发明背景
将纤维素质、例如棉纤维加工成服装制造原料要牵涉若干步骤:将纤维纺成纱;从纱构造纺织或编织的织物;随后进行制备、染色和整理操作。制备过程可能牵涉退浆(用于纺织物)、脱胶和漂白,生产出适合于染色或整理的纺织品。

本发明方法是这样进行的,使纤维素质与(i)酶系统和(ii)漂白系统接触;在含有所要处理的纤维素质的同一溶液中加入酶系统和漂白系统,在酶处理与漂白步骤之间不排空浸浴液(without emptying the bath)或者清洗纤维素质,也就是单浴过程。可以向溶液同时加入酶系统和漂白系统。作为替代选择,可以向溶液先后加入酶系统和漂白系统,其中(i)使纤维素质在适当条件下与酶系统接触充分时间,以便有效的生物脱胶(bioscouring)和/或退浆,然后(ii)向含有纤维素质和酶系统的溶液直接加入漂白系统,没有排空浸浴液或洗涤纤维素质。
在本发明的一个方面中,公开了处理纤维素质的方法,包含使纤维素质(i)与酶系统接触,用于退浆和/或脱胶,和(ii)与漂白系统接触,包含至少一种过酸化合物,其中在单浴处理中酶系统和过酸漂白系统同时或先后加入到同一溶液中。
在本发明的一种实施方式中,所述酶系统和过酸漂白系统在单浴过程中依次加入,通过首先(i)将含纤维素质的溶液与酶系统接触并温育溶液成分足够的时间,并在适合促进有效生物脱胶和/或退浆的条件下,然后通过(ii)将过酸漂白系统加入到同一含纤维素质和酶系统的溶液中并温育完成处理。
在另一种实施方式中,酶系统和过酸漂白系统同时加入到含纤维素质的溶液中,即在或大约同一时间或没有中间的温育步骤。
本发明的方法和组合物提供了这样一种产品,它表现为吸湿度(wettability)高、白度(whiteness)高和除尘(mote removal)均匀,同时具有优于常规制备过程的优点,包括:(i)加工时间更短;(ii)保存了水分;和(iii)减少了废液流出。
发明详述
本发明方法中,在单浴处理中生物脱胶和/或退浆结合漂白。
在优选实施方案中,通过同时加入酶系统和漂白系统到水溶液或冲洗液(wash liquor)中进行单浴处理,包含(i)同时加入酶系统和漂白系统到水溶液或冲洗液中,其包含或接触纤维素质,和(ii)根据所选酶系统,在适合达到有效脱胶和/或退浆,有效漂白的条件下温育足够的时间。
在另一优选实施方案中,先后加入酶系统和漂白系统到水溶液或冲洗液(wash liquor)中进行单浴处理,包含(i)同时加入酶系统和漂白系统到水溶液或冲洗液中,其包含或接触纤维素质,和(ii)根据所选酶系统,在适合引发和引起达到有效脱胶和/或退浆,有效漂白的条件下足够的时间进行第一次温育;(iii)将过酸漂白系统加入到包含纤维素质和酶系统的同一溶液中,温育完成处理。
纤维素质
本文所用的“纤维素质”表示所要处理的纤维素底物,非限制性地包含棉花、亚麻布、亚麻、苎麻、人造丝、大麻、黄麻和它们与其他天然或合成纤维的掺合物。纤维素质还可以非限制性地包含粗纤维、纱、纺织或编织的纺织品或织物或者服装或成品。
酶系统
本发明所用的酶系统表示生物脱胶酶系统和/或退浆酶系统。因此,酶系统可以包含一种或多种生物脱胶酶,有无一种或多种退浆酶均可,或者一种或多种退浆酶,有无一种或多种生物脱胶酶均可。
退浆酶:
任何适合的退浆酶都可以用在本发明中。优选地,退浆酶是淀粉分解酶。更优选地,退浆酶是α-或β-淀粉酶及其组合。
适合于本发明的α-和β-淀粉酶包括细菌或真菌来源的那些。在这一点上也包括这类淀粉酶在化学或遗传上经过修饰的突变体。优选的α-淀粉酶例如包括可从芽孢杆菌属菌种得到的α-淀粉酶,具体为地衣形芽孢杆菌菌株,如GB 1296839所详述。更优选的淀粉酶包括DuramylTM,TermamylTM,FungamylTM与BANTM(均可从Novozymes AIS,Bagsvaerd,Denmark获得)和Rapidase与Maxamyl(可从Gist-Brocades,Holland获得)。其他优选的淀粉分解酶有CGT酶(环糊精葡聚糖转移酶,EC2.4.1.19),例如从芽孢杆菌属、Thermoanaerobactor或Thermoanaero-bacterium菌种得到的那些。
