Abstract
本发明公开了虎纹蛙抗菌肽cathelicidin‑2及其编码序列和应用。本发明通过筛选虎纹蛙肝脏转录组文库,获得了虎纹蛙抗菌肽cathelicidin‑2编码基因。采用固相化学合成方法获得纯度达95%以上的cathelicidin‑2蛋白,该蛋白对多种病原微生物具有较强的抗菌活性,尤其对白色念珠菌、铜绿假单胞菌、大肠杆菌、变形假单胞菌具有较强的抑杀活性。cathelicidin‑2来源于蛙类,既可作为水产养殖饲料添加剂应用,也可进一步研制为抗菌剂、抗菌药物等,具有广泛的应用前景。

技术领域
本发明涉及分子生物学技术领域,特别涉及虎纹蛙(Hoplobatrachus rugulosus)抗菌肽cathelicidin-2基因克隆及其应用。
背景技术
近年来,抗生素不仅用于人类、畜禽和水产养殖业的疾病治疗和预防,而且在提高畜禽和水产养殖业的经济效益和饲料利用率方面发挥了不可替代的作用。然而,抗生素的滥用污染了养殖土壤和水体,导致耐药菌的出现;此外,残留的抗生素通过食物链进入人体,严重威胁人类的生命健康。因此,寻找抗生素的替代品已成为当前亟待解决的健康问题。在众多抗菌活性物质中,抗菌肽具有广谱抗菌活性,且不易产生耐药性,是抗生素的理想替代品。
抗菌肽(Antibacterial peptides,AMPs)是一类宿主防御肽,能够直接杀死微生物并刺激免疫系统产生响应。抗菌肽具有多种生物学活性,如对多种免疫细胞具有趋化作用、促进伤口愈合和淋巴细胞活化等,是机体抵抗病原菌感染的重要防线。截至2024年1月,抗菌肽数据库Antimicrobial Peptide Database(http://aps.unmc.edu/AP/)中收录的已知编码抗菌肽的基因序列达3940个,其中动物源抗菌肽超过2400个。天然抗菌肽是一般小于80个氨基酸的一类小分子肽,富含正电荷,具有较好的热稳定性和水溶性等性质,其结构特点主要为α-螺旋和β-折叠。抗菌肽的抗微生物活性机制,主要分为直接杀菌及间接杀菌:直接杀菌机制是抗菌肽通过与带负电荷的膜相互作用,导致膜通透性增加、细胞膜裂解或细胞内含物释放,最终导致细胞死亡;间接杀菌机制指抗菌肽渗入细胞质并与细胞内物质相互作用,如抑制DNA、RNA和蛋白质的合成、抑制蛋白质的折叠、抑制酶的活性和细胞壁的合成、促进裂解酶的释放以破坏细胞结构。近年来研究表明,一些抗菌肽对病毒、真菌、寄生虫和癌细胞等也有杀灭作用,甚至能提高免疫力、加速伤口愈合。迄今,越来越多耐药性微生物被发现,而天然抗菌肽或化学合成抗菌肽因其特殊的结构及生物学活性,在医药、养殖、饲料等领域具有广阔的开发和应用前景,现已成为国内外学者的研究热点。随着对抗菌肽的进一步改良,将会有更多新型、高效、有针对性的抗菌肽及其产品在畜禽和水产养殖领域得到广泛应用。
发明内容
本发明于虎纹蛙肝脏转录组文库中首次筛选获得抗菌肽cathelicidin-2基因序列,进一步利用PCR方法对其核苷酸序列进行了验证,采用固相化学合成方法获得了cathelicidin-2多肽,实验发现其具有广谱抗细菌、真菌活性。本发明为来源于虎纹蛙的抗菌肽cathelicidin-2的首次报道。
因此,本发明的第一目的在于提供虎纹蛙抗菌肽cathelicidin-2基因。

所述虎纹蛙抗菌肽cathelicidin-2的氨基酸分子式为C854H1346N268O279S9,分子量为20120.25道尔顿。抗菌肽cathelicidin-2由187个氨基酸组成,来源于虎纹蛙,具有抗菌功能的多肽。该抗菌肽的成熟肽预测分子量为4231.42道尔顿,所含52个氨基酸中包含11个带正电荷的氨基酸残基,没有带负电荷的氨基酸残基,根据氨基酸残基电荷预测该抗菌肽等电点为12.65。该抗菌肽的亲水性平均系数为-1.213,具有亲水水溶性,是一种带有正电荷的阳离子多肽。
采用固相化学合成方法即可获得纯度达95%以上的虎纹蛙抗菌肽cathelicidin-2。
