发明背景
单纯疱疹病毒(HSV)经常作为神经系统的合适的基因传递的载体,因为它为神经营养生活型并具有在神经元中保持细胞终生的潜伏能力。这一独特能力暗示,通过合适的开发这种载体系统仅仅施用一次就可对一定的疾病具有终生治疗益处,如帕金森氏病其中酪氨酸羟化酶或者GDNF在人脑中的表达显示了优势。
然而,虽然不完整疱疹病毒显示了在体内有效地向神经系统和其它组织传送基因,但由疱疹基因组表达的异源基因的转录常常只持续很短的时间(<1周)。当疱疹基因组处于病毒潜伏状态期间所保持的转录失活状态,转录就停止。这样,尽管疱疹载体DNA在治疗的细胞中有可能保持至细胞终生,然而由于异源基因通常不被转录(目前),所以治疗效果仅仅在短期内得以显示。

单纯疱疹病毒(造成唇疱疹(HSV1)或生殖疱疹(HSV2))通过皮肤或粘膜表面的磨损感染感觉神经元的轴索的末端,接着,它们在细胞体中迁移至核,并建立潜伏感染或发生裂解复制。影响决定潜伏或裂解感染的因素尚未得以充分了解。然而,在不完整病毒载体的情况下只有潜伏途径是可能的。
在潜伏期间,疱疹基因组很大程度上为转录失活。然而在该基因组的长重复片段中的一个小区域转录后产生与潜伏有关的转录物(LAT),本文描述的是HSV1(HSV2类似)。LATs分为两类,分别为由含TATA启动子(参见Coffin和Latchman,1996;本文叙述为LAT p1)转录的低丰度的共线性的较大(约8千碱基(kb))的转录物,和丰度稍高的约2kb和1.5kb的转录物(其被认为是由较大转录物剪接的嵌套的稳定内含子)。1.5kb转录物仅仅在神经元中检测到,并且在潜伏期间二者的丰度增加。在LAT p1和2kb LAT的起点之间鉴定出第二个具有十分弱的启动子活性的区域(参见Goins等,1994;本文叙述为LAT P2),这说明在它一些条件下该区可以单个独转录物的形式进行表达。

发明概要

本发明也涉及用于研究真核细胞(尤其是哺乳动物的细胞)的基因的功能的方法中,例如,鉴定基因互补细胞机能障碍,或者在野生型或突变体细胞中研究突变基因表达的影响。本发明的方法特别适用于疾病相关基因的功能研究。
因此本发明提供了一种包含按该顺序有效连接的HSV潜伏相关转录物P2区,启动子以及异源基因的表达盒。优选的所说的启动子是与潜伏非相关的转录启动子。更优选的启动子是病毒的启动子或哺乳动物的启动子。优选的启动子是使得异源基因在哺乳动物细胞中得以表达的病毒启动子或哺乳动物的启动子,其中所说的细胞优选的是中枢或外周神经系统的细胞,或眼的细胞,心脏或骨骼肌的细胞,更优选的是中枢或外周神经系统的细胞。启动子也可以是诱导型和/或组织特异性。
这样表达盒或载体(优选的是病毒毒株,更优选的是HSV毒株及包含本发明的表达盒)可用于将异源基因传送至它将加以表达的哺乳动物细胞内。这样的表达盒和载体在多种应用中有用,例如,基因治疗,疫苗,基因功能的体外或体内分析方法或研究。
术语异源基因旨在包括任何在HSV基因组中未发现的基因。异源基因可以是任何野生型基因的等位基因的变体,或者是突变基因。异源基因优选地编码具有治疗用途的多肽,包括具有细胞毒性的或能够把前药转化成为细胞毒性化合物的多肽。
基因治疗和其它治疗应用可能都要求施用多基因。多基因的表达对多种疾病的治疗有利,例如利用多神经营养因子。由于HSV没有其它病毒载体系统的包装能力限制,因此HSV格外适用。这样可将多个异源基因置于它的基因组内部。本发明,例如,至少有两种方法可完成这一目的。例如,可将多个本发明的表达盒引入到特定HSV毒株中。每一表达盒可以包含一个异源基因。另一方法是利用成对的启动子,它们以相反的方向背向位于中间的LAT P2元件(相同或不同的启动子),这些启动子分别驱动异源基因(相同或不同的异源基因)的表达。

本发明进一步提供表达盒与载体的利用,优选的是包含表达盒的HSV毒株在人类和动物的治疗上的用途。例如,包含表达盒的HSV毒株可用于治疗神经系统的疾病或损伤(包括帕金森氏病,脊柱损伤或中风),或者眼疾,心脏或骨胳肌疾病,或恶性肿瘤的治疗。减毒处理这些HSV毒株,使得它们不能在人和动物细胞中经历裂解循环。