退浆酶也可以优选地从上述酶衍生,其中一个或多个氨基酸已被加入、删去或取代,包括杂种多肽,只要所得多肽表现退浆活性即可。可用于实施本发明的这类变体可以利用常规的诱变工艺产生,例如利用高产量筛选工艺加以鉴别,例如琼脂平板筛选工艺。
向水溶液或洗液(即处理组合物)加入退浆酶的量足以使纤维素质退浆。通常,向处理组合物加入退浆酶、例如α-淀粉酶的量为0.00001%至2%酶蛋白,按组合物的重量计,优选为0.0001%至1%酶蛋白,按组合物的重量计,更优选为0.001%至0.5%酶蛋白,按组合物的重量计,进而更优选为0.01%至0.2%酶蛋白,按组合物的重量计。退浆酶的使用水平优选为约2-30,000KNU/I,更优选为20-30,000KNU/I,最优选为200-300KNU/I,或者约3-50,000NAU/I,优选为30-5,000NAU/I,最优选为350-500NAU/I。
生物脱胶酶:
任何适合的生物脱胶酶都可以用在本发明中。优选的生物脱胶酶非限制性地包括果胶酶、蛋白酶、脂肪酶、角质酶及其组合,更优选地,生物脱胶酶是果胶酶,进而更优选地,生物脱胶酶是果胶酸盐裂合酶(pectate lyase)。
果胶酶:任何具有降解植物细胞壁果胶组合物的能力的果胶分解酶组合物都可以用于实施本发明。适合的果胶酶非限制性地包括真菌或细菌来源的那些。也涵盖化学或遗传修饰的果胶酶。优选地,用在本发明中的果胶酶是重组产生的,是单组分酶。
果胶酶可以根据它们的优先底物——高甲基酯化的果胶或低甲基酯化的果胶和聚半乳糖醛酸(polygalacturonic acid)(果胶酸盐(pectate)),和它们的反应机理,β-消去或水解,加以分类。果胶酶可以是以内-作用性为主,在链内随机位置切割聚合物,得到寡聚物的混合物,或者它们可以是外-作用性的,从聚合物的一个末端攻击,生成单体或二聚物。若干作用于果胶平滑区(smooth region)的果胶酶活性包括在由Enzyme Nomenclature(1992)提供的酶分类中,例如果胶酸盐裂合酶(EC 4.2.2.2)、果胶裂合酶(EC 4.2.2.10)、聚半乳糖醛酸酶(EC3.2.1.15)、外-聚半乳糖醛酸酶(exo-polygalacturonase)(EC3.2.1.67)、外-聚半乳糖醛酸盐裂合酶(EC 4.2.2.9)和外-聚-α-半乳糖醛酸苷酶(EC 3.2.1.82)。
在本发明的优选实施方式中,果胶酶是果胶酸盐裂合酶。本文所用的果胶酸盐裂合酶的酶活性表示借助转移消去作用对果胶酸(也称聚半乳糖醛酸)中α-1,4-糖苷键随机裂解的催化作用。果胶酸盐裂合酶也被称为聚半乳糖醛酸盐裂合酶和聚(1,4-α-D-半乳糖醛酸化物)裂合酶。
任何果胶酸盐裂合酶都可以用于实施本发明。在优选的实施方式中,该方法利用在约70℃以上的温度下表现最大活性的果胶酸盐裂合酶。果胶酸盐裂合酶也可以优选地在约8以上的pH下表现最大活性,和/或在没有加入二价阳离子的存在下表现酶活性,例如钙离子。用在本发明中的果胶酸盐裂合酶的非限制性实例包括已从不同细菌属克隆的果胶酸盐裂合酶,例如欧文氏菌属、假单胞菌属、克雷白氏杆菌属和黄单胞菌属,以及枯草芽孢杆菌(Nasser et al.(1993)FEBS Letts.335:319-326)和芽孢干菌YA-14(Kim et al.(1994)Biosci.Biotech.Biochem.58:947-949)。也已描述了在8-10的pH范围内具有最大活性的果胶酸盐裂合酶的纯化,它们是由下列细菌产生的:短小芽孢杆菌(Dave and Vaughn(1971)J.Bacteriol.108:166-174)、多粘芽孢杆菌(Nagel and Vaughn(1961)Arch.Biochem.Biophys.93:344-352)、嗜热脂肪芽孢杆菌(Karbassi and Vaughn(1980)Can.J.Microbiol.26:377-384)、芽孢杆菌(Hasegawa and Nagel(1966)J.Food Sci.31:838-845)和芽孢杆菌RK9(Kelly and Fogarty(1978)Can.J.Microbiol.24:1164-1172)。任意上述以及二价阳离子独立性和/或热稳定性果胶酸盐裂合酶都可以用于实施本发明。在优选的实施方式中,果胶酸盐裂合酶包含如Heffron et al.,(1995)Mol.Plant MicrobeInteract.8:331-334和Henrissat et al.,(1995)Plant Physiol.107:963-976所公开的果胶酸盐裂合酶的氨基酸序列。
