Abstract
一种新的超嗜热性微生物,更具体地说是属于新的属的超嗜热性微生物Caldothrix satsumae,它从有机废物在85℃或以上发酵而得的堆肥中获得,并能在80℃以上生长。该超嗜热性微生物在50℃以下不会生长,但是在70-85℃下生长和增殖活跃。它是革兰阴性菌,是绝对的好氧菌,且不形成孢子。它在高温时使有机废物发酵,可用作发酵微生物来产生堆肥。另外,它还可用于产生耐热性酶。
Description
技术领域
本发明涉及能在80℃及以上增殖的从堆肥衍生获得的新的超嗜热菌(hyperthermophile)。
背景技术
以往,使嗜热性微生物对家畜粪便、粪尿、污泥和城市垃圾等有机废弃物发生作用,使其有氧发酵和无臭化干燥,从而制得堆肥。而且,作为这些嗜热性微生物,已知有属于高温放线菌属或高温单孢菌属的嗜热放线菌(JP 55-121992A),芽孢杆菌属、Geobacillus属、产乳酸菌等的嗜热好氧且形成孢子的细菌混合物(JP 51-129759A),好氧性枯草芽孢杆菌(JP 6-5197A),属于水生栖热菌的有溶解木质素能力的细菌(JP 6-105679A),好氧性纤维素降解菌热纤维端孢菌、水生栖热菌(JP 6-191977)等。然而,尽管使用了这些微生物,由于发酵时的发酵热使发酵温度上升至70℃,最多为80℃,所以不能杀死杂菌尤其是形成芽孢的杂菌。另外,所得肥料中有用的菌体数最多为每克(干肥料)约1亿个,所以当用作肥料时,不能充分发挥肥料的效果。
为了解决污泥处理中的这些问题,本发明者进行了深入的研究,使污泥在85℃以上、较佳的在95℃以上的高温下发酵处理,来灭除杂菌、草种子等而净化污泥,且获得含大量有用菌体的发酵物。结果,发明人发现了一种获得只含大量有用菌体的污泥发酵物的方法,该方法是将从日本鹿儿岛县雾岛火山地带的土壤中得到的在85℃以上生长的细菌培养物加入生污泥(未经处理的污泥)中混合,进行有氧发酵,在85℃以上的发酵温度下杀灭污泥中含有的杂菌和种子,使污泥净化。该方法获得了专利(JP 3064221B)。因此,该污泥发酵物可用作堆肥,堆肥中发现有大量属于芽孢杆菌属或Geobacillus属的中温性好氧性芽孢菌、嗜热性(高温性)好氧性芽孢菌和嗜热菌等。
即,每克污泥发酵物中含有约10亿个细菌,这些菌主要包括如表1所示的好氧性细菌、高温性细菌、耐热性芽孢。

另外,在培养中,选择培养板上优势生长的菌落获得分离菌,对分离菌进行形态学观察等,寻找与发酵有关的微生物,结果显示其中有这样的微生物。

发现主要存在多形性无芽孢革兰阳性杆菌、好氧性芽孢菌(中温性和高温性)。
另外,根据R&D Planning出版由山里一英等3人编著的《微生物的分离方法》中的记载进行嗜热菌的测定。嗜热菌中的优势菌是好氧性芽孢菌(高温性)。
另外,对在上述微生物寻找中分离的优势的中温性好氧性芽孢菌(分离菌a)、高温性好氧性芽孢菌(分离菌b)和嗜热菌(分离菌c)进行了形态观察、生理学性状试验和菌体内DNA的GC含量测定。结果示于表3。