发明详述
A.表达盒-LAT P2区,启动子,异源基因
本发明的表达盒本质上包括按该顺序有效连接的LAT P2区,启动子以及异源基因。术语″有效连接″是指并列连接,其中的组分处于可使他们按所计划的方式起作用的关系中。这样,例如,与异源基因序列有效连接的启动子以可使异源基因在一定条件(其适合由启动子控制的表达活化)下可完成表达的方式进行连接。

术语″长期表达″是指在被本发明的单纯疱疹病毒感染的细胞中即使单纯疱疹病毒进入潜伏状态后的异源基因的表达。优选的,至少两周,更优选的在感染后至少一个或两个月,最优选的是细胞终生。
表达盒的构筑可通过本领域技术人员熟知的常规克隆技术(参见,例如,Sambrook等,1989,分子克隆实验室手册;冷泉港出版社)。
1.LAT P2域
本文定义LAT P2域为HSV1核苷酸118,866-120,219(基因库HE1CG:从PstI-BstXI位点)或者该区域的片段或衍生物,其中所说的区域包括能使异源基因在本发明的表达盒中长期表达的与HSV2同源的区域。
2.启动子
启动子是指非来源于HSV的LAT P2区域的转录启动子。术语启动子为本领域人员所熟知,其包括在大小和复杂度上从最小的启动子至包含上游元件和增强子的启动子的核酸区域。将启动子与LAT P2区域和其下游有效连接。将第二启动子以相反的取向有效连结在LATP2区域的上游也是可能的,这样LAT P2区域授与两个启动子具有长期活性。
启动子从在哺乳动物(优选的是人类)的细胞中具有功能的启动子中进行选择。启动子可来源于病毒或真核基因的启动子序列。例如,它可以是来源于在其内的异源基因的表达能发生的细胞的基因组的启动子,优选的是哺乳动物的中枢或外周神经系统的细胞。关于真核启动子,它们可以是以普遍方式起作用的启动子(如α-肌动蛋白的,β-肌动蛋白,微管蛋白的启动子),或者,还有以组织特异性方式起作用的启动子(如丙酮酸激酶基因的启动子)。尤其优选的是仅仅在特定神经元的细胞类型内活化的启动子(例如酪氨酸羟化酶(TH)的,L7的,或神经元特异性的烯醇化酶(NSE)的启动子)。它们也可以是对特定刺激作出反应的启动子,例如结合甾族激素受体的启动子。也可利用病毒的启动子,例如Moloney鼠白血病病毒长末端重复序列,(MMLV LTR)启动子,HSVI或HSV2 LAT p1启动子,rous肉瘤病毒(RSV)LTR启动子或人类巨细胞病毒(CMV)IE启动子。
启动子为诱导型的更有利,这样在细胞生命期内可调节异源基因的表达水平。诱导性是指可以调节利用启动子所获得的表达水平。例如,在优选的实施方案中其中两个启动子以相反的方向位于单个LATP2区域的侧翼,其中一个启动子包含了对以前报导的tet阻遏物/VP16转录激活融合蛋白质作出响应的启动子(Gossen和Bujard,1992;Gossen等,1995),其并且驱动要调节的异源基因的表达。第二启动子将包含一个强启动子(例如CMV IE启动子),其驱动tet阻遏物/VP16融合蛋白质的表达。这样在这一例子中第一异源基因的表达将取决于四环素的存在或缺乏。
此外,这些启动子的之任一可以通过增加另外的调节序列(例如增强子序列)进行修饰。也可以利用嵌合的启动子,其包含来自上述两个或多个不同启动子的序列元件。
3.异源基因

优选的是异源基因编码具有治疗用途的多肽或其功能可能与疾病过程相关的多肽。例如,上述蛋白质中的酪氨酸羟化酶和胶质细胞来源的神经营养因子可用于治疗帕金森氏病,视紫红质可用于治疗眼疾,肌营养不良蛋白可用来治疗肌肉萎缩,热休克蛋白质可用来治疗与局部缺血紧张相关的心脑病。治疗用途的多肽也可包括细胞毒性多肽(如蓖麻毒蛋白)或利用(例如,定位病毒的酶前药治疗或定位基因的酶前药治疗的方法,能够将前药转化成为细胞毒性的化合物的酶。在后者情况下,要求保证酶具有合适的信号序列以定位到细胞表面,优选的是使得酶暴露在细胞表面的外部但锚定在细胞膜上的信号序列。合适的酶包括细菌的硝基还原酶(如WO93/08288描述的大肠杆菌硝基还原酶)或羧肽酶,尤其是WO88/07378描述的羧肽酶CPG2。其它酶在参考EP-A-415731中可找到。合适的前药包括氮芥前药和其它化合物如本文一并参考的WO88/07378,WO89/10140,WO90/02729和WO93/08288中所描述的那些化合物。