在酶水溶液或洗液中掺入果胶酶的量可以是0.00001%至2%酶蛋白,按组合物的重量计,优选为0.0001%至1%酶蛋白,按组合物的重量计,更优选为0.001%至0.5%酶蛋白,按组合物的重量计,进而更优选为0.01%至0.2%酶蛋白,按组合物的重量计。果胶酶的使用水平优选为约2.5至500,000APSU/g织物,更优选为约25至50,000APSU/g织物,最优选为约250至5,000APSU/g织物。

也可用于本发明的是蛋白酶变体,例如公开在EP 130,756(Genentech)、EP 214,435(Henkel)、WO 87/04461(Amgen)、WO 87/05050(Genex)、EP 251,446(Genencor)、EP 260,105(Genencor)、Thomas etal.,(1985),Nature.318,p.375-376、Thomas et al.,(1987),J.Mol.Biol.,193,pp.803-813、Russel et al.,(1987),Nature,328,p.496-500、WO 88/08028(Genex)、WO 88/08033(Amgen)、WO 89/06279(Novozymes A/S)、WO 91/00345(Novozymes A/S)、EP 525,610(Solvay)和WO 94/02618(Gist-Brocades N.V.)中的那些。蛋白酶的活性可以如″Methods of Enzymatic Analysis″,第3版,1984,Verlag Chemie,Weinheim,vol.5所述加以测定。

角质酶
可以使用任何适用于本发明的角质酶,例如包括从Humicolainsolens角质酶菌株DSM 1800衍生的角质酶,如美国专利No.4,810,414实施例2所述。
向酶水溶液掺入角质酶的量优选为0.00001%至2%酶蛋白,按组合物的重量计,优选为0.0001%至1%酶蛋白,按组合物的重量计,更优选为0.001%至0.5%酶蛋白,按组合物的重量计,进而更优选为0.01%至0.2%酶蛋白,按组合物的重量计。
适合的生物脱胶酶例如还包括从上述酶衍生的生物脱胶酶,其中一个或多个氨基酸已被加入、删去或取代,包括杂种多肽,只要所得多肽表现生物脱胶活性即可。可用于实施本发明的这类变体可以利用常规的诱变工艺产生,例如利用高产量筛选工艺加以鉴别,例如琼脂平板筛选工艺。例如,果胶酸盐裂合酶可以这样测量,在琼脂板(例如LB琼脂)中冲压4mm孔,涂上供试溶液,其中含有0.7%w/v聚半乳糖醛酸钠(SigmaP1879)。然后将平板在特定温度(例如75℃)下温育6小时。然后将平板(i)在1M CaCl2中浸泡0.5小时,或者(ii)在1%混合的溴化烷基三甲铵(MTAB,Sigma M-7635)中浸泡1小时。这两种工艺都导致聚半乳糖醛酸盐沉淀在琼脂内。在所沉淀的半乳糖醛酸盐背景内出现澄清的区域,检测果胶酸盐裂合酶的活性。使用标准果胶酸盐裂合酶制备物的稀释液校准测定灵敏度。
利用根据AATCC试验方法39-1980的滴液试验测量,有效的脱胶通常提高吸湿度。优选地,漂白后的织物的吸湿度为20秒(seconds)或以下,最优选为10秒或以下。
用在本发明中的退浆与生物脱胶酶可以是从它们的细胞来源衍生的,或者可以是重组产生的,并且可以是纯化的或分离的。本文所用的“纯化的”或“分离的”酶是这样一种酶,它已被处理,以清除非酶材料或其他从细胞衍生的酶,其中它是合成的,能够干扰酶活性。通常,退浆与生物脱胶酶是从细菌或真菌微生物分离的,其中它是作为内源性成分或重组产物而被产生的。如果酶被分泌到培养基中,那么纯化可以包含利用常规方法借助离心、过滤或沉淀来分离培养基与生物质。作为替代选择,可以借助细胞破坏和生物质的分离而从宿主细胞中释放出酶。在有些情况下,可以借助常规的蛋白质纯化方法实现进一步的纯化,非限制性地包括硫酸铵沉淀;酸或离液剂萃取;离子交换,分子筛,和疏水性色谱,包括FPLC和HPLC;制备型等电点聚集;和制备型聚丙烯酰胺凝胶电泳。作为替代选择,可以利用亲合色谱实现纯化,包括免疫亲合色谱。例如,可以使用杂种重组果胶酸盐裂合酶,它具有额外的氨基酸序列,充当亲合性“标记”,这有利于利用适当的固相基质进行纯化。