所述虎纹蛙抗菌肽cathelicidin-2对多种革兰氏阴性菌、革兰氏阳性菌及真菌有显著的抗菌活性。其中,对铜绿假单胞菌的最小抑菌浓度为16-32μM;对大肠杆菌的最小抑菌浓度为16-32μM;对变形假单胞菌最小抑菌浓度为16-32μM;对白色念珠菌的最小抑菌浓度为32-64μM。扫描电子显微镜结果显示未处理的菌体形状规则,边界清晰,虎纹蛙抗菌肽cathelicidin-2处理后的大肠杆菌和铜绿假单胞菌出现菌体凹陷、皱缩和内容物外泄。与许多已知的两栖动物抗菌肽比较,cathelicidin-2抗菌效果较好、杀菌速率较快,显示出极大的应用价值,在制备防霉防腐剂、抗菌剂方面有良好的应用前景。
本发明通过固相化学合成方法获得了一种具有广谱且高效抗菌的抗菌肽,该抗菌肽来源于蛙类,既可用于畜禽养殖业作为饲料添加剂,也可研制用于动物的抗菌药物,具有广泛的应用前景。

以下实施例可以使本专业技术领域的技术人员更加全面的了解本发明,本发明并不限于下述实施例。
本实施例涉及的菌种有:铜绿假单胞菌(Pseudomonas aeruginosa)、嗜水气单胞菌(Aeromonashydrophila)、维氏气单胞菌(Aeromonas veronii)、迟缓爱德华氏菌(Edwardsiella tarda)、副溶血弧菌(Vibrio parahaemolyticus)、大肠埃希氏菌(Escherichia coli)、变形假单胞菌(Pseudomonas plecoglossicida)、单核细胞李斯特菌(Listeria monocytogenes)、金黄色葡萄球菌(Staphylococcus aureus)、表皮葡萄球菌(Staphylococcus epidermidis)、枯草芽孢杆菌(Bacillus subtilis)、白假丝酵母(Candida albicans)、黑曲霉(Aspergillus niger)等,均购自中国科学院微生物研究所菌种保藏中心。
虎纹蛙来源于具有人工养殖虎纹蛙资质的企业。
实施例1虎纹蛙抗菌肽cathelicidin-2核苷酸序列扩增
通过对虎纹蛙肝脏组织转录组进行测序,筛选获得了虎纹蛙抗菌肽cathelicidin-2基因全长cDNA序列。虎纹蛙抗菌肽cathelicidin-2基因的开放阅读框(SEQID NO:1)为564bp(含终止密码子TAA)。

根据所获得的cathelicidin-2基因的全长cDNA序列,设计基因特异性引物(表1),通过RACE PCR技术对其序列进行进一步验证。以第一个引物为例,PCR反应体系如下:
混合均匀,进行PCR反应。反应程序如下所述:
1)95℃预变性5min;
2)95℃变性30s,55℃退火30s,72℃延伸1min,30个循环;
3)72℃延伸5min;
4)4℃停止。
将PCR产物进行琼脂糖凝胶电泳分离,切下含有目的片段的琼脂糖凝胶,放入1.5mL离心管中。使用Wizard SV Gel and PCR Clean-Up System回收试剂盒回收目的DNA片段。具体步骤如下:
(1)称量凝胶重量,按照每100mg琼脂糖凝胶对应100μL溶液BD的比例,向离心管中加入溶液BD;
(2)60℃水浴10min,直至凝胶完全融化;
(3)将溶液置于DNA纯化柱中,静置1min;16 000g离心1min,弃溶液;
(4)将纯化柱置于原收集管中,加入700μL Membrane Wash Solution,16 000g离心1min,弃滤液;
(5)重复1次步骤(4);
(6)将纯化柱置于原收集管中,16 000g再次离心1min,彻底去除纯化柱中残留的液体;
(7)将离心柱至于新的1.5mL离心管中,向纯化柱的中央处滴加30~50μLNuclease-Free Water;16 000g离心1min,管底部即为目的DNA片段,置于-20℃保存。
(8)取部分回收的DNA样品进行1.5%琼脂糖凝胶电泳检测,用凝胶成像系统对电泳结果拍照。
PCR产物与载体连接并转化
(1)使用pGEM-T Vector试剂盒,在离心管中配置如下反应体系:
(2)16℃反应30min;
(3)全量加入到感受态细胞中,在冰水中放置30min;
(4)42℃热激45s,然后迅速放置在冰水中1min;
(5)加入890μL LB液体培养基,37℃、200rpm振荡培养2h,然后4000rpm离心10min,从底部吸取100μL菌液,在含Amp的固体培养基上培养过夜(14h);