异源基因也可编码作为疫苗利用的抗原多肽。优选地,这些抗原多肽来自致病有机体例如细菌或病毒或肿瘤。
异源基因也可包括标记基因(例如编码β-半乳糖苷酶或绿色荧光蛋白)或其产物调节其它基因的表达的基因(例如,上述的转录调节因子包含tet阻遏物/VP16转录激活融合蛋白)。
B.载体
表达盒可以以裸露的核酸构建体形式进行利用。此外,可将它导入到各种核酸载体中。载体包括质粒和病毒载体,优选的是HSV载体。载体还可包含位于表达盒侧翼的序列,它们含有与真核基因组序列同源的序列,优选的是哺乳动物的基因组序列,或病毒的基因组序列。这可把表达盒通过同源重组而导入真核细胞或病毒的基因组内。具体地说,包含侧翼为病毒序列(优选的是HSVI或HSV2序列)的表达盒的质粒载体可用来制备适于向哺乳动物的细胞传送表达盒的病毒载体(优选的是HSV载体)。下文详述单纯疱疹病毒。然而,所采用的技术是技术人员所熟知的技术,并且其适于其它病毒如腺病毒。其它的合适的病毒载体的例子包括能够将其它们的染色体组整合到宿主细胞的基因组上的病毒载体,例如逆转录病毒,包括慢病毒和腺伴随病毒。
C.单纯疱疹病毒载体
1.
病毒毒株
本发明的包含表达盒的HSV毒株可来自例如HSV1或HSV2毒株,或者其衍生物,优选的是HSV1。衍生物包括包含HSV1和HSV2毒株的DNA的类型内的重组体。优选地,衍生物具有与HSV1或HSV2基因组至少70%的序列同源性,更优选的至少90%,尤其优选的95%。
在治疗方法中利用HSV毒株要求减毒毒株处理,这样它们不能建立裂解循环。具体地说,如果将HSV载体用于人类基因治疗中,表达盒应该优选地插入一个必需基因。这是因为如果载体病毒遇到野生型病毒,通过重组可以发生异源基因转移到野生型病毒。然而,只要异源基因中插入了一个必需基因,重组转移将使受体病毒中缺失必需基因,并且阻止异源基因“逃跑”成为感受态野生型病毒的复制。
减毒毒株可用来产生本发明的HSV毒株,本文仅仅给出其例子,包括在ICP34.5或ICP27有突变的毒株,例如均发生ICP34.5突变的毒株1716(MacLean等,1991),毒株R3616和R4009(Chou和Roizman,1992)和R930(Chou等,1994),以及ICP27缺失的d27-1(Rice和Knipe,1990)。此外,ICP4,ICP0,ICP22,ICP6,ICP47,vhs或gH缺失的毒株,在VMW65发生失活突变的毒株,或上述的任何组合均可用来产生本发明的HSV毒株。
用于描述各种HSV基因的术语参见Coffin和Latchman,1996。
2.
互补细胞系
ICP27缺损的HSV病毒在表达ICP27细胞株中进行繁殖,例如V27细胞(Rice和Knipe,1990),2-2细胞(Smith等,1992)或B130/2细胞(参见实施例),优选的是B130/2细胞。
通过用一种包含功能HSV ICP27基因(其能在所说的细胞中被表达)的载体(优选的是质粒载体)和编码选择性标记(例如新霉素抗性)的载体(优选的是质粒载体)共转染哺乳动物的细胞(例如Vero或BHK细胞)可制备表达ICP27的细胞系。然后筛选具有选择性标记的克隆,并进一步确定哪些克隆也表达有功能的ICP27,例如利用为本领域技术人员所熟知的方法(例如Rice和Knipe,1990所描述的方法)根据他们支持ICP27~突变HSV毒株的生长的能力确定。
如上述制备不会使ICP27~突变HSV毒株回复突变成具有有功能的ICP27毒株的细胞系,确保含有功能ICP27基因的载体不含有与ICP27~突变体病毒重叠(即同源)的序列。
当本发明的HSV毒株包含对其它必需基因(例如ICP4)的失活修饰时,互补细胞系将还包含一个有功能的HSV基因,其以ICP27的所描述的相同方式互补被修饰的必需基因。
3.
