Abstract
Verfahren
zum Nachweis eines G-Protein-gekoppelten Rezeptor (GPCR) Liganden
in einer Testprobe, umfassend die Schritte:
(a) Bereitstellen einer Zelle umfassend einen GPCR und ein Arrestin;
(b) Exponieren der Zelle gegenüber der Testprobe; und
(c) Bestimmen der zellulären Verteilung des GPCR oder Arrestins in Anwesenheit der Testprobe, wobei die Testprobe eine biologische Probe, eine Umweltprobe oder eine davon abgeleitete Probe ist.
(a) Bereitstellen einer Zelle umfassend einen GPCR und ein Arrestin;
(b) Exponieren der Zelle gegenüber der Testprobe; und
(c) Bestimmen der zellulären Verteilung des GPCR oder Arrestins in Anwesenheit der Testprobe, wobei die Testprobe eine biologische Probe, eine Umweltprobe oder eine davon abgeleitete Probe ist.
Claims (64)
- Verfahren zum Nachweis eines G-Protein-gekoppelten Rezeptor (GPCR) Liganden in einer Testprobe, umfassend die Schritte: (a) Bereitstellen einer Zelle umfassend einen GPCR und ein Arrestin; (b) Exponieren der Zelle gegenüber der Testprobe; und (c) Bestimmen der zellulären Verteilung des GPCR oder Arrestins in Anwesenheit der Testprobe, wobei die Testprobe eine biologische Probe, eine Umweltprobe oder eine davon abgeleitete Probe ist.
- Verfahren nach Anspruch 1, wobei die zelluläre Verteilung des GPCR oder des Arrestins in Anwesenheit der Testprobe mit der zellulären Verteilung des GPCR oder Arrestins in Abwesenheit der Testprobe verglichen wird.
- Verfahren nach Anspruch 1, wobei der GPCR oder das Arrestin nachweisbar markiert ist, ein anderes endogenes Molekül nachweisbar markiert ist oder ein anderes exogenes Molekül nachweisbar markiert ist.
- Verfahren nach Anspruch 3, wobei die Verteilung der nachweisbar markierten Moleküle der zellulären Verteilung der GPCR- oder der Arrestinproteine entspricht.
- Verfahren nach Anspruch 1, wobei die zelluläre Verteilung des GPCR oder Arrestins zu verschiedenen Zeitpunkten nach der Exposition gegenüber der Testprobe bestimmt wird.
- Verfahren nach Anspruch 1, wobei die zelluläre Verteilung des GPCR oder Arrestins nach der Exposition gegenüber verschiedenen Konzentrationen der Testprobe bestimmt wird.
- Verfahren nach Anspruch 4, wobei die zelluläre Verteilung der nachweisbar markierten Moleküle quantifiziert wird.
- Verfahren nach Anspruch 7, wobei die Konzentration des Liganden in der Testprobe durch Vergleichen der zellulären Verteilung des GPCR oder Arrestins in Anwesenheit der Testprobe mit der zellulären Verteilung des GPCR oder Arrestins in Anwesenheit einer bekannten Konzentration des Liganden, quantifiziert wird.
- Verfahren nach Anspruch 1, wobei die biologische Probe Serum, Gewebe, Blut oder Harn oder davon abgeleitet ist.
- Verfahren nach Anspruch 1, wobei der GPCR CCK-B oder CCK-A ist.
- Verfahren nach Anspruch 1, wobei der Ligand Gastrin, Präprogastrin, gespaltenes Präprogastrin, Gastrin-34, Gastrin-17, Pentagastrin, Progastrin, Glycin-verlängertes Gastrin-17, Glycin-verlängertes Gastrin-34, Gastrin-71, Gastrin-6, hG17, eine Verbindung mit einem amidierten Tetrapeptid der Sequenz Trp-Met-Asp-Phe-NH2 oder eine andere bioaktive Isoform von Gastrin, ist.
- Verfahren nach Anspruch 1, wobei der GPCR ein Muskarinrezeptor ist.
- Verfahren nach Anspruch 1, wobei der Ligand Acetylcholin ist.
- Verfahren nach Anspruch 3, wobei das markierte Molekül im Cytosol, in der Plasmamembran, in Clathrin-beschichteten Vertiefungen, in endozytischen Vesikeln oder Endosomen angeordnet ist.
- Verfahren nach Anspruch 4, wobei ein Anstieg in der lokalen Konzentration des markierten Moleküls zu einem Anstieg in der lokalen Signalintensität führt.
- Verfahren nach Anspruch 15, wobei die Signalintensität des markierten Moleküls in der Plasmamembran, den Clathrin-beschichteten Vertiefungen, den endozytischen Vesikeln oder Endosomen bezüglich des Grades an Signalintensität im Cytosol ansteigt.
- Verfahren nach Anspruch 15, wobei die lokale Signalintensität in Anwesenheit einer erhöhten Konzentration des Liganden in der Testprobe ansteigt.
- Verfahren nach Anspruch 8, wobei die Konzentration des Liganden in der Testprobe einen Krankheitszustand anzeigt.
