Description
Ein Großteil der gegenwärtig angebotenen Immersionsöle für die
Mikroskopie mit einem Brechungsindex von ne=1,518 enthält als
Hauptbestandteile Chlorparaffine. Chlorparaffine sind vor allem
aufgrund ihrer problematischen Umweltverträglichkeit seit
einiger Zeit als gefährliche Arbeitsstoffe eingestuft.
Neben den Immersionsölen auf Basis von Chlorparaffinen sind
auch halogenfreie Immersionsöle erhältlich. Das Hauptproblem
bei der Herstellung halogenfreier Immersionsöle ist das
ungünstige Verhältnis von Brechzahl und Dispersion bei den
meisten halogenfreien Flüssigkeiten mit Brechungsinidizes, die
einen Wert von <1,5 aufweisen. Die die Dispersion
beschreibende Abbe-Zahl weicht bei solchen Immersionsölen
häufig deutlich von den durch die DIN 58 884 bzw. ISO 8036/1
spezifizierten Bereichen für die Abbe-Zahl von 44 ±5 (DIN)
bzw. 44 ±3 (ISO) ab. Bekannte halogenfreie Immersionsöle,
deren Abbe-Zahl <41 ist, sind entweder hinsichtlich ihrer
UV-Durchlässigkeit unbefriedigend oder weisen eine zu große
Eigenfluoreszenz auf.
In der EP 0 209 621 sind neben halogenhaltigen auch
Ausführungsbeispiele für halogenfreie Immersionsöle angegeben.
Zwei Ausführungsbeispiele für halogenfreie Immersionsöle
enthalten Tricyclodecanol als Nebenkomponente, jedoch stets in
Abmischung mit Flüssigpolyolefinen. Flüssigpolyolefine weisen
ein schlechtes Transmissionsverhalten im nahen UV-Bereich und
eine hohe Restfluoreszenz auf. Bezüglich der UV-Durchlässigkeit
(bei Wellenlängen unter 400 nm) sind in der Schrift keine
Angaben enthalten und selbst bei einer Wellenlänge von 400 nm
wird die Transmission nur durch den relativ bescheidenen Wert
von 95% spezifiziert. Außerdem ist die Viskosität mit Werten
von 1000 bis 2000 mm2/sec bei 25°C für die meisten Anwendungen
zu groß, d. h. beim Aufbringen auf das Präparat kann es sehr
leicht zum Einschluß von Luftblasen kommen. Desweiteren sind
diese Immersionsöle nicht mit den üblicher Weise eingesetzten
polaren Lösungsmitteln wie Äthanol oder Isopropanol lösbar, so
daß zum Entfernen des Immersionsöls Petrolether oder
vergleichbare Benzin-Kohlenwasserstoffe eingesetzt werden
müssen, die wiederum ein gesundheits- und umweltgefährdendes
Potential aufweisen.
In der noch nicht veröffentlichten deutschen Patentanmeldung
Nr. 197 05 978 der Anmelderin sind halogenfreie Immersionsöle
beschrieben, deren Abbe-Zahlen überwiegend im oberen Bereich
der durch die DIN 58 884 bzw. ISO 8036/1 definierten Bereiche
liegen und die gleichzeitig eine hohe UV-Durchlässigkeit und
eine geringe Eigenfluoreszenz aufweisen. Hauptbestandteil
dieser Immersionsöle sind Tricyclodecanester. Da
Tricyclodecanester gegenwärtig keine großtechnische Anwendung
haben, muß diese Esterkomponente in den für die
Immersionsölabmischung benötigten Mengen synthetisiert werden,
wodurch relativ hohe Herstellkosten resultieren.
Ziel der Erfindung ist es, ein halogenfreies Immersionsöl für
Mikroskope anzugeben, das aus großtechnisch hergestellten
Bestandteilen zusammengesetzt ist und dadurch zu den für
Immersionsöle üblichen Herstellkosten hergestellt werden kann.
Zusätzlich soll das Immersionsöl eine gute UV-Durchlässigkeit
und eine geringe Restfluoreszenz aufweisen.
Dieses Ziel wird erfindungsgemäß durch ein Immersionsöl mit den
Merkmalen des Anspruches 1 gelöst. Vorteilhafte Ausgestaltungen
der Erfindung ergeben sich aus den Merkmalen der abhängigen
Ansprüche.
Das erfindungsgemäße Immersionsöl enthält einen ersten
Bestandteil, der aus der Gruppe der Tricyclodecanalkohole
(TCD-Alkohole), Tricyclodecanketone (TCD-Ketone) oder
Decahydronaphthole stammt und als weitere Bestandteile
ausschließlich Weichmacher und/oder Poly-ε-Caprolactone. Der
erste Bestandteil aus der Gruppe der TCD-Alkohole, TCD-Ketone
oder Decahydronaphthole dient primär zur Realisierung einer
ausreichend großen Abbe-Zahl, während der zweite Bestandteil
bestehend aus Weichmacher und/oder Poly-ε-Caprolactone zur
Einstellung des Brechungsindex und der Viskosität dient.
