Abstract

本发明公开了一种治疗尖锐湿疣的药物组合物，其特征在于该药物组合物是由下列重量份的原料药制成的：苦参50～150份、水杨酸50～150份、鬼臼毒素2.5～7.5份、冰片1～5份，本发明还公开了其制备方法。

Description

一种治疗尖锐湿疣的药物组合物及其制备方法
本发明涉及一种治疗尖锐湿疣的药物组合物及其制备方法。
尖锐湿疣是当前常见的性传播疾病之一，中医称之为“臊瘊”、“尿瘊”。中医认为尖锐湿疣是由于肝肾亏损，交合不洁，外感湿热淫毒，湿毒下注所致，其病机要点为湿毒瘀结。治疗以清热解毒、祛湿散结、腐蚀疣赘为主。生殖器疱疹是由单纯疱疹病毒HSV引起的一种性传播疾病，属于中医“热疮”、“疳疮”的范畴。中医认为是本病乃交媾不洁，感染秽浊湿热毒邪，湿热下注，蕴结肌肤而成。治疗以清热利湿、凉血解毒为主。
本发明目的在于提供一种疗效显著的具有清热解毒，化瘀散结等功效的外用药物及其制备方法。
本发明技术方案是这样实现的：
如下所述重量份的原料药：
苦参50～150份        水杨酸50～150份
鬼臼毒素2.5～7.5份   冰片1～5份
苦参用95％乙醇回流提取2次，第一次2小时，第二次1.5小时，合并提取液，滤过，浓缩。另取冰片、水杨酸、鬼臼毒素分别用适量乙醇溶解，加入至提取浓缩液中，再加入250～750ml聚乙二醇400，混匀，调整至全量，分装，即得。
经过实验观察，结果表明本发明药物组合物具抗菌作用，对各菌都具有抑制作用；较低剂量就可起到抗菌作用；对各菌种具有不同程度的最低抑制浓度；对金黄色葡萄球菌有一定的杀菌作用。具有杀滴虫作用，其抗滴虫作用与其剂量呈正相关。高、低剂量均有明显抑制大鼠足跖肿胀的作用。高低剂量对二甲苯所致的小鼠耳廓肿胀均有明显的抑制作用。对HSV-2有灭活作用，灭活指数达166.0以上；可降低实验感染HSV-2的小鼠的致死率。
本发明药物组合物具有热解毒，化瘀散结等功效，可用于尖锐湿疣，生殖器疱疹等病症的治疗。
实验例1：抗菌试验：
1)无菌试验：取本发明药物组合物用生理盐水稀释并加入细菌和霉菌营养琼脂中使药物终浓度成1∶10，1∶100，1∶1000，倾注平板，每个浓度做三组。置37℃温箱培养7天，观察有无菌生长和计菌落数。
结果：各稀释度无菌生长，未检出细菌。
2)抑菌试验：取各菌种分别涂布细菌、霉菌或淋球菌琼脂平板，用孔径5mm打孔器打孔，取本发明药物组合物加满各孔内，每菌分做三组。置37℃培养24小时后测抑圈直径。
结果：本发明药物组合物对各菌都具有抑制作用。
3)最低抑菌浓度(MIC)试验：营养琼脂稀释法，用生理盐水稀释本发明药物组合物，并加入营养琼脂使药物终浓度成1/10、1/20、1/30、1/40、1/50，混匀后倾注平板。用生理盐水稀释各菌成3亿/ml，各取一接种环种于各浓度药平板上，每个菌做三组。37℃培养24小时观察结果。
结果：以三组试验均完全无菌生长的药物最高稀释度为其最低抑菌浓度(MIC)。本发明药物组合物对各菌种具有不同程度的最低抑制浓度。
4、最低杀菌浓度(MBC)试验，割取MIC试验中无菌生长的接种细菌处的琼脂种于营养琼脂平板，37℃培养24小时观察结果。以完全无菌生长的药物最高稀释度为其最低杀菌浓度(MBC)。
结果：本发明药物组合物对金黄色葡萄球菌有一定的杀菌作用。
实验例2：抗滴虫试验
用培养基稀释本发明药物组合物成90％、50％、25％、10％、5％浓度。取滴虫加入各浓度本发明药物组合物培养基中，于不同时间观察结果。以药物在最短时间内完全杀死滴虫的药物最高稀释度判定其杀滴虫效果。
结果：本发明药物组合物具有杀滴虫作用，其抗滴虫作用与其剂量呈正相关。
实施例1：
苦  参100g    水杨酸100g
鬼臼毒素5g    冰  片2g
苦参用95％乙醇回流提取2次，第一次2小时，第二次1.5小时，合并提取液，滤过，浓缩。另取冰片、水杨酸、鬼臼毒素分别用适量乙醇溶解，加入至提取浓缩液中，再加入500ml聚乙二醇400，混匀，调整至1000ml，分装，即得。
实施例2：
苦  参120g    水杨酸80g
鬼臼毒素5g    冰  片3g
苦参用95％乙醇回流提取2次，第一次2小时，第二次1.5小时，合并提取液，滤过，浓缩。另取冰片、水杨酸、鬼臼毒素分别用适量乙醇溶解，加入至提取浓缩液中，再加入500ml聚乙二醇400，混匀，调整至1000ml，分装，即得。

1、一种治疗尖锐湿疣的药物组合物，其特征在于该药物组合物是由下列重量份的原料药制成的：
苦参50～150份                      水杨酸50～150份
鬼臼毒素2.5～7.5份                 冰片1～5份。
2、按照权利要求1所述的治疗尖锐湿疣的药物组合物，其特征在于：该药物组合物是由如下重量份的原料药制成的：
苦参100份                          水杨酸100份
鬼臼毒素5份                        冰片2份。
3、如权利要求1、2所述的治疗尖锐湿疣的药物组合物的制备方法，其特征在于该方法为：苦参用乙醇回流提取2次，第一次2小时，第二次1.5小时，合并提取液，滤过，浓缩；另取冰片、水杨酸、鬼臼毒素分别用适量乙醇溶解，加入至提取浓缩液中，再加入适量辅料，混匀，调整至全量，分装，即得。
