinigung des Rückstandes
(Kieselgel/Dichlormethan: Aceton = 95:5) 2"-(6-Carboäthoxyhexyl)
-3-/3-hydroxy-3~methyl-4- (4-chlorphenoxy) ■
1-butenyl7-cyclopentanol als Öl,

Man tropft zu 2,88 g 2-(6-Carboäthoxyhexyl)-3-(3-oxo-4-phenoxy-1-butenyl)-cyclopentanol,
gelöst in 150 ml trockenem Diäthyläther unter Stickstoff, Feuchtigkeitsausschluß
und Rühren eine G-rignardlösung, hergestellt aus 2,27 g Methylbromid und 520 mg Magnesiumspänen in 80 ml
trockenem Diäthyläther und rührt 45 Minuten bei Raumtemperatur. Man hydrolysiert durch Zutropfen einer eisgekühlten
wässerigen Ammoniumchloridlösung und arbeitet die abgetrennte organische Phase wie in Beispiel 19 beschrieben auf.
Man erhält 2-(6-Carb οäthoxyhexy1)-3-(3-hydroxy-3-me thy1-4-phenoxy-1-butenyl)-cyclopentanol
als Öl,

und Rühren eine Grignardlösung, hergestellt aus 3,42 g

Diäthyläther. Man rührt das Reaktionsgemisch 1 Stunde bei 0°, hydrolysiert durch Eingießen in Eiswasser, trennt die
organische Phase ab, wäscht mit gesättigter wässeriger Natriumchloridlösung, trocknet über Magnesiumsulfat,

zu einem Gemisch aus 343 mg trockenem Chromtrioxid, 0,545 ml trockenem Pyridin und 10 ml trockenem
Dichlormethan, rührt 15 Minuten bei Raumtemperatur, dekandiert das Lösungsmittel vom Rückstand ab und extrahiert
diesen noch zweimal mit Dichlormethan. Die vereinigten
organischen Phasen werden filtriert, mit eiskalter 1 %iger Schwefelsäure, Wasser, gesättigter Natriumbicarbonatlösung und nochmals mit Wasser gewaschen und über Magnesiumsulfat getrocknet. Das Lösungsmittel wird abdestilliert und man
erhält nach chromatographischer Reinigung des Rückstandes (Kieselgel/Dichlormethan:Aceton = 19:1) 2-(6-Carboäthoxyhexyl)-3-£3-hydroxy-3-methyl-4-(4-fluorphenoxy)-1-butenyl7-cyclopentanon als Öl,
Dichlormethan, rührt 15 Minuten bei Raumtemperatur, dekandiert das Lösungsmittel vom Rückstand ab und extrahiert

Man gibt 830 mg Natriumborhydrid innerhalb von 15 Minuten portionsweise zu 1,03 g 2-(6-Carboäthoxyhexyl)-3-//5-0X0-4-(4-fluorphenoxy)-1-pentenylj-cyclopentanol,
gelöst in einem Gemisch aus 30 mi trockenem Methanol und 20 ml trockenem THF unter Rühren, Feuchtigkeitsausschluß
und Stickstoff, rührt weitere 15 Minuten bei Raumtemperatur, stellt unter Eiskühlung mit 0,1 η Salzsäure auf
pH 5 ein, verdünnt mit Wasser und extrahiert mit Äther. Die organische Phase wird mit Wasser, 1 ^iger wässeriger
Natriumcarbonatlösung und nochmals mit Wasser gewaschen,
über Magnesiumsulfat getrocknet und das Lösungsmittel
abdestilliert. Nach chromatographischer Reinigung des Rückstandes (Kieselgel/Dichlormethan:Aceton = 9:1) erhält
man 2-(6-Carboäthoxyhexyl)-3-/3-hydroxy-4-(4-fluorphenoxy
)-1-butenyl7-cyclopentanol.
Analog Beispiel 81 sind die in den nachstehenden Beispielen 82 bis 110 genannten Dihydroxyverbindungen der

