发明背景
树突细胞(DC)是最有潜力的抗原呈递细胞,能够有效诱导针对甚至免疫系统已耐受的抗原的反应。因此,对于其中引发对抗肿瘤的免疫反应的肿瘤免疫疗法来说,如果能使DC呈递肿瘤特异性抗原,那么使用DC将是理想的。DC也可以用于呈递衍生自传染原(如细菌、病毒或寄生虫)的抗原,为这样的疾病提供保护性或治疗性疫苗。然而,为这些靶中的任何一个将抗原有效地转移进DC已经证明是关于该方法的最大问题。
为实现产生对抗肿瘤抗原或其它疾病相关抗原的治疗免疫反应,需要满足几个条件。首先,需要鉴定其表达是肿瘤特异性或疾病特异性(或至少是选择性)、并因此能够作为免疫反应的靶起作用的分子。已经证明这项工作对于大多数普通肿瘤来说是非常困难的,但是例如宫颈癌因为在大多数情况下存在病毒癌基因E6和E7而得到解决,而对于其它肿瘤,已经开始鉴定出好的候选抗原。例如,MUC-1基因产物在许多肿瘤、包括90%的卵巢癌中过度表达。也已经鉴定了各种其它肿瘤相关抗原,所述肿瘤相关抗原中的任何一种都可以用于癌症的免疫治疗。第二,在鉴定所述一种抗原/多种抗原后,需要将所述抗原以免疫原形式传递到免疫系统。为产生对于肿瘤排斥来说关键的细胞免疫反应,这意味着必须或者将所述蛋白传递进入宿主细胞的细胞质(对于高分子量蛋白抗原来说是一项困难的工作),或者在基因送递或DNA免疫接种后由所述宿主细胞本身合成所述蛋白。已经考虑用于该目的的病毒载体包括牛痘、腺病毒或逆转录病毒。

我们以前已经测试并报道了单纯疱疹病毒(HSV)能够有效感染并送递基因到树突细胞(Coffin等,1998),但未测量相关毒性。在此目的上HSV比其它载体系统具有许多优点,因为它能感染广泛的细胞类型(包括一些用其它载体系统非常难感染的细胞类型,如Dilloo等,1997;Coffin等,1998)、易于操作、并且能够接受大DNA插入片段,以允许多个基因的表达(由Coffin和Latchman 1996综述)。对于一些癌症和传染性疾病的治疗来说,送递多个基因到树突细胞、然后将其重新引入体内可能是一种特别有前途的方法。
发明简述
我们以前已经发现:与我们所掌握的其它载体不同,HSV可以有效地转导树突细胞(Coffin等,1998),但没有测试所转导的树突细胞的任何毒性效应以及功能大小(functional capability)。目前我们已经测试了多种HSV突变体,包括从基本上是野生型的病毒到具有防止在一些细胞类型中复制的突变(在非必需基因中的突变)或防止在所有细胞类型中复制的突变(在必需基因上的突变)的病毒,以及一些必需基因和非必需基因上的均有缺失的突变体。我们已经发现这些各种突变体在基因送递效率以及所显示的毒性水平(根据树突细胞死亡估计)上有相当大的差异。
令人惊奇的是,我们已经发现:(1)在ICP34.5、VMW65、vhs以及UL43上具有失活突变的病毒与所测试的其它能够复制的病毒相比,其毒性更低并且对树突细胞的基因送递效率更高,包括以前报道的病毒(Coffin等,1998)以及许多测试过的不能复制的病毒。这是特别令人吃惊的,因为ICP34.5、VMW65、vhs以及UL43通常都被认为是非必需基因。例如,包括一个必需基因缺失(ICP34.5、VMW65、vhs以及ICP27突变)的相似病毒与该病毒相比,显示低得多的基因送递效率。此外,与测试过的其它病毒突变体不同,在所述靶细胞内证实表明有效的抗原加工的结果。这暗示转导树突细胞保留了在体内刺激有效免疫反应的功能能力。我们还发现(ii)与其它仅除去一个立即早期基因的突变体不同,在靶细胞中仅仅表达最低水平的立即早期基因(因此一般预期在靶细胞中仅有非常低水平的HSV基因表达)的病毒产生相似的结果。
这些结果显示:虽然使用HSV载体可以相对容易地获得对树突细胞的有效基因送递,但为使所述细胞保持它们的抗原加工能力,必须仔细选择在所述病毒中特定突变组合。因此,本发明第一次提供了针对树突细胞的病毒载体,所述病毒载体提供高效的基因送递,同时不有害地影响所感染的树突细胞的抗原加工能力。
因此,本发明提供能够有效感染树突细胞并且不会防止所述感染细胞内有效的抗原加工的减毒疱疹病毒。所述疱疹病毒最好是人单纯疱疹病毒。所述疱疹病毒更优选是单纯疱疹病毒(HSV),例如HSV1或HSV2或同源的病毒株。
在一个实施方案中(上面的实施方案(i)),本发明的疱疹病毒如果是HSV株的话,通常缺少有功能的UL43基因以及有功能的vhs基因或它们在其它病毒物种中的同功物。本发明的病毒最好还缺少有功能的HSV ICP34.5基因或它在其它病毒物种的同功物。本发明的病毒还可以缺失有功能的VMW65基因或它在其它病毒物种中的同功物,尤其是由于在所述基因中完全破坏其转录激活活性的突变而造成的缺乏。在第二个实施方案(上面的实施方案(ii)),本发明的疱疹病毒包含在不能互补(contemplate)所述病毒中所述缺失的细胞中最小化立即早期基因表达的突变。这样的病毒包括,例如,在编码ICP27和ICP4的基因上有失活突变、在vmw65编码基因上具有去除其反式激活功能的失活突变(如vmw65突变,如在Ace等,1989或Smiley等1997中)的病毒。这样的病毒还包括在每一个单独的调节性立即早期基因(编码ICP4、ICP27、ICP0和ICP22)上具有失活突变并且可选地失活剩下的非调节性立即早期基因ICP47的病毒。