用在本发明方法中的退浆与生物脱胶酶还可以是经过化学修饰的,以增强一种或多种性质,赋予它们进而更多的优点,例如提高溶解度、降低不稳定性或二价离子依赖性等。修饰作用非限制性地包括磷酸化、乙酰化、硫酸化、酰化或本领域技术人员已知的其他蛋白质修饰作用。
漂白系统
本发明所述“过酸漂白系统”包含一或多种过酸漂白化合物,任选包含,碱试剂,以及任选包含至少一种漂白稳定剂。
过酸漂白化合物
本发明所述“过酸漂白化合物”或“过氧酸漂白化合物”是包含至少一个过羟基(-OOH)的酸。优选,所述过酸漂白化合物选自几类有机过酸物质。优选所述过酸是过甲酸,或包含单个过羧酸基和少于11个碳原子的直链或支链饱和烷基链的羧酸脂族过氧酸(carboxylicaliphatic peroxyacids)。包含少于6个碳原子的直链饱和烷基链的脂族羧酸过氧酸也是优选的。这样的过氧酸例子是过乙酸,过丙酸,过-n-丁酸和过-n-戊酸。特别优选过乙酸因为其有效性和相对简单的制备方法。
本发明另一具体实施方式中,有机过氧酸选自含少于16个碳原子的直链或支链烷基链和,在位于相对alpha-omega位的碳原子有两个过羧酸基团取代的二过羧酸(diperoxycarboxylic acid)。这样的过氧酸例如1,6-己二过氧二酸(1,6-hexanediperoxydioic acid),1,8-辛二过氧二酸,1,10-癸二过氧二酸,1,12-十二烷二过氧二酸(1,12-dodecanediperoxydioic acid)。本发明另一具体实施方式中,有机过氧酸选自芳香族过氧酸,其每一个苯核含有至少一个过羧基基团。优选每一个苯核只含有一个过羧基基团的芳香族过氧酸。这样的过氧酸例如是过苯甲酸(peroxybenzoic acid)。本发明另一具体实施方式中,有机过氧酸被一或多个卤素原子或任何其它有机功能取代基取代,如羰基(酮,醛或羧酸),醇基,含氮基团如腈基,硝基,氨基和酰胺基,含硫基团如磺基,巯基。例如过氧单硫酸(peroxymonosulphuric acid)。
过酸漂白化合物以有效量加入到水溶液或洗涤液(即处理组合物)中,以除去有色杂质,改进吸收性,达到合适的白度和/或可染性(dyeability)。优选的,过酸漂白化合物加入到处理组合物中的量为约0.01g/l-约15g/l组合物,更优选约0.1g/l-约10g/l组合物,最优选约0.5g/l-约5g/l组合物。
碱试剂
碱试剂是本领域熟知的。优选用在本发明中的碱剂包括氢氧化钠、碳酸钠、碳酸氢钠、过硼酸钠、硫化钠和亚硫酸钠。优选加入处理组合物的量为约0.1g/l-约10g/l组合物,更优选约0.5g/l-约5g/l组合物。
漂白稳定剂:
在本发明的另一种优选实施方式中,漂白系统另外含有一种或多种漂白稳定剂。漂白稳定剂包含能够吸附、粘合或配位化合痕量重金属的添加剂。按照本发明能够使用的、具有漂白稳定作用的添加剂的实例有多阴离子化合物,例如多磷酸盐、多羧酸盐、多羟基多羧酸盐、可溶性硅酸盐,它们是完全或部分中和的碱金属或碱土金属盐,确切为中性的Na或Mg盐,它们是相对弱的漂白稳定剂。按照本发明能够使用的强漂白稳定剂的实例有配合剂,例如乙二胺四乙酸盐(EDTA)、二亚乙基三胺五乙酸(DTPA)、次氨基三乙酸(NTA)、甲基甘氨酸二乙酸(MGDA)、β-丙氨酸二乙酸(ADA)、乙二胺-N,N’-二琥珀酸盐(EDDS)和膦酸盐类,例如乙二胺四亚甲基膦酸盐、二亚乙基三胺五亚甲基膦酸盐(DTMPA)或羟基亚乙基-1,1-二膦酸的酸形式或者部分或完全中和的碱金属盐形式。
向处理组合物加入漂白稳定剂的量通常为约0.1至约5g/升组合物,更优选为约0.5至约2g/l,最优选为约1g/l。
