发明背景
发明领域
本发明涉及组织特异性和靶RNA特异性核酶用于治疗癌症和细菌、寄生虫和病毒感染的应用。更具体的,本发明涉及在前列腺上皮特异性启动子(probasinpromoter)控制下寻靶RNA聚合酶1(A)的核酶。
背景技术
核酶是断裂RNA特异序列的催化性RNA分子。已经指出核酶可作为哺乳动物细胞中的有潜力的基因调节器和抗病毒试剂,但其仍面临着技术可行性的严重问题。例如,还不知道核酶是怎样被引到靶细胞上的,或它们是怎样被指向作为它们的靶RNA的同一亚细胞区室的。其它问题是关于靶RNA二级结构对核酶活性的影响。该领域中还没有成功地解决这些问题。
而且,由于核酶是反义技术的一种,反义技术用于疾病治疗时遇到的问题也在核酶技术应用中遇到。例如,发现反义RNA在转基因小鼠中的表达始终使靶RNA分子减少,而且当靶RNA分子减少时,它并不与蛋白质的减少平行,是不可预测的。有时甚至当蛋白质水平降低时,没有检测到有生物效应(Whitton,J.Lindsay"(反义治疗病毒感染)Antisense Treatment of Viral Infection"Adv.in VirusRes.Vol.44,1994)。
该领域中的经验表明,核酶在游离时或包埋在不相关的大RNA分子中时是是否最有效的,这点是不清楚的(Whitton,1994)。如今,也没有获得足够的体外或细胞培养中的数据来系统地比较游离的核酶和其包埋型的反式活性(transeactivities)。
有些研究侧重于核酶技术在治疗癌症方面的潜力。在这些研究中,核酶被直接作用于细胞培养物中的c-fos和c-ras致癌基因,并在移植入小鼠时表现出抑制恶性的活性。因此,这些核酶可特异性寻靶致癌基因。
没有任何文献指出,通过将有组织特异性启动子的核酶输递至组织上并且寻靶至细胞存活所必需的RNA上,可治疗组织特异性癌症或其它组织特异性疾病。本发明提供了这样一种可治疗组织特异性癌症和其它组织特异性疾病的核酶。
大多数前列腺癌症问题不需要作很多介绍。每年有大约44000个男子死于前列腺癌症,约10000000个男子有前列腺癌症前期的疾病症状。很明显,新的治疗方法是急需的。用动物模型来测试医疗方法刚刚变成可能,并且还需要数年才能有效化。然而,本发明提供了解决这一问题的重要的试剂。
用基因治疗法来治疗前列腺癌症的一个难点是靶特异性输递的长时间持久问题。然而,最近研究出的前列腺上皮特异性启动子提供了靶特异性,其可全身输递本发明的编码核酶的载体(Greenburg等人,Mol.Endocrinol.8:230-239,1994)。本发明的载体包括串联排列的3个锤头状核酶,其5'和3'端设计成可将其从初级转录物上自催化断裂下来。这一新颖的结构消除了延伸残基侧翼序列带来的问题,否则侧翼序列的存在将妨碍催化活性和特异性。本发明的结构将前列腺特异性的前列腺上皮特异性启动子与可寻靶前列腺细胞生长关键必需的mRNA的三连体核酶(triple-ribozyme)连接起来。
在组织特异性或其它启动子控制下的核酶的内源性输递由于“渗漏(leakiness)”(体外组织中发生低水平的转录)而复杂。这将根据所选的细胞靶的不同而产生相当严重的治疗问题。本发明的核酶通过寻靶有高水平产物的细胞靶(即RNA聚合酶Ⅰ产生大量的细胞核糖体RNA)来弥补了该问题。因此,当启动子渗漏发生在不希望的组织中时细胞就不会死亡。因此,这种选择提供了所需程度的安全性,且polⅠ的寻靶适用于许多采用其它启动子的选择性组织。