Besonders bevorzugt sind dabei Drei- oder Vierstoffgemische,
die sowohl ein Poly-ε-Caprolactone als auch einen Weichmacher
enthalten.
Das Immersionsöl besteht ausschließlich aus Substanzen, die
mehr oder weniger stark ausgeprägt polar sind. Damit ist das
Immersionsöl mit polaren Lösungsmitteln verdünnbar bzw. lösbar,
so daß Immersionölreste problemlos mit den üblichen
Lösungsmitteln wie Äthanol und/oder Isopropanol entfernt werden
können. Insbesondere enthält das Immersionöl keine
Flüssigpolyolefine.
Als TCD-Alkohole kommen insbesondere das 8(9)-Hydroxymethyl
tricyclo[5.2.1.02,6]decan (TCD-Alkohol M) und das 3(4),
8(9)-Dihydroxymethyl-tricyclo[5.2.1.02,6]decan (TCD-Alkohol DM) in
Frage. Als TCD-Keton kommt insbesondere das 8-Oxotricyclodecan
und aus der Gruppe der Decahydroxynaphthole das Decahy
dro-2-naphthol in Betracht. Aus der Gruppe der Poly-ε-Caprolactone
sind insbesondere Poly-ε-Caprolactontriole und Poly-ε-Capro
lactondiole bevorzugt. Da beide Stoffe großtechnisch in
der Polyurethan- bzw. Polyesterherstellung eingesetzt werden,
sind diese neben den Weichmachern aufgrund ihrer
großtechnischen Herstellung relativ kostengünstig und in guter
Qualität verfügbar. Als Weichmacher werden bevorzugt
Phosphorsäurearylester, Phthalsäurepolyester,
Adipinsäurepolyester, Dibenzylether, Butylbenzylphthalat,
Dicyclohexylphthalat oder Benzylbenzoat eingesetzt.
Von Bedeutung ist, daß alle eingesetzten Substanzen hochsiedend
sind und demzufolge in der Anwendung kaum Brechzahländerungen
durch Verdunstung auftreten können. Weiterhin ist ein Teil der
eingesetzten Substanzen - die Substanzen des ersten
Bestandteils und die Weichmacher - vakuumdestillierbar und
damit in einer für eine geringe Restfluoreszenz erforderlichen
hohen Reinheit darstellbar. Die Poly-ε-Caprolactone sind zwar
nicht vakuumdestillierbar, werden jedoch mit hinreichender
optischer Reinheit hergestellt und angeboten.
Der Anteil des ersten Bestandteiles aus der Gruppe der
TCD-Alkohole, TCD-Ketone oder Dekahydronaphthole sollte
vorzugsweise zwischen 8 und 25 Gewichtsprozent liegen. Bei
Abmischungen unter 8 Gewichtsprozent dieses Bestandteils ist
deren Auswirkung auf die Abbe-Zahl zu gering und bei einem
Anteil von über 25 Gewichtsprozent können Abmischungen mit
reizenden Wirkungen resultieren.
Der Anteil der Poly-ε-Caprolactone beträgt vorzugsweise mehr
als 25 Gewichtsprozent und die Poly-ε-Caprolactone und der
erste Bestandteil aus der Gruppe TCD-Alkohole, TCD-Ketone oder
Decahydroxynaphthole beträgt zusammen vorzugsweise zwischen 40
und 80 Gewichtsprozent, während der Anteil der Weichmacher
vorzugsweise zwischen 20 und 60 Gewichtsprozent beträgt.
Als besonders gut geeignet unter den Weichmachern hat sich
insbesondere das Diphenylkresylphosphat herausgestellt.
Die konkreten Zusammensetzungen für bevorzugte
Ausführungsbeispiele erfindungsgemäßer Immersionsöle sind in
der Tabelle I gegeben. Der TCD Alkohol M und der TCD Alkohol DM
werden unter diesen Bezeichnungen von der Firma Hoechst AG,
Frankfurt angeboten. Die verschiedenen Poly-ε-Caprolactontriole
werden von der Firma Union Carbide unter den Bezeichnungen Tone
Polyol 305 und Tone Polyol 301 und die Poly-ε-Caprolactondiole
von der selben Firma unter den Bezeichnungen Tone Polyol 0201
und Tone Polyol 2221 angeboten. Der Adipinsäurepolyester wird
von der Firma Baier AG unter der Bezeichnung Ultramoll III
angeboten. Die Herstellung der Immersionsöle erfolgt einfach
durch Abmischen der angegebenen Substanzen in den angegebenen
Mengen.
Wie der Tabelle I entnehmbar ist, liegen die Brechungsindizes
ne und Dispersion ve für alle angegebenen 10
Ausführungsbeispiele innerhalb der durch die DIN 58 884
vorgegebenen Bereiche. Und lediglich die Ausführungsbeispiele
Nr. 4, 8 und 10 liegen bezüglich der Dispersion mit Werten von
ve=40,5 bzw. ve=40,8 geringfügig unter dem durch die ISO
8036/1 definierten Wertebereich. Durch geringfügige Änderungen
der Zusammensetzungen, beispielsweise geringfügige Anhebung des
Anteils des TCD-Alkohols oder des 8-Oxotricyclodecans lassen
sich auch bei Verwendung dieser Ausgangssubstanzen
Immersionsöle mit einer Abbe-Zahl ve<=41 herstellen.
