1. Experiment for clearing heat and abating fever
1. Method of
As a result of a large number of studies, LPS is considered to be an active ingredient of endotoxin of gram-negative bacteria, has high water solubility, is a bacterial heat source with a strong effect, and is similar to the type of fever caused by clinical infectious inflammation in that the type of fever induced by LPS is shown as biphasic fever or triphasic fever. Therefore, 80 healthy Kunming mice with normal development and no fever lesions are selected and randomly divided into 10 groups (blank group, model group, control group and 1-7 groups), 8 mice in each group have a weight of about 18-22 g, and other groups of mice except the blank group adopt LPS to induce fever modeling.
And the water is not forbidden after fasted for 8 hours before molding. The blank group and the model group are filled with physiological saline according to the ratio of 0.2mL/10g, the body temperature is measured for 1 time every 0.5h, and the continuous monitoring is carried out for 10h. Except for the blank group, the other groups respectively inject LPS (20 mug/kg) into the abdominal cavity of the mice to induce animals to generate heat, and the stomach is irrigated by different medicines while molding; the control group was dosed with acetaminophen at 1.5mg/20 g; 1 to 7 groups of Chinese medicinal decoction are infused according to groups, and the dosage is 0.2mL/10g. The body temperature was measured 1 time every 0.5h after LPS injection, and the monitoring was continued for 10h, and the maximum body temperature elevation DeltaT (difference between the body temperature at the time of fever and the basal body temperature) was recorded.
2. Results
The specific results are shown in tables 1 to 3
TABLE 1 measurement of DeltaT (. Degree. C.) data at 0.5h intervals
TABLE 2 measurement of DeltaT (. Degree. C.) data at 0.5h intervals
TABLE 3 measurement of DeltaT (. Degree. C.) data at 0.5h intervals
Note that: p is less than or equal to 0.05 compared with a blank group; the ratio of the material to the model group is #p is less than or equal to 0.05; compared with the #1 group, the p is less than or equal to 0.05.
From the above results, it can be seen that: for mice with a model of fever induced by LPS, the prescription provided by the invention can play a role in reducing temperature within 1.5 hours and lasts for 8.5 hours, and has definite effects of reducing body temperature, relieving fever and bringing down fever, and is quick in action speed and strong in action. After the components of the invention are split differently, the antipyretic and antipyretic effects of the respective split items are not obvious. Therefore, the prescription has the effects of clearing heat, reducing fever, nourishing yin, cooling blood and dispersing wind-heat through specific medicinal components.
2. Phlegm resolving experiment
1. Method of
72 Healthy Kunming mice with normal development and no fever lesions are selected, the weight of the mice is about 18-22 g, the mice are randomly divided into 9 groups (blank group, control group and 1-7 groups), and 8 mice in each group are subjected to a tracheal phenol red excretion experiment. The experimental principle is that after phenol red is used as an indicator, if a mouse takes a medicine with the function of resolving phlegm after the phenol red is injected into the abdominal cavity of the mouse, the bronchogenic secretion of the mouse is increased, so that the excretion amount of the phenol red in the mucous membrane of the respiratory tract is increased, the excretion of the phenol red is more, the expectoration of the sputum is predicted to be promoted, and the phlegm resolving function of the medicine can be deduced from the influence of the experimental medicine on the excretion amount of the intratracheal phenol red.
The blank group is filled with normal saline according to 0.2mL/10g, the control group is compound fresh bamboo juice, and 1-7 groups are filled with corresponding traditional Chinese medicine decoction according to grouping, and the dosage is 0.2mL/10g. Fasted for 8h before the experiment, 30min after administration, each mouse was intraperitoneally injected with 0.7mL of 2.5% phenol red solution, immediately sacrificed after 30min, fixed in position, the trachea was separated, 0.5mL of 5% NaHCO 3 solution was extracted with a 1mL syringe, the trachea was flushed 3 times by inserting about 0.3-0.5 cm into the trachea from the thyroid cartilage, 3 times of aspiration back and forth each time, the 3 times of eluates were combined, centrifuged for 15min, the supernatant was diluted 5 times with distilled water, and the phenol red absorption (A) was measured at a wavelength of 546nm with an ultraviolet spectrophotometer.
2. Results
The specific results are shown in Table 4
Table 4 comparison of tracheal phenol Red excretion in mice of each group
Note that: p is less than or equal to 0.05 compared with a blank group; compared with the #1 group, the p is less than or equal to 0.05.