本发明还提供了带有一个异源基因/多个异源基因的本发明的病毒。术语异源基因包括未在所述病毒基因组中发现的任何基因。所述异源基因可以是野生型基因的任何等位变异体,或者可以是突变基因。异源基因最好有效连接到允许所述异源基因在树突细胞、最好是人类树突细胞中表达的控制序列。因此,本发明的病毒可用于送递异源基因到其将在其中表达的树突细胞。
所述一个异源基因/多个异源基因最好编码治疗用途的多肽。所述一个异源基因/多个异源基因尤其可以编码能够改变免疫反应的多肽,例如细胞因子或其它免疫调制多肽。所述一个异源基因/多个异源基因也可以编码在肿瘤细胞中存在但在非肿瘤细胞中不存在的抗原性多肽,或者与非肿瘤细胞相比在肿瘤细胞中向上调节的多肽。这在癌症治疗中是有用的,因为本发明的感染树突细胞可以用于刺激宿主免疫系统对肿瘤特异性或在肿瘤中广泛存在的一种抗原/多种抗原起反应,导致肿瘤减小(tumor reduction)/肿瘤消退(tumorregression)。所述多肽抗原最好是细胞表面多肽或已经工程化以便能将它转运到细胞表面的多肽(例如通过与信号肽融合)。所述异源基因也可以编码寄生虫来源、病毒来源或细菌来源的一种多肽/多种多肽,使得例如用本发明病毒感染的树突细胞可用于在病原体感染宿主前或感染后刺激所述宿主的免疫系统产生针对所述病原体的免疫反应。所述一个异源基因/多个异源基因也可以编码与其它疾病(如神经变性疾病,如亨廷顿氏病、Alzheimer氏病或帕金森病)有关的一种蛋白/多种蛋白,针对所述蛋白诱导免疫反应可能是有利的。
本发明还提供了带有一个异源基因/多个异源基因的本发明的病毒,所述病毒用于人类和动物治疗,尤其是通过免疫治疗的人类和动物治疗。例如,这样的病毒可用于治疗或预防肿瘤或寄生虫感染、细菌感染或病毒感染。
还提供了用本发明的病毒转导的树突细胞。这样的细胞可用于治疗疾病的来自体内的方法,所述疾病如恶性肿瘤或病毒或细菌病原体的感染。发明详述A.病毒
本发明的减毒病毒能够有效感染树突细胞而不妨碍在所感染的细胞内发生的抗原加工。本发明的病毒最好能够以感染复数1感染至少40%的细胞、更优选至少50、60、70或80%的细胞。使用本发明的病毒在个体细胞内获得的表达水平/效率将根据所表达的多肽以及有效连接到编码所述多肽的基因的控制序列而有所不同,但所述水平/效率至少与使用野生型病毒获得的水平/效率一样。此外,本发明的病毒显示对感染细胞的低毒性。感染后细胞存活率最好在感染后第一天有至少50%,更优选在感染后第一天有至少60、70、80或90%。
为获得降低的毒性同时保持对树突细胞的有效感染以及树突细胞中的抗原加工,本发明的病毒一般将缺少有功能的UL43基因和vhs基因(在HSV中)或其在其它病毒物种中的同源物或同功物。可以制造其它突变以进一步降低所述病毒的毒性,例如通过编码vmw65的基因突变以便最小化所述蛋白的反式激活活性,以及/或者通过编码ICP34.5的基因的失活突变。或者,或另外,所述病毒将包括最小化所述病毒立即早期基因表达的突变,可选地还包括其它突变。因此,本发明的病毒通常也可以发生ICP27、ICP4突变并且发生最小化所述蛋白的反式激活活性的vmw65突变,或用另一种方式在编码ICP4、ICP27、ICP0和ICP22的每一个基因上都发生突变。本发明的病毒另外可以在编码ICP47的基因上发生突变。
虽然本发明已经使用单纯疱疹病毒进行了举例描述,但应当理解:可以修饰疱疹病毒科的其它病毒,以获得降低的毒性同时保持对树突细胞的有效感染以及树突细胞中的抗原加工。尤其是当上面所述HSV基因(特别是UL43和vhs)的基因同源物存在于其它疱疹病毒物种中时,那么将修饰这些同源物。或者或加之,带有所述立即早期基因上的突变或导致立即早期基因表达降低的突变的病毒将是特别优选的。“同源物”意味着一个基因与对应的HSV基因具有序列同源性,所述同源性或者是氨基酸序列同源性或者是核酸序列同源性。HSV基因的同源物一般将与对应的HSV基因在氨基酸水平上至少15%相同、最好至少20%相同。例如猪疱疹病毒、假狂犬病病毒的prv43基因与UL43有23%相同(Powers等,1994)。
测量核酸和蛋白质同源性的方法是本领域内众所周知的。例如,UWGCG软件包提供可以用于计算同源性的BESTFIT程序(Devereux等(1984)Nucleic Acids Research 12:387-395)。
同样,可以用PILEUP和BLAST算法排列序列(例如在AltschulS.F.(1993)J.Mol.Evol.36:290-300;Altschul,S.F.等(1990)J.Mol.Biol.215:403-10中所述)。进行BLAST分析的软件可以通过国立生物技术信息中心(http://www.ncbi.nlm.nih.gov/)公开地获得。
对这样的程序可以有许多不同的设置。依照本发明,可以使用默认设置。
