Abstract

技术领域
本发明涉及含有喹啉并异喹啉衍生物或其药学上可接受的酸加成盐作为有效成分的学习或记忆改善剂。
背景技术
学习是指学到通过经验或练习产生的较永久的行为变化,或其过程,记忆是指将通过经验得到的情报保持一定的时间,根据需要再现该情报。但是,该两概念的定义并不单纯,与其说彼此独立,不如说大部分是重复的。因此,在行为药理学方法中,很多情况是学习能力和记忆能力通过同样的试验测定。

作为R1,优选氢、碳原子数1~5的烷基,碳原子数4~7的环烷基烷基,碳原子5~7的环烯基烷基,碳原子数7~14的芳烷基,碳原子数4~5的反式烯基,烯丙基,呋喃基-2-基烷基,噻吩基-2-基烷基,碳原子数1~5的烷酰基,苯甲酰基,乙烯氧羰基,三氯乙氧羰基,苄氧羰基或碳原子数8~14的芳基烷酰基,特别优选氢,甲基,乙基,环丙基甲基,烯丙基,苯乙基,呋南-2-基乙基,噻吩-2-基乙基。
作为R2,优选氢或OR6(这里,R6表示氢,碳原子数1~5的烷基,或碳原子数1~5的烷酰基),特别优选氢,羟基,甲氧基,乙氧基。

