Claims (6)

1. Ein Apparat zur Phosphatbestimmung nach dem Ablauf einer PCR-Reaktion (Polymerasekettenreaktion) in einem sog. Thermocycler, dadurch gekennzeichnet, daß dieser Apparat nach vorheriger Zugabe eines Phosphatreagenz spektroskopisch photometrisch die Extinktion des gebildeten Phosphatkomplexes mißt.

Ein Photometer (Elisa-Reader) zur Phosphatmessung in Multititterplatten (multi- well-platten, 96 well platten) nach zuvor durchgeführter PCR-Reaktion (Polymerasekettenreaktion) dadurch gekennzeichnet, das dieses Photometer mit einem Mikroprozessor ausgestattet ist und die gemessene Phosphatextinktion anhand von parallel mitbestimmten Eichproben direkt in die entsprechende DNA-Ausgangsmenge umrechnet.
3. Ein Apparat nach Schutzanspruch 2) zusätzlich dadurch gekennzeichnet, daß dieser Apparat insbesondere bei einer der Wellenlängen der gängigen Phosphatreagenzkomplexe Phosphomolybdänblau 680 nm (oder 880 nm), Phosphomolybdovanadat 345 nm (oder 420 nm) und Malachitgrün 650 nm mißt.
4. Ein Apparat nach Schutzanspruch 1) und 2) zusätzlich dadurch gekennzeichnet, daß dieser Apparat geeicht ist und die gemessene Extinktion direkt in die entsprechende Phosphatmenge umrechnet.
5. Ein Apparat nach Schutzanspruch 1), 2) und 3) zusätzlich dadurch gekennzeichnet, daß dieser Apparat aus der gemessenen Extinktion (bzw. Phosphatkonzentration) anhand einer zuvor bestimmten Eichkurve die Ursprungskonzentration an untersuchter DNA errechnet.
6. Ein PCR-Reaktionsgefäß bzw. eine Multititerplatte (multi-well-platte, 96-well-platte) dadurch gekennzeichnet, daß dieses lichtdurchlässig für die zur Phosphatmessung verwendeten Wellenlängen ist und aus einem entsprechenden Material gefertigt ist. (z. B. Glas, Quarzglas)
