Abstract

Adrenocorticotrophin preparations having a prolonged action are prepared, in the form of a stable suspension suitable for implantation or injection intramuscularly or subcutaneously, by adding a protamine sulphate and a water-soluble zinc salt to an aqueous solution of adrenocorticophin and adjusting the pH of the solution to between 6 and 8 by means of a buffer, e.g. trisodium phosphate. The protamine sulphate addition may take the form of an aqueous solution of concentration greater than one per cent and preferably greater than two per cent. The specific activity of the hormone is greater than 2 I.U. per mg. and the concentration of the hormone solution is such that the concentration in I.U. pre ml. exceeds the amount corresponding to five times the specific activity in I.U. per mg. The suspension may also be formed by dissolving the hormone in a portion of the protamine sulphate solution and adding a corresponding quantity of the zinc salt e.g. zinc sulphate, and buffer solution to attain a pH between 6 and 8. The remainder of the protamine sulphate solution is then added, followed by the remainder of the zinc and buffer solution. Glucose and tricresol may also be present.