可以使用多种方法鉴定HSV基因的同源物,例如在中到高严格性条件下(例如0.03M氯化钠和0.03M柠檬酸钠,在约50℃到约60℃下),用包含全部或部分所述HSV基因的探针探测由其它病毒制得的基因组文库或cDNA文库。或者,也可以使用简并PCR获得物种同源物,其中使用设计为靶向所述变异体和同源物内编码保守氨基酸序列的序列的引物。所述引物将包含一个或多个简并位点,并在严格条件下使用,所述严格条件低于用于使用单序列引物针对已知序列克隆序列的条件(例如0.03M氯化钠和0.03M柠檬酸钠,在约40℃下)。
为安全性的目的,本发明的病毒是减毒的,一般它们在靶细胞中不能有效复制。可以或者除去为减毒目的改变的病毒区(完全或部分),或者可以使所述病毒区变得没有功能,或者用其它序列、尤其是异源基因序列取代所述病毒区。已经描述了用作病毒载体的所有病毒组的减毒突变。例如,可以通过在ICP34.5和/或必需基因如ICP4和ICP27上的突变使HSV变得无毒。1.单纯疱疹病毒
i.病毒
当本发明的所述病毒是单纯疱疹病毒时,所述病毒可以衍生自例如HSV1或HSV2株或它们的衍生物,最好是HSV1。衍生物包括含有HSV1和HSV2株的DNA的类型间(inter-type)重组体。一般按照本文所述的方法测试,衍生物与HSV1或HSV2基因组优选有至少70%序列同源性,更优选有至少80%序列同源性,甚至更优选有至少90或95%序列同源性。可以用于获得本发明的病毒的其它衍生物包括已经在需要在本发明的病毒中功能性失活的基因中有突变的株,例如在ICP34.5上具有突变的1716株(MacLean等,1991)、R3616株和R4009株(Chou和Roizman,1992)以及R930株(Chou等,1994)、在ICP4上具有缺失的d120株(DeLuca等,1985)、在ICP27上具有缺失的d27-1株(Rice和Knipe,1990)或在ICP27和ICP4上都具有缺失的d92株(Samaniego等,1995)。使用这些株将减少产生本发明的突变HSV株所需的步骤数目。
在描述各种HSV基因时使用的术语与在Coffin和Latchman,1996中的一样。
ii.互补细胞系
当本发明的单纯疱疹病毒缺少特定有功能的必需基因(例如编码ICP4或ICP27的基因)时,可以使用表达该必需基因的细胞系繁殖本发明的所述病毒。例如,当所述病毒缺乏有功能的ICP27基因时,可以使用V27细胞(Rice和Knipe,1990)、2-2细胞(Smith等,1992)或B130/2细胞(WO98/30707)繁殖所述病毒,最好使用B130/2细胞繁殖所述病毒。当所述病毒缺少有功能的ICP4基因时,可以使用表达ICP4的细胞系例如E5细胞(DeLuca等,1985)繁殖所述病毒。当所述病毒缺乏有功能的ICP4基因和有功能的ICP27基因时,可以使用表达ICP4和ICP27的细胞系(如E26细胞;Samaniego等,1995)繁殖所述病毒,当所述病毒另外还缺少有功能的vmw65基因时,可以使用还包含vmw65的非HSV同源物(如马疱疹病毒基因12或牛疱疹病毒的BTIF)的细胞系繁殖所述病毒。
可以共转染哺乳动物细胞,产生表达ICP27的细胞系,如用包含能在所述细胞中表达的有功能的HSV ICP27基因的载体(最好是质粒载体)以及编码选择标记(如新霉素抗性)的载体(最好是质粒载体)共转染哺乳动物细胞,如Vero细胞或BHK细胞。然后使用本领域内技术人员已知的方法(例如Rice和Knipe,1990中所述方法),例如根据其支持ICP27-HSV株生长的能力,进一步筛选含有选择标记的克隆,以确定哪些克隆还表达有功能的ICP27。
如上产生不允许ICP27-突变HSV株回复到具有有功能的ICP27的株的细胞系,确保包含有功能的ICP27基因的载体不包含与保留在ICP27-突变病毒中的序列有重叠(即同源)的序列。
当本发明的HSV株在其它必需基因(例如ICP4)上包含失活修饰时,互补细胞系将以如针对ICP27所述的同样方式互补所述修饰的必需基因。例如,在包含ICP27以及ICP4突变的HSV株的情况下,使用同时表达ICP27和ICP4的细胞系(如E26细胞(Samaniego等,1995))。可以以针对ICP27所述的相似方式构造表达其它必需基因的HSV株。B.突变方法
可以通过本领域内众所周知的几种技术使所提到的各种病毒基因在功能上失活。例如,可以通过缺失、置换或插入使它们在功能上失活,其中最好通过缺失。缺失可以除去所述基因的部分或整个基因。例如,可以造成仅一个核苷酸的缺失,以造成移码。然而,最好制造更大的缺失,例如缺失至少25%、更优选缺失至少50%的整个编码序列和非编码序列(或者,以绝对数表示,至少10个核苷酸、更优选至少100个核苷酸、最优选至少1000个核苷酸)。尤其优选除去整个基因以及一些侧翼序列。插入的序列可以包括下面描述的异源基因。尤其优选在ICP27或ICP4中插入异源序列。在VMW65基因的情况下,由于它编码必需的结构蛋白,因此并不缺失整个基因,而是制造完全破坏VMW65转录激活IE基因的能力的小的失活突变(如在Ace等,1989或Smiley等,1997中所述)。
通过本领域技术人员众所周知的同源重组方法在所述疱疹病毒中制造突变。例如,用包含侧翼有同源HSV序列的突变序列的载体、最好是质粒载体与HSV基因组DNA一起转染。所述突变序列可以包含通过常规技术构造的缺失、插入或取代。插入可以包括选择标记基因,如lacZ或GFP,以通过例如α-半乳糖苷酶活性或荧光筛选重组病毒。C.异源基因和启动子