本发明的通式(I)化合物,例如可按照特开平4-275288或国际公开专利9902157号中记载的方法,以酮(III)为原料,在酸催化剂共存下,与溶剂中的邻氨基苯甲醛衍生物(IVa)或邻氨基苯乙酮衍生物(IVb)或邻氨基苄腈(IVc)缩合得到,如果原料使用光学活性体,可得到光学活性的化合物(流程1)。
通式(I)所示的异喹啉衍生物在记忆障碍的治疗或预防中是否有效,如实施例所示,可通过行为药理学方法在动物中确认。更具体地说,被动的逃避反应试验,主动的条件逃避反应试验,或使用水迷宫或放射迷路学习的空间记忆试验法等在抗痴呆药物的开发中常被使用。
本发明化合物可作为伴有学习·记忆障碍的痴呆性疾病的治疗或预防中有用的药物使用。特别是可在伴有脑血管障碍,神经变性疾病,内分泌疾病,营养和代谢障碍,缺氧性脑病,肿瘤,感染症,金属代谢异常和药物中毒等引起的记忆障碍的痴呆性疾病的治疗或预防中使用。作为显示痴呆症状的具有脑血管障碍的疾病,有脑梗塞,脑出血,多发梗塞性痴呆,云雾病,脑动静脉畸形,全身性红斑狼疮(SLE),颞动脉炎等血管炎等。作为同样显示痴呆症状的神经变形疾病,有阿耳茨海默氏病,皮克氏病,帕金森病,亨廷顿氏舞蹈症,进行性核上麻痹,Shy-Drager综合征,Ramsey-Hunt综合征,家族性大脑基底核钙化症等,作为内分泌疾病,有甲状腺机能减退症,甲状旁腺机能减退症,库兴氏病,阿狄森氏病,反复低血糖发作,作为营养和代谢障碍,有韦尼克氏脑病,糙皮病脑症,维生素B12缺乏症,肝功能衰竭或肾功能衰竭慢性代谢性障碍,低钠血症,作为缺氧性脑病有一氧化碳中毒等,作为肿瘤,有脑肿瘤,癌性髓膜炎等,作为感染症有髓膜炎,脑炎,脑肿瘤,迟发性病毒感染,AIDS,神经梅毒等。另外,作为其他的显示痴呆症状的疾病,有正常压脑积水,慢性硬膜下血肿,脑挫伤,肌强直性营养不良和线粒体肌病。但是,本发明化合物的应用并不仅限于这些疾病例。
本发明的学习·记忆改善剂,可与例如,其他的显示痴呆症状的疾病(例如,脑血管障碍、神经变性疾病,内分泌疾病,营养障碍和代谢障碍,缺氧性脑症,恶性肿瘤,感染症等)的预防·治疗药,或对于痴呆症状中伴随的问题行为(夜间谵妄,夜间不安静,抑郁,睡眠障碍,人格变化)使用的药物组合使用。
作为脑血管障碍的预防·治疗药,可列举例如ATP,细胞色素C,甲氯芬酯盐酸盐,艾地苯醌,丙戊茶碱,γ-氨基丁酸,γ-氨基-β-羟基丁酸,高泛酸钙,茴拉西坦,金刚烷胺盐酸盐,麦角乙脲马来酸盐,二苯美仑盐酸盐,茚氯嗪盐酸盐,二氢麦角毒碱甲磺酸盐,艾芬地尔酒石酸盐,莫西赛利盐酸盐,苄环烷富马酸盐,溴长春胺富马酸盐,依布拉特,长春西汀,尼麦角林,桂哌齐特马来酸盐,己酮可可碱,曲匹地尔,克冠草盐酸盐,氟桂利嗪盐酸盐,桂利嗪,尼卡地平盐酸盐,尼伐地平,血管舒缓素等脑循环代谢改善药,氯噻嗪,依他尼酸,可乐定,利血平,普萘洛尔,哌唑嗪,肼屈嗪,罂粟碱,卡托普利,硝苯地平等降压药,华法令等抗凝血药,尿激酶等血栓溶解药,奥扎格雷,贝前列素等抗血小板药。
作为神经变性疾病的预防·治疗剂,可列举他克林等抗痴呆药,左旋多巴,苯托品,苄甲炔胺,比哌立登,异丙嗪,苯海拉明等帕金森氏病治疗药和前述脑循环代谢改善药等。
对于伴随痴呆症状的问题行为的预防·治疗药,可列举阿米替林,度硫平,洛非帕明,丙咪嗪,氟西汀,氟伏沙明,米安色林,曲唑酮,马普替林,哌异丙肼等抗抑郁药,地西泮,眠尔通等抗焦虑药,卤沙唑仑,三唑仑等催眠药,氯丙嗪,硫利达嗪,氟奋乃静等神经阻断药等。
作为内分泌疾病的预防·治疗药,可列举例如抗甲状腺药(thionamide,甲硫咪唑,碘等)甲状腺激素,抗糖尿病药(胰岛素制剂,甲苯磺丁脲,格列吡嗪,甲福明,阿卡糖等),肾上腺皮质激素(氢化可的松,氢化泼尼松,倍地米松,地塞米松等糖皮质激素,醛甾酮等无机肾上腺皮质激素等),抗肾上腺皮质激素药(美替拉酮等)。
作为对恶性肿瘤的预防·治疗剂,可列举例如,环磷酰胺,苯丁酸氮芥等烷基化试剂,阿霉素等细胞毒性抗生素,长春新碱等长春花属生物碱,甲氨蝶呤,氟尿嘧啶,阿糖孢苷,巯基嘌呤等代谢拮抗药和顺铂等。
本发明的学习·记忆障碍的治疗或预防剂作为药物在临床使用时,药剂可以是游离碱或其盐本身,或与赋形剂,稳定剂,防腐剂,缓冲剂,溶解助剂,乳化剂,稀释剂,等渗剂等添加剂适当混合。作为给药形式,可以是非口服给药或口服给药使用。作为给药剂型,可列举注射剂,片剂,液剂,胶囊剂,颗粒剂,粉剂等,这些剂型可通过公知的制剂技术制备。给药量可根据患者的症状或年龄,体重,给药方法等适当选择,对于成人,有效成分量为1天0.0001mg~10g,优选0.001mg~1g,可分一次或多次给药。

