电介质微球或平面波导环或圆盘形共振器在传感应用方面(包括生物传感)越来越引起人们的注意。这些类型的共振器尺寸通常对于微球而言为约20μm到几毫米,对于环或圆盘形共振器而言为5μm到几十微米。这些微球和环或圆盘形共振器通常称作微谐振器。在大部分常见的微谐振器基生物传感器结构中,微谐振器接近于光波导(如光纤),其几何形状经过特别调整,例如为锥形或蚀刻到尺寸为1-5μm。
对波导的锥形修改导致在波导外部具有大量光场,因此光可能耦合进微谐振器并激励它的本征模式,通常称作回音壁模式(WGM)。当使用由低损耗材料制成并具有高表面质量的微谐振器时,在WGM中光传播的传播损耗极低,而质量指标极高,也称作Q-指标:其值高达109。由于较高的Q-指标,光可以在微球共振器内部循环极长时间,从而在腔模式中产生极大的场增强和极长的有效光路。这使得这种装置适用于非线性光学和传感应用中。在传感应用中,待感测的样品置于球表面上,在那里它们与微谐振器外部的WGM的渐消部分相互作用。由于场增强以及光和样品之间的相互作用长度增大,因而微谐振器基光学传感器具有高灵敏度和/或低检测限的特征。
发明概述
存在不同的应用需要提高引入到微谐振器表面上的材料的量。使用多孔表面可以使更大量的材料被捕获到微谐振器表面上或与其接近,从而提高材料和微谐振器中的光传播间的光学相互作用。因此,本发明的一部分涉及至少在微谐振器的表面上包括多孔材料。

待检测的分析物样品被加到微谐振器的表面。在一个方法中,至少微谐振器表面的多孔区域,或可选择地全部微谐振器,与含有分析物的流体接触。例如,流体可以是含有分析物的液体溶液或悬浮液,或可以是含有分析物的气态混合物。通过渐消耦合实现WGM和微谐振器表面上的分析物之间的光-物相互作用。
因为光-物相互作用与WGM的局部电场的强度成比例,所以提高灵敏度的一种方法是使样品经受最大电场,因此与WGM发生最大相互作用。此外,微谐振器内部和外部间的折射率变化取决于吸附表面积和样品分子量。需要较大的相互作用面积,特别是对于检测形成单层或极薄层的小分子量分子而言。
根据本发明,在微谐振器中,至少使用多孔材料形成微谐振器的表面。例如,微谐振器可以完全由多孔材料形成,或从非多孔材料的芯形成,同时在非多孔芯包围有多孔材料涂层。在任一种情况下,有利的是,多孔材料的孔尺寸小于探针光的波长,从而避免对Q-指标有任何明显的影响。
层状多孔微谐振器300的一个特定实施方案示意性地表明在图3中,其具有在芯304外面的多孔层302。芯304可以由非多孔材料形成,或者可以是中空的芯。与多孔层302被空气(或溶液,如水)所代替的情况相比,WGM的电场朝向多孔层302移动。因此,电场有较大部分在多孔层302中,如图3所示,在进行感测时,可以使分析物接触到高水平电场,由此提高了光和分析物之间的相互作用。此外,使用多孔层302可以使分析物在WGM 306的区域中更为密集,从而进一步提高了光-分析物相互作用。分析物的密度至少部分地取决于微谐振器的表面积:表面积增加可以使更多位点粘附分析物,因为分析物倾向于在微谐振器上形成单层。使用多孔表面使表面积增大。在一个例子中,非多孔粒子的表面积约为1m2/克材料(1m2/g)。多孔材料的表面积可以是1000m2/g或更大。如果多孔材料是芯周围的多孔材料层,那么多孔层的厚度可以小于λ,其中λ是被用于激励微谐振器的WGM的光的波长。多孔层的厚度也可以小于λ/10。
