Description
The The present invention relates to dietary supplements containing alpha-keto analogs of branched-chain amino acids to help build muscle, increase muscle performance and improve overall well-being while at the same time Relief of metabolism for nitrogen detoxification by reduced nitrogen supply and improved nitrogen retention in the body.
Alanin ist weiterhin ein wichtiger Vorläufer für die Gluconeogenese und Glutamin wirkt als Stickstofftransporter zwischen den Organen.physical Physical inactivity is a risk factor leading to a decreased physical Performance and thus a reduced quality of life can lead. To the decay of physical performance prevent or rebuild is a physical Training essential, being a set of cellular Processes, such as B. muscle damage and degradation, muscle regeneration, muscle hypertrophy and muscle fiber transformation, expire. In the cellular Processes of energy or protein metabolism play a crucial role. Consequently, the supply of amino acids plays a crucial Role with regard to the metabolic processes occurring in the muscle tissue. In particular, the branched-chain amino acids valine, leucine and isoleucine are essential substrates and important regulators in protein biosynthesis and the main source of nitrogen for the glutamine and alanine synthesis in skeletal muscle. Alanin is still on important precursor for gluconeogenesis and glutamine acts as a nitrogen transporter between the organs.
Of the Average protein requirement is about 660 mg / kg Body weight, however, by physical Training can be increased significantly. The need for protein can usually be covered by a balanced diet which is not easy to reach. Body training leads, caused by increased protein degradation and decreased protein synthesis, to a changed need of nutrients, continues to result in a changed Metabolic position, the z. B. on the influence of physical Exercises due to the hormone system and ultimately there is a lack of knowledge about a suitable one Diet with increasing physical stress especially in relation to age, so that fast a malnutrition can occur.
Out For these reasons, the use of a dietary supplement appears in individuals who are under physical strain, to be meaningful. In this context, studies have already been performed with different results that the Influence of creatinine supplementation on performance the subjects had the subject. Furthermore, it is known that high carbohydrate intake promotes muscle recovery can be.
Alpha-keto acids Branched-chain amino acids also play a role important role in amino acid metabolism, especially in the Skeletal muscle and in the liver. One third of the muscle protein exists from the branched-chain amino acids released by the body can not be formed, but taken with food Need to become. In the muscle become particularly with physical effort Proteins are constantly being built up and broken down, whereby during degradation the amino acids the corresponding alpha-keto acid formed by transfer of the amino group to a carrier becomes. The obtained keto acid can then enzymatically for energy be further oxidized. The carrier is transported to the liver and releases there toxic ammonia, which is converted into urea and must be excreted via the kidney.
There is a need in the art for nutritional supplements, which promote the well-being after sporting activities, the Strengthen muscle building or the performance of the muscle and permanently reduce the nitrogen load of the metabolism.
The Task is by providing nutritional supplements dissolved, the at least one alpha-keto acid selected from the group alpha-ketoisocaproate (KIC), alpha-ketoisovalerate (KIV) and alpha-keto-beta-methylvalerate (KMV) and / or a salt thereof contain alpha-keto acids, wherein the supplement optionally additionally one or more amino acids, especially leucine, isoleucine and / or valine, but may contain otherwise nitrogen-free.
As in essential nitrogen-free "are dietary supplements to see, in addition to the amino acids mentioned no nitrogenous Contain formulation additives, in particular no nitrogen-containing Cations, such as. B. cationic amino acids or dibasic Amino acids from the group arginine, lysine, histidine or Ornithine, or other nitrogenous additives in appreciable Scope. Essentially nitrogen-free, however, are dietary supplements, which comprise nitrogenous additives, which in a very small amount Amount of less than 5 mg (daily dose) of the dietary supplement can be added, such as. B. nitrogenous vitamins. However, dietary supplements are preferred have no nitrogen-containing constituents, especially none cationic amino acids. The term "nitrogen-free" on the supplement itself. Foods (Functional Foods) containing the claimed dietary supplement can of course also nitrogenous ingredients exhibit. Preference is given in the food with the inventive Dietary supplements are modified, no further Food supplements included, especially none Supplements that increased an additional Include supplementation with nitrogenous compounds. In particular, there should be no artificial increase in salary done on cationic amino acids in the food.
In addition to the alpha-keto acids KIC, KIV and KMV, their salts, if they contain nitrogen-free cations, can also be present in the dietary supplement according to the invention. Suitable salts are in particular the alkali or alkaline earth metal salts, in particular the Na + , K + , Ca 2 + and Mg 2 + salts, of the mentioned alpha-keto acids.
