Description

含1-羟甲基吡唑和一种 防腐剂的协同作用的 抗微生物组合物
本发明涉及某种组合物以及控制一种或多种微生物生长和防止各种产品,材料或介质,尤其是工业产品、材料或介质中的细菌和/或真菌引起的腐败的方法。这些材料或介质包括例如木桨、木屑、木材、粘合剂、覆盖层、包括鸟和爬行类的动物皮毛,造纸厂溶液、纸、光学增亮剂、药物和化学制剂,美容和化妆用品制剂,地质钻探润滑剂和辅助品,农用化学组合物、涂料、皮革、木制品、金属加工流体、工业加工助剂制剂、冷却塔水、废水、巴氏杀菌器、鞣革液、淀粉、染料、石油化学品,表面活性剂,消泡剂、含聚合物的乳剂或水,脂基材料,碳水化合物基材料,蛋白质材料,丙烯酸胶乳涂料乳剂和纺织品。本发明的新的方法和混合物显示了意想不到的抗包括细菌和真菌的微生物的协同作用活性。具体地,本发明涉及含二甲基羟甲基吡唑和至少一种防腐剂的组合物使用。
当许多上述的产品、材料或介质在水中经过处理或置于潮湿的环境中而湿润时,其易于受细菌和/或真菌影响而变质或降解,除非采取步骤控制这种变质或降解。
为控制微生物引起的变质或降解,使用了各种杀微生剂。贸易工作者在不断地寻找改进的杀生物剂,这种杀生物剂应具有低毒性并且以常规量使用时能显示具有延长期的杀生物效果。
可单独使用防腐剂控制微生物,但是其中的许多防腐剂除非使用极高的浓度否则抗细菌和真菌的有效性很低。1-羟甲基吡唑也能以低浓度作为杀生物剂单独使用。
本发明提供了能在延长期阶段控制至少一种微生物,尤其是真菌和细菌生长的杀微生物组合物。另外,本发明提供的组合物使用起来比较经济。对至少一种微生物生长的控制方法也是本发明的目的。
本发明提供的含1-羟甲基吡唑和至少一种防腐剂的组合物,其中各组分是以对控制至少一种微生物的生长起到协同地有效的组合量存在。
本发明还提供了控制易受微生物攻击的材料内部或表面上至少一种微生物生长的方法。该方法包括的步骤是将本发明组合物以可协同有效的量加入到材料或介质以控制至少一种微生物的生长。
本发明还包括分别将1-羟甲基吡唑和至少一种防腐剂添加到上述产品、材料或介质中。根据这个方案,各组分是被分别添加到该系统中的,结果是使用时系统中1-羟甲基吡唑和至少一种防腐剂的最终量是对控制至少一种微生物的生长协同有效量。

当两种化学杀微生物剂结合在一种产品中或者被分别加入时,可能有三种结果:
1)得到的产品将产生相加(中性)效果。
2)产品中的该产品将产生拮抗效果,或者
3)产品中的该化学品将产生协同效应。
与各单独组分相比,相加效果不具有经济上的优点。拮抗效果将产生负面影响。而仅仅是可能性小于相加和拮抗效果的协同作用效果将会产生正面效果并因此具有经济上的优点。
含1-羟甲基吡唑和至少一种防腐剂的杀微生物组合物相比于各单独组分显示具有一种意想不到的协同效应。因此,这些组合物达到了低浓度下抗多种微生物的杰出的(即高于相加的)杀微生物活性。微生物的实例包括,但不限于,真菌和细菌如黑色曲霉(ATCC 9642)和铜绿假单胞菌(ATCC15442)。优选地,本发明的组合物具有低毒性。

1-羟甲基吡唑的制备描述在美国专利4,801,361和5,335,432以及这些专利的参考文献中,所有这些均被作为参考引入本文。市售的3,5-二甲基-1-羟甲基吡唑是在商品名Busan1504或Busan1104(BuckmanLaboratories(Buckman国际实验室)International,Memphis,Tennessee)下,并且该产品容易从市售的原材料合成。