分离菌a与任一菌种的性状均不一致,其种类没有确定。分离菌b在弱碱性培养基(pH8.0-8.5)中生长良好,在pH7.0的培养基中不生长,但从其它性状试验结果认为它是与栗褐芽孢杆菌(Bacillus badius)或短芽孢杆菌(Bacillus brevis)相接近的菌种。然而,分离菌b具有两者的非典型性状,因此没有确定其种类。另外,由于分离菌c表现出与嗜热脂肪芽孢杆菌(Bacillus stearothermophilus)一致的细菌学性状,认为它们是同一菌种。然而,它们的GC含量有很大差异,所以认为它们是近缘菌种。
这些分离菌已被保藏于工业技术院生命工业技术研究所(目前为产业技术综合研究所微生物保藏中心)中。分离菌a(YM-01)的保藏号为FERM P-15085,分离菌b(YM-02)的保藏号为FERM P-15086,分离菌c(YM-03)的保藏号为FERM P-15087。
本发明者对这些堆肥中在高温下生长的微生物的存在进行了进一步的研究,结果惊奇地发现了属于新的菌属的在75℃以上高温下生长活跃、在85℃下仍有增殖、但在50℃下没有增殖的超嗜热菌。
发明的揭示
本发明的目的是从污泥在85℃以上发酵所得的堆肥中获得一种新的嗜热菌(尤其是超嗜热菌)。
为了解决上述问题,本发明者尝试着在污泥于85℃以上温度下发酵所得堆肥(商品名为Satsuma Soil;由鹿儿岛市水道局制造)中存在的嗜热性微生物进行检索,结果发现了在通常的细菌培养温度(30-40℃)下不增殖而在70-85℃、尤其是80℃以上的温度下生长增殖活跃的专性好氧菌。然而,本发明者基于该菌的16Sr DNA的碱基序列进行了系统分类分析。结果,本发明者发现,尽管该专性好氧菌是革兰阴性且不能形成孢子,但是它与如图1所示能形成孢子的革兰阳性土壤细菌芽孢杆菌属、Geobacillus属是相近,是至少在属水平上独立的细菌。本发明者将其命名为Caldothrix satsumaeYM081菌,并保藏于产业技术综合研究所专利生物保藏中心,保藏号为FERM-18598,随后,该菌被转移至国际保藏部门并给予保藏号FERM BP-8233。
即,本发明涉及一种属于Caldothrix属的新的超嗜热菌,它在80℃以上能生长。
即,本发明涉及一种属于Caldothrix satsumae属的新的超嗜热菌。
具体而言,本发明涉及新的超嗜热菌Caldothrix satsumae YM081菌(FERM BP-8233)。
另外,该菌的16SrDNA的全部碱基序列如序列表SEQ ID NO:1所示。

发明最佳实施方案
本发明的超嗜热菌是按照前述日本专利JP 3064221号公报中记载的方法从日本鹿儿岛县鹿儿岛市的粪尿等有机废弃物在高温下发酵所得堆肥(商品名为SatsumaSoil)中分离获得的。作为分离方法,采用以下方法。
在具有下表4所示组成的5毫升培养基中加入约0.1克上述堆肥,80℃下保温,反复传代培养经浓缩后,在加有吉兰糖胶的相同培养基的平板中反复分离和纯化。

所得细菌的微生物学性质和分类学位置如下所示。
1)在形态上,它是宽0.5微米、长3微米的长杆菌。革兰氏染色的结果显示阴性。菌体的超薄切片的电子显微镜观察也显示细胞表面结构为革兰阴性型。即,除细胞膜(质膜)和细胞壁外,还存在外膜。它没有形成孢子的能力。
2)发现该菌在70-85℃下生长活跃,但在50℃以下没有增殖。发现其在75℃以上的高温下增殖活跃,即便在85℃下仍然有增殖。该菌是专性需氧的。
3)增殖的最适pH为中性。能增殖的pH范围为6-9。另外,它显示有弱的嗜盐性。
4)对白蛋白和酪蛋白等各种蛋白质和淀粉有同化能力。
5)有脲酶生产性。不能还原硝酸。不能产生硫化氢和吲哚。
6)DNA的G+C含量为70.0%。
7)基于16SrDNA的碱基序列进行系统分类分析。其结果示于图1和表6。另外,16Sr DNA的全部碱基序列如序列表SEQ ID NO:1所示。
如上所述,虽然本发明的细菌为革兰阴性且不能形成孢子,但它与能形成孢子的革兰阳性土壤菌芽孢杆菌属、Geobacillus属近缘。然而,该菌至少在属的水平上是独立的细菌。
根据这些,判断本发明的微生物属于真细菌,是超嗜热菌。另外,根据16S rDNA的碱基序列,判断出它与Geobacillus stearothermophilus近缘,但是形成独立的属。
实施例
下面根据本发明的参考例和实施例进行说明。然而,本发明不应解释为局限于这些参考例和实施例。
参考例1
1.细菌培养物的制备
将日本鹿儿岛县始良郡牧园町的雾岛火山带的硫磺地带的37-40℃的土壤与附近长青苔的水稻田土壤混合。将蔗糖溶解在500-1000倍的水中,在混合物中加入该蔗糖水溶液,每1立方米加3-4升,在40-50℃下放置培养30-50日。将该培养物与数批生污泥(未经处理的污泥)混合,通入空气同时在有氧条件下发酵,用发酵温度为85℃以上的批次作为菌体培养物。
2.生污泥的处理
在动物粪便、来自污水的污泥、淀粉残渣和垃圾的混合物中加入消石灰进行除臭处理,将80重量份该物质与20重量份上述步骤1得到的细菌培养物混合,在发酵槽内通气条件下进行发酵。这样,发酵产物的温度约在1日内从室温升高至85-95℃。在该温度下维持发酵3日,在发酵开始后的第5日将产物搅混(搅拌)。经搅拌的发酵物的温度降低至约60℃,并在1日内温度上升至85-95℃。在该温度下维持发酵5日。重复发酵和搅拌操作数次,搅拌时的温度和发酵温度逐步降低。重复该操作4次,当发酵产物在搅拌时的温度降低至为35℃时,为最终发酵日。干燥所得发酵物,形成棕色颗粒,它可用作有机肥料。
3.原料堆肥的制备
将80重量份来自日本鹿儿岛县鹿儿岛市公共下水道污泥经压缩脱水后的含68%水分的生污泥与20重量份上述步骤2所得的细菌培养物混合,加入发酵围场内,在从下吹入空气的同时进行发酵。发酵开始后7日,温度达到98℃。进行发酵10日,在发酵温度开始从98℃下降时进行混合,以使其再次发酵。在首次达到99℃的温度后(即混合后第10日),温度迅速降低至60-70℃。此时,使发酵产物在发酵围场内散开,并使温度迅速降低至常温,得到棕色污泥发酵粉末。该污泥发酵粉末可用作堆肥或用作进行前述发酵的细菌培养物或培养基。
实施例1
超嗜热菌的分离
将约0.1克参考例得到的堆肥作为样品接种到5毫升的表4所示的培养基中,在80℃下保温,反复传代培养和浓缩,在加入了吉兰糖胶的相同组成培养基的平板上进行反复分离和纯化,得到本发明的超嗜热菌。
将堆肥样品加入蛋白胨/酵母提取物培养基(各为0.5%,0.3%,pH7.2)中,70℃下培养,在琼脂板(pH7.2,70℃)上分离增殖的细菌,结果发现了迄今认为导致堆肥在70℃以上温度下发酵的Geobacillus stearothermophilus和其它许多新的嗜热性细菌。
实施例2
超嗜热菌的微生物学性质
将实施例1所得的本发明的超嗜热菌接种到含0.3%酪蛋白、0.2%酵母提取物、0.1%淀粉、5%NaCl组成的pH7-8的琼脂培养基中,80℃下培养24小时,研究其微生物学性质。该结果示于表5中。
另外,该超嗜热菌的显微照片示于图2-5中。