根据本发明的漂白组合物优选地含有至少一种漂白稳定剂,更优选为至少一种上述强漂白稳定剂。有效的漂白通常带来一种或多种下列性质:(i)所需的白度(借助Ganz白度测量法测定,例如使用Macbeth coloreye);(ii)除尘均匀性令人满意(借助肉眼检查评估)。优选地,织物的白度是50Ganz单位或更高,最优选为60Ganz单位或更高。
额外的组分:
在有些发明实施方式中,水溶液或洗液进一步包含其他组分,非限制性地包括其他酶,以及表面活性剂、防沫剂、润滑剂、增效系统(builder system),它们增强脱胶和/或漂白过程,和/或提供优异的效果,例如与强度、抗成丸性(resistance to pilling)、吸水性和染色能力有关。
适合用在本发明中的酶非限制性地包括如上所述的果胶酶、蛋白酶和脂肪酶;和纤维素酶。纤维素酶归入一系列酶家族,涵盖内-与外-活性以及纤维二糖水解能力。用于实施本发明的纤维素酶可以是从微生物衍生的,已知它们能够产生纤维素分解酶,例如腐质霉属、Thermomyces、芽孢杆菌属、木霉属、镰孢属、毁丝霉属、Phanerochaete、耙菌属、Scytalidium、裂褶菌属、青霉属、曲霉属或地霉属的菌种,确切为Humicola insolens、尖孢镰孢或Trichoderma reesei。适合的纤维素酶的非限制性实例公开在美国专利No.4,435,307、欧洲专利申请No.0495 257、PCT专利申请No.WO 91/17244和欧洲专利申请No.EP-A2-271004中。
酶可以是从它们的细胞来源分离的,或者可以是重组产生的,并且可以是在化学或遗传上经过修饰的。通常,在水溶液中掺入酶的水平为约0.0001%至约1%酶蛋白,按组合物的重量计,更优选为约0.001%至约0.5%,最优选为0.01%至0.2%。不言而喻的是,利用常规的测定法容易确定关于每种额外与特定生物脱胶酶联合用在本发明方法中的酶的酶活性单位数。
适合用于实施本发明的表面活性剂非限制性地包括非离子型(美国专利No.4,565,647);阴离子型;阳离子型;和两性离子型表面活性剂(美国专利No.3,929,678);它们的浓度通常在约0.2%至约15%之间,按重量计,优选从约1%至约10%,按重量计。阴离子型表面活性剂非限制性地包括直链烷基苯磺酸盐、α-烯烃磺酸盐、烷基硫酸盐(脂肪醇硫酸盐)、醇乙氧基硫酸盐、仲烷烃磺酸盐、α-磺基脂肪酸甲酯、烷基-或烯基-琥珀酸和皂类。非离子型表面活性剂非限制性地包括醇乙氧基化物、壬基苯酚乙氧基化物、烷基多苷、烷基二甲基氧化胺、乙氧基化脂肪酸单乙醇酰胺、脂肪酸单乙醇酰胺、多羟基烷基脂肪酸酰胺和葡糖胺的N-酰基N-烷基衍生物(葡糖酰胺(glucamide))。
增效系统非限制性地包括硅铝酸盐、硅酸盐、多羧酸盐、脂肪酸、诸如乙二胺四乙酸盐等材料和金属离子螯合剂,例如氨基多膦酸盐,确切为乙二胺四亚甲基膦酸和二亚乙基三胺五亚甲基膦酸,它们的浓度在约5%至80%之间,按重量计,优选在约5%与约30%之间,按重量计。
防沫剂非限制性地包括硅酮(美国专利No.3,933,672);DC-544(DowCorning),它们的浓度通常在约0.01%与约1%之间,按重量计。
组合物还可以含有污垢悬浮剂、污垢释放剂(soil-releasingagent)、光亮剂、研磨剂和/或杀菌剂,它们是本领域公知的。
加工条件
使含有酶和漂白系统的水溶液与纤维素质接触的方式将依赖于加工制度是否是连续的、不连续的pad-batch或分批的。例如,关于连续的或不连续的pad-batch加工,酶水溶液优选地包含在饱和浴中,随着纤维素质穿过浴而连续涂在其上,在该过程期间纤维素质通常吸收0.5-1.5倍于其重量的加工液。在分批操作中,使纤维素质暴露于酶溶液达约5分钟至24小时,加工液与织物之比为5∶1-50∶1。
水溶液或洗液的pH通常在约4与约11之间。优选地,处理组合物的pH在约5与约10之间,优选在约7至约9之间,最优选为约8至约9。进行联合脱胶和/或退浆与漂白过程的温度浆依赖于所用的过程。在冷pad-batch过程的情况下,脱胶和/或退浆与漂白温度优选地在约15℃与约45℃之间,最优选地在约25℃与约35℃之间。关于连续的和其他分批(batch)过程,脱胶和/或退浆温度优选地在约35℃与约75℃之间,最优选地在约45℃与约65℃之间;漂白温度可以在约30℃与约100℃之间,优选地在约50℃与约100℃之间,最优选地在约60℃与约90℃之间。