基因治疗中的一个常见问题是将核酶输递至正确的组织很困难。本发明通过使核酶寻靶到细胞(核酶构建物可有效输递至其上)中非细胞类RNA来避免了这一困难。Ⅳ脂质体输递对治疗HBV肝炎是有效的。Ⅳ和/或体外治疗有效地将构建物输递至红细胞上来治疗疟疾传染。表面给药(有或没有ⅳ)有效地将核酶构建物输递至发育不良/癌症前期/癌症的HPV 16颈损伤的颈上皮上。后一个例子对治疗发育不良/癌原位损伤(经异常的巴氏污点(Pap smear)诊断得出)特别重要。非细胞类靶核酶的第二个优点是,即使不希望的细胞中发生启动子渗漏和/或体外输递和/或表达,核酶也不会断裂任何细胞RNA。
发明概述

发明详述
核酶
本发明提供了组织特异性和靶RNA特异性的核酶。这些核酶可用来破坏靶特异性肿瘤和治疗病毒、细菌或寄生虫感染及其它用途。本发明的核酶包括5'端自催化断裂的核酶序列、含靶RNA特异性结合位点的催化性核酶和3'端自催化断裂核酶。本发明的核酶的一个例子以其DNA编码序列显示在图2和SEQ IDNO:1中。1-164位的核苷酸编码了包括5'端和3'端自催化核酶序列的核酶。这种典型核酶的5'端自催化断裂核酶、催化性核酶和3'端自催化断裂核酶分别显示在SEQ ID NO:1中。
或者,5'端自催化断裂核酶可用转录的RNA的另一段序列来代替。在该RNA中,最初的20-30个(或更长)核苷酸后的序列是最初20-30个核苷酸反向补体。这样,构建物估计可能仍在5'端与polⅡ转录物一样形成帽结构,但是最初核苷酸不会改变寻靶中间核酶的5'侧上核苷酸的特异性。由于转录物将形成帽结构,它可有效地输送至细胞质。对于本发明有5'和3'自催化核酶的三连体核酶来说,希望有一些扩散将内部寻靶的核酶介导输送至细胞质,尽管这一替换的5'端应当提高细胞质比例。
本发明提供了有独特特征的核酶,其特征是在其催化反应中有靶RNA特异性,并被组织特异性表达。在图1和SEQ IN NO:1所示的例子中,可与RNA聚合酶Ⅰ(A)(核糖体RNA聚合酶)单一序列特异性结合的RNA结合位点提供了靶RNA特异性。应当理解任何RNA的单一RNA序列可以作为本发明的靶RNA特异性核酶的靶。单一序列的测定方法是常规的,只要有大量的计算机数据库(GenBank)和计算机程序(Genetics Computer Group,PCGENE and BLAST)来检索查找任何与核酸序列配合的序列即可。位于一个数据库上的单一DNA序列会有相应的单一RNA序列。

本发明也提供了编码本发明的核酶的核酸。这些核酸可用来在选择的位点上表达本发明的核酶。在治疗组织特异性癌症时,这些位点可以是组织特异性的,或者在核酶防止感染性试剂来治疗感染(如肝炎、疱疹、疟疾、结核病等)时可以是靶特异性的。
本发明的核酸包括一个组织特异性启动子的结合位点,位点在编码5'端自催化断裂核酶序列、包括靶RNA特异性结合位点的催化性核酶和3'端自催化断裂核酶序列的序列上游。
产生核酶的构建物中的组织特异性启动子结合位点使得核酶在组织中组织特异性表达,从选择的启动子开始活性转录RNA。因此,组织中只有利用启动子的靶RNA会被核酶断裂。图1和SEQ ID NO:1中显示的典型的核酶采用了Probasin启动子(一种前列腺上皮特异性启动子)(Greenburg等人,1994)的结合位点。该组织特异性启动子结合位点的序列显示在(Greenburg等人,1994)中。
如所预料的,本发明的核酸构建物中可采用其它组织特异性启动子。这些启动子的例子包括前列腺特异性抗原(前列腺)、白蛋白(肝)、脂肪酸结合蛋白(骼骨)、乳清酸性蛋白(乳房)、平滑肌运动(平滑肌)等的结合位点。还未被鉴定出的靶特异性启动子可用来使本发明的核酶表达定向到选择的组织上。一旦鉴定出靶特异性启动子,其结合序列可用常规方法如序列分析方法来测得。启动子通过缺失分析、诱变、足迹法、凝胶移位和转染分析来确定(Sambrook等人,分子克隆实验手册“Molecular Cloning:A Laboratory Manual”,2nd Ed.Cold Spring HarborLabortory,Cold Spring Harbor,New York,1989)。