可以修饰本发明的病毒以携带一个异源基因/多个异源基因。术语“异源基因”包括任何基因。虽然异源基因一般是在疱疹病毒基因组中不存在的基因,但可以使用疱疹病毒基因,只要所述编码序列不与其天然连接的病毒控制序列有效连接。所述异源基因可以是野生型基因的任何等位变异体,或所述异源基因可以是突变基因。术语“基因”包括至少能被转录的核酸序列。因此,该定义包括编码mRNA、tRNA和rRNA的序列。然而,本发明涉及多肽的表达,而不是tRNA和rRNA的表达。编码mRNA的序列将可选地包括与翻译的编码序列天然地或以其它方式连接的部分或全部5′和/或3′转录但不翻译的侧翼序列。它还可以可选地包括与转录序列正常连接的相关转录控制序列,例如转录终止信号、聚腺苷酸化位点和下游增强子元件。
可以通过HSV株与例如携带侧翼有HSV序列的一个异源基因/多个异源基因的质粒载体的同源重组,将所述一个异源基因/多个异源基因插入病毒基因组中。使用本领域内众所周知的克隆技术,可以将所述一个异源基因/多个异源基因导入包含疱疹病毒序列的合适质粒载体。可以将所述一个异源基因/多个异源基因在任何位点插入病毒基因组,只要所述病毒仍然能够繁殖。优选将所述一个异源基因/多个异源基因插入必需基因。异源基因可以插入病毒基因组内的多个位点。
所述一个异源基因/多个异源基因的转录序列最好有效连接到允许所述一个异源基因/多个异源基因在树突细胞中表达的控制序列,所述树突细胞最好是哺乳动物树突细胞、更优选是人类树突细胞。术语“有效连接”指并列(juxtaposition),其中所述组成部分处于允许它们按照它们所固有的方式起作用的关系。与编码序列“有效连接”的控制序列是以这样一种方式连接:在所述方式下,在适合所述控制序列的条件下完成所述编码序列的表达。
所述控制序列包含允许所述一个异源基因/多个异源基因表达的启动子以及转录终止的信号。从在哺乳动物树突细胞、最好是人类树突细胞中有功能的启动子中选择所述启动子。所述启动子可以衍生自真核基因的启动子序列。例如,它可以是衍生自在其中发生异源基因表达的细胞的基因组,所述细胞最好是哺乳动物树突细胞,更优选是人类树突细胞。至于真核启动子,它们可以是以遍在的方式作用的启动子(如β-肌动蛋白启动子、微管蛋白启动子)或者是以组织特异性方式作用的启动子(例如丙酮酸激酶基因的启动子)。它们也可以是应答特定刺激的启动子,例如结合类固醇激素受体的启动子。也可以使用病毒启动子,例如莫洛尼鼠类白血病毒长末端重复(MMLV LTR)启动子或疱疹病毒基因的启动子。
HSV LAT启动子以及包含LAT启动子区的元件的启动子是特别优选的,因为有可能获得所述异源基因在潜伏期内的长期表达。尤其这样的表达盒是特别优选的:所述表达盒主要包含LAT P2区,它本身并不作为启动子,但是与启动子和异源基因以该顺序连接。
术语“长期表达”是用来指在用本发明的单纯疱疹病毒感染的细胞中表达异源基因,甚至在所述单纯疱疹病毒已经进入潜伏期后表达所述异源基因。所述表达最好在感染后持续至少两周,更优选持续至少一个月或两个月,甚至更优选在细胞的生存期内持续。
表达盒还可以包含以该顺序有效连接到所述HSV LAT P2区的第二启动子和第二异源基因,而且所述第二启动子和第二异源基因处于与所述第一启动子和第一异源基因相反的取向,其中所述第二启动子和第二异源基因与所述第一启动子和第一异源基因相同或不同。由此构建了处于相反取向、侧翼有单一LAT P2区的一对启动子/异源基因,所述单一LAT P2区允许异源基因对的长期表达,所述异源基因对可以是相同或不同的,由相同或不同的启动子驱动。此外,所述第一异源基因的产物可以在合适生理条件下调节所述第二异源基因的表达(反之亦然)。
可以使用本领域内技术人员熟知的常规克隆技术制造包含一个异源基因/多个异源基因和控制序列的表达盒以及其它合适的构造物(见例如,Sambrook等,1989,Molecular Cloning-a laboratory manual;Cold Spring Harbor Press)。所述LAT P2区在此定义为HSV1株17+的HSV1核苷酸118866-120219(GenBank HE1CG:从Pst I-BstXI位点起)、并包括该区的片段或衍生物,其中包括其它HSV1株和HSV2株的同源区,所述LAT P2区能够为它们所连接的启动子提供长期表达能力。
所述启动子是诱导型也是有利的,以便可以在所述细胞的生存期内调节所述异源基因的表达水平。诱导型意味着使用所述启动子获得的表达水平是可以调节的。例如,在将多于一个异源基因插入HSV基因组的优选实施方案中,一个启动子将包含对以前报道的tet阻抑物/VP16转录激活物融合蛋白应答的启动子(Gossen和Bujard,1992,Gossen等,1995),并驱动需要调节的异源基因的表达。所述第二启动子将包含驱动所述tet阻抑物/VP16融合蛋白表达的强启动子(如CMV IE启动子)。因此,在这个例子中,所述第一异源基因的表达将取决于四环素存在与否。