不言而喻的是,酶、漂白化合物、漂白稳定剂和碱剂(如果使用的话)的最佳剂量与浓度、水溶液或洗液的体积和pH与温度将因下列因素而异:(i)纤维的属性,也就是粗纤维、纱或纺织品;(ii)是否同时或先后进行脱胶和漂白;(iii)所用特定的酶和酶的比活度;(iv)进行加工的温度、pH、时间等条件;(v)洗液中其他组分的存在;和(vi)所用加工制度的类型,也就是连续的、不连续的pad-batch或分批的。加工条件的优化可以利用惯用实验方法加以确定,例如建立基础条件,再测试基础中的不同点。例如,酶量、接触温度和总加工时间可以各不相同,之后评价所得纤维素质或纺织品的(a)果胶清除效率;(b)脱胶性质,例如吸湿度;和(c)漂白的质量,例如白度。
在优选的实施方式中,可以调节条件或处理组合物,以有利于退浆、脱胶或漂白过程,例如通过调节pH、润湿剂的浓度或二价阳离子螯合剂--例如乙二胺四乙酸盐--的浓度,进一步促进漂白过程。在优选的实施方式中,先后模式可以进一步包含在步骤(ii)与(iii)之间调节水溶液或洗液组合物的一种或多种性质。例如,可以在步骤(ii)与(iii)之间调节pH、润湿剂的浓度或二价阳离子螯合剂--例如乙二胺四乙酸盐--的浓度,以进一步促进漂白过程。第一次与第二次温育的条件也可以在温度、搅拌速率、时间等方面有所不同。
实施例
下面是对本发明的非限制性举例说明。
实施例1:利用H2O2单浴同时生物脱胶和漂白
A.生物脱胶和漂白:从连结型针织织物(interlock knit fabric)(4600型,Ramseur Co.,NC)切取重约25克的45cm×21.5cm织物。将织物装入Labomat烧杯(Mathis Labomat,Werner Mathis USA,Inc,NC),然后倒入250ml 20mM碳酸氢钠-碳酸钠缓冲溶液(pH 9.2),其中含有3000APSU/kg纤维的果胶酸盐裂合酶、0.5g/l润湿剂(Kierlon Jet B,BASF)、1.7g/l H2O2和0.75g/l稳定剂(Calgon,Dexter)。将织物在55℃下处理15分钟,之后按5℃/分钟升温至70℃达1小时。然后将织物用自来水彻底洗涤,以清除残留的化学品,在室温下干燥过夜。
B.分析:借助Macbeth color eye测量织物的白度,以Ganz单位表示。借助滴液试验测定吸湿度,测量一滴水被织物吸收的时间,以秒表示。
结果列在表1中。织物的白度和吸湿度都是非常低的。本例表明,在没有碱存在下单用过氧化氢漂白编织的织物仅有限地提高白度、吸湿度和除尘效率。
实施例2:利用H2O2/NaOH单浴同时生物脱胶和漂白
使用与上例1相同的织物和设备。按照基本上与例1相同的方式进行实验,除了向生物脱胶/漂白溶液加入2g/l NaOH以外。
结果如表1所示。碱的存在戏剧性地提高织物的白度。但由于碱使酶变性,吸湿度很差。
实施例3:利用H2O2/NaOH单浴顺序(sequential)生物脱胶和漂白
A.生物脱胶:从连结型针织织物(4600型,Ramseur Co.,NC)切取重约25克的45cm×21.5cm织物。将织物装入Labomat烧杯(MathisLabomat,Werner Mathis USA,Inc,NC),然后倒入250ml 20mM磷酸钠缓冲溶液(pH 9.2),其中含有3000APSU/kg纤维的果胶酸盐裂合酶和0.5g/l润湿剂(Kierlon Jet B,BASF)。将织物在55℃下处理15分钟。
B.漂白:向同一烧杯加入H2O2、NaOH和硅酸钠。H2O2、NaOH和硅酸钠的最终浓度与上例1相同。按5℃/分钟升高Labomat温度至70℃达1小时,之后排干水。然后将织物用自来水彻底洗涤,以清除残留的碱,在室温下干燥。
结果如表1所示。吸湿度好于同时模式。
实施例4:用H2O2/NaOH双浴(two-bath)生物脱胶和漂白
按照基本上与上例3相同的方式进行实验,除了在生物脱胶阶段之后排干脱胶溶液并用水代替以外。