在本发明的编码核酶的核酸中,编码5'端自催化断裂核酶的核酸有SEQ IDNO:1中显示的核苷酸1-54的序列。在本发明的编码核酶的核酸中,编码3'端自催化断裂核酶的核酸有SEQ ID NO:1中显示的核苷酸111-164的序列。
应当理解,可以获得本发明核酸编码的其它5'端和3'端自催化断裂核酶。这些核酶可根据Taira等人的方法来获得(Nuc.Acids Res.19(19):5125-5130,1991)。
本发明的核酸编码的催化性核酶含有两个功能区:一个高度保守的断裂靶RNA的催化性序列(也称为“催化核心”),以及包括靶RNA特异性结合位点的侧翼区域。通过核酸互补性,结合位点使核酶定向断裂靶RNA分子上的特异性位点。侧翼序列的长度不仅意味着特异性,也意味着单个核酶分子的断裂效率。在本发明的催化性核酶中,侧翼序列对靶RNA有高的特异性,但一旦发生断裂很容易从靶RNA上解离下来。这就使得核酶可以循环(估计的Kcat约为1断裂/分钟)并减少了达到效果所需的核酶量。结合/解离值的范围在16-21Kcal内应当是有效的。
人RNA的复杂度比人DNA要低约100倍,且特异性只通过12-15碱基对就可实现。RNA-DNA双链体受几个因素的影响,如GC含量、温度、pH、离子浓度和结构。最近邻规则可提供对双链体稳定性的有用评价(Castanotto等人,作为医疗试剂的反义催化RNA“Antisense Catalatic RNAs as Therapeutic Agents”Advances in Pharmacol.25:289-317,1994)。
如上所述,编码的RNA结合位点是单一的,因此核酸编码序列将是相应的单一DNA序列。RNA结合位点可包含与核糖体RNA聚合酶Ⅰ(A)亚单位的单一RNA序列结合的序列。编码核糖体RNA聚合酶结合位点的DNA的序列如图3所示。该序列来自RNA聚合酶Ⅰ(A)亚单位。
本发明的催化性核酶也包括催化性序列,它在靶RNA特异性结合位点结合的位点中间的附近处将靶RNA断裂。在锤头状核酶中,催化性序列通常是高度保守的。保守的催化核心残基是5'CUGANGA3'和5'GAAA3',两者通过进化上保守的茎-环结构来连接。
靶RNA上最保守的、且可能最有效的断裂序列是5'GUC3'。然而,XUN(N=A、U或C)也可有效断裂。该断裂位点普遍存在于大多数RNA中,使得基本上所有的RNA能被寻靶(Whitton,J.Lindsay,病毒感染的反义治疗“AntisenseTreatment of Viral Infection”Adv.in Virus Res.Vol.44,1994)。
关于靶RNA上合适位点的选择,已知靶位点的二级结构对体外断裂有影响(Whitton,1994)。因此,可用程序RNAFOLD(来自PCGENE组程序或国际网Internet上获得)对选择的靶分子序列来常规筛选潜在的二级结构。因此,可对靶可及性进行合理地预测。计算机辅助RNA折叠(Castanotto等人,1994)和RNA三维模型计算机分析(Major等人,Science 253:1255-1260,1991和Castanotto等人,1994)在指导断裂位点的选择上是相当有效的。
内部核酶可寻靶至寄生虫、病毒生命周期等所必需的非细胞类RNA上,且表达可由组织特异性或病毒特异性启动子来驱动。申请中特别指出的三个重要的例子是:
A)肝炎B型病毒靶(选择用相同核酶靶部位来断裂病毒RNA前基因组、S蛋白和聚合酶/反向转录酶转录物)的白蛋白启动子的应用;
B)在红细胞中有活性的普通启动子的应用,用寻靶EMP-1蛋白家族(来自P.falciparum,其是疟疾中细胞粘连和抗原变化所必需的)高度保守区的核酶;和