突变方法
通过本领域中熟知的几种技术可使HSV基因的功能失活。例如,通过缺失,取代或插入(优选的是缺失)可使它们功能失活。缺失可除去部分或整个基因。插入的序列可以包括上述的表达盒。
HSV毒株的突变可通过本领域技术人员熟知的同源重组的方法得到。例如,用含有突变序列(其侧翼为HSV的同源序列)的载体(优选的是质粒载体)转染HSV基因组DNA。突变序列可包含缺失,插入或取代,所有这些可通过常规技术来构建。插入可包括选择性标记基因,例如lacZ,例如,通过β-半乳糖苷酶活性来筛选重组病毒。
也可在其它HSV基因中获得突变,例如基因ICP0,ICP4,ICP6,ICP22,ICP47,VMW65,gH或vhs。为VMW65基因时,整个基因不能全缺失,因为它编码一种必需的结构蛋白,但是可进行小的失活插入其可消除VMW65转录激活IE基因的能力(Ace等,1989)。
4.包含表达盒的HSV毒株
表达盒可插入到HSV基因组中的任何位置,只要病毒仍然可以繁殖,如果异源基因插入在必需基因内,那么要求利用携带另一个HSV必需基因的细胞系(如2.所描述的)。例如,如果异源基因插入在HSV毒株的ICP27基因内,那么需要表达ICP27的细胞系。表达盒优选插入到ICP27突变的区域内,因为在不太可能事件中突变通过与野生型病毒的重组得到修复,这种修复除去了插入的表达盒。
例如,如同上述的导入突变,通过HSV毒株与具有表达盒(其侧翼是HSV序列)的质粒载体的同源重组可将表达盒插入到HSV基因组中。利用本领域熟知的克隆技术可将表达盒导入到包含HSV序列的合适的质粒载体中。
在病毒基因组内插入多个表达盒是可能的,这样一个病毒载体可包含多个本发明的表达盒。
通过克隆包含了内源性LAT P2区域的下游区有效连接了异源基因的启动子的构建体来利用内源性LAT P2区域也是可能的。另一个构建体可以相反的取向克隆到LAT P2区域的上游区。这样所形成的病毒将包含本发明的表达盒,但是其通过与上述稍不同的方法来制备。
D.施用
本发明的表达盒可用来向需要治疗的人或动物传送治疗基因。具体地说,单纯疱疹病毒的神经营养性质使得利用包含本发明的表达盒的减毒HSV毒株非常适于治疗某些疾病,例如帕金森氏病,神经系统疾病,脊椎损伤,中风或恶性肿瘤,例如神经胶质瘤。此外,本发明的表达盒可用来传送作为疫苗用途的编码潜在免疫原性的多肽的基因。
本发明的表达盒可以以裸露的核酸构建体(优选的还包含侧翼的与宿主细胞基因组同源的序列)形式直接施用。通过几种已知的技术(包括biolistic转化和脂转染技术)促进哺乳动物细胞摄取裸露的核酸构建体。
此外,表达盒可以作为核酸载体的一部分来施用,这些载体包括质粒载体或病毒载体,优选的是HSV。
优选地,送递载体(即,例如裸露的核酸构建体或包含表达盒的病毒载体)以及药学上可接受的载体或稀释剂相结合以产生药物组合物。合适的载体和稀释液包括等渗盐溶液,例如磷酸缓冲盐水。可将组合物制成肠胃外,肌内,静脉内,颅内,皮下,眼内,以及透皮施用的制剂。
将含有用于基因治疗的治疗基因的表达盒以其会在适当的区域整合到细胞内的方法来施用药物组合物。例如,当基因治疗的靶是中枢或外周神经系统并且表达盒由单纯疱疹病毒载体进行传送时,可在突触末端定位的区域中施用组合物,这样末端可摄取这些病毒并且通过逆行轴索运输以一种逆行的方式经轴突运输至轴索的细胞体中。通常通过趋实体性接种方法将药物组合物施用至人脑。当施用药物组合物至眼时,视网膜下注射是通常采用的技术。
当用病毒载体将表达盒传送至细胞时,所施用的病毒的量通常在103至1010pfu的范围内,优选105至108pfu,更优选106至107pfu。注射时,通常施用1-10μl位于药学上可接受的合适的载体或稀释剂中的病毒。
当表达盒以裸露的核酸形式施用时,所施用的核酸的量通常在1μg-10mg的范围内,优选100μg至1mg。
当异源基因处于诱导型启动子的控制下时,在治疗整段时期内只需诱导基因的表达。一旦疾病治愈了,除去诱导物,异源基因的表达即停止。这点具有很大的临床优点。例如,这样一种系统包括上述的施用抗生素四环素,其通过影响tet阻遏物/VP16融合蛋白质而激活基因表达。