- Verfahren nach Anspruch 8, wobei die Konzentration des Liganden in der Testprobe die Anwesenheit einer Verbindung in der Testprobe anzeigt, welche die Ligandenkonzentration verändert.
- Verfahren nach Anspruch 8, wobei die Konzentration des Liganden in der Testprobe die Anwesenheit einer Verbindung in der Testprobe anzeigt, die Acetylcholin modifiziert.
- Verfahren nach Anspruch 8, wobei die Konzentration des Liganden in der Testprobe die Anwesenheit einer Verbindung anzeigt, die eine Acetylcholinesterase inhibiert.
- Verfahren nach Anspruch 3, wobei das nachweisbare Molekül ein Radioisotop, ein Epitopetag, eine Affinitätsmarkierung, ein Enzym, eine Fluoreszenzgruppe oder eine Chemilumineszenzgruppe ist.
- Verfahren nach Anspruch 3, wobei das Molekül nachweisbar markiert ist aufgrund seiner Interaktion mit einem anderen Molekül, das nachweisbar markiert sein kann.
- Verfahren zum Überwachen eines GPCR Liganden in einem Säugetier, umfassend die Schritte: (a) Bereitstellen einer Zelle umfassend einen GPCR und ein Arrestin; (b) Exponieren der Zelle gegenüber der Testprobe; und (c) Bestimmen der zellulären Verteilung des GPCR oder Arrestins in Anwesenheit der Testprobe, wobei die Testprobe eine biologische Probe darstellt, die von dem Säugetier abgeleitet ist.
- Verfahren nach Anspruch 24, wobei die Konzentration des Liganden in der Testprobe durch Vergleichen der zellulären Verteilung des GPCR oder Arrestins in Anwesenheit der Testprobe mit der zellulären Verteilung des GPCR oder Arrestins in Anwesenheit einer bekannten Konzentration des Liganden, quantifiziert wird.
- Verfahren nach Anspruch 24, wobei ein klinischer Zustand überwacht wird.
- Verfahren nach Anspruch 26, wobei der klinische Zustand die Anwesenheit eines Krankheitszustandes anzeigt.
- Verfahren nach Anspruch 26, wobei der klinische Zustand anzeigt, dass das Subjekt eine Störung aufweist oder gefährdet ist, eine Störung zu entwickeln.
- Verfahren nach Anspruch 26, wobei der klinische Zustand gastrointestinaler Krebs, Hypergastrinämie, atrophische Gastritis, gastritische Geschwüre, bösartige Tumore oder eine andere GPCR-verwandte Krankheit ist.
- Verfahren nach Anspruch 24, wobei das Säugetier einer andauernden säureunterdrückenden Behandlung unterzogen wird.
- Verfahren nach Anspruch 1, wobei die Zelle ein Protein exprimiert, das die Internalisierung des GPCR erhöht.
- Verfahren nach Anspruch 14, wobei der GPCR modifiziert ist, was zu einer erhöhten Konzentration des markierten Moleküls an der Plasmamembran, den Clathrin-beschichteten Vertiefungen, den endozytischen Vesikeln oder den Endosomen führt.
- Verfahren nach Anspruch 1, wobei die Zelle weiterhin eine G-Proteingekoppelte Rezeptorkinase (GRK) aufweist.
- Einzelzellbiosensor umfassend eine Zelle, die Arrestin oder wenigstens einen GPCR überexprimiert, wobei der GPCR, das Arrestin oder die Zelle nachweisbar für die Überwachung der Internalisierung des GPCR markiert ist.
- Verfahren zum Nachweis eines GPCR Liganden in einer Testprobe, umfassend die Schritte: (a) Bereitstellen eines Einzelzellbiosensors umfassend eine Zelle, die Arrestin und wenigstens einen GPCR überexprimiert; (b) Exponieren des Biosensors gegenüber der Testprobe; und (c) Bestimmen der zellulären Verteilung des GPCR oder Arrestins in Anwesenheit der Testprobe, wobei die Testprobe eine biologische Probe, eine Umweltprobe oder eine davon abgeleitete Probe ist.
- Einzelzellbiosensor nach Anspruch 34, wobei der GPCR ausgewählt ist aus der Gruppe bestehend aus CCK-A, CCK-B und Muskarinrezeptor.
- Einzelzellbiosensor nach Anspruch 34, wobei das Arrestin an ein grün fluoreszierendes Protein (GFP) konjugiert ist.
- Einzelzellbiosensor nach Anspruch 34, in dem der Biosensor eine erhöhte Sensitivität aufgrund längerer Inkubationszeit, erhöhter Konzentration der Testprobe, GPCR Mutation oder CPCR Antikörpern aufweist.
- Verfahren zum Verändern der GPCR Internalisierung, umfassend Bereitstellen einer wirksamen Menge eines Antagonisten von CCK-B an die Zellen.
- Verfahren nach Anspruch 1, wobei die zelluläre Verteilung durch Flusszytometrie sichtbar gemacht wird.
- Verfahren nach Anspruch 1, wobei die zelluläre Verteilung durch konfokale Fluoreszenzmikroskopie sichtbar gemacht wird.
- Verfahren nach Anspruch 1, wobei ein Computer ein Bild der zellulären Verteilung auswertet.