层状微谐振器400设计的变形示意性地表明在图4中。在此实施方案中,多孔层402的厚度在芯404周围并不总是恒量。这种实施方案的一个优点在于,当多孔层402的最薄部分406接近于波导408时,当在WGM 410和波导408间耦合光时发生的散射损耗减小。然后,耦合进WGM 410的光绝热地传递到多孔层402中,用于感测。
微谐振器的多孔表面可以被改性,以吸引所需的分析物。一旦所需的分析物被吸引到多孔表面上,或接近于多孔表面,那么微谐振器的光谱性能就发生改变。例如,微谐振器的表面可以被处理,以吸引蛋白,DNA分子,病毒,或细菌。在制备用于吸引蛋白的微谐振器的例子中,微谐振器的表面首先可以用抗体或抗原处理,从而当置于分析物溶液中时,抗体和抗原中的另一个被吸引到微谐振器上。抗原可以在细菌的细胞壁上表达,在这种情况下,细菌可以被吸引到微谐振器上。
在制备用于吸引DNA分子的微谐振器的例子中,微谐振器首先被处理成在其多孔表面上固定有特定DNA链。固定在微谐振器表面上的DNA链具有高度选择性,并仅与其互补链(cDNA)结合。病毒生物传感器可以具有被吸引到其多孔表面上的互补DNA片段。该片段与通常较大病毒的DNA分子的一部分互补。cDNA片段优选足够长,从而其可与病毒DNA分子牢固地结合。然后,可以光学测定病毒DNA分子的存在。带有微谐振器的DNA的用途公开在S.Chan等人,″Nanoscalesilicon microcavities for biosensing″,Materials Science and Engineering C15,页277-282(2001),在此引入作为参考。
任何适合类型的材料都可以用于微谐振器。在微谐振器包括覆盖在芯上的多孔层的技术中,芯可以是任何适合类型的玻璃,例如:氧化硅玻璃;改性的硅酸盐玻璃,如碱金属硅酸盐玻璃;重金属氧化物玻璃;卤化物玻璃,如氟化物玻璃;卤氧化物玻璃;硫族化物玻璃;和磷酸盐玻璃。此外,芯也可以是中空的,不需要完全是实心的。因此,芯可以是中空体,例如中空球体。
多孔材料层可以由任何适合的多孔材料形成,其在用于分析的波长下是透明的。例如,多孔层可以由溶胶-凝胶涂层,表面活性剂模板材料,微粒涂层,液晶层等形成。
整个微谐振器也可以由多孔材料制成,例如多孔玻璃VycorTM,从Corning Inc.,NY得到。
此外,多孔层可以用光学活性的材料掺杂。例如,多孔层的材料可以用光学可激励的物质来掺杂,如稀土离子,从而产生光学增益。因此,微谐振器内的光传播可用于泵激光学可激励的物质,从而在光学可激励的物质放大的波长下得到增益和/或激光振荡。微谐振器可以在光学可激励的物质放大的波长下振荡。因此,使用不同于从波导耦合进微谐振器的光的波长,微谐振器可用于感测分析物。
可以理解,不同类型的光学活性材料可以被引入到微谐振器的表面上,以与微谐振器的WGM内的光传播相互作用。相互作用例如可以包括吸收光,发射光,或一些其他类型的相互作用。例如,有机染料可以引入到微谐振器的表面,而微谐振器内的光传播使染料发荧光。这样甚至在加到微谐振器表面上的材料放大的光的波长下,也可以在WGM上产生振荡。
实施例1:多孔层
使用溶胶-凝胶技术,其中包括成孔剂(如表面活性剂),用于在芯外形成多孔层。这样可以控制孔尺寸,这样可以提供一定的尺寸识别,例如,单峰孔的直径约为2~50nm,直径控制到<10%。当使用Stober球时,直径范围为约2nm~1000nm或更大。
简单系统包括混合四乙氧基硅烷,非离子表面活性剂,酸,水,以及乙醇,形成涂覆溶液。可以通过浸渍或喷溅将该溶液涂覆到芯球上。如果需要可以通过液体萃取或加热来除去表面活性剂。可以通过硅烷偶联剂使孔壁官能化。