C)HPV启动子的应用,核酶寻靶E6蛋白翻译起始位点附近的特异性位点,翻译起始位点是E6和E7蛋白质(它们密切参与颈致癌)表达的关键位点。
本发明的一个核酸例子有序列表SEQ ID NO:1中所示的核苷酸序列。该典型的核酸包括前列腺上皮特异性启动子,其在编码5'端自催化断裂核酶(有SEQ IDNO:1中所示序列),编码核糖体RNA结合位点的DNA(有序列表SEQ ID NO:1所示序列),和3'端自催化断裂核酶(有SEQ ID NO:1所示序列)的序列上游。
或者,启动子结合位点和核酶编码序列中可进行沉默碱基取代,以在相同的组织内表达相同的核酶。因此,本发明提供了基本上有SEQ ID NO:1所示核苷酸序列的核酸,其可编码SEQ ID NO:1所示的核酶。核酸可根据所选的启动子的特征/定义而不同,且与SEQ ID NO:1有80%-99%的序列相同性,更佳的,其与SEQ ID NO:1有90%-99%的序列相同性,其它改进包括例如,为克隆目的而插入的核苷酸(图2,其包括-1至-8,+69至+76)的变化(或缺失),在图3中,框包括作为克隆选择的外部核苷酸,因此,它可作改进。未配对的碱基可以是任何碱基,其只由所选的克隆方案来确定。如果一对碱基中的一个改变,则另一个必需以互补方式改变。而且,编码核酶的序列可以以改变核酶序列但不影响核酶的靶RNA特异性或使断裂活性失去的方式来变化。例如,在5'、3'端和内部核酶(图2)的茎环区内的变化可以结合入其它构建物而维持催化活性(Fedor and Uhlenbeck,1990)。
核酶生成构建物的合成
典型地,RNA结合序列和核心序列合成反向互补寡核苷酸,并克隆人载体,从而生成含有核酶的有关RNA。本发明的核酶通过寡核苷酸(5'GGA AGA TCTTTC AAA GAC TGA TGA CTC CGT GAG GAC GAA ACG AGG ATC AGA TCTTCC 3')和其反向互补体的合成来制得。克隆中所用的BglⅡ位点用下划线表示。在合适的限制酶消化后,在这一具体的BglⅡ例子中,双链DNA寡核苷酸被克隆入亲代载体的多克隆位点(图1)。
功能测试
一旦测序后,测定这些核酶的功能。测试可包括用两种可能的细菌启动子中的一种来转录核酶,在这一例中是SP-6或T-7(在痕量放射性存在下),然后通过电泳来评价核酶的自催化断裂。实施例中提供了来自这些测试的数据。
其它测试步骤包括体外转录的核酶与体外合成的靶RNA转录物或细胞质RNA制备物的培育。在培育后,RNA用标准的Northern印迹分析来检验靶RNA转录物的特异性分解。
构建的三连体核酶还可通过将其亚克隆到一个组织特异性启动子启动子之后来进行进一步测试,其中组织特异性启动子可以组织特异性方式驱动靶上的载体表达。实施例中提供了来自这些测试的数据。
最后,三连体核酶实验方法通过在小鼠体内实验来进一步有效化。两组这样的研究如实施例描述的那样进行。第一个例子用了比polⅠ核酶更容易监测的对照载体,其中对照载体包括驱动藻类绿色荧光蛋白质(pBGFT)表达的前列腺上皮特异性启动子。该测试质粒用来确认我们的体内输递系统的有效性。第二个实验也在有polⅠ核酶加入的小鼠中进行。在两个例子中,无论是加入三连体核酶还是绿色荧光蛋白,动物均在操作后不同时间处杀死、解剖,并用免疫组织化学法测定各个组织的载体活性。
输递
本发明的核酸可以是用来将核酸输递至表达核酶的部位处的载体。载体可以是一种商业上可获得的制品,如pGM质粒(Promega),载体输递可以通过脂质体,用商业上可获得的脂质体制品或新研究出的有本发明脂质体特点的脂质体。也可采用其它常规输递方法在这里的方法中测试。用脂质体输递的方法例子在实施例中有进一步描述。