Wie der Tabelle I weiterhin entnehmbar ist, weisen sämtliche
Immersionsöle hervorragende Transmissionseigenschaften auf. Die
Transmission durch eine 10 mm dicke Küvette beträgt für alle
Ausführungsbeispiele bei einer Wellenlänge von 400 Nanometer
über 95% - bei dem besonders bevorzugten Beispiel Nr. 2 sogar
über 99% Prozent - und selbst im nahen Uv-Bereich bei einer
Wellenlänge von 350 Nanometer beträgt die Transmission noch
zwischen 54,9 und 93,6 Prozent. Für die Anwendung von
besonderer Bedeutung ist dabei die Transmission bei 365
Nanometer (bei der sogenannten i-Linie). Die hier erreichten
Transmissionswerte zwischen 77,6% beim Ausführungsbeispiel Nr.
3 und 97,3% beim Ausführungsbeispiel Nr. 4 liegen wesentlich
höher als bei bekannten chlorparaffinhaltigen Immersionsölen
und sind mindestens vergleichbar, wenn nicht gar besser,
gegenüber bekannten chlorparaffinfreien Immersionsölen.
Gleichzeitig sind auch gute Werte für die Restfluoreszenz, die
bei Anregungs-/Meßwellenlängen von 405/485 nm zwischen 1,8 und
17,5 mg/l Chininsulfatäquivalenten und bei Anregungs-/Meß
wellenlängen von 365/450 nm zwischen 0,15 und 0,36 mg/l
Chininsulfatäquivalenten liegt, erreicht worden, die zum Teil
wesentlich niedriger sind, als die entsprechenden Werte für
bisher erhältliche chlorfreie Immersionsöle, die gleichzeitig
die ISO 8036/1 erfüllen und eine Viskosität zwischen 200 und
1200 mm2/s aufweisen.
Claims (12)
1. Immersionsöl für Mikroskope, das einen ersten Bestandteil
aus der Gruppe der Tricyclodecanalkohole,
Tricyclodecanketone oder Decahydronaphthole und einen
zweiten Bestandteil aus einem Weichmacher und/oder einem
Poly-ε-Caprolacton enthält.
2. Immersionsöl nach Anspruch 1, wobei ausschließlich
Substanzen mit polaren Eigenschaften enthalten sind.
3. Immersionsöl nach Anspruch 1 oder 2, wobei der zweite
Bestandteil sowohl mindestens einen Weichmacher als auch
mindestens ein Poly-ε-Caprolacton enthält.
4. Immersionsöl nach einem der Ansprüche 1-3, wobei der
Anteil des ersten Bestandteils zwischen 8 und 25
Gewichtsprozent beträgt.
5. Immersionsöl nach einem der Ansprüche 1-4, wobei der
Anteil der Poly-ε-Caprolactone mindestens 25
Gewichtsprozent beträgt.
6. Immersionsöl nach Anspruch 5, wobei der Anteil der Poly-ε-Capro
lactone und der erste Bestandteil aus der Gruppe der
Tricyclodecanalkohole, Tricyclodecanketone oder
Decahydronaphthole zusammen mindestens 40 Gewichtsprozent
beträgt.
7. Immersionsöl nach einem der Ansprüche 1-6, wobei der
Anteil der Weichmacher zwischen 20 und 60 Gewichtsprozent
beträgt.
8. Immersionsöl nach einem der Ansprüche 1-7, wobei ein
oder mehrere Weichmacher aus der Gruppe
Phosphorsäurearylester, Phthalsäurepolyester,
Adipinsäurepolyester, Dibenzylether, Polybenzylphthalat,
Benzylbenzoat oder Dicyclohexylphthalat enthalten sind.
9. Immersionsöl nach Anspruch 8, wobei der Weichmacher
Diphenylkresyplphosphat ist.
10. Immersionsöl nach einem der Ansprüche 1-9, wobei das
Immersionsöl ein Drei- oder Vierstoffgemisch ist.
11. Immersionsöl nach einem der Ansprüche 1-10, das eine
Zusammensetzung entsprechend der in der Tabelle II
angegebenen Beispiele 1-10 oder eine durch Mischung der
Beispiele 1-10 resultierende Zusammensetzung aufweist.
12. Verwendung einer Abmischung aus einem
Tricyclodecanalkohol, Tricyclodecanketon oder
Decahydronaphthol in Kombination mit einem Poly-ε-Capro
lacton und/oder einem Weichmacher zur Herstellung
eines Immersionsöles für Mikroskope, wobei die Viskosität
des Immersionsöles bei einer Temperatur von 20°C zwischen
200 und 900 mm2/sec. liegt.