此外,可以通过添加其它调节序列来修饰这些启动子中的任何一个,所述调节序列如增强子序列(包括HSV LAT区的元件)。也可以使用包含来自上面所述的两个或更多个不同启动子的序列元件的嵌合启动子,例如MMLV LTR/LAT融合启动子(Lokensgard等,1994)或包含LAT区的元件的启动子(见上文)。
异源基因一般编码治疗用途的多肽。例如,为促进特异性针对特定肿瘤的免疫反应,需要用指导一种肿瘤抗原/多种肿瘤抗原表达的本发明的病毒转染树突细胞。肿瘤抗原可以对一种肿瘤细胞是特异性的,或者由于所述抗原的表达的向上调节,所述抗原在该肿瘤细胞中以比非该类型肿瘤细胞中水平更高。尤其优选所述肿瘤抗原在所述肿瘤细胞表面表达,例如细胞表面受体或细胞粘着蛋白。肿瘤抗原的例子包括在多种肿瘤(包括卵巢癌)中过量表达的MUC-1基因产物(Gendler等,1990)、与宫颈癌有关的人乳头瘤病毒蛋白E6和E7。尤其优选引发T细胞反应的肿瘤抗原。
异源基因也可以编码能够改变免疫反应的多肽,例如细胞因子(如α、β或γ-干扰素、白细胞介素(包括IL-1、IL-2)、肿瘤坏死因子或胰岛素样生长因子I或II)或其它免疫调制蛋白。
异源基因也可以编码用作疫苗的抗原性多肽。这样的抗原性多肽最好衍生自致病生物,例如寄生虫、细菌或病毒。这样的抗原性多肽的例子包括丙型肝炎病毒抗原、乙型肝炎表面抗原或核心抗原、HIV抗原、疟疾抗原、百日咳毒素、霍乱毒素或白喉毒素。
异源基因也可以包括标记基因(例如编码β-半乳糖苷酶或绿荧光蛋白的基因)或其产物调节其它基因表达的基因(例如转录调节因子,包括上文所述的tet阻抑物/VP16转录激活物融合蛋白)。
基因治疗和其它治疗应用很可能要求给予多种基因。多基因的表达对于治疗多种病理状况是有利的。疱疹病毒是唯一合适的,因为它们没有其它病毒载体系统的受到限制的包装能力。因此在它的基因组中可以容纳多个异源基因。例如,有至少两种达到该目的的方法。例如,可以将多于一个的异源基因和相关控制序列或者在病毒基因组中的单一位点或者多个位点引入特定HSV株。还有可能如上所述,使用以相反取向相对的启动子对(相同或不同的启动子),这些启动子每一个都驱动异源基因(相同或不同的异源基因)的表达。D.树突细胞
可以通过多种方法分离/制备树突细胞,例如或者直接从外周血纯化,或者例如通过用G-CSF处理CD34+前体细胞转移进外周血后产生,或者直接从骨髓中纯化它们。可以用GM-CSF/IL-4混合物处理来自外周血的粘附性前体(Inaba等,1992),或者可以用GM-CSF和TNF-α处理来自骨髓的非粘附性CD34+细胞(Caux等,1992)。可以从人类志愿者的外周血中常规制备DC,制备方法类似Sallusto和Lanzavecchia,1994的方法,使用纯化的外周血单核细胞(PBMC)并用GM-CSF和IL-4处理粘附性细胞2小时。然后使用磁珠从这些细胞中去除CD19+B细胞和CD3+、CD2+T细胞(见Coffin等,1998)。也可以用其它方法制备树突细胞。E.治疗用途
本发明的病毒以及用本发明的病毒感染的树突细胞可用于治疗方法。尤其是表达肿瘤抗原的本发明的病毒以及用本发明的病毒感染的树突细胞可用于治疗癌症的方法中。具体地说,本发明的病毒以及用本发明的病毒感染的树突细胞可用于抑制哺乳动物、包括人类的各种肿瘤的生长,例如卵巢、宫颈和子宫内膜的肿瘤以及癌症,例如乳癌、肺癌、膀胱癌和结肠癌。其它生长可能被抑制的肿瘤包括肉瘤(如软组织肉瘤和骨肉瘤)以及血液性恶性肿瘤如白血病。可用表达肿瘤抗原的本发明的病毒和/或用本发明的病毒感染的树突细胞治疗的癌症的特定例子包括黑素瘤、白血病、宫颈癌和卵巢癌。
本发明的病毒以及用本发明的病毒感染的树突细胞可用于治疗或预防致病性感染的方法中,所述致病性感染如寄生虫感染、细菌感染或病毒感染。在这种情况下,在感染前可以给予所述病毒/树突细胞以刺激所述宿主中的保护性免疫反应,或在感染后给予所述病毒/树突细胞,刺激宿主免疫系统以克制所述感染。F.给予
因此,本发明的疱疹病毒可以用于向需要治疗的人或动物送递治疗性基因。使用本发明的疱疹病毒送递治疗性基因可用于治疗,例如,恶性肿瘤以及病原感染。
一种进行治疗的方法涉及如上所述在本发明的疱疹病毒的基因组中插入治疗性基因,然后将得到的重组病毒与药学上可接受的载体或稀释剂结合以产生药用组合物。合适的载体以及稀释剂包括等渗盐溶液,例如磷酸盐缓冲液。配制所述组合物以进行肠胃外给予、肌肉内给予、静脉内给予、皮下给予或经皮给予。
可以通过将包含本发明的病毒的组合物给予患者,在体内用本发明的病毒感染树突细胞。以这样一种方式给予所述药用组合物:所述方式使得包含治疗性基因的所述病毒可以在合适区域引入细胞。所给予的病毒的量在从104到1010pfu的范围内,最好在105到108pfu的范围内,更优选在约106到107pfu的范围内。当进行注射时,一般给予从10μl到1ml、最好给予100μl到1ml的在药学上可接受的合适载体或稀释剂中的病毒。
另一种方法涉及从外周血或骨髓中分离/制备树突细胞,然后在体外用本发明的病毒感染所述细胞。然后一般将转导后的树突细胞给予患者,其中给予方法是肌肉内注射、腹膜内注射、皮下注射或静脉内注射,或者直接注射进所述患者的淋巴结,最好直接注射进淋巴结。一般给予所述患者104到108的转导树突细胞,最好给予所述患者105到107细胞,更优选给予所述患者约106细胞。
