Abstract

一种鹿茸药用成分的水解提取方法,取新鲜的鹿茸加入缓冲液,用组织绞碎机绞碎,调pH值为1.0-3.0,将溶液的温度调至20℃-30℃,加入复合的生物催化剂进行水解,然后离心或硅藻土过滤,得到澄清金黄色的鹿茸水解液。鲜鹿茸经过水解方法处理后,其平均水解率达70%左右(水解率根据鲜鹿茸的质量不同在60-75%之间);水解后的残余物为鹿茸钙化的部分及部分皮毛,所有的有机质基本都变成溶液状态,变成了易于人体吸收的溶液状态。得到的产品可应用于制药、保健品、食品的诸多方面,应用极其方便,同时利用本技术对鹿茸产品进行开发可衍生很多新的开发途径,得到系列新产品。

技术领域:
本发明涉及中药有效部位成分的提取方法,尤其是提供一种鹿茸药用成分的水解提取方法及产品,属于中药有效成分提取技术领域。
背景技术:
虽然鹿茸有较好的医用价值和较强的保健作用,但由于鹿茸的组织结构独特一通过鹿茸组织结构研究表明,鹿茸(除茸皮外)从顶部到基部是由发育程度不同的骨组织构成;因此鹿茸中含有大量构成骨及前骨的材料—硫酸软骨素和胶原纤维,因此对鹿茸并没有好的利用方法;在传统的中医医疗中,鹿茸作为中药配伍的一部分,只能进行水煎,在利用过程中利用率只有2%左右,绝大部分都废弃;而作为传统的补品,一般只能用于泡酒等,利用率则更低。因此,传统鹿茸的利用方式,对鹿茸资源是很大的浪费。
发明内容:
本发明提供一种鹿茸药用成分的水解提取方法及产品,充分利用宝贵的鹿茸资源,更好的发挥鹿茸的药用价值。
本发明的技术解决方案包括以下步骤:
取新鲜的鹿茸加入缓冲液,用组织绞碎机绞碎,调溶液的pH值为1.0-10.0,按鹿茸重量的0.005~0.01加入生物催化剂,包括:胃蛋白酶、HAP碱性蛋白酶、木瓜蛋白酶、胰酶、碱性内切蛋白酶(Alcalase)、风味酶(Flavourzyme)六种酶中的一种或多种,同时加入Ca2+、Mg2+、Zn2+、Mg2+等氯化物酶激活剂,在温度20℃-60℃,pH1.0-pH10.0条件下进行水解反应2小时-5小时,然后离心或硅藻土过滤,得到澄清金黄色的鹿茸水解液,水解率在60%-70%。
缓冲液:磷酸盐、柠檬酸盐、乳酸盐缓冲液。
生物催化剂包括:胃蛋白酶、HAP碱性蛋白酶、木瓜蛋白酶、胰酶、碱性内切蛋白酶(Alcalase)、风味酶(Flavourzyme)。
激活剂:Ca2+、Mg2+、Zn2+、Mg2+等氯化物。
水解液中含有多种有效物质:经过分析,鹿茸水解后的溶液中含有大量的小分子多肽、脂类(主要有磷脂、甾体化合物、脂肪酸、中性脂和前列腺素等)、游离氨基酸、微量元素、糖类等多种有效成分。
本发明的积极效果在于:鲜鹿茸经过水解方法处理后,其平均水解率达70%左右(水解率根据鲜鹿茸的质量不同在60-75%之间);水解后的残余物为鹿茸钙化的部分及部分皮毛,所有的有机质基本都变成溶液状态,鹿茸中几乎所有的有效成分都变成了易于人体吸收的溶液状态。得到的产品可应用于制药、保健品、食品的诸多方面,应用极其方便,同时利用本技术对鹿茸产品进行开发可衍生很多新的开发途径,得到系列新产品。
具体实施方式:
实施例1
取一定量新鲜的鹿茸1kg,加入3000ml磷酸盐的缓冲液,用组织绞碎机绞碎,加入生物催化剂:胃蛋白酶10g、激活剂氯化锌0.1g,调pH值为1.5,将溶液的温度调至25℃,放入反应罐反应4小时,离心去残渣,收集透明澄清的液体3200ml。
实施例2:
取一定量新鲜的鹿茸1kg,加入3000ml柠檬酸盐的缓冲液,用组织绞碎机绞碎,加入生物催化剂:木瓜蛋白酶8g、激活剂氯化钙0.08g,调pH值为7.0,将溶液的温度调至37℃,放入反应罐反应5小时,离心去残渣,收集透明澄清的液体3300ml。
实施例3
取一定量新鲜的鹿茸1kg,加入3000ml乳酸盐的缓冲液,用组织绞碎机绞碎,加入生物催化剂:胰酶10g、激活剂氯化锰0.15g,调pH值为9.0,将溶液的温度调至52℃,放入反应罐反应3小时,离心去残渣,收集透明澄清的液体3340ml。

Claims (1)

1、一种鹿茸药用成分的水解提取方法,包括以下步骤完成的:取新鲜的鹿茸加入缓冲液,用组织绞碎机绞碎,按比例为鹿茸重量的0.005-0.01加入生物催化剂,再加入氯化物作为激活剂,在温度20℃-60℃,pH1.0-pH10.0的条件下进行水解反应2小时-5小时,然后离心或硅藻土过滤;
所述的缓冲液选自磷酸盐缓冲液、柠檬酸盐缓冲液、乳酸盐缓冲液中的一种或多种;
所述的生物催化剂选自胃蛋白酶、HAP碱性蛋白酶、木瓜蛋白酶、胰酶、碱性内切蛋白酶、风味酶中的一种或多种;
所述的激活剂选自Ca2+、Mg2+、Zn2+、Mn2+的氯化物。