结果如下表1所示。双浴处理模式(实施例4)的过氧化物漂白优于单浴模式(实施例3):吸湿度更好,白度更高,除尘更有效。
实施例5:用H2O2/NaOH双浴(two-bath)脱胶和漂白
按照基本上与上例4相同的方式进行实验,除了脱胶溶液中不含有果胶酸盐裂合酶以外。
结果如下表1所示。由于不含有果胶酸盐裂合酶,织物的吸湿度很差。此实施例表明生物脱胶酶对于改进织物的吸湿度是非常重要的。

实施例6:用过乙酸(PA)单浴同时生物脱胶和漂白
从连结型针织织物(4600型,Ramseur Co.,NC)切取重约25克的45cm×21.5cm织物。将织物装入Labomat烧杯(Mathis Labomat,WernerMathis USA,Inc,NC),然后倒入250ml 20mM磷酸钠缓冲溶液(pH 9.2),其中含有3000APSU/kg纤维的果胶酸盐裂合酶、0.5g/l润湿剂(KierlonJet B,BASF)、50mM过乙酸和0.75g/l稳定剂(Calgon,Dexter)。将织物在55℃下处理15分钟,之后按5℃/分钟升温至70℃达1小时。然后将织物用自来水彻底洗涤,以清除残留的化学品,在室温下干燥过夜。
结果如下表2所示。证据表明在此试验条件下,过乙酸是比过氧化氢(peroxide)更为有效的漂白剂(实施例6相比实施例1)。
实施例7:采用过乙酸(PA)单浴顺序生物脱胶和漂白
A.生物脱胶:从连结型针织织物(4600型,Ramseur Co.,NC)切取重约25克的45cm×21.5cm织物。将织物装入Labomat烧杯(MathisLabomat,Werner Mathis USA,Inc,NC),然后倒入250ml 20mM磷酸钠缓冲溶液(pH 9.2),其中含有3000APSU/kg纤维的果胶酸盐裂合酶和0.5g/l润湿剂(Kierlon Jet B,BASF)。将织物在55℃下处理15分钟。
B.漂白:向同一烧杯加入过乙酸和Calgon。过乙酸和Calgon的最终浓度与上例6相同。按5℃/分钟升高Laboma t温度至70℃达1小时,之后排干水。然后将织物用自来水彻底洗涤,以清除残留的碱,在室温下干燥。
结果如表2所示。试验表明单浴顺序模式进行生物脱胶和过乙酸漂白得到的织物吸湿度好于同时模式。
实施例8:采用过乙酸(PA)单浴顺序脱胶和漂白
按照基本上与上例7相同的方式进行实验,除了脱胶溶液中不含有果胶酸盐裂合酶以外。
结果如下表2所示。试验进一步证明了果胶酸盐裂合酶可以改进织物的吸湿度。
实施例9:采用过乙酸(PA)和NaOH单浴顺序生物脱胶和漂白
按照基本上与上例7相同的方式进行实验,除了漂白溶液中加入2g/L NaOH外。
结果如下表2所示。试验证明了漂白浴中加入碱可以改进织物的白度和吸湿度。
实施例10:采用过乙酸(PA)双浴生物脱胶和漂白
按照基本上与上例7相同的方式进行实验,除了生物脱胶步骤后排干脱胶液并用水替代。
结果如下表2所示。令人惊奇的是,双浴处理(实施例9)的白度和灰尘去除不如单浴顺序模式(实施例7)好,这与在生物脱胶和过氧化物漂白中观察到的非常不同。
实施例11:采用过乙酸(PA)双浴脱胶和漂白
按照基本上与上例10相同的方式进行实验,除了脱胶溶液中不含有果胶酸盐裂合酶以外。
结果如下表2所示。织物的吸湿度很差。试验进一步证明了果胶酸盐裂合酶可以改进织物的吸湿度。

实施例12:利用H2O2/NaOH单浴顺序生物脱胶和漂白
按照基本上与上例3相同的方式进行实验,除了用25克退浆编织织物(woven fabric)(428U,Testfabric,INC.,PA)代替针织织物(knitted fabric)。
结果如表3所示。单浴顺序生物脱胶和过氧化物漂白。得到的编织织物白度很低。
实施例13:利用H2O2/NaOH双浴生物脱胶和漂白
按照基本上与上例4相同的方式进行实验,除了用25克退浆编织织物(woven fabric)(428U,Testfabric,INC.,PA)代替针织织物。
结果如表3所示。试验进一步表明双浴顺序模式的过氧化物(peroxide)漂白要好于单浴模式。
实施例14:利用过乙酸(PA)/NaOH单浴顺序生物脱胶和漂白