- Verfahren nach Anspruch 4, wobei die Verteilung des nachweisbaren Moleküls quantifiziert wird.
- Verfahren nach Anspruch 1, wobei die Testprobe einen Liganden des GPCR aufweist.
- Verfahren nach Anspruch 1, wobei die Testprobe einen Antagonisten des GPCR aufweist.
- Verfahren zum Nachweis einer Verbindung, die einen GPCR Liganden in einer Testprobe moduliert, umfassend die Schritte: (a) Bereitstellen einer Zelle umfassend einen GPCR und ein Arrestin; (b) Exponieren der Zelle gegenüber der Testprobe; und (c) Bestimmen der zellulären Verteilung des GPCR oder Arrestins in Anwesenheit der Testprobe, wobei die Testprobe eine biologische Probe, eine Umweltprobe oder eine davon abgeleitete Probe ist.
- Verfahren nach Anspruch 46, wobei die zelluläre Verteilung die Anwesenheit einer Verbindung, die einen GPCR Liganden moduliert, anzeigt.
- Verfahren zum kontinuierlichen Screenen von GPCR Liganden in einer Testprobe, umfassend die Schritte: (a) Bereitstellen einer Zelle umfassend einen GPCR und ein Arrestin; (b) Exponieren der Zelle gegenüber der Testprobe; (c) Bestimmen der zellulären Verteilung des GPCR oder Arrestins in Anwesenheit der Testprobe, wobei die Testprobe eine biologische Probe, eine Umweltprobe oder eine davon abgeleitete Probe ist; und (d) Ersetzen der Zelle durch eine andere Zelle, die einen GPCR oder ein Arrestin aufweist.
- Verfahren zum Nachweis eines Inhibitors von Acetylcholinesterase in einer Testprobe, umfassend die Schritte: (a) Bereitstellen einer Zelle, die einen Muskarinrezeptor und ein Arrestin exprimiert; (b) Bereitstellen einer Mischung, die eine Testprobe, Acetylcholinesterase und einen Agonisten des Muskarinrezeptors aufweist, wobei der Agonist sensitiv gegenüber Acetylcholinesterase ist; (c) Exponieren der Zelle gegenüber der Mischung; und (d) Bestimmen der zellulären Verteilung des Muskarinrezeptors oder Arrestins in Anwesenheit der Testprobe, wobei die Testprobe eine biologische Probe, eine Umweltprobe oder eine davon abgeleitete Probe ist.
- Verfahren nach Anspruch 49, wobei der Agonist Acetylcholin ist.
- Verfahren nach Anspruch 1, wobei die Testprobe weiterhin Acetylcholin und Acetylcholinesterase aufweist.
- Verfahren nach Anspruch 1, wobei die Testprobe weiterhin einen Agonisten aufweist.
- Verfahren nach Anspruch 1, wobei der Ligand identifiziert wurde.
- Verfahren nach Anspruch 1, wobei multiple bioaktive Isoformen der GPCR Liganden in der Testprobe nachgewiesen werden.
- Verfahren nach Anspruch 10, wobei die Testprobe von einem Patienten mit Hypergastrinämie abgeleitet ist.
- Verfahren nach Anspruch 11, wobei die Gastrinkonzentration weniger als 10 nM beträgt.
- Verfahren nach Anspruch 1, wobei die Testprobe heterogen ist.
- Verfahren nach Anspruch 1, wobei die zelluläre Verteilung nach 15–30 Minuten Exposition gegenüber der Testprobe bestimmt wird.
- Verfahren nach Anspruch 1, wobei die zelluläre Verteilung nach 1 Stunde Exposition gegenüber der Testprobe bestimmt wird.
- Verfahren nach Anspruch 1, wobei die Zelle der Testprobe bei einer Temperatur von 37°C exponiert wird.
- Verfahren zum Nachweis einer Verbindung, welche die GPCR Internalisierung in einer Testprobe moduliert, umfassend die Schritte: (a) Bereitstellen einer Zelle umfassend einen GPCR und ein Arrestin; (b) Exponieren der Zelle gegenüber der Testprobe; und (c) Bestimmen der zellulären Verteilung des GPCR oder Arrestins in Anwesenheit der Testprobe, wobei die Testprobe eine biologische Probe, eine Umweltprobe oder eine davon abgeleitete Probe ist.
- Verfahren zum Nachweis einer Verbindung, welche die GPCR Internalisierung in einer Testprobe moduliert, umfassend die Schritte: (a) Bereitstellen einer Zelle umfassend einen GPCR und ein Arrestin; (b) Exponieren der Zelle gegenüber einem Agonisten; (c) Exponieren der Zelle gegenüber der Testprobe; und (d) Bestimmen der zellulären Verteilung des GPCR oder Arrestins in Anwesenheit der Testprobe, wobei die Testprobe eine biologische Probe, eine Umweltprobe oder eine davon abgeleitete Probe ist.
- Bioarray umfassend wenigstens einen Biosensor nach Anspruch 34.
- Bioarray nach Anspruch 63, wobei multiple GPCR Liganden nachgewiesen werden.