在利用本发明的表达盒的治疗中,组织特异性启动子具有协助作用。例如,几种神经疾病是由于仅仅在小子集细胞中的特定基因产物的异常表达而引起的。仅在相关影响的细胞类型中表达治疗基因将是有利的,尤其在这样的基因在其它细胞类型中表达时有毒性的情况下。
所描述的施用途径和剂量仅仅作为指导,因为熟练的医师能够针对任何具体的患者与疾病决定最佳施用途径和剂量。
E.分析方法
本发明的表达盒也可用于科学研究的方法中。这样,本发明的另一方面还涉及在体外或体内分析真核细胞(优选的是哺乳动物的细胞)中的基因功能的方法。异源的功能可由下列方法来确定:
(a)将本发明的包含所说异源基因的表达盒导入到真核细胞内;并且
(b)检测所说异源基因在所说的细胞中的表达作用。
例如,某些细胞具有细胞分裂时的温度敏感缺陷。若按照本发明将包含异源基因的表达盒导入到缺陷细胞内并且这些细胞在限定的温度下生长,技术人员将易于检测出异源基因是否能弥补细胞分裂中的缺陷。类似地,如果异源基因的表达能纠正细胞中可观察到的突变表型,那么可以应用其它已知的技术进行检测。
这种方法也可用于进行异源基因的系统突变以确定由该基因编码的蛋白质的哪一个区域与恢复突变表型有关。
本方法也可用于具有所谓的″基因剔除″的动物,例如小鼠。利用本发明的表达盒将野生型异源基因导入到动物体内,并利用本领域已知的各种行为学的,组织化学的,以及生化的分析方法确定对动物的影响。此外,可将突变的异源基因导入到野生型或″基因缺失″的动物内,从检测是否诱导疾病相关的病理学。这一应用的例子是在啮齿类动物中枢神经系统中利用编码朊病毒的基因诱导Creutzfeld-Jacob和其它朊病毒类型的疾病。其它疾病模型可包括老年痴呆症,运动神经元疾病或帕金森氏病模型。
由于利用一种或多种表达盒将至少两个不同的异源基因导入到细胞内是可能的,研究两种或多种基因产物之间的相互作用也是可能的。
这样,本发明的方法尤其可用于研究疾病相关基因的功能。利用本发明的方法来研究基因功能的一个优点是:控制特定基因的暂时表达的能力意味着在细胞发育或修复中确定哪个阶段需要表达的基因(即使是完全表达)是可能的。
参照下列实施例描述本发明,这些实施例是说明性的但不是限制性的。
实施例
实施例1至8,描述了下列构建体的可引起长期表达的LAT P2区域的利用:
1.驱动lacZ长期表达的MMLV LTR启动子,其来自ICP34.5缺失的并且VMW65反式激活活性消失的病毒(病毒毒株1764/pR14)中的LAT区域。本发明中,MMLV LTR/LacZ盒直接插入在LAT区域中LAT P2之后。
2.驱动lacZ表达的CMV启动子,其来自ICP27缺失的病毒(病毒毒株17+/D27/pR19lacZ)中的LAT区域。本发明中,CMV IE启动子lacZ盒直接插入在LAT区域中LAT P2之后。
3.驱动lacZ的LAT P2/CMV启动子,其来自ICP27基因座(即又有ICP27缺失的病毒)(病毒毒株17+/D27/pR20)。本发明中,利用授与非LAT启动子长期活性的LAT P2区域与其它LAT序列隔离。
4.一段LAT P2区域,其下游侧翼有MMLV LTR启动子/GFP盒且上游端有相反取向的LAT p1启动子/lacZ盒(pR20.9盒),并且其被插入到ICP34.5缺失的并且具有VMW65失活突变的病毒(病毒毒株1764/pR20.9)的UL43基因。
材料和方法
病毒和细胞系
通过同源重组的标准方法(例如,参见Coffin以及Latchman,1996;Coffin等,1996)接着通过X-gal染色检测lacZ活性,噬斑纯化和Southern印迹检查重组病毒的正确基因组结构从而制备所有病毒。
(a)亲本病毒和ICP27-互补细胞系:
17+/D27w
用标准的方法,首先通过同源重组的方法从以前所产生的ICP27缺失的病毒(其中lacZ已插入在ICP27基因座内)中除去lacZ基因,然后X-gal染色之后选择白噬斑从而可获得不包含标记基因的ICP27缺失的突变体。该病毒命名为17+/D27w,它除了缺失核苷酸113,273-117,854外其它均为野生型,这样它没有编码ICP27(UL54)的序列并且无非必需的基因UL55和UL56,因此它必须培育在表达ICP27的细胞系内。核苷酸编号是指HSV1毒株17+序列(Genbankno.HE1CG)。