Man läßt eine Lösung τοη 0,5 g 2-(6-Carboäthoxyhexyl)-3-/3-hydroxy-3-methyl-4-(4-fluorphenoxy)-1-butenyl7-cyclopentanol
in 9 ml Dioxan mit 4,5 ml 1n wässeriger KOH 4 Stunden unter Stickstoff "bei Raumtemperatur stehen,
tropft anschließend unter Rühren und Eiskühlung 1n wässerige
HCl zu, "bis ein pH-Wert von 3 erreicht ist. Nach Zugabe von 17 ml Wasser wird mit Äther extrahiert. Die
Ätherphase wird mit Wasser gewaschen, über Natriumsulfat getrocknet und das Lösungsmittel abdestilliert. Nach
chromatographischer Reinigung des Rückstandes erhält man
2-(6-Carboxyhexyl)-3-ZJ-kydroxy-3-methyl-4-(4-fluorphenoxy)-1-butenyl7-cyclopentanol.

ab. Nach chromatographischer Reinigung des Rückstandes (Kieselgel/Diehlormethan:Aceton = 9:1) erhält man 2-(6-Carbomethoxyhexyl)-3-ZJ-hydroxy-3-methyl-4-(4-fluorphenoxy)-1-butenyi7-cyclopentanol.

luorphenoxy )-1 butenyl7-cyclopentanols ab.
Man rührt 4,8 g 2-(6-Carboäthoxyhexyl)-3~/jT-hydroxy-3-me
thy 1-4- (4-f luorphenoxy )-1 -buteny]_7-1 -tetrahydropyranyl
oxy-cyclopentan (erhältlich aus 2-(6-Carboäthoxyhexyl)-3-/5-0X0-4-(4-fluorphenoxy)-1-butenyl7~cyclopentanol
durch Umsetzen mit Dihydropyran und anschließende Reaktion des erhaltenen Produktes mit Methylmagnesiumjodid) 2 Stunden
unter Stickstoff bei 60° in 40 ml wässeriger 1n HCl,
extrahiert das Reaktionsgemisch zweimal mit Methylenchlorid, wäscht die organische Phase mit Wasser, trocknet
über Natriumsulfat, destilliert das Lösungsmittel ab und erhält nach chromatographischer Reinigung des Rückstandes

Die Wirkstoffe der Formel I lassen sich nach Methoden, die aus der Literatur bekannt sind, zu pharmazeutischen
Zubereitungen verarbeiten, wie in den folgenden Beispielen gezeigt wird:

Beispiel A: Tabletten
Ein Gemisch bestehend aus 0,2 kg 2-(6-Carboxyhexyl)-3-/3-hydroxy-3-me
thy 1-4- (4-fluorphenoxy) -1 -but enjl/-cyc lopentanol-natriumsalz,
0,5 kg Lactose, 0,18 kg Maisstärke, 0,02 kg Cellulosepulver und 0,02 kg Magnesiumstearat wird
in üblicher Weise zu Tabletten gepreßt, derart, daß jede Tablette 200 mg des Wirkstoffs enthält.
Beispiel B: Dragees
Analog Beispiel A werden Tabletten gepreßt, die anschließend in üblicher Weise mit einem Überzug bestehend aus
Zucker, Maisstärke, Talk und Tragant überzogen werden.
Analog den Beispielen A und B sind Tabletten und Dragees erhältlich, die einen oder mehrere der anderen Wirkstoffe
der Formel I oder ihrer physiologisch unbedenklichen Salze enthalten.

1. Verbindungen der allgemeinen Formel I
OH=GH-O (0H)R2-B-R3
worin
A -CO- oder -OHOH-,
B -CH2-, -CH2CH2- oder -CH2O-,
R H oder Alkyl mit 1 bis 4 C-Atomen, R2 H, Methyl oder Äthyl, und
R Phenyl oder ein- bis dreifach durch P, Cl, CUP,,,
OH, OCH5, OC2H5 oder Alkyl mit 1 bis 3 C-Atomen
substituiertes Phenyl
bedeuten,
und ihre physiologisch unbedenklichen Metall- und Ammoniumsalze.

Verfahren zur Herstellung von pharmazeutischen Präparaten, dadurch gekennzeichnet, daß man eine Verbindung der allgemeinen
Formel I und/oder eines ihrer physiologisch unbedenklichen Salze zusammen mit mindestens einem festen,
flüssigen oder halbflüssigen Träger- oder Hilfsstoff und gegebenenfalls zusammen mit einem weiteren Wirkstoff in
eine geeignete Dosierungsform bringt.
13. Pharmazeutische Zubereitung, enthaltend eine Verbindung
der allgemeinen Formel I und/oder eines ihrer physiologisch unbedenklichen Salze.