按照基本上与上例12相同的方式进行实验,除了漂白液包含50mM过乙酸(Aldrich),2g/L NaOH和0.75g/L Calgon(BASF)。
结果如表3所示。试验表明过乙酸(实施例14)是比过氧化物(实施例12)更有效的漂白剂。
实施例15:利用过乙酸(PA)/NaOH单浴顺序生物脱胶和漂白
按照基本上与上例13相同的方式进行实验,除了漂白液包含50mM过乙酸(Aldrich),2g/L NaOH和0.75g/L Calgon(BASF)。
结果如表3所示。与从针织织物实施例(6-10)观察到的类似,此实施例表明单浴顺序模式在改进白度和灰尘去除方面,要大大好于双浴处理模式(实施例9)。

Claims (26)
1.处理纤维素质的方法,包含酶脱胶和漂白,其中酶脱胶和漂白是按下述实施:将纤维素质与用于生物脱胶纤维素质的酶系统和包含至少一种过酸漂白化合物的漂白系统接触,其中向含有纤维素质的单一溶液先后加入酶系统和漂白系统。
2.权利要求1的方法,其中向含有纤维素质的溶液先后加入酶系统和漂白系统,包含加入酶系统,温育,随后加入漂白系统,温育。
3.权利要求1的方法,其中使纤维素质与酶系统和漂白系统接触,形成吸湿度为20秒或以下、白度为至少50Ganz单位的织物。
4.权利要求1的方法,其中使纤维素质与酶系统接触,形成吸湿度为20秒或更低的织物,之后向含有纤维素质的溶液加入漂白系统。
5.权利要求2的方法,进一步调节溶液的性质,所述性质选自pH、离子强度、温度、表面活性剂的浓度、二价阳离子螯合剂的浓度或任意上述的组合。
6.权利要求1的方法,进一步包含用至少一种退浆酶处理纤维素质以退浆纤维素质,该退浆酶选自α-淀粉酶、β-淀粉酶或其组合。
7.权利要求1的方法,其中该酶系统是生物脱胶酶系统。
8.权利要求1的方法,其中该酶系统包含至少一种生物脱胶酶,选自果胶酶、蛋白酶、脂肪酶或任意上述的组合。
9.权利要求1的方法,其中该酶系统包含至少一种生物脱胶酶,选自果胶酸盐裂合酶、果胶裂合酶、聚半乳糖醛酸酶、外-聚半乳糖醛酸酶、外-聚半乳糖醛酸盐裂合酶或外-聚-α-半乳糖醛酸苷酶。
10.权利要求1的方法,其中该酶系统包含果胶酸盐裂合酶。
11.权利要求1的方法,其中该酶系统包含蛋白酶,选自氨基肽酶、丝氨酸内肽酶、半胱氨酸内肽酶、天冬氨酰内肽酶或金属内肽酶。
12.权利要求1的方法,其中该酶系统包含脂肪酶,选自三酰甘油脂肪酶或磷脂酶。
13.权利要求1的方法,其中该酶系统包含这样一种果胶酸盐裂合酶,在70℃以上的温度下表现最大的果胶酸盐裂合酶的酶活性。
14.权利要求1的方法,其中该酶系统包含这样一种果胶酸盐裂合酶,在8以上的pH下表现最大的果胶酸盐裂合酶的酶活性。
15.权利要求1的方法,其中该纤维素质包含纺织品。
16.权利要求1的方法,其中所述纤维素质包含棉。
17.权利要求1的方法,其中所述单一溶液进一步包含一种或多种缓冲剂、表面活性剂、螯合剂和/或润滑剂,或任意上述的盐。
18.权利要求1的方法,其中所述至少一种过酸漂白化合物是有机过氧酸化合物。
19.权利要求1的方法,其中所述至少一种过酸漂白化合物是过乙酸。
20.权利要求1的方法,其中所述至少一种过酸漂白化合物选自下类有机过氧酸:过甲酸和羧酸脂族过氧酸;二过氧羧酸;芳族过氧酸;一或多个卤素原子或任何其它有机官能团取代的有机过氧酸。
21.权利要求1的方法,其中所述纤维素质与至少一种过酸漂白化合物的接触是与至少一种漂白稳定剂一起进行的。
22.权利要求21的方法,其中所述漂白稳定剂选自乙二胺四乙酸盐(EDTA)、二亚乙基三胺五乙酸(DTPA)、次氨基三乙酸(NTA)、甲基甘氨酸二乙酸(MGDA)、β-丙氨酸二乙酸(ADA)、乙二胺-N,N’-二琥珀酸盐(EDDS)、乙二胺四亚甲基膦酸盐、二亚乙基三胺五亚甲基膦酸盐(DTMPA)或羟基亚乙基-1,1-二膦酸。
23.权利要求1的方法,其中所述纤维素质与至少一种过酸漂白化合物的接触是与至少一种碱试剂一起进行。
24.权利要求23的方法,其中所述碱试剂是氢氧化钠。
25.权利要求1的方法,其中所述纤维素质与至少一种过酸漂白化合物的接触是与至少一种漂白稳定剂和至少一种碱试剂一起进行。
26.权利要求1-25任一项的方法产生的织物。