通过在以前制备的ICP27互补BHK细胞系B130/2中培育可产生ICP27缺失病毒并获得其储备,其中互补BHK细胞系是通过质粒pSG130BS(Sekulovich等,1988)DNA与编码新霉素抗性的质粒pMamNeo(Invitrogen)共转染到BHK细胞内然后选择具有新霉素抗性的克隆来获得的。选择能使HSV1 ICP27缺失突变体高度生长的克隆(B130/2)用于培育病毒。pSG130BS含有来自HSV1(核苷酸113,322-115,743)的BamHI/SacI片段,其编码完全的ICP27与部分UL55序列。
1764
以前已描述过HSV毒株1764(Coffin等,1996)。它缺失了编码ICP34.5的基因(MacLean等,1991)并且在编码VMW65(毒粒逆激活蛋白)的基因上发生了失活突变(Ace等,1989)。体外病毒生长ICP34.5是非必需的,而且VMW65可通过在培养基中加入环己烷-二乙酰胺得以补充(McFarlane等,1992),因此可培育在BHK细胞上。
实施例1-1764/pR14的构建
用纯化的1764的基因组DNA与质粒pR14共转染到BHK细胞内,然后X-gal染色之后选择蓝噬斑,这样就可产生病毒毒株1764/pR14。pR14的制备通过将MMLV LTR/lacZ序列插入到pNot 3.5内而获得。pNot 3.5含有的来自克隆在pGem5(Promega)的NotI位点内的HSV1的LAT区域(核苷酸118439-122025)的3.5Kb NotI片段。MMLV LTR/lacZ的插入由两个步骤来完成:
1:将来自pCH110(Phannacia)的lacZ基因(HindIII-BamHI)插入到pJ4(其包含MMLV LTR启动子/聚连接物/SV40的聚A序列;(Morgenstern和Land,1990)的HindIII位点内,得到pJ4lacZb。
2:通过用NheI和PstI切割pJ4lacZ,将来自pJ4lacZ的MMLVLTR/lacZpolyA插入在pNot 3.5的BbsI位点(在LAT P2后端)上,得到pR14。取向:LAT P1/LAT P2/LTR/lacZ。
实施例2-17+/D27/pR19lacZ的构建
用纯化的17+/D27w基因组DNA与质粒pR19lacZ共转染进B130/2细胞内,然后X-gal染色后选择蓝噬斑,来产生病毒毒株17+/D27/pR19lacZ。pR19lacZ是通过将CMV IE启动子/lacZ/polyA盒插入到pNot 3.5的BstXI位点内(即在LAT P2之后)而制备的。首先将来自pCH110(Pharmacia)的lacZ基因(HindIII-BamHI)克隆在pcDNA3(Invitrogen,包含CMV IE启动子/聚连接物/polyA序列)的BamHI和HindIII位点之间。然后用NruI和BbsI切割CMV IE启动子/lacZ/polyA盒,并插入到pNot 3.5中的BstXI位点内。取向:LAT P1/LAT P2/CMV/lacZ/polyA。
实施例3-17+/D27/pR20的构建
用纯化的17+/D27w基因组DNA与质粒pR20共转染到B130/2细胞内,然后X-gal染色后选择蓝噬斑,来产生病毒毒株17+/D27/pR20。pR20是通过将LAT P2/CMV IE启动子/lacZ/polyA盒(来自pR19lacZ的PstI-SrfI。SrfI位点恰好在pNot 3.5内BstXI位点后。)插入到pDMN内而获得的。pDMN通过缺失克隆在pACYC184(NBL)内的HSVI基因组的EcoR1 B片段的NotI/XmnI片段,留下包含ICP27基因和侧翼序列(HSV1毒株17+核苷酸11095-118439)进行制备。通过MluI消化和再连接除去一对编码整个ICP27编码序列的MluI片段与非必需基因UL55和56(核苷酸113273-116869)。然后在MluI位点插入LAT P2/CMV IE启动子/lacZ/polyA盒。
