Abstract

Το γονιδίωμα των μη ανθρώπινων, μεταλλαγμένων θηλαστικών, τα οποία δεν διαθέτουν έναν ενδογενή Σίγμα υποδοχέα, περιέχει μια μετάλλαξη, η οποία περιλαμβάνει μια ρήξη σε ένα ενδογενές γονίδιο Σίγμα υποδοχέα, όπου η αναφερθείσα ρήξη γονίδιου προκαλεί ένα μετάλλαγμα, το οποίο δεν διαθέτει ανιχνεύσιμα επίπεδα ενδογενούς Σίγμα υποδοχέα. Το μετάλλαγμα μπορεί να χρησιμοποιηθεί ως ζώο ελέγχου για in vivo δοκιμές, καθώς και ως πηγή κυττάρων, τα οποία μπορούν να χρησιμοποιηθούν σε in vitro δοκιμές. Τα μεταλλάγματα τα οποία δεν διαθέτουν Σίγμα-1 υποδοχέα μπορούν να χρησιμοποιηθούν ως μοντέλα για την in vivo μελέτη των διαταραχών του κεντρικού νευρικού συστήματος, των διαταραχών της μνήμης, των καταστάσεων στρες και εθισμών σε ουσίες, των διαδικασιών αναλγησίας και νευροπροστασίας. Τα μεταλλάγματα που δεν διαθέτουν Σίγμα-2 υποδοχέα μπορούν να χρησιμοποιηθούν για τη μελέτη διαγνωστικών ή θεραπευτικών εργαλείων για την καταπολέμηση του καρκίνου και/ ή εκφυλιστικών διαδικασιών και/ ή τον σχεδιασμό ενώσεων που μπορούν να προλάβουν, να μειώσουν ή να μετριάσουν τη δευτερογενή παθολογία που σχετίζεται με τη χορήγηση νευροληπτικών αντιδραστηρίων.