实施例2:多孔层
表面活性剂作为模板的氧化硅薄膜被浸渍涂覆在微球共振器上和涂覆在硅基底上。共振器和硅基底在400℃下煅烧15分钟,以除去表面活性剂,留下纳米直径的单峰孔。薄膜的多孔性为~25到~50vol.%,折射率为1.21-1.34。
在此实施例中,设计了两组四个样品(8个条件)。一组的薄膜厚度为40-55nm;第二组的薄膜厚度为230-280nm。涂覆溶液中的氧化硅浓度是用于控制厚度的关键因素。每组四个样品中,有两个样品具有较小(~3nm)孔直径,两个样品具有较大(~10nm)孔直径。表面活性剂的化学式量被用于控制孔直径。应注意到,两组中的孔直径都比探测光(>400nm)的波长小。可以预期到,这些多孔涂层有助于通过增大分析物的表面加载来提高共振生物传感器的灵敏度。在此实施例中所述的涂层是氧化硅基的,尽管原理上可以是金属氧化物、金属硫化物、金属或聚合物基的。

制备涂覆的共振器(以有硅基底)的第一步是合成2.16M基于四乙氧基硅烷的溶液。由此溶液,通过加入表II所示的适合量的表面活性剂和乙醇溶剂来制备8种涂覆溶液。
通过在9L烧瓶中,混合892mL无水乙醇(Aaper Alcohol andChemical Company,Shelbyville,KY),892mL四乙氧基硅烷(Alfa Aesar,Ward Hill,MA),71.97mL去离子水(18MΩ),和0.0210mL浓HCl(JTBaker,Phillipsburg,NJ;29%水溶液)来制备2.16M氧化硅溶胶原溶液。用顶置式气动搅拌器搅拌溶液,并在程序加热板(带有温度探针)上在60℃下加热90分钟。溶液冷却后,在聚丙烯瓶中在0℃下保存。
为制备表II中的酸调节的TEOS溶液,将上一段落中的34.48mL2.16M TEOS原溶液,4.14mL 0.07N HCl,和1.38mL去离子水(18MΩ)在60mL聚丙烯瓶中混合。
使用表II所示的制剂制备8种涂覆溶液。用微量吸管将预定量的酸调节的TEOS溶液转移到60mL聚丙烯瓶中。将表面活性剂和乙醇加到瓶中,剧烈摇动瓶中内含物,直到形成透明均匀溶液。表面活性剂是Pluronic P123(EO20PO70EO20)(BASF,Mount Olive,NJ)和十六烷基三甲基溴化铵(CH3C15H30N(CH3)3Br或CTAB;Aldrich,Milwaukee,WI)。

在晶片样品冷却后,对煅烧和制得的薄膜进行椭圆偏光分析。用Gaertner L116A椭圆偏光仪(Skolde,IL)在50°和70°下,测量硅晶片上的薄膜的数据(ψ和δ),使用从单色HeNe激光器发出的632.8nm光。每种样品至少测量三个光斑。记录平均值。通过椭圆偏光仪的软件测量单层薄膜的厚度值t和折射率n。基底的参数是Ns=3.850;吸收系数Ks=-0.020;波长632.8nm。结果记录在表III和IV和图5和6中。

Note:使用α值0.1定义显著性;用t-试验检测表面活性剂类型对折射率的影响;p=0.079。注意,样品6的收缩不能被计算,因为不能测量制造的薄膜的折射率。括号表明在α=0.1时的效果分析中,显著性不令人满意。
如所预料的那样,涂层厚度和氧化硅浓度之间有较强的相关性。对于任何x′s,涂层多孔性或折射率没有令人满意的显著相关性。收缩和表面活性剂类型/浓度之间有相关性。具体而言,使用较大表面活性剂和较低反应物浓度或较小表面活性剂和较高反应物浓度,易于使收缩值最大。
实施例3:使用多孔微谐振器的实验结果