脂质体的给药方式可根据治疗疾病以及寻靶组织的不同而不同。对于脂质体在其中会下沉的肺(如结核病和癌症)和肝(如肝炎和癌症)来说,静脉给药是合理的。对于许多其它局限性病理疾病,如癌症、感染(如肝炎、膀胱炎、直肠炎、子宫颈炎等)以及癌症前期疾病来说,在器官上游动脉插入导管是较佳的输递方式,因为其避免了肺和肝大量地清除脂质体。对于其它许多部位的损伤(如皮肤癌、人乳头瘤病毒感染、疱疹(嘴或生殖器)和癌症前期颈发育不良),表面输递是有效的且较佳的,因为它很方便。
白血病和其它疾病如疟疾,也很容易通过核酶来自体内给药来治疗。
脂质体可以表面给药、非肠道给药(如静脉给药)、肌肉间注射、腹膜内注射、经皮肤给药、外体(excorporeally)给药等,但是Ⅳ或表皮给药是特别佳的。所需脂质体的确切量根据患者种类、年龄、体重和大致疾病状况,待治疗疾病的严重程度,所用的特殊的化合物,其给药方式等而不同。因此,不可能确定确切的量。然而,合适的量可由该领域普通技术人通过这里提及的常规实验法来测得。通常,剂量与实施例中给出的典型例子相近。
非肠道给药,如果用的话,通常是指注射。注射制品可制成常规形式,如液体溶液或悬浮液、适用于在注射前溶解或悬浮在液体中的固体形态或乳剂。一种最近改进的非肠道给药方法是用缓慢释放或持续释放的系统,使得维持恒定水平的剂量。见如美国专利No.3,710,795,其结合人本文作参考。
表面给药可以通过乳膏、凝胶、栓剂等。来自体内(外体)输递可以典型地用于其它上下文中。
转基因动物
本发明提供了一种转基因非人类动物,其胚芽或体细胞中含有核酸,核酸包括一个靶特异性启动子结合位点,位点在编码5'端自催化断裂核酶序列、含靶RNA特异性结合位点的催化性核酶和3'端自催化断裂核酶序列的序列上游,其中动物表达的核酸含有5'端自催化断裂核酶序列、含靶RNA特异性结合位点的催化性核酶和3'端自催化断裂核酶序列。
核酸可以是图1和SEQ ID NO:1中所示的核酸。或者,可在启动子结合位点和核酶编码序列中进行沉默碱基取代,以在相同组织中表达相同的核酶。例如这些取代可如上所述那样。
本发明的转基因非人类动物是有用的,因为动物不表达与靶RNA相关的表型(如与其编码的蛋白质相关)。这里所用的术语“表型”包括受失效影响的形态学、生物化学曲线(如RNA或表达的蛋白质的变化量等)和其它参数。例如,健康细胞的死亡可以是由于核酶表达而改变表型的衡量标准。
转化的宿主细胞
本发明的核酶可在转化的细胞系中表达。转化细胞可用来使核酶表达特异性和对靶RNA断裂活性的特异性有效。有如此筛选功能的例子在实施例中有所描述。
筛选方法
本发明的转基因动物和转化的宿主细胞可用于筛选可使动物或宿主细胞表达与靶RNA有关的表型的化合物的方法。方法需要对动物/细胞用化合物给药,并评价化合物引起表型表达的能力。如果表型恢复,则认为化合物是有效的。例如可制备L-多巴功能失效(L-dopa functional knockout)转基因动物并用来筛选使L-多巴相关表型恢复的药物。
治疗增生型疾病
本发明提供了一种治疗患有特异组织增生疾病的患者的方法。治疗通过抑制特异组织中的细胞增生来进行治疗,且这是通过对患者用核酸给药来实现的,核酸可编码寻靶细胞存活或复制所必需的RNA的核酶,并含有对病患组织有特异性的靶特异性启动子结合序列。核酸编码的核酶在病患组织中表达,组织中必需RNA的生成被抑制,组织中的细胞增生被抑制,随即引起细胞死亡而治疗了增生疾病。
用本发明的方法来治疗的增生疾病包括靶特异性启动子来治疗的几乎所有的癌症,如前列腺、乳房、结肠、胰、肺和肝中的癌症。
例如,本发明提供了一种治疗患有前列腺癌症的患者的方法,方法包括对患者用SEQ ID NO:1中所示的核酸来给药,从而使核酸编码的核酶在前列腺中表达来治疗前列腺癌症。
治疗病毒感染