可基于与材料或介质的相容性来选择防腐剂。相容性的确定可通过如在流体系统中的溶解度和与所讨论的流体材料或介质没有反应性作为判据。考虑到本发明,该相容性是容易确定的,其可通过本领域内的普通技术人员添加(例如施用于其表面,掺入到液体、材料或介质和类似物中)该防腐剂到所用的材料或介质中即可。当用于一种流体系统中时,优选地,该防腐剂可大量地溶于该特定的流体中,得到均匀的溶液或分散体。

在下述优选实施方案的讨论中,组分(a)是3,5-二甲基1-羟甲基吡唑而组分(b)是一种防腐剂。
如上所述,组分(a)和组分(b)的使用量是协同作用有效量。(a)和(b)的重量比依据微生物的类型、该组合物施用的材料或介质而定。通常,优选地,组分(a)与组分(b)的比在0.1到99的范围内,更优选从1∶30到30∶1,最优选从1∶5到5∶1。

根据具体的应用,可通过将该组合物溶于有机溶剂中而制备液体形式的该组合物。乳剂形式防腐剂的制备是通过将其在水中乳化,如果需要,再加入一种表面活性剂。附加的化学品如防腐蚀剂、执行剂(performing agents)和杀虫剂也可加入到前述制剂中。
采用以下举例说明的标准的实验室技术进一步证实了上述并用的协同作用活性。下述实施例的目的是举例说明,而不是限制本发明。微生物测定A.真菌的测定
所用的是矿物盐-葡萄糖培养基。为制备该培养基,将下列组分加入到1升去离子水中:0.7g KH2PO4,0.7gMgSO4·7H2O,1.0g NH4NO3,0.005GNaCl,0.002g FeSO4·7H2O,0.002g ZnSO4·7H2O,0.001g MnSO4·7H2O,10g葡萄糖。用1N的NaOH调节该培养基的pH值到6。将该培养基以5ml的量分入测试管中,在121℃高压灭菌20分钟。真菌黑色曲霉在马铃薯葡萄糖琼脂斜面培养基上生长7到10天,并且通过将孢子从斜面洗入到无菌盐溶液中而制备出孢子的悬浮液。在以合适的浓度向该无菌矿物盐-葡萄糖培养基中添加该杀生物剂后,加入该真菌孢子悬浮液(0.1ml种菌),最终的孢子浓度为大约106cfu/mL。将已接种的培养基在28℃培养10-14天。B.细菌的测定
所用的是营养性肉汤生长培养基,为制备这种培养基,将下列组分加入到1升去离子水中:8.0g NaCl,1.0g葡萄糖和1.0g胰蛋白栋。用1N的HCl调节该培养基的pH值到6。将它以5ml的量分配入测试管中,在121℃高压灭菌20分钟。在以所需的浓度向营养性液体培养基中加入该杀生物剂后,加入大约为106cfc/mL的铜绿假单胞菌的悬浮液100毫升,在37℃培养5-7天。
在实施例中,使用上述方法通过在一系列实验中分别测定不同比例的和不同浓度的指定为组分A的3,5-二甲基-1-羟甲基吡唑和指定为组分B的对应的防腐剂或其盐的抗真菌黑色曲霉和抗细菌铜绿假单胞菌从而验证了协同作用效果。
采用完全防止真菌生长两星期(10-14天)和细菌生长一星期(5-7天)的每种混合物或化合物的最低浓度作为端点用于协同作用计算。然后将各种混合物的端点与在同时制备的烧瓶或测试管中单独的纯活性成分的端点比较。
通过Kull E.CEisman P.CSylwestrwicz H.D.和Mayer R.L.1961在《应用微生物学》9:538-541中所述的方法验证了协同作用,其中

用来验证本发明组合物的协同作用的方法是被广泛使用的并且为可接受的方法。在Kull等人的文章上提供了更详细的信息。涉及这个方法的进一步的信息包括在美国专利号3,231,509中,其被引入本文作为参考。
但是,通常当采用浓度为大约0.1到10,000ppm,优选大约0.1到5,000ppm,最优选大约1到2,000ppm的1-羟甲基吡唑,以及浓度为大约0.1到10,000ppm,优选大约0.1到5,000ppm,最优选大约1到3,000ppm的的防腐剂协同并用时,能得到有效的杀细菌和杀真菌的反应。