参考例1
(4aR,12aR)-2-甲基-4a-(3-羟基苯基)-1,2,3,4,4a,5,12,12a-八氢-喹啉[2,3-g]异喹啉

本化合物按照国际公开专利9902157号中记载的方法合成。
参考例2
2-甲基-4aα-(3-甲氧苯基)-11-甲基-1,2,3,4,4a,5,12,12aβ-八氢-喹啉并[2,3-g]异喹啉

将2-甲基-4aα-(3-甲氧苯基)-6-氧代-1,2,3,4,4a,5,6,7,8,8aβ-八氢异喹啉150mg(0.55mmol)和邻氨基苯乙酮100mg(0.74mmol)加入乙酸5ml中,加热回流3小时。放冷后,加入饱和碳酸氢钠水溶液,用乙酸乙酯萃取。有机层用饱和食盐水洗净,用无水硫酸镁干燥后浓缩。所得残渣用硅胶柱色谱分离纯化(氯仿∶甲醇∶28%氨水=20∶1∶0.1~10∶1∶0.1),得到211mg标题化合物(收率100%)。

2-甲基-4aα-(3-羟基苯基)-11-甲基-1,2,3,4,4a,5,12,12aβ-八氢-喹啉并[2,3-g]异喹啉3盐酸盐
将参考例2得到的2-甲基-4aα-(3-甲氧苯基)-11-甲基-1,2,3,4,4a,5,12,12aβ-八氢-喹啉并[2,3-g]异喹啉210mg(0.56mmol),正丙烷硫醇0.29ml(3.20mmol)在氩气氛围下溶于DMF溶剂7ml中,加入叔丁醇钾320mg(2.85mmol),在120度加热搅拌20小时。在冰冷却下加入1当量盐酸4ml,调至酸性后,加入饱和碳酸氢钠水溶液再次调成碱性,用氯仿∶甲醇(4∶1)的混合液萃取。有机层用水洗涤,用无水硫酸镁干燥后浓缩。所得残渣用二氯甲烷-甲醇混合液重结晶,得到131mg标题化合物(收率65%)。将其悬浮于甲醇中,加入盐酸盐化,浓缩后,加入醚,过滤固体得到标题化合物的盐酸盐143mg。

将2-甲基-4aα-(3-甲氧苯基)-6-氧代-1,2,3,4,4a,5,6,7,8,8aβ-八氢异喹啉150mg(0.55mmol)和邻氨基苄腈130mg(1.10mmol)加入乙酸5ml中,加热回流44小时。放冷后,加入饱和碳酸氢钠水溶液,用氯仿∶甲醇(4∶1)混合液萃取。有机层用饱和食盐水洗净,用无水硫酸镁干燥后浓缩。所得残渣用胺包覆的硅胶柱色谱分离纯化(氯仿∶甲醇=50∶1),得到83mg标题化合物(收率41%)。

2-甲基-4aα-(3-羟基苯基)-11-氨基-1,2,3,4,4a,5,12,12aβ-八氢-喹啉并[2,3-g]异喹啉

将参考例4得到的2-甲基-4aα-(3-甲氧苯基)-11-氨基-1,2,3,4,4a,5,12,12aβ-八氢-喹啉并[2,3-g]异喹啉83mg(0.22mmol),正丙烷硫醇0.10ml(1.060mmol)在氩气氛围下溶于DMF溶剂7ml中,加入叔丁醇钾106mg(0.95mmol),在120度加热搅拌20小时。在冰冷却下加入1当量盐酸4ml,调至酸性后,加入饱和碳酸氢钠水溶液再次调成碱性,用氯仿∶甲醇(4∶1)的混合液萃取。有机层用水洗涤,用无水硫酸镁干燥后浓缩。所得残渣用二氯甲烷-甲醇混合液重结晶,得到35mg标题化合物(收率44%)。将其悬浮于甲醇中,加入盐酸盐化,浓缩后,加入醚,过滤固体得到标题化合物的盐酸盐40mg。

