Abstract
Η παρούσα εφεύρεση αφορά σε μια άποψη μια μέθοδο για ανάλυση της συχνότητας αλληλεπίδρασης μιας αλληλουχίας νουκλεοτιδίων στόχου με μια ή περισσότερες αλληλουχίες νουκλεοτιδίων που ενδιαφέρουν (π.χ. έναν ή περισσότερους γονιδιωματικούς τόπους) η οποία περιλαμβάνει τα στάδια: (α) παροχής ενός δείγματος διασταυρούμενα συνδεδεμένου DNA, (β) πέψης του διασταυρούμενα συνδεδεμένου DNA με ένα πρωτογενές ένζυμο περιορισμού, (γ) σύνδεσης των διασταυρούμενα συνδεδεμένων αλληλουχιών νουκλεοτιδίων, (δ) αντιστροφής της διασταυρούμενης σύνδεσης, (ε) πέψης των αλληλουχιών νουκλεοτιδίων με ένα δευτερογενές ένζυμο περιορισμού, (στ) σύνδεσης μιας ή περισσότερων αλληλουχιών DNA γνωστής νουκλεοτιδικής σύνθεσης στη διαθέσιμη τοποθεσία(-ες) δευτερογενούς ενζύμου περιορισμού που είναι παρακείμενες στη μια ή περισσότερες αλληλουχίες νουκλεοτιδίων που ενδιαφέρουν, (ζ) ενίσχυσης της μιας ή περισσότερων αλληλουχιών νουκλεοτιδίων που ενδιαφέρουν χρησιμοποιώντας τουλάχιστον δυο ολιγονουκλεοτιδικούς˙ εκκινητές, όπου κάθε εκκινητής υβριδίζεται προς τις αλληλουχίες DNA οι οποίες είναι παρακείμενες στις αλληλουχίες νουκλεοτιδίων που ενδιαφέρουν, (η) υβριδισμού της ενισχυμένης αλληλουχίας(ών) προς μια συστοιχία, και (θ) προσδιορισμού της συχνότητας αλληλεπίδρασης μεταξύ των αλληλουχιών DNA.