Claims (16)

1.一种组合物,其包括
(a)一种具有式(Ⅰ)的化合物,

其中R和R'是分别选自氢和有1-4个碳原子的烷基,R″是一个选自(a)氢,(b)一个选自氟、氯、溴和碘的卤素原子,和(c)硝基的部分,或者为所述化合物的盐酸盐;以及
(b)至少一种选自六氢化-1,3,5-三(2-羟乙基)-硫代-三嗪,双1,4-(溴乙酰氧基)-2-丁烯;2-溴-2-硝基丙烷-1,3-二醇;二甲基乙内酰脲;咪唑烷基脲;二唑烷基脲;5-氯-2-甲基-4-异-噻唑啉-3-酮和2-甲基4-异-噻唑啉-3-酮的混和物;1-(3-氯烯丙基)-3,5,7-三氮-1-氮鎓金刚烷氯化物;或者C1-C6烷基羟基苯甲酸酯的防腐剂,其中所述的组分(a)和(b)的存在量为对控制至少一种微生物的生长起到协同有效的并用量。
2.权利要求1的组合物,其中所述的式Ⅰ化合物是3,5-二甲基-1-羟甲基吡唑。
3.权利要求1的组合物,其中所述的微生物是一种细菌或真菌。
4.权利要求3的组合物,其中所述的细菌或真菌是黑色曲霉或铜绿假单胞菌。
5.权利要求1的组合物,其中(a)与(b)的重量比是大约0.1∶99到99∶0.1。
6.权利要求5的组合物,其中(a)与(b)的重量比是大约1∶30到30∶1。
7.权利要求6的组合物,其中(a)与(b)的重量比是大约1∶5到5∶1。
8.控制易受微生物攻击的材料或介质内部或表面上的至少一种微生物生长的方法,包括向材料或介质中加入包含下列组分的组合物的步骤
(a)一种具有式(Ⅰ)的化合物其中R和R'分别选自氢和有1至4个碳原子的烷基,R″是一个选自(a)氢,(b)一个选自F、Cl、Br和碘的卤素原子,和(c)硝基的部分,或者为所述化合物的盐酸盐;和
(b)至少一种选自六氢化-1,3,5-三(2-羟乙基)-硫代-三嗪,双1,4-(溴乙酰氧基)-2-丁烯;2-溴2-硝基丙烷-1,3-二醇;二甲基乙内酰脲;咪唑烷基脲;二唑烷基脲;5-氯-2-甲基-4-异-噻唑啉-3-酮和2-甲基4-异-噻唑啉-3-酮的混合物;1-(3-氯烯丙基)-3,5,7-三氮-1-氮鎓金刚烷氯化物;和C1-C6烷基羟基苯甲酸酯的防腐剂,它们以对控制所述的生长协同有效的并用量存在。
9.权利要求8的组合物,其中所述的式Ⅰ化合物是3,5-二甲基-1-羟甲基吡唑。
10.权利要求8的方法,其中的微生物是细菌或真菌,或者为它们的混合物。
11.权利要求10的组合物,其中所述的细菌或真菌是黑色曲霉或铜绿假单胞菌。
12.权利要求9的方法,其中所述的材料或介质是木桨、木屑、木制品、木材、涂料、染料、皮革、粘合剂、覆盖层、动物皮毛、鞣革液、造纸厂溶液、金属加工液体、淀粉、石油化学品、丙烯酸胶乳涂料乳剂、药物制剂、冷却塔水、美容和化妆用品制剂、纺织品、工厂进出水、废水、地质钻探润滑剂或农用化学品组合物。
13.权利要求9的方法,其中该材料或介质是固体、分散体、乳剂或溶液剂的形式。
14.防止细菌和/或真菌引起的材料或介质的腐败的方法,包括将含下列成分的组合物加入到一种材料或介质中
(a)一种具有式(Ⅰ)的化合物