本发明提供了一种治疗患者病毒感染的方法,方法包括用本发明核酸对患者给药,其中编码的靶RNA特异性结合位点对感染因素(infectious agent)单一RNA有特异性,从而使核酸编码的核酶表达并杀死感染因素。转录可用病毒特异性启动子或组织特异性启动子来驱动,启动子可在感染病毒的组织中选择性地定向表达核酶,即用肝特异性自蛋白启动子来定向表达抗肝炎B型病毒的核酶。
在测定抗病毒效应中,表达核酶的细胞系可与可支持病毒感染/复制/生成的负对照核酶比较。以上述相同方法可获得表达核酶的细胞系并对其测定;而且在所有情况下可测得核酶防止感染的能力。
本发明将在下列非限制性的实施例中进行进一步详细地描述。
实施例
核酶基因治疗前列腺癌症的测定
本实施例指出了用于采用锤头状核酶的前列腺定向表达来杀死普通的和肿瘤型前列腺上皮的载体和新的方案。核酶是非常新颖的三连体核酶,其可通过攻击必需RNA来定向破坏细胞。核酶的5'和3'端被设计成在转录时可自催化断裂,使内部核酶(高水平地)游离存在于细胞中。
锤头状核酶的细胞内表达并定向寻靶细胞的必需RNA(如RNA聚合酶Ⅰ(A))致使细胞死亡。如果核酶是以限制性方式寻靶特异性组织,则只有该组织中的细胞受核酶表达的作用。选择性地寻靶前列腺可通过鼠前列腺上皮特异性启动子(pb)(或前列腺特异性抗原启动子)来实现。由于用前列腺上皮特异性启动子可寻靶组织特异性前列腺,因此载体全身输递或直接将载体引到前列腺上可使前列腺癌靶死亡,而在剩余的正常前列腺上皮上也观察到有一些伴随损伤。由于前列腺上皮特异性启动子的单一特异性,预计在体内其它部位不会有伴随性损伤。这预计也适于前列腺特异性抗原。
靶的例子包括RNA聚合酶Ⅰ和Ⅱ的Ⅰ(A)亚单位。其它内部寻靶的核酶可用所述方法来进行体外和体内活性测试。
合成
构建图1所示的初级双连体核酶载体。两个侧翼核酶(碱基1至54和66至120)可以自身断裂。将第三个寻靶polⅠmRNA的核酶(图2)(碱基64-105)克隆入侧翼核酶之间。内部核酶有两个区域内的19个碱基(TTCAAAGA-催化核心-ACGAGGATCAG),它们与polⅠ信息反义并通过最小二级结构区域内碱基配对相互作用来实现断裂。
内部polⅠ核酶通过寡核苷酸的合成来制得,寡核苷酸为5'GGA AGA TCTTTC AAA GAC TGA TGA CTC CGT GAG GAC GAA ACG AGG ATC AGA TCTTCC3'(只有核酶的有义链显示在其反向补体中)。下划线是克隆所用的BglⅡ位点。在用BglⅡ进行合适地限制性消化后,将双链寡核苷酸克隆人本发明载体的BglⅡ位点中(图1)。
输递
当前列腺特异性启动子与三连体核酶构建物偶联时,它将通过脂质体全身输递至前列腺。各种引入血管系统的途径(一些通过肺和肝)如所述的进行评价。也可用常位途径(orthotopic route)。预计脂质体赋形剂是有效的,由于高度血管化,其足以将分子输递至前列腺癌细胞上。因此,通过这种基因治疗方法来治愈或至少减少肿瘤负担的结果是合理的。
在这些研究中用了下列脂质体制剂:(a)脂质转染胺试剂(GIBCO BRL,Gaithersburg,MD)是由正电荷液体DOSPA和中性液体DOPE以3∶1摩尔比组成的聚阳离子液体;(b)阳离子液体DDAB与DOPE以2∶1或0.6∶1.0的比例组合(Brunette等人,Mol.Cell.Biol.14∶2411-2418,1994);和(c)DMRIE与DOPE以1∶1摩尔比组合(Felgner等人,Methods(Orlando)5∶67-75,1995)(从VICAL Corp.(SanDiego,CA)获得)。脂质体试剂在转染前保藏在4℃下。
测试
一旦测序后就测试这些核酶的功能。测试机理包括用两种可能的细菌启动子中的一种来转录核酶,在这一例中是pCRⅡ载体中的SP-6或T-7(Invitrogen,SanDiego,Ca)(在痕量放射性存在下),然后通过电泳来评价核酶的自催化断裂。这是用polⅠ核酶来进行的,并发现有断裂,即首先产生包括内部寻靶核酶和3'端核酶的113bp片段,然后出现含有内部polⅠ核酶的74bp的片段。