实施例4-1764/pR20.9的构建
用纯化的1764基因组DNA与质粒pR20.9/43共转染,使得pR20.9盒插入在病毒毒株1764的UL43基因内从而产生病毒毒株1764-pR20.9。该构建体包含两个异源基因,第一基因绿色荧光蛋白(GFP)处于MMLV LTR启动子的控制下,而第二基因lacZ处于LAT p1启动子的控制下。它们以相反的方向位于LAT P2区域的侧翼,这样各基因(lacZ和GFP)从中心位置的LAT P2区开始以相反的方向转录。
质粒pR20.9/43由下列步骤进行构建:
(i)将GFP插入在质粒pcDNA3(Invitrogen)内,其通过用AgeI和NotI切割下质粒pEGFP-N1(Clontech)的GFP基因并将其插入到质粒pcDNA3的EcoR1和NotI位点之间来完成,这样得到质粒pcDNA3GFP。GFP基因的起始处定位在CMV启动子之后。
(ii)将来自pJ4(NheI-Hind III)的MMLV启动子插入到pcDNA3GFP的BamHI位点内的,得到了pcMMLVGFP,其中pJ4的NheI位点位于CMV启动子之后。
(iii)将来自pcMMLVGFP的MMLV/GFP/pA盒插入到pNot3.5内,其通过用HindIII和BbsI进行切割并插入在pNot3.5的BstXI位点之间(即在LAT P2序列之后)而完成,其中MMLV启动子位于LAT P2区之后,这样得到了质粒p3.5MG。
(iv)将来自Ddel-StyI的LAT P1启动子(nt 118,179-118,877)插入在pGem5(Promega)的EcoRV和SpeI位点之间(方向:StyI连接EcoRV,DdeI连接SpeI),得到pGemSPI。
(v)将来自pCH110(Pharmacia)的lacZ(HindIII-BamHI)插入在pGEM5PI的NcoI位点内(方向:HindIII在LAT P1序列之后),得到pP1/lacZ。
(VI)将编码一个Srf1位点(5′-GCCCGGGCCATG)的寡核苷酸插入在pP1/lacZ的SphI位点内,得到pP1/lacZSrf。
(vii)将来自p3.5MG的LAT P2/MMLV/GFP/pA盒(PstI片段)插入在pP1/lacZSrf的NsiI位点内,其中来自p3.5MG的LAT P2区位于pP1/lacZSrf的LAT P1区之后,得到质粒pR20.9。
(viii)将pR20.9盒插入在UL43侧翼区中,其通过用SrfI切割并插入在p35唯一的NsiI位点内而完成,得到pR20.9/43。p35含有克隆在pGeml(Promega)内的HSV1 nts 91,610-96,751(BamHI-EcoRI)。
实施例5-用1764/pR14/TH接种小鼠/大鼠
外周神经系统
通过爪垫接种小鼠(1×107pfu),接着处死小鼠并解剖腰背根神经节来检测1764/pR14。用标准方法(Coffin等,1996)进行固定和X-gal染色,随后分别在2天,2周,1个月和2个月(检测的最长时间)可观察到腰神经节L4和L5处为蓝色,这表明了长期的启动子活性。
中枢神经系统
在给大鼠纹状体(200-220g路易斯大鼠)进行趋实体性接种(2μl5×105pfu)后,接着分别在2天,2周,1个月和2个月(检验的最长时间)进行固定和X-gal染色后,1764/pR14为蓝色。这再次表明长期的启动子活性。
实施例6-用17+/D27/PR19接种大鼠
中枢神经系统
在给大鼠纹状体(200-220g路易斯大鼠)进行趋实体性接种(2μl5×106pfu)后,并分别在2天,2周,1个月和2个月(检验的最长时间)进行固定和x-gal染色后,17+/D27/pR19显示了强的蓝色。这表明CMV IE启动子当放置在LAT P2之后时,能象MMLV LTR一样显示出长期活性。
实施例7-用17+/D27/pR20接种大鼠
中枢神经系统