(+)-7,9-二溴-4a-(3-甲氧苯基)-2-甲基-1,2,3,4,4a,5,12,12a-八氢-喹啉[2,3g]异喹啉

将(+)-4a-(3-甲氧苯基)-2-甲基-6-氧代-1,2,3,4,4a,5,6,7,8,8a-八氢异喹啉382.4mg和2-氨基-3,5-二溴苯甲醛1.95g溶于乙醇中,加入甲磺酸0.454ml,加热回流2小时。放冷后,加入饱和碳酸氢钠水溶液,用乙酸乙酯萃取。有机层用饱和食盐水洗净,用无水硫酸钠干燥。过滤,浓缩后的残渣用硅胶柱色谱纯化(氯仿∶甲醇=40∶1),得到标题化合物683.7mg(收率95%)。

(+)-7,9-二溴-4a-(3-羟基苯基)-2-甲基-1,2,3,4,4a,5,12,12a-八氢-喹啉[2,3g]异喹啉

将实施例6得到的(+)-7,9-二溴-4a-(3-甲氧苯基)-2-甲基-1,2,3,4,4a,5,12,12a-八氢-喹啉[2,3g]异喹啉669.6mg溶于二氯甲烷15ml中,在冰冷却下,滴加入三溴化硼二氯甲烷溶液(1mol/l)6.49ml。在0℃搅拌4小时后,加入饱和碳酸氢钠水溶液,在室温剧烈搅拌30分钟。用氯仿萃取后,有机层用饱和食盐水洗净,用无水硫酸钠干燥。将过滤,浓缩后的残渣用硅胶柱色谱纯化(氯仿∶甲醇=20∶1~5∶1),得到标题化合物381.5mg(收率59%)。

(+)-7,9-二溴-4a-(3-羟基苯基)-2-甲基-1,2,3,4,4a,5,12,12a-八氢-喹啉[2,3g]异喹啉甲磺酸盐

将实施例7得到的(+)-7,9-二溴-4a-(3-羟基苯基)-2-甲基-1,2,3,4,4a,5,12,12a-八氢-喹啉[2,3g]异喹啉381.5mg悬浮于甲醇中,加入甲磺酸1.522ml,搅拌10分钟,浓缩反应液,将残渣用甲醇、乙酸乙酯和乙醚再沉淀,得到标题化合物348.2mg。

(+)-9-氟-4a-(3-甲氧苯基)-2-甲基-1,2,3,4,4a,5,12,12a-八氢-喹啉[2,3g]异喹啉

将(+)-4a-(3-甲氧苯基)-2-甲基-6-氧代-1,2,3,4,4a,5,6,7,8,8a-八氢异喹啉380.3mg和二甲基甲酰胺二甲基乙缩醛0.924ml在甲苯溶剂中加热回流5.5小时。放冷后,浓缩反应液,减压干燥。将残渣溶于8ml,加入4-氟苯胺0.395ml和三氟乙酸0.536ml,加热回流11小时。放冷后,加入饱和碳酸氢钠水溶液,用乙酸乙酯萃取。有机层用饱和食盐水洗净,用无水硫酸钠干燥。将过滤,浓缩得到的残渣用硅胶柱色谱纯化(氯仿∶甲醇=15∶1~10∶1),得到标题化合物249.9mg(收率48%)。

(+)-9-氟-4a-(3-羟基苯基)-2-甲基-1,2,3,4,4a,5,12,12a-八氢-喹啉[2,3g]异喹啉

将参考例9得到的(+)-9-氟-4a-(3-甲氧苯基)-2-甲基-1,2,3,4,4a,5,12,12a-八氢-喹啉[2,3g]异喹啉249.9mg溶于二氯甲烷9ml中,在冰冷却下,滴加入三溴化硼的1当量二氯甲烷溶液3.32ml,搅拌2小时。慢慢加入饱和碳酸氢钠水溶液,剧烈搅拌30分钟。将反应液用氯仿萃取,有机层用饱和食盐水洗净,用无水硫酸钠干燥。过滤,浓缩,将残渣用硅胶柱色谱纯化(氯仿∶甲醇∶异丙胺=97∶2.7∶0.3),进一步用薄层色谱纯化(氯仿∶氨饱和的氯仿=1∶1),得到标题化合物65mg(收率27%)。

