Description

Gegenstand des Hauptpatents -ist die Verwendung von Glutathion als Stabilisierungsmittel für Di- oder Triphosphopyridinnucleotid in der reduzierten Form in enzymatischen Testreagenzien.
In Erweiterung dieser Erfindung wurde festgestellt, daß nicht nur Glutathion, sondern auch andere SH-gruppenhaltige Verbindungen, welche die Testreaktion nicht stören, eine Stabilisierung dieser enzymatischen Präparate bewirken.
Gegenstand des vorliegenden Zusatzpatents ist somit die Verwendung von die Testreaktion nicht störenden Sulfhydrylgruppen enthaltenden Verbindungen mit Ausnahme von Glutathion als Stabilisierungsmittel für Di- oder Triphosphopyridinnucleotid in der reduzierten Form in enzymatischen Testreagenzien.
Besonders vorteilhaft ist die Verwendung von Cystein, Cystein-Carbamid und Acetyl-Cystein als Stabilisierungsmittel in kombinierten Testreagenzien, welche insbesondere DPNH enthalten. Die Stabilisierungsmittel dürfen die Testreaktion als solche natürlich nicht stören oder beeinflussen, was durch einen einfachen Vorversuch ohne Schwierigkeiten von jedem Fachmann festgestellt werden kann.
Ein Vorteil gegenüber dem Gegenstand des Hauptpatents ist darin zu sehen, daß die beim vorliegenden Erfindungsgegenstand bevorzugt verwendeten Stoffe wie Cystein, Cystein-Carbamid und Acetyl-Cystein im Preise wesentlich niedriger liegen als das beim Hauptpatent verwendete Glutathion.
Die folgenden Beispiele zeigen mehrere Kombinationen, welche über längere Zeiträume hinaus stabile lyophilisierte Präparate ergaben.
1. Bestimmung
der Glutamat-pyruvat-transaminase (GPT)
2,43 g Na0HPO4-2 H.,O,
0,21 g NaH2PO4 · 2 H2O,
2,05 g DL-Alanin,
0,35 g a-Ketoglutarat,
0,30 g Cystein,
0,30 g Serum-Albumin,
0,05 g NADH,
0,0015 g Lactat-Dehydrogenase,
werden in 90 ml bidest. Wasser gelöst und der pH-Wert mit verdünnter Natronlauge auf 7,6 eingestellt. Dann wird die Lösung mit bedest. Wasser auf 100 ml aufgefüllt, anschließend eingefroren und gefriergetrocknet.
" 2. Bestimmung der Lactat-Dehydrogenase (LDH)
2,43 g Na2HPO4-2H2O,
0,21g NaH2PO4-2 H2O,
0,004 g Natriumpyruvat,
0,30 g Cystein-Carbamid, 0,30 g Serum-Albumin,
0,05 g NADH.
Aufbereitung wie im Beispiel 1 angegeben.
3. Bestimmung von Aldolase (ALD)
2,43 g g Na2HPO4-2 H0O,
0,21 g g NaH2PO4-2H2O,
0,14 g g Fructose-1,6-diphosphat,
0,005 g Jodacetat,
0,30 Cystein,
0,30 Serum-Albumin,
0,05 NADH,
0,0015 g Glycero-phosphat-dehydrogenase, 0,0015 g Triose-phosphat-isomerase.
Aufbereitung wie im Beispiel 1 angegeben.

Claims (2)

Patentansprüche:
1. Verwendung von die Testreaktion nicht störenden Sulfhydrylgruppen enthaltenden Verbindungen mit Ausnahme von Glutathion als Stabilisierungsmittel für Di- oder Triphosphopyridinnucleotid in der reduzierten Form in enzymatischen Testreagenzien.
2. Verwendung von Cystein, Cystein-Carbamid und Acetyl-Cystein als Stabilisierungsmittel nach Anspruch 1. *