(6)在平板培养基中挑选5个白色菌落加入5mL含Amp的LB液体培养基中,37℃、200rpm,培养时间约3.5h。
PCR法快速鉴定阳性克隆
利用通用引物M13F-47和M13R-48,对克隆的cathelicidin-2cDNA序列进行验证。
(1)使用2×HS Mix试剂构建PCR反应体系,菌落PCR体系如下:
(2)混合均匀后,按以下程序进行PCR反应:
94℃、3min;
94℃、30sec,55-68℃、30sec,72℃、1min,30个循环;
72℃、10min;
4℃停止反应。
(3)吸取5μL扩增产物进行琼脂糖凝胶电泳检测,用凝胶成像系统在紫外灯下观察电泳结果,并拍照。
核酸序列测定与分析
将鉴定含有重组阳性克隆的大肠杆菌按照15%的比例加入灭菌甘油进行保种,样品送至生物公司进行核苷酸序列测定。利用生物信息学软件对所测的序列进行拼接和比对。
综上分析,所述cathelicidin-2基因来源于虎纹蛙,其氨基酸序列为:SEQ ID NO:2。
所述虎纹蛙抗菌肽cathelicidin-2的分子式为C854H1346N268O279S9,分子量为20120.25道尔顿。抗菌肽cathelicidin-2由187个氨基酸组成,来源于虎纹蛙,具有抗菌功能的多肽。该抗菌肽的成熟肽预测分子量为4231.42道尔顿,所含52个氨基酸中包含11个带正电荷的氨基酸残基,没有带负电荷的氨基酸残基,根据氨基酸残基电荷预测该抗菌肽等电点为12.65。该抗菌肽的亲水性平均系数为-1.213,具有亲水水溶性,是一种带有正电荷的阳离子多肽。
采用固相化学合成方法即可获得纯度达95%以上的虎纹蛙抗菌肽cathelicidin-2。
本实施例中虎纹蛙抗菌肽cathelicidin-2委托江苏金斯瑞生物科技有限公司通过固相化学合成方法获得,并提供多肽分子量、HPLC、质谱等监测信息。
实施例2虎纹蛙抗菌肽cathelicidin-2最小抑菌浓度(MIC:minimuminhibitionconcentration)和最小杀菌浓度(MBC:minimum bactericidal concentration)的测定
(a)将保种的铜绿假单胞菌、嗜水气单胞菌、维氏气单胞菌、迟缓爱德华氏菌、副溶血弧菌、大肠埃希氏菌和变形假单胞菌在MH肉汤平板上划线,在相应培养最适温度倒置培养12-16h;单核细胞李斯特菌、金黄色葡萄球菌、表皮葡萄球菌和枯草芽孢杆菌在LB肉汤平板上划线,在相应培养最适温度倒置培养12-16h;白假丝酵母、黑曲霉在YM平板上划线,于37℃培养3-7d。从各平板上挑取菌落接种于相应培养基斜面上,细菌继续培养10-16h,真菌继续培养1-3d后,用10mM磷酸盐缓冲液(pH 7.4)冲洗斜面,革兰氏阴性菌用MH液体培养基:10mM磷酸缓冲液=2:3的混合培养基稀释至OD600=0.003;革兰氏阳性菌用LB液体培养基:10mM磷酸缓冲液=2:3的混合培养基稀释至OD600=0.003;真菌用YM液体培养基:10mM磷酸缓冲液=2:3的混合培养基调整稀释至OD600=0.00067。准备好的菌液须在20min内使用。
(b)将抗菌肽cathelicidin-2经0.22μm滤膜过滤后,利用Bradford法测定蛋白浓度,倍比稀释蛋白浓度为0.5、1、2、4、8、16、32、64μM,4℃保存备用。
(c)在96孔细胞培养板上进行最小抑菌浓度检测实验,每种被测菌按照以下操作设置空白对照组、阴性对照组和待测样品实验组,每组设置3个平行样:
阴性对照组:加100μL灭菌超纯水和100μL菌悬液;
空白对照组:加100μL待测蛋白样品和100μL磷酸盐缓冲液;
样品实验组:加100μL待测蛋白样品和100μL菌悬液。
对于细菌,将培养板置于各菌种相对应的最适培养温度下,培养18-24h,观察结果;
对于真菌,将培养板置于28℃培养1-2d,观察结果。
将96孔细胞培养板待测实验组中肉眼观察未有菌生长的孔中培养液混匀后,吸取2μL接种于相应固体培养基平板上,置于各最适生长温度继续培养24-48h,观察MBC(minimum bactericidal concentration)结果。