使用Morris水迷宫的学习·记忆改善作用的评价
<试验步骤>
在试验中使用6周龄的ICR系雄性小鼠。试验装置使用Morris水迷宫试验装置,将到逃避到池内设置的平台的时间作为指标,评价空间学习·记忆能力。另外,1次试验最多120秒,从池子中不同3点开始游泳的小鼠进行3次试验作为一个训练期间,算出其平均逃避时间,进行评价。第一天(第1训练期间),不使用药物或其它任何物质给药,第2训练期间(第2天)以后,使用化合物

8或溶剂给药,将试验重复5天时间(5训练期间)。

1给药的大鼠的学习课程和与使用生理盐水给药的大鼠的比较结果。在图中,纵轴表示到达平台的潜伏期(和SE),横轴表示训练次数(*表示P<0.05,**表示P<0.01)。通过化合物
1给药,在Morris水迷宫试验中,显示出鼠的空间学习·记忆能力被改善了。

实施例2
跳下型(Step-Down)被动逃避学习试验中学习改善效果
<试验步骤>
在试验中使用6周龄的ICR系雄性小鼠。试验装置使用在25×25×30cm(宽×深×高)塑料制盒的底部上具备为给与电刺激的不锈钢制栅栏,在底部栅栏的一角设置了4.5×4.5×3.0(宽×深×高)的木制平台的装置。
训练试验60分钟前使用化合物
1以100和300μg/kg或作为其溶剂的生理盐水皮下给药。30分钟后,使用引起学习·记忆障碍的化合物,NMDA受体的非竞争拮抗药MK-801以0.1mg/kg腹腔内给药。训练试验的进行是将小鼠放置在平台上,测定小鼠从平台跳下到底部栅栏的时间,即跳下潜伏期。在小鼠的四肢跳下到底部的同时,施加2秒钟6mA的电刺激,使之学习一跳下到底部就受到电刺激。另外,在训练试验进行中,只使用跳下潜伏期显示3~30秒的小鼠进行试验。
在训练进行24小时后进行试验。将小鼠再次放在平台上,测定跳下潜伏期最大为300秒。

1的评价>
其结果在图2显示。只用溶剂给药的对照组进行试验时的跳下潜伏期比训练进行时延长,表明记住了一跳下到底部会受到电刺激。另一方面,用MK-801单独给药组进行试验时的跳下潜伏期比对照组明显缩短,诱发了学习·记忆障碍。该MK-801的跳下潜伏期缩短作用通过化合物
1给药被明显抑制,确认化合物
1对学习·记忆障碍显示改善作用。
另外,图2显示的是关于只用溶剂给药的对照组和,MK-801给药组,和化合物
1给药组的跳下潜伏期(每个柱内的棒是中位值)。MK-801给药组,与对照组相比明显被抑制了被动的逃避反应。该抑制通过化合物
1给药恢复,化合物
1显示对学习·记忆障碍有改善作用。(**为相对于对照组P<0.01,##为相对于MK-801单独给药P<0.01)。
产业上的可利用性
本发明的学习或记忆改善剂,在伴有脑血管障碍、阿耳茨海默氏病等神经变性疾病,内分泌疾病,营养·代谢障碍,肿瘤,感染症,药物中毒等引起的记忆障碍的痴呆症的治疗中有用。

1.通式(I)所示的异喹啉衍生物或其药学上可接受的盐在制备学习或记忆改善剂中的应用,
其中,R1为甲基,环丙基甲基或烯丙基;R2是氢或羟基;R3,R3’独立地是甲基,氢,氯,氟,溴或氨基;R4是羟基或甲氧基;X是碳;并且R5是甲基,氢,氯,氟,溴或氨基。