所述的给予途径以及剂量仅仅作为参考,因为有经验的执业医师将能够依据例如患者的年龄、体重和病况容易地确定对于任何特定患者的最佳给予途径以及剂量。

所有病毒株衍生自HSV1株17+,其核苷酸序列保藏于GenBank(登录号HE1CG)。除在B130/2细胞(用HSV1 ICP27基因稳定转染的BHK细胞)上培养17+/27-/pR20株和1764/27-/pR20.5/vhs株、用编码HSV1 ICP27、ICP4的基因和马疱疹病毒基因12稳定转染的细胞繁殖1764/27-4-/pR20.5之外,所有株都使用BHK C-21细胞(ECACC No.8501143)产生和繁殖。EHV基因12是HSV vmw65的EHV同源物,我们已经发现当将它包括入细胞系中时,它可以补偿与vmw65突变有关的生长缺陷,但EHV基因12蛋白产物随后并不包装进得到的HSV病毒体,这与在用于培养病毒的细胞中含有的未改变的HSV vmw65时不同。如下产生这样的细胞:在10cm平板中,用包含处于CMV启动子和BGHpA序列(pcDNA3/E)控制下的EHV-基因12(Lewis JB等,(1997)Virology,230,369-375)的质粒以及编码ICP27的质粒(pSG130BS[Sekulovich等1988])转染(通过Gorman,1985的方法)BHK细胞(在DMEM+10%FCS中于37℃/5%CO2下培养,其中DMEM和FCS都来自Gibco)。在用新霉素筛选后,根据它们支持在ICP27和vmw65上发生突变的HSV的生长的能力选择和评估新霉素抗性集落,并选出高度允许(permissive)的集落。为引入ICP4,从包含EHV基因12和ICP27的那些BHK细胞中产生腐草霉素/新霉素抗性细胞系,所述细胞系中ICP4由地塞米松诱导型MMTV启动子和SV40 polyA驱动(使用质粒pMAMzeo/ICP4)。为构造pMAMzeo/ICP4,用作为来自pVgRXR(Invitrogen)的BamH I片段的腐草霉素抗性基因取代质粒pMAMneo(Invitrogen)中的新霉素抗性基因(作为BamH I片段切除)。然后将ICP4编码区(HSV1核苷酸127,167-131,187[Mse I-BstE II]GenBank file helcg)在Xho I位点插入MMTV启动子之后。在转染进包含ICP27/EHV基因12的细胞并用腐草霉素筛选后,随后筛选高度允许在ICP4、ICP27和vmw65上发生突变的HSV的生长的集落(Zeocin;Cayla,Toulouse,France)。
对于在VMW65上具有突变的病毒,在用于培养病毒的培养基中加入3mM六亚甲基二乙酰胺(hexamethylene-bisacetamide)(HMBA)(McFarlane等,1992)。
(i)17+/pR20.5/UL43
使用标准方法,将一个来自质粒pR20.5的盒通过与纯化的HSV117+株基因组DNA的同源重组插入UL43座位,所述盒包含处于相反的背对背取向并由HSV LAT区序列(核苷酸118,866-120,219)分开的RSV/lacZ/pA序列以及CMV/GFP/pA序列。首先将所述pR20.5盒在单一Nsi I位点插入包含UL43侧翼区的质粒(Coffin等,1996),产生质粒pR20.5/43。由于在所述20.5盒的一侧插入了编码Srf I的寡核苷酸,因此可以用Srf I将所述盒从它的pGEM5(Progema)质粒主链上切除下来。从pRc/RSV(Invotrogen)上切下RSV启动子,从pCH110(Pharmacia)上切下lacZ/pA,从pcDNA3(Invitrogen)上切下CMV/pA,并从pEGFP-N1(Clontech)上切下GFP,以构造质粒pR20.5。
(ii)17+/pR20.5/US5
通过将所述pR20.5盒插入US5侧翼区(质粒pΔUS5),将所述pR20.5盒插入HSV1的US5座位,产生质粒pR20.5/US5,随后将其与纯化的HSV1 17+株基因组DNA一起同源重组进入BHK细胞,产生病毒株17+/pR20.5/US5。通过将来自HSV1的包含US5编码区的BamH I-EcoN I片段(核苷酸136,289-131,328)克隆进质粒pAT153,产生质粒pΔUS5。将pR20.5插入US5基因中在核苷酸137,945的单一Sac I位点。
(iii)17+/27-/pR20
质粒pR20用于同源重组进纯化的HSV 17+株DNA。质粒pR20含有插入ICP27侧翼区(pACYC184[NBL]中HSV1核苷酸11095-113273和核苷酸116869-118439允许在连接所述两个片段的单一MluI位点上进行插入)的LAT P2(HSV1核苷酸118866-120219)/CMV/lacZ盒。使用B130/2细胞纯化表达LacZ的噬菌斑。
(iv)1764/pR20.5/UL43
质粒pR20.5/43用于同源重组进纯化的HSV 1764株DNA(Coffin等,1996)并纯化表达GFP和LacZ的噬菌斑。HSV 1764株是其中两个ICP34.5拷贝都已缺失并且在VMW65基因上带有失活突变的17+株(Ace等,1989)。