A preferred embodiment is nutritional supplements containing a combination of alpha-ketoisocaproate and alpha-ketoisovalerate or alpha-keto-beta-methylvalerate or a combination of alpha-ketoisovalerate and alpha-keto-beta-methylvalerate or a combination of all three alpha-keto acids or their salts. Preferably, a KMV / KIV weight ratio or KIC / KIV weight ratio or a KIC / KMV weight ratio of 50: 1 to 1 :: 50, preferably from 5: 1 to 1: 5 set in the dietary supplement. Preferred weight ratios are between 3: 1 and 1: 1 for KIC / KIV, between 3: 1 and 1: 1 for KIC / KMV, and between 2: 1 and 1: 2 for KIV / KMV. Corresponding quantitative ratios are also advantageous for a KIC / KIV / KMV mixture. A particularly suitable combination of KIC / KIV / KMV represents a weight ratio of 2: 1: 1, wherein said proportions can be varied +/- 10%. The weight ratios mentioned are particularly suitable for rapid regenerative muscle growth. The daily dose of alpha-ketoacids taken up by the dietary supplement should not exceed the levels of 2000 mg / kg body weight for KIC, 1000 mg / kg body weight for KIV and 1500 mg / kg body weight for BMV, preferably doses of between 2.5 mg / kg and 500 mg / kg body weight for each of the three alpha-keto acids. Particularly preferred dosages are in the range of 25 mg / kg to 250 mg / kg body weight for KIC, KIV and KMV, so that in adults (eg at a body weight between 50 and 100 kg) an approximate preferred total amount per ingested alpha Keto acid from 1.25 g to 25 g. Specifically, the following preferred dosages are given for the alpha-keto acids: for KIC between 50 mg / kg to 200 mg / kg, for KIV between 25 mg / kg to 150 mg / kg and for KMV between 25 mg / kg to 100 mg / kg. Usual daily dosages are in the range of 0.5 g to 50 g per alpha-keto acid per day. With regard to the amount of cations contained in the dietary supplement should preferably not more than 10 g, more preferably between 0.5 g and 2 g of Na + , K + ions, preferably not more than 5 g, more preferably between 0.2 and 2.5 g of Ca 2+ ions and not more than 2 g, more preferably between 0.2 and 1 g, of the corresponding Mg 2+ ions.
Farther can use the dietary supplement described here also the branched-chain amino acids leucine, isoleucine and valine are added. Preference is given to the amino acids mentioned but used in small quantities. With regard to the present Invention should be added to the dietary supplement Amount a ratio of 1: 0.5, better of 1: 0.1 (alpha-keto acid: corresponding Amino acid), should not be exceeded. Especially However, preferred dietary supplements contain no leucine, isoleucine or valine.
Especially preferred dietary supplements contain more of naturally occurring amino acids derived Keto acids or their salts, more preferably alpha-ketoglutaric acid or their salts, especially alpha-ketoglutarate (AKG). The Use of the dietary supplements or Food can basically be possible all day, But especially during or after a physical Recommended load. The physical training effects a muscular adaptation including muscle damage, muscle hypertrophy and muscle transformation. Here is a training session as a combination from a training phase and a regeneration phase. A suboptimal embodiment of the training unit may, for. B. too an overtraining syndrome that results in a long-lasting fatigue with reduced physical Efficiency expresses. Such an overtraining syndrome is often caused or intensified by malnutrition.
With Help the claimed supplements will be the Ammonia detoxification in the muscle, which is caused, inter alia, by the protein and amino acid removal in muscle becomes necessary. By transfer of released amino groups on the keto acids the corresponding amino acids are generated and stand in turn available for muscle growth and the energy-consuming nitrogen detoxification and excretion Liver and kidney is reduced. Accordingly, the throughput nitrogen-containing degradation products, such as. As urea, in the blood or Reduced urine. At the same time, the performance becomes the musculature increased or the muscle build-up by the dietary supplements supported as transaminated by the administered Converted keto acids in the muscle into the corresponding amino acids who are there for anabolic reactions be available. Finally, it turns a faster regeneration of muscle tissue, and physical performance as well as the general well-being is improved. Both the support protein synthesis in muscle as well as ammonia detoxification particularly supported by the mentioned alpha-keto acids, as these are selectively absorbed by the muscle tissue. The Ammonia detoxification is particularly efficient when the claimed Dietary supplement additionally alpha-ketoglutarate (AKG) included.
Durch eine schnellere Regeneration lässt sich auch ein erhöhter Trainingsumfang bzw. eine erhöhte Trainingsintensität oder -häufigkeit realisieren, ohne die oben genannten negativen Effekte eines Übertrainings zu provozieren.consequently is the nitrogen balance of the body, which under one physical stress is, with the invention Dietary supplements improved in two ways: For one thing, the loss of nitrogen is due to muscle loss processes reduced at a physical load, resulting in a improved muscle regeneration or muscle repair, on the other hand, the ammonia detoxification is accelerated, resulting in a faster Muscle recovery or reduced muscle fatigue leads. By a faster regeneration leaves also an increased training scope or an increased Realize training intensity or frequency, without the above negative effects of overtraining to provoke.
In summary can be said that the substitution with the inventive Dietary supplements of a malnutrition prevents, to Satisfying the increased need for nutrients, which promotes muscular regeneration and overtraining prevents, which leads to a strengthening of the musculature and the performance and well-being of a person elevated.

In order to evaluate the muscle function in general, the maximum oxygen uptake (VO 2max ) is measured under load of a subject. For this purpose, the load on a cycle ergometer is increased by means of a ramp protocol (for example, beginning 0 watt, continuous increase in power at 50 watt / min until subjective exhaustion occurs). In addition, the heart rate can be determined synchronously by means of electrocardiography. Oxygen uptake is calculated from online minute ventilation and the difference in oxygen concentration between inspiratory and expiratory gases.
To determine the endurance capacity, the Individual Anaerobic Aerobic Threshold (IAAS) is determined. This is done by measuring a lactate power curve with a treadmill test (training phase protocol: start 6 km / h, increase 2 km / h, which corresponds to an increase of about 25-50 watts / min, stage duration 3 min.) or with a bicycle ergometer (initial stage 50 watts, increase 25 watts, stage duration 3 min, until subjective exhaustion is reached). Blood samples are taken before and after a 30-second break (eg whole blood (hemolyzed) from hyperemic capillary blood (earlobe)) and the glucose and lactate concentration using a YSI 2300 STAT plus analyzer from YSI Life Sciences, Yellow Springs, USA and the maximum oxygen uptake (VO 2 max value) was determined spirometrically using a K4 measuring instrument from Cosmed (Rome, Italy).
The measurement of the bounce improvement can be determined by means of a bounce plate of the company Kistler, Winterthur, Switzerland.