在给大鼠纹状体(200-220g路易斯大鼠)进行趋实体性接种(2μl5×106pfu)后,并分别在2天,2周,1个月(检验的最长时间)进行固定和X-gal染色后,17+/D27/pR20也显示了强的蓝色。重要地是,这表明当插入在疱疹基因组中的其它位置时(即不在LAT区域内),LAT P2能使邻近的启动子具有长期活性。
实施例8-用1764/pR20.9接种小鼠
外周神经系统
通过爪垫接种小鼠(1×107pfu),接着处死小鼠并解剖腰背根神经节来检测1764/pR20.9。固定后在荧光显微镜(荧光光学设备)下能观察到在腰神经节L4和L5处(以及较低程度在其它DRG中)有强的绿色荧光,X-gal染色后在2天,2周,1个月和2个月(检测的最长时间)时在相同细胞中可观察到蓝色。这表明两个启动子在长时间内均处于激活状态,并显示了利用本发明的盒可有效地表达成对基因。

Claims (35)

它们按该顺序有效连结,其中所述LAT P2区能够使异源基因在一被含有该表达盒的HSV株感染的真核细胞中在感染后表达至少一个月。
2.按照权利要求1的表达盒,其中LAT P2区是HSV-2 LAT P2区。
3.按照权利要求1的表达盒,其中LAT P2区是HSV-1 LAT P2区。
4.按照权利要求3的表达盒,其中LAT P2区是HSV-1株17+的118,866-120,219位核苷酸。
5.按照权利要求1的表达盒,其中所说的启动子是病毒启动子。
6.按照权利要求1的表达盒,其中所说的启动子是使所说的异源基因在哺乳动物细胞中表达的哺乳动物的启动子。

12.按照权利要求1-7之任一的表达盒,其中所说的异源基因选自编码下列物质的基因:与细胞分裂调节有关的蛋白质、与细胞代谢途径有关的酶、转录因子和热休克蛋白质。
13.按照权利要求1-7之任一的表达盒,其中所说的异源基因编码含有至少一个表位的多肽。
14.按照权利要求13的表达盒,其中所说的多肽来自病原体。
15.按照权利要求1的表达盒,其还包含第二启动子和第二异源基因,它们按该顺序与HSV LAT P2区有效连接,并以与第一启动子和第一异源基因取向相反的方式连接,其中所说的第二启动子与第一启动子相同或不同,所说的第二异源基因与第一异源基因相同或不同。
16.按照权利要求15的表达盒,其中所说的第二启动子是如权利要求1和5-8之任一所限定的,所说的第二异源基因是如权利要求9-14之任一所限定的。
17.按照权利要求15或16的表达盒,其中所说的第一异源基因的产物在合适的生理条件下调节所说的第二异源基因的表达。
18.包含权利要求1-12和15-17之任一所限定的表达盒的核酸载体。
19.按照权利要求18的载体,其还包含位于所说表达盒侧翼的哺乳动物的基因组序列。
20.按照权利要求18的载体,其还包含位于所说表达盒侧翼的HSV基因组序列。
21.一种包含按照权利要求13或14的表达盒的载体。
22.包含上述权利要求1-12和15-17之任一所限定的表达盒的HSV毒株。
23.按照权利要求22的HSV毒株,它是HSV-1毒株。
24.按照权利要求22的HSV毒株,它是HSV-2毒株。
25.按照权利要求22或23的HSV毒株,其中所说的LAT P2区是内源性HSV LAT P2区。

(a)将按照权利要求1-12和15-17之任一的表达盒,按照权利要求18-20之任一的载体或按照权利要求22-25之任一的HSV毒株导入至真核细胞内;
(b)检测所说的一个或多个异源基因在所说的真核细胞中的表达作用。
31.按照权利要求30的方法,其中所说的异源基因是与致病相关的野生型或突变基因。
32.按照权利要求30或31的方法,其中所说的真核细胞有机能障碍,并且所说的异源基因是野生型的。
33.按照权利要求30或31的方法,其中所说的真核细胞具有一个或多个通过突变失活的内源性基因。
34.一种产生按照权利要求22-26之任一的HSV毒株的方法,该方法包括将按照权利要求1-17之任一所限定的表达盒导入到单纯疱疹病毒的基因组内。
35.按照权利要求34的方法,其中通过所述单纯疱疹病毒的基因组与权利要求18-20之任一定义的载体之间的同源重组将所述表达盒导入到所说的基因组内。