(v)1764/pR20.5/US5
质粒pR20.5/5用于同源重组进纯化的HSV 1764株DNA,并纯化表达GFP和LacZ的噬菌斑。
(vi)in1814/pR15
质粒pR15用于同源重组进in1814株中编码病毒体宿主中断蛋白(virion host shutoff protein)(UL41)的基因(Ace等,1989)。质粒pR15包含vhs侧翼区,同时在UL41基因中的单一Nru I位点插入了由HSV LAT/MMLV LTR嵌合启动子(基本与Lokensgard等,1994所报道的相同)驱动的lacZ基因。纯化表达LacZ的噬菌斑。
(vii)1764/pR15
质粒pR15用于同源重组进纯化的HSV 1764株DNA,并纯化表达LacZ的噬菌斑。
(viii)1764/27-/pR20.5/vhs
将所述pR20.5盒在单一Nru I位点插入vhs侧翼区,将得到的质粒(pR20.5/vhs)用于同源重组进纯化的HSV 1764/27-株DNA。使用B130/2细胞纯化表达lacZ以及GFP的噬菌斑。通过同源重组除去lacZ基因、缺失ICP27座位周围的核苷酸113322-115743,从1764/27-/pR20株(通过将质粒pR20同源重组进HSV 1764株DNA并选择表达lacZ的噬菌斑而产生)产生1764/27-株。
(ix)1764/pR15/MSVGFP/UL43
将MSV LTR启动子/GFP盒在单一NsiI位点插入UL43侧翼区,产生质粒pMSVGFP/43。然后pMSVGFP/43用于同源重组进纯化的HSV 1764/pR15株基因组DNA,并纯化表达lacZ和GFP的噬菌斑。
(x)1764/27-/4-/pR20.5

这些试验目标在于确定使用基本是野生型的HSV病毒时,HSV能否感染树突细胞并送递基因到树突细胞。使用0.1到10之间的感染复数(MOI)。在本文中每种情况下,如下感染1×105树突细胞:温和沉淀树突细胞,重悬浮于约100μl DMEM病毒悬浮液,37℃温育1小时,然后转移到盛有2ml RPMI/10%FCS+100ng/ml GM-CSF,50ng/ml IL-4的24孔板中。在试验的其余时间内将这些板在37℃/5%CO2下培养。这些试验使用病毒17+/pR20.5/UL43和17+/pR20.5/US5,每种病毒都通过在不同的基因(UL43或US5)插入pR20.5盒而发生了突变。UL43和US5以前都鉴定为是HSV在培养物中的生长所非必需的,并且不影响在小鼠模型体内感染的动力学。通过在感染24小时后在荧光显微镜下计算GFP阳性细胞的数目,评估基因送递的效率。下面表1所示的结果显示HSV可以有效地送递基因到树突细胞。

为确定实施例1中观察到的有效基因送递是否是由于所述病毒的裂解性复制,如实施例1,在以0.1到10的MOI感染树突细胞的情况下绘制生长曲线,并根据时间定量病毒生长。以24小时的间隔,将培养基样品在BHK细胞上测定滴度(BHK细胞允许HSV的生长),并计算噬菌斑的数目。

如上面表2所示,这些试验显示:虽然在用HSV感染的树突细胞培养物中发生有限的增殖性感染,但这仅在低MOI时是明显的(尤其是缺失US5的病毒)。UL43的失活而不是US5的失活进一步降低了这种有限的增殖性感染。看起来在所述树突细胞培养物中可以发生野生型HSV的缓慢转换,这由于UL43的失活而被降低,但这并不伴随着显著的树突细胞死亡(见实施例3)。以前没有发现UL43的失活影响HSV的生长动力学(Maclean等,1991)。实施例3:不同的HSV突变体显示不同水平的基因送递效率以及在树突细胞中不同水平的毒性
如实施例1,将上面所述的病毒株(i)-(x)以0.1到10的MOI感染树突细胞。定期取两份样品(培养物的2×100μl),一份样品根据所测试的具体病毒中插入的种类,观察GFP表达(荧光显微镜检术)和/或用X-gal染色,如下进行X-gal染色:温和地沉淀细胞并重悬浮于固定液(0.8%戊二醛10分钟),然后用X-gal缓冲液处理(见Coffin等,1996)并在37℃温育2小时。另一份样品用锥虫蓝染色(活细胞不能被染色),将未染色细胞的数目作为细胞在原始培养物中的数目的百分率进行计数。在每一组单独试验中,试验重复两次并且一般一次测试三种病毒,病毒(i)用作内部对照以估计(allow for)树突细胞制备间的任何变化。结果以MOI=1显示。

这些结果显示:不同的基因缺失组合产生各种基因送递效率以及毒性缺乏组合的病毒,与原始病毒(i)和(ii)相比,一些组合改善了一种特性或另一种特性或两种特性,而在其它组合中基因送递效率或细胞存活率降低。去除一个必需的立即早期基因(ICP27)看起来产生明显最小毒性的病毒,但基因送递效率显著降低。发现带有失活的非必需基因的组合(ICP34.5、VMW65、vhs和UL43)的病毒实现基因送递效率(至少对于GFP如此,见实施例4)以及缺乏毒性的最好组合,该基因送递效率明显大于所测试的其它病毒中的任何一种,其中所述非必需基因中的每一种以前都已分别显示在体内降低毒性(除UL43外)。然而,也具有最小毒性的病毒(x)显示产生高水平的GFP和lacZ RNA表达(见下),虽然通过荧光可以检测到更少GFP,但其表达水平与病毒(ix)相似。然而,也具有最小毒性的病毒(x)产生高水平的GFP和lacZ RNA表达(见下),虽然通过荧光可以检测到更少GFP,但其表达水平与病毒(ix)相似。使用抗HSV ICP27、ICP4、ICP0、ICP22和ICP47抗体的蛋白质印迹分析以前已经显示:该病毒在不互补所述病毒中的缺陷(即ICP27、ICP4和vmw65上的突变)的细胞中仅表达非常低水平的任何立即早期基因。实施例4:UL43、ICP34.5、vhs和VMW65失活突变的HSV产生这样的结果:该结果表明在转导树突细胞中发生所送递抗原的有效蛋白水解加工