进行实验检测根据实施例2所述步骤制造的微谐振器的有效Q-指标。实验装置与图1A所示的相似。在每个实验中,纤维108与微谐振器110接触,二者都浸渍在水中。可调的二极管激光器(Velocity 6304,从New Focus得到)用作光源102,中心波长约630nm。通过改变驱动激光器内压电激励器的电压,来调节激光器所发出的光的波长。当波长与微谐振器的一种或多种回音壁模式112共振时,耦合进微谐振器中的激光功率的量增大,导致检测器106的功率下降。
图7表明检测器中的检测信号随激光调节的变化,其中根据表II中的样品号6形成微谐振器。从这些结果估计出微谐振器的Q-指标约为1.4×106。图8表明检测器中的检测信号随激光调节的变化,其中根据表II中的样品号7形成微谐振器。从这些结果估计出微谐振器的Q-指标约为2.8×106。
这两个结果证实,在具有多孔外层的微谐振器中仍可得到高Q-指标。
如上所述,本发明涉及微谐振器,并被认为特别适用于使用微谐振器的传感应用中。本发明不应被认为限于上述的特定实施例,而是应该被理解成覆盖所附权利要求所述的本发明所有方面。在阅读本说明书后,本发明可以适用的各种修改、等价方法及多种结构对于本发明领域所属技术人员是相当清楚的。权利要求也意图覆盖这些修改和装置。
Claims (19)
1.一种微谐振器系统,包括:
产生光的光源;
耦合的第一波导,用于从所述光源接收光;和
至少一个安装的微谐振器,用于从所述第一波导将光耦合进微球,所述微谐振器限定了回音壁模式,并具有至少一个多孔表面区域。
2.如权利要求1所述的系统,其中所述回音壁模式被通过波导从光源耦合进微谐振器的光所激励,且所述回音壁模式光学耦合到所述多孔表面区域。
3.如权利要求1所述的系统,其中所述微谐振器完全由多孔材料形成。
4.如权利要求1所述的系统,其中所述微谐振器包括置于芯上的多孔外层。
5.如权利要求1所述的系统,还包括光学耦合的检测器,用于从所述微谐振器接收光。
6.如权利要求5所述的系统,其中光学耦合所述检测器,以通过所述第一波导从所述微谐振器接收光。
7.如权利要求5所述的系统,还包括第二波导,用于耦合所述微谐振器和所述检测器间的光。
8.如权利要求1所述的系统,其中所述微谐振器的多孔表面区域被改性以吸引分析物。
9.如权利要求8所述的系统,其中所述微谐振器的多孔表面区域含有抗原和相关的抗体之一。
10.如权利要求1所述的系统,还包括在液体介质和气态介质之一中的分析物,所述多孔表面区域与所述液体介质接触。
11.如权利要求1所述的系统,其中所述微谐振器包括涂敷在在所述多孔区域上的表面活性剂模板。
12.如权利要求1所述的系统,其中所述多孔表面区域其厚度小于光源所提供的光的波长的十分之一。
13.如权利要求1所述的系统,在所述多孔表面区域上还包括一种光学活性材料。
14.一种检测分析物的方法,所述方法包括:
使光进入第一波导;
使从所述第一波导来的光耦合进具有多孔表面区域的微谐振器;
使所述多孔耦合区域接触含有分析物的流体;
检测来自所述微谐振器的光;和
从检测的光确定分析物的存在。
15.如权利要求18所述的方法,还包括扫描耦合进所述微谐振器中的光的波长。
16.一种微谐振器,其包括:作为微谐振器操作的主体,所述主体在至少第一波长处限定回音壁模式,至少所述主体的表面部分是多孔的。
17.如权利要求16所述的微谐振器,其中所述主体基本上是球形的。
18.如权利要求16所述的微谐振器,其中所述主体基本上是平面的。
19.如权利要求16所述的微谐振器,其中所述主体完全由多孔材料形成。