随后核酶通过将其瞬时转染人C3H10T 1/2小鼠成纤维细胞来测试。其证实了当诱导三连体核酶表达时polⅠRNA被分解。因此测得并证实了分子的两阶段活性的产生。顺式核酶的自催化断裂和反式polⅠmRNA的功能性降解如预计地那样发生。
随后是将三连体核酶在组织特异性启动子(如前列腺上皮特异性或前列腺特异性抗原的启动子,其以组织特异性的方式使表达定向)控制下引入载体。这是通过用含有整个3'、5'端和内部核酶的Not1片段(图2)并将其插入含有前列腺上皮特异性启动子区的载体(-426至+28(Greenburg等人,1994))来实现的。该启动子被证明可使基因表达定向到前列腺上。
在表达三连体核酶抗polⅠ构建物的转基因小鼠体内测试载体。转基因小鼠用Hogan描述的标准前核注射方法(鼠胚胎操作实验手册“Manipulating themouse embryo:a laboratory manual”,Cold Spring Harbor,NY 1986)来获得。在注射前,用限制性酶(HindⅢ和SacⅡ)消化质粒,然后用蔗糖梯度分级来将构建物从载体DNA上分离下来。在注射前,分离获得的构建物用10mm Tris pH 8.0,0.1mm EDTA来透析。在这些小鼠中,一旦前列腺上皮特异性启动子表达变得明显,则在产生后的3-5周阶段就会发生前列腺上皮的破坏。在26只输递的小鼠中,发生转基因的小鼠的比例为10-15%,即有2-3只转基因小鼠。另一种确定本发明的核酶功能的方法是将载体引入组织培养物如人PC3前列腺癌细胞中,并观察响应核酶激活后的细胞死亡情况。已经确认C3H10T 1/2小鼠成核细胞对polⅠ三连体核酶敏感。
通过体内研究可进一步证实该基因治疗方法的有效性。下面进行两组研究。在第一个例子中,采用比polⅠ核酶更容易监测的对照载体,其中对照载体包括驱动藻类绿色荧光蛋白质表达的前列腺上皮特异性启动子,以确认脂质体的体内输递系统的有效性。步骤包括使小鼠麻醉并用外科手术使降主动脉暴露。将30导管径计置于降主动脉中,然后引入300μg载体/千克体重(载体∶脂质体之比为10μg/40μl)。结果表明,绿色荧光蛋白质在背外侧前列腺上皮中有表达而在肺(脂质体正常的靶)中没有。进一步的研究要确定在肺和其它组织如肾、肾上腺和脑中是否发现有表达,第二个实验也在用上述方法在体内有polⅠ核酶加入的小鼠中进行。在两个例子中,无论是加入三连体核酶还是绿色荧光蛋白,动物均在操作后不同时间处杀死、解剖,并用免疫组织化学法测定组织的载体活性。数据表明,在给药7天后前列腺上皮中有程序性细胞死亡(apoptotic cell death)。
在产生前列腺肿瘤的转基因小鼠中进行体内研究。肿瘤通过前列腺上皮特异性启动子控制的基因如EcoRl(一种限制性酶)、cfos(一种原癌基因)或lamin(一种核基质分子)改进型的表达来诱导产生。核酶如上所述进行给药。
靶选择
各种分子被选择用来寻靶。第一种是RNA聚合酶Ⅰ(A)。它是很好的靶,因为其是一种高丰度RNA。如果在前列腺细胞除外的细胞内有前列腺上皮特异性启动子渗漏(低水平转录),预计它们可使限制水平的polⅠ核酶存活。测定其它潜在的靶,它们包括果糖磷酸激酶(根据包括的组织,核酶靶部位为核苷酸178、121或162)、RNA polⅡ亚单位14.4kd(核酶靶部位为核苷酸83或884)、mRNApolⅡ140kd亚单位(核酶靶部位为核苷酸204)和RNA polⅡ23kd亚单位(核酶靶部位为核苷酸143)。