From the above results, it can be seen that: the prescription can promote the secretion of the tracheal phenol red of the mice, and has obvious phlegm-resolving effect and reduces phlegm dampness, and after the prescription is split differently, the phlegm-resolving effect of the split items is not obvious. Therefore, the prescription of the invention can take the functions of spleen and stomach transportation and transformation into account through specific medicine composition, and has the effects of dredging channels, resolving dampness and removing cataract.
3. Acute upper respiratory tract infection simulation experiment
1. Method of
Acute upper respiratory tract infection belongs to the category of external infection of traditional Chinese medicine, no matter whether the external infection is wind-cold or wind-heat or epidemic disease, the external pathogenic factors disturb the lung qi to lose the benefit, exterior cold can enter the interior to transform heat, and the heat burns the body fluid to form phlegm heat, and the pathogenic heat flows along the channel to form lung heat. The nature of exogenous pathogenic factors is considered by traditional Chinese medicine to be due to the internal cause of 'deficiency of healthy qi'. Therefore, the Chinese medicinal composition aims at strengthening body resistance to eliminate pathogenic factors, improving the immunity of the organism, inhibiting bacteria and resisting inflammation, and controlling the illness state, relieving symptoms and shortening the illness course by regulating the immunity of the organism.
80 Healthy Kunming mice with normal development and no fever lesions are selected, the weight of the mice is about 18-22 g, the mice are randomly divided into 10 groups (blank group, model group, control group and 1-7 groups), 8 mice in each group except the blank group, 50 mu l of staphylococcus aureus bacterial liquid is slowly dripped from the nasal cavity of the mice in the rest groups under the state of diethyl ether anesthesia, and after the mice are automatically inhaled, the mice begin to be treated by medicaments after molding for 1 hour. Wherein the blank group and the model group are filled with physiological saline according to 0.2mL/10g, and the control group is filled with azithromycin according to 1.5mg/20g.1 to 7 groups of Chinese medicinal decoction are infused according to groups, and the dosage is 0.2mL/10g. Fasted for 8 hours before the experiment, the administration is carried out for 1 time/d for 7 days, and the materials are taken for 7 days after the modeling to detect TNF-alpha and IL-6.
2. Results
The specific results are shown in tables 5 to 6.
TABLE 5 Lung TNF-alpha content (pg/ml) in mice of each group
Note that: p is less than or equal to 0.05 compared with a blank group; the ratio of the material to the model group is #p is less than or equal to 0.05; compared with the #1 group, the p is less than or equal to 0.05.
TABLE 6 Lung IL-6 content (pg/ml) in mice of each group
Note that: p is less than or equal to 0.05 compared with a blank group; the ratio of the material to the model group is #p is less than or equal to 0.05; compared with the #1 group, the p is less than or equal to 0.05.
TNF-alpha has various activities such as inflammation, immunity, internal environment stabilization and the like, and TNF-alpha and IL-6 are important links in an organism immunity regulation system after infection, have the effect of improving organism immunity response and have the important effect of resisting infection. Experimental results show that compared with healthy and active mice in a blank group, the concentration and content of TNF-alpha and IL-6 in a model group are improved, the mice are poor in mental state, are curled, have poor response to the surrounding environment, have upright and matt hair, have increased conjunctival secretion of partial mice, have symptoms of cough caused by shortness of breath, and are positively correlated with the degree of fever caused by clinical verification infection. In mice treated by the drug lavage, the content of TNF-alpha and IL-6 in the lung tissue homogenate of mice in the control group and the group 1 is obviously reduced compared with that in the model group, and the mice have gradually improved spirit, stable respiration and no cough symptom. Compared with other experimental groups after the group prescription is disassembled, the group 1 is obviously superior to other groups, which proves that the formula of the invention can activate the immune mechanism of the organism in a short time, strengthen the anti-inflammatory and immunoregulatory effects of the organism, shorten the anti-inflammatory process of the organism and accelerate the recovery of respiratory tract infectious diseases, but other formulas after the group prescription is disassembled have no obvious effect.
From the experimental results, it can be seen that: the prescription has the effects of detoxification, vital qi nourishing, vital qi strengthening and evil eliminating through specific medicine composition, has advantages in improving clinical symptoms of cough with fever and phlegm, and has outstanding advantages in enhancing organism anti-inflammatory and immunoregulation.