缺失UL43、ICP34.5、vhs和VMW65的病毒产生高度有效的GFP送递。另一方面,LacZ活性虽然在许多细胞中存在,但感染后24小时仅以非常低的水平存在,处于X-gal染色的极限值上。这提高了lacZ和GFP都在所述细胞中有效产生的可能性,但lacZ活性由于对lacZ抗原的有效蛋白水解加工而明显降低了,正如在完全有功能的树突细胞中所预期的那样。所述lacZ基因从相同的启动子开始转录,与在树突细胞中产生强X-gal染色的病毒相同。因此,假如lacZ的加工在所述细胞内发生,那么通过RNA印迹杂交分析应当容易地检测到lacZ mRNA和GFP mRNA处于相当的水平,但通过蛋白质印迹分析检测到的lacZ蛋白水平应当显著降低。
使用来自树突细胞的RNA和蛋白质(在每种情况下以1×105细胞/泳道提取)进行RNA印迹分析和蛋白质印迹分析,所述树突细胞用17+/pR20.5/UL43或者1764/pR15/MSVGFP/UL43以MOI为1感染。然后用或者抗LacZ抗体(Promega)或者抗GFP抗体(Quantum)探测蛋白质印迹。用LacZ基因(用HindIII和BamH I从pCH110(Pharmacia)切下)或GFP基因(用Not I和HindIII从pEGFP-N1(Clontech)切下)探测RNA印迹。使用标准方法(Sambrook等,1989),通过放射性(RNA印迹)或非放射性(蛋白质印迹;ECL,Amersham)方式进行RNA印迹和蛋白质印迹。
结果
蛋白质印迹显示:
(i)在用17+/pR20.5/UL43感染的树突细胞中有效检测到lacZ以及GFP。在每种情况下,在预期分子量处观察到单条确定的带,同时还可见一些更小更弱的带。
(ii)如用X-gal染色所预期的,虽然对1764/pR15/MSV/GFP/UL43容易地检测到GFP,但在预期分子量处的lacZ蛋白水平显著降低。
在每种情况下,蛋白质印迹显示预期大小的强而确定的带,不论细胞用17+/pR20.5/U143或用1764/pR15/MSVGFP/UL43感染。
这些结果显示:虽然lacZ mRNA在用1764/pR15/MSVGFP/UL43和17+/pR20.5/UL43感染后的树突细胞中有效产生,但对于1764/pR15/MSVGFP/UL43,该RNA或者未被翻译,或者lacZ蛋白在翻译后迅速被降解,以致通过蛋白质印迹可检测到的全长蛋白的水平显著降低。考虑到树突细胞在抗原加工中的作用,后者可能是实际情况,因为假如这样一种送递的抗原以有效方式受到加工,这就是所预期的情况。因此,这样的有效抗原加工不会在其它测试的突变体中发生(除了在下面的实施例5中),因为在每种情况下可以见到强X-gal染色。因此,为了使病毒递送的抗原的抗原加工在树突细胞中发生,必须仔细选定用于递送的正确病毒突变体。实施例5:仅有可能表达最小立即早期基因的HSV也允许对树突细胞的有效、低毒的基因送递并产生这样的结果:该结果表明在转导树突细胞中发生所送递抗原的加工
如上面实施例4,比较蛋白质印迹以及RNA印迹(这里对于RNA使用狭线印迹法)。比较在用17+/pR20.5/UL43、1764/pR15/MSVGFP/UL43或1764/27-/4-/pR20.5感染的树突细胞的提取物中LacZ和GFP的蛋白水平以及RNA水平。
结果
除了1764/27-4-/pR20.5感染细胞不同外,该试验的结果如实施例4中一样。这里,虽然与17+/pR20.5/UL43和1764/pR15/MSVGFP/UL43感染的细胞一样可以检测到相等水平的GFP和lacZ RNA,但通过蛋白质印迹仅检测到相当低水平的GFP,该水平显著低于用17+/pR20.5/UL43或1764/pR15/MSVGFP/UL43感染的细胞的水平。因此在1764/27-/4-/pR20.5的情况下,可以检测到相对于根据lacZ和GFP特异性RNA的量所预期的蛋白水平而言更低水平的lacZ蛋白以及GFP蛋白(而不是如前面实施例4中对于1764/pR15/MSVGFP/UL43而言仅仅更低水平的lacZ蛋白),而所述lacZ和GFP特异性RNA的量就实施例4和实施例5中测试的所有病毒而言是相似的。该结果指出:与测试过的其它病毒中的任何一种不同,在用1764/27-/4-/pR20.5送递基因后,lacZ和GFP都承受蛋白水解加工,例如在有功能的树突细胞中可以预期的蛋白水解加工。这暗示了除使用1764/pR15/MSVGFP/UL43送递基因后提供的功能性外,还有其它功能性。