虎纹蛙抗菌肽cathelicidin-2MIC、MBC抗菌谱如表2所示:

注:MIC:最小抑菌浓度(μM),用a-b表示。a:肉眼可见菌体生长的最高蛋白浓度;b:肉眼未见菌体生长的最低蛋白浓度。
MBC:最小杀菌浓度(μM),能够杀死99.9%菌体的最低蛋白浓度。
实施例3虎纹蛙抗菌肽cathelicidin-2杀菌动力学曲线
本部分选取两种革兰氏阴性菌(大肠杆菌)及(铜绿假单胞菌)作为被测菌进行虎纹蛙抗菌肽cathelicidin-2杀菌动力学曲线的测定。
(a)将保种的大肠杆菌及铜绿假单胞菌涂布于MH营养肉汤平板上在最适培养温度倒置培养。
(b)从平板上挑取菌落接种于斜面上继续培养,用10mM磷酸盐缓冲液(pH 7.4)冲洗斜面,用MH液体培养基:10mM磷酸缓冲液=2:3的混合培养基调整菌悬液浓度至OD600=3.3×104cfu/mL。
(c)将cathelicidin-2蛋白经过0.22μm滤膜过滤后,稀释蛋白至所需实验浓度,置于冰上待用。
(d)在96孔细胞培养板上,设置阳性对照组、待测实验组,每组设置三个平行样:
阳性对照组:加100μL菌悬液和100μL 10mM磷酸缓冲液。
待测实验组:加100μL菌悬液和100μL待测蛋白样品。
将96孔细胞培养板置于28℃培养箱中继续培养,于不同时间点取适量培养液稀释后涂布MH营养肉汤平板,平板于菌种最适温度继续培养至单克隆生长,计数每个平板上克隆数目,统计分析并作杀菌动力学曲线。如图1所示,结果表明:2倍MBC浓度cathelicidin-2可在作用2h内杀死50%以上的大肠杆菌,在10h后杀菌指数达到100%;2倍MBC浓度的cathelicidin-2与铜绿假单胞菌作用13h后,杀菌指数达到100%。
实施例4虎纹蛙抗菌肽cathelicidin-2与微生物作用后形态变化
(a)将大肠杆菌和铜绿假单胞菌在MH培养基中培养,收集对数生长期的菌液,使用DPBS洗涤菌体两次并重悬,调整菌悬液浓度至1×107cfu/mL。
(b)将多肽稀释至4×MBC,与等体积的菌悬液混合至终浓度2×MBC,在室温条件下孵育2h。孵育结束离心收集菌体,用2.5%的戊二醛溶液重悬菌体,在4℃条件下固定2h;结束后5000g,10min离心收集菌体,并用PBS重悬菌体后洗涤2次,用10μLPBS重悬菌体制成高浓度菌悬液。
(c)将悬液滴加到裁剪成4mm×4mm带有多聚赖氨酸涂层的玻片上,4℃条件下放置2-4h使菌体与玻片充分粘附;用裁剪后的滤纸吸净玻片上残余的水分。
(d)将玻片放置在自制的支架上用PBS清洗3次,每次10min;分别用30%(v/v)乙醇溶液5min,50%(v/v)乙醇溶液5min,70%(v/v)乙醇溶液10min,上述三步均在冰上进行。然后80%(v/v)乙醇溶液脱水10min,无水乙醇处理15min,再次用无水乙醇处理15min脱水,结束后立即将玻片放入临界点干燥仪中干燥2h。
干燥后的玻片样品用导电胶贴附于样品台后进行喷金并使用扫描电子显微镜观察和拍照,结果如图2所示。

2.编码如权利要求1所述的虎纹蛙抗菌肽cathelicidin-2的核苷酸序列。
3.虎纹蛙抗菌肽cathelicidin-2的开放阅读框序列,如SEQ ID NO:1所示。
4.如权利要求1所述的抗菌肽cathelicidin-2的制备方法,其特征在于所述抗菌肽cathelicidin-2通过固相化学法合成获得。
5.如权利要求1所述的抗菌肽cathelicidin-2在制备抗菌剂中的应用。
6.如权利要求5所述的应用,其特征在于,所述的抗菌包括抗白色念珠菌、铜绿假单胞菌、大肠杆菌、变形假单胞菌中的至少一种。
7.如权利要求1所述的抗菌肽cathelicidin-2在制备饲料添加剂中的应用。
8.如权利要求1所述的抗菌肽cathelicidin-2在制备防霉防腐剂中的应用。
9.如权利要求1所述的抗菌肽cathelicidin-2在制备抗菌药物中的应用。
10.如权利要求9所述的应用,其特征在于,所述的抗菌包括抗白色念珠菌、铜绿假单胞菌、大肠杆菌、变形假单胞菌中的至少一种。
