Abstract

本发明提供了一种采用半导体激光辐照果胶酶产生菌—黑曲霉,以选育果胶酶高产菌株的方法。该方法的特点是采用不同参数的半导体激光辐照果胶酶产生菌—黑曲霉孢子悬液。分析所采用的激光参数与黑曲霉致死率,产酶正变率以及酶活之间的关系,定向筛选产酶活高的适合于工业化生产的高产酶活菌株,并确定所采用的最佳激光参数。

Description

半导体激光辐照选育果胶酶高产菌株
本发明涉及采用半导体激光辐照微生物育种,该方法特别适用于采用半导体激光辐照果胶酶产生菌—黑曲霉孢子,以选育果胶酶高产菌株。
果胶酶在果汁、果酒、果蔬加工生产咖啡、可可、烟草及食用菌加工处理中广泛应用。选育高酶活的果胶酶生产菌的方法是果胶酶生产的关键。国内曾有人采用红外线、紫外线等光源辐照发酵期果胶酶产生菌或果胶酶产生菌的白色孢子突变株,取得一定成效。随着激光技术的发展,各种新型激光器件相继出现,特别是半导体激光器具有众多输出谱线,且可以调谐波长。我们首先将其引入激光辐照选育果胶酶高产菌株这一领域。本发明的目的是找到可以选育果胶酶高产菌株的半导体激光器的参数,以及所采用的方法。
为此,本发明是这样实现的:采用不同参数的半导体激光(包括不同波长、剂量、功率密度、偏振方式、激光模式、脉冲或连续激光输出等),辐照果胶酶产生菌—黑曲霉的孢子悬液。分析所采用的激光参数、生物条件与黑曲霉致死率、黑曲霉产酶正变率及酶活之间的关系,从中定向筛选产酶活力高的菌株。再经过初筛、复筛,选育出适合工业化生产的高产酶活菌株,并确定所采用的最佳的半导体激光参数与实验条件。
采用上述方法选育的果胶酶产生菌高酶活菌株,已稳定应用于工业化生产。实现这一方法所采用的半导体激光的一组参数为:波长830nm、照射剂量每平方厘米59焦尔、非偏振、多横模连续输由的半导体激光。

Claims (3)

1.采用半导体激光辐照选育果胶酶高产菌株,本发明的特征在于采用不同参数的半导体激光辐照果胶酶产生菌—黑曲霉孢子悬液,定向筛选产酶活力高的菌株,再经过初筛、复筛,选育出适合工业化生产的高产酶活菌株,并确定选育所采用的最佳半导体激光参数。
2.根据权利要求1规定的采用不同参数的半导体激光,其特征在于所说的不同参数,包括半导体激光的波长、剂量、功率密度、偏振方式、激光模式、脉冲或连续激光输出等。
3.根据权利要求1规定的确定选育所采用的一组最佳半导体激光参数是:波长830nm、照射剂量每平方厘米59焦尔、非偏振、多横模连续输出的半导体激光。