其它潜在的感兴趣靶是复制蛋白A的70kD亚单位。它是DNA复制中点/焦点(centers/foci)形成所需的,因此它应破坏DNA的复制而没有引入错误的危险。序列参考Kim等人的分子细胞生物学Mol.Cell.Biol.12:3050-3059,1992。
寄生虫感染的核酶基因治疗法
上述方法适用于本文,除非另有特指。例如,对于寄生虫感染,其给药方式与前列腺癌症不同,并与组织部位有关。
对于疟疾(恶性疟原虫),对EMP-1蛋白进行寻靶,它是细胞粘连所必需的并且带来迅速改变抗原的问题。特别地,对外显子Ⅱ中的高度保守的GTC进行定向。相关的EMBL登记号为L42246、L42244、L42245、L42247、L40600-L40609、L42636。见Smith等人,Cell 82:101-110,1995,和Su,X等人,细胞学Cell 82:89-100,1995。可用在红细胞中有活性的启动子,治疗也可在体外进行。
细菌感染的核酶基因治疗法
上述方法适用于本文,除非另有特指。例如,对于细菌感染,其给药方式与前列腺癌症不同,并与组织部位有关。
对于结核分支杆菌,对细胞进入所必需的转录片段进行寻靶。EMBL登记号为X70901。见Arruda,S.等人,科学Science 26l:1454-1457,1993。靶在1535bp片段的靠近编码52kD蛋白质的开放读框N端处。
病毒感染的核酶基因治疗法
上述方法适用于本文,除非另有特指。例如,对于病毒感染,其给药方式与前列腺癌症不同。
人乳头瘤病毒型16E6和E7蛋白质从双顺反子mRNA翻译获得。E6的翻译起始位点的反义寡核苷酸抑制了E6和E7的合成。靶可以是GTT,其在核苷酸108处断裂(Tan等人,基因病毒杂志J.Gen.Virol.75:2663-2670,1994)。原始数目来自Seedort等人,病毒学Virology 145:181-185,1985。Ⅳ和表面给药应有效组合。表面给药应对发育不良/癌症前期损伤。
肝炎B型病毒是部分为单链DNA的病毒,现在认为病毒基因组的整合不是关键。而病毒基因组形成延伸的RNA中间体,然后中间体反向转录成DNA。选择有多个有利点的靶。它将在第一位置处切开病毒RNA前基因组,其位于S(包膜)和聚合酶/逆转录酶区。切口在HBV亚型ayw的核苷酸438后GTC处。EMBL登记号是X02496,其原始序列参考Galibert等人,Nature 281,646-650,1979。表达通过白蛋白启动子来驱动,Ⅳ给药是非常有效的。
尽管前述的发明为了清楚和便于理解,在一些细节上进行了描述,但是该领域技术人员应当理解,根据本公开,在不脱离发明和权利要求的范围内可作出各种形式和细节上的变动。

Claims (24)

19.一种转基因非人类动物,其生殖细胞或体细胞中含有权利要求8所述的核酸,其中动物表达的核酶包括5'端自催化断裂核酶序列、含靶RNA特异性结合位点的催化性核酶和3'端自催化断裂核酶序列。
20.根据权利要求17所述的转基因非人类动物,其中动物不表达与靶RNA相关的表型。
21.一种筛选化合物活性的方法,化合物可终止权利要求20所述的动物表达与靶RNA相关的表型,方法包括对动物用化合物给药并评价其使表型表达终止的能力。
22.一种通过抑制组织中细胞增生来治疗患有特异性组织增生疾病的患者的方法,方法包括为患者用权利要求5所述的核酸给药,其中靶特异性启动子结合序列对疾病组织有特异性,从而使核酸编码的核酶表达,组织中核糖体RNA的产生被抑制,细胞增生被抑制,从而来治疗增生疾病。
23.一种治疗患有前列腺癌症的患者的方法,方法包括对患者用权利要求7所述的核酸给药,从而使核酸编码的核酶在前列腺中表达来治疗前列腺癌症。
24.一种治疗患者的感染型疾病的方法,方法包括对患者用权利要求1所述的核酸给药,其中编码的靶RNA特异性结合位点对感染因素单一RNA有特异性,从而使核酸编码的核酶被表达并除去感染因素。
