KAISERLICHESIMPERIAL
PATENTAMT.PATENT OFFICE.
Dabei hat man in erster Linie diejenige Substanz ins Auge gefaßt, auf derenIt is understandably not on. No efforts are made to protect the ergot (secale cornutum), a drug so interesting from a toxicological point of view, which is used as a medicinal product For almost a hundred years, the active ingredients have played a prominent role in to withdraw more or less pure condition. In the first place you have that one Substance envisaged on whose
Für keine dieser Substanzen ist ein überzeugender Beweis ihrer chemischen Individualität geliefert worden.ίο Presence is the most important therapeutically Property of the ergot, namely the ability to make the uterus contract to stimulate is based. This substance (or substances) that act on the uterus is alleged in a large number of differently presented ergot extracts in more concentrated form Form as included in the drogue. Whether and to what extent this applies to the individual This applies to preparations belonging here, there are manifold differences of opinion on this. Only the fact that none of these pharmaceutical preparations may be regarded as a chemically pure substance. The same thing but also applies to those substances which in the course of strictly scientific pharmacological Experimental studies have been found and associated with certain effects of this drug are. None of these substances is convincing evidence of their chemical individuality has been delivered.
It is. we have now succeeded in producing a nicely crystallizing substance from the ergot, who exhibits and is capable of all the properties of chemical uniformity to stimulate contractions in the pregnant uterus. The following are properties and representation of this substance mentioned C la viii.
To obtain the clavin one uses an aqueous ergot extract. This is precipitated with saturated barite solution, as long as there is still a precipitate. After allowing it to settle, the After removing the barite with carbonic acid, the filtrate was evaporated to the syrup and mixed with hot absolute alcohol, about 500 ecm to as much extract as one gets from 500 gr ergot, extracted. The alcoholic solution is cautious until crystallization begins evaporated. The crystals are sucked off and recrystallized from alcohol. The yield An Clavin varies, of course, depending on the quality of the ergot, but is always several grams for the kilo.
The clavin is a nitrogenous, in Substance readily soluble in water. An aqueous clavin solution reacts neutrally and is neither precipitated by caustic alkalis or carbonated alkalis. Is in absolute alcohol the clavin not at all in the cold, very little soluble in the heat. Spirit of wine solves that The more dilute it is, the lighter the clavin. Crystallized from 75 percent alcohol by volume the clavin in colorless needles.
In Äther, Essigäther, Petroläther ist es unlöslich. It is insoluble in ether, vinegar ether, petroleum ether.
. Stoffe sind keine chemischen Individuen. When injected into pregnant animals, the clavin causes violent uterine contractions.
All so far represented from the ergot and with the physiological we-. related effects of this drug. Substances are not chemical individuals.
1. Sphacelinsäure [Kobert, Archiv f. ίο exper. Pathol. u. Pharmak., Bd. 1. Sphacelic acid [Kobert, Archiv f. Ίο exper. Pathol. u. Pharmak., Vol. XVIII (1884) 327] is an amorphous, water-insoluble, nitrogen-free acid. She becomes the ergot which had previously been extracted with dilute hydrochloric acid, water and finally with ether was withdrawn with absolute alcohol. The alcoholic solution is after removal evaporated the dye, the residue dissolved in sodium carbonate and then the Sphacelic acid excreted by hydrochloric acid.
2. Chrysotoxin und Sphacelotoxin [Jacobj, Arch. f. exper. Pathol. u. Pharmak., Bd. Chrysotoxin and Sphacelotoxin [Jacobj, Arch. F. Exper. Pathol. u. Pharmak., Vol. XXXIX (1897), 85] are yellow, insoluble in water, soluble in alcohol and ether Resins that have the well-known uterine effect of ergot. Except for these substances Jacobj found an equally effective, nitrogenous body of in large and properties similar to those of those nitrogen-free resins. Jacobj denotes this nitrogenous body as secalin toxin and regards it as a compound of the effective nitrogen-free sphacelotoxins with one of them recognized as ineffective Base, the Secalin. The spasmotin is the water-soluble compound of the chrys ο toxin with sodium. The representation of the substances mentioned is based on their solubility in Ether and its insolubility in petroleum ether.
They are removed by ether from the ergot, which has been defatted with petroleum ether the ethereal solution obtained by fractional precipitation with petroleum ether.
3. Ergotinsäure [Kobert, Archiv f. exper. Pathol. u. Pharmak., Bd. Ergotic acid [Kobert, Archiv f. Exper. Pathol. u. Pharmak., Vol. XVIII (1884) 316] is an amorphous, nitrogen-containing, water-soluble Acid, whose representation from ergot on the precipitability by ammoniacal Lead acetic acid based. To obtain this .acid is ergot, which was previously with Ether then extracted with alcohol (both slightly acidified) with water moved out. The water extract is first precipitated and off with neutral lead acetate The ergotic acid is precipitated in the filtrate with ammonia and lead acetic acid.
4. Ecbolin und Ergotin Wenzell [cf. Kobert, Arch. f. exper. Pathol. u. Ecbolin and Ergotin Wenzell [cf. Kobert, Arch. F. Exper. Pathol. u. Pharmak., Vol XVIII (1884) 359] are amorphous basic substances obtained from concentrated aqueous ergot extracts after precipitation with neutral lead acetate, respectively in the filtrate with sublimate (Ecbolin). with phosphomolybdic acid (ergotine) were eliminated.
5. Ergotinin Tanret [vergl. Ergotin Tanret [cf. Roscoe-Schorlemmers Textbook of organic chemistry by J. W. Brühl, VI. Part 424], an in Alkaloid, which is hardly soluble in water, was obtained in such a way that ergot with 95 percent Alcohol extracted, the alcohol after adding baking soda to the weakly alkaline Reaction distilled off and the residue was extracted with ether. The essential Solution the alkaloid was removed by tartaric acid-containing water, then with potassium carbonate made free again and absorbed into ether.
6. Cornutin [Kobert, Arch. f. exper. Pathol u. Pharmak., Bd. XVIII (1884) 358]. 6. Cornutin [Kobert, Arch. F. Exper. Pathol and Pharmak., Vol. XVIII (1884) 358]. The representation of this alkaloid is based on its easy solubility in alcohol and on its property of being shaken out of the alkaline aqueous solutions with vinegar ether. To obtain the cornutin, ergot is extracted with dilute g ^ hydrochloric acid, this extract is evaporated after the acid has been blunted, and the residue is extracted with alcohol. The residue obtained after evaporation of the alcoholic solution is made alkaline with sodium carbonate go and extracted with ethyl ether.