结果表明,本发明的细菌属于真细菌,呈革兰阴性,且不能形成孢子。然而,它与能形成孢子的革兰阴性土壤细菌枯草杆菌属、Geobacillus属是近缘,而是属于新的属。另外,对于DNA的GC含量,如表1所示,没有菌株显示出达到90%的相同性,而且本发明的菌株与两个菌属(芽孢杆菌属和Geobacillus属)相距等距离(各为85%),因此判断为新的属(参见表6和图1)。因此,将该属命名为Caldothrix属。由于其最适增殖温度为80℃,这表明该菌是超嗜热菌。
本发明的Caldothrix satsumae YM081和枯草芽孢杆菌的生化性质的比较如表7所示。

将上述属于新属的本发明菌株命名为Caldothrix satsumae YM081菌株,并保藏于产业技术综合研究所专利生物保藏中心,保藏号为FERM-18598。随后,该菌被转移至国际保藏部门并给予保藏号FERM BP-8233。
实施例3
Caldothrix satsumae YM081菌株的温度与生长的关系
将Caldothrix satsumae YM081菌株接种于含有淀粉/酪蛋白/酵母提取物的培养基中,在各个预定温度下以120rpm振荡培养,测定倍增时间(细胞数增加至2倍的时间)。其结果如图6所示。
从图6可以看出,该菌的最适增殖温度为78℃,78℃时的倍增时间为26分钟,而82℃下的倍增时间约为55分钟。即使在82℃下,该菌仍以最适条件下的1/2的速度增殖。
另外,在培养基中加入来自堆肥的水提取液,如表8所示,发现即便在85℃也有增殖。

产业上的利用可能性
当用粪尿等有机废物作为原料接种发酵培养物进行发酵时,发酵温度依靠大多数属于芽孢杆菌属、Geobacillus属的中温性嗜热菌等而上升。随后,随着发酵温度的提高,本发明的超嗜热菌Caldothrix satsumae开始参与有机废物的分解和发酵。因此,本发明的超嗜热菌可用作高温下分解和发酵有机废物来生产堆肥的种子菌或培养基。
另外,该超嗜热菌产生的蛋白酶和淀粉酶在高温下有活性,因此预计利用该性质能制造耐热性酶。

1.一种超嗜热菌,它属于Caldothrix属,且能在80℃以上增殖。
2.根据权利要求1所述的超嗜热菌,它属于Caldothrix satsumae。
