背景技术
由于长期反复不当的使用外用激素或广泛使用含激素的护肤品导致激素依赖性皮炎发病呈逐年上升趋势,顽固难治愈;病症具体表现为外用糖皮质激素之后原发皮损消失,但是停用后再次出现炎性损害,需要反复使用糖皮质激素来控制症状并且皮炎逐步加重。长期用药后出现多毛、色素沉着等症状。多毛症是指女性在面部、上下肢和躯干等部位出现男性类型的终(粗)毛,粗毛过分增多而超出正常范围或毳毛部位长出粗黑的毛。研究更为有效的方法来消除激素依赖性皮炎,抑制多毛一直是皮肤科医生的努力方向。
目前治疗多毛的方法并不理想。药物治疗,比如依氟鸟氨酸副作用严重,治疗效果不佳。通过光脱毛和电解等物理疗法去除不必要的毛发是痛苦和反复的,这会破坏毛囊并耗尽皮肤干细胞库,不利于伤口愈合。长期来看,这种疾病会给患者造成心理困扰。因此,开发一种更安全、更有效的策略显得尤为重要。

本发明内容还包括所述的原位生成的金纳米簇材料或金纳米簇材料在制备针对皮肤损伤相关的多毛症的抑毛药物中的应用。
有益效果:与现有技术相比,本发明用金离子作为前驱体,利用受损皮肤中的OSM原位合成金纳米簇,实现对正常皮肤与受损皮肤的成像鉴别;在损伤模型中原位合成的金纳米簇进一步还可抑制毛囊再生,对皮肤损伤相关多毛症起抑毛作用。这种简便、安全、无毒的原位生物合成方法在未来应用于抑制皮肤损伤相关的多毛症具有很大的潜力。

具体实施方式
下面结合附图和实施例,对本发明的技术方案进行详细说明。
本实验的氯金酸、氯金酸钠、八氯化四金、三氯化金、一氯化金等试剂购于国药集团化学试剂有限公司。
实施例1氯金酸溶液在拔毛损伤模型中原位合成金纳米簇及其表征
1、金前驱体溶液的制备
将氯金酸溶于PBS缓冲液(pH=7,10mmol/L),制得浓度为1.5mmol/L的氯金酸溶液。
2、拔毛损伤模型和未拔毛正常小鼠模型的构建
选取处于毛囊静止期的C57小鼠(7-8周龄),背部皮肤呈粉红色,无皮肤病且无抓痕损伤。采用松蜡合剂拔去背毛,形成拔毛损伤模型。用眼科剪小心剪去小鼠的背毛(裸露出背皮即可,不要损伤皮肤),作为未拔毛正常小鼠模型。
3、金前驱体溶液注射到拔毛损伤皮肤中得到原位合成的金纳米簇
拔毛30分钟后,将浓度为1.5mmol/L的氯金酸溶液25μl皮内注射进入拔毛损伤小鼠体内,作为实验模型组(DAu组)。注射25μl PBS缓冲液(pH=7,10mmol/L)进入拔毛损伤小鼠皮肤内,作为对照模型组(DPBS组);同时将浓度为1.5mmol/L的氯金酸溶液25μl皮内注射进入未拔毛正常小鼠体内,作为实验对照组(HAu组)。每天一次,连续注射三次,注意要注射至皮肤内浅表部位且形成一个临时性的小皮丘。
4、活体荧光成像、安全性分析和相关蛋白表达等结果表征
活体荧光成像仪观察,分别选取3小时、8小时、24小时不同时间点。用5%异氟烷将小鼠进行气体麻醉后置于小动物活体成像仪暗箱内,选择480nm作为激发波长采集荧光图像。
分别提取各组注射部位皮肤组织中的金纳米材料做分析表征:荧光光谱、高分辨透射电镜(HR-TEM)、X射线光电子能谱(XPS)、能谱分析(EDS);解剖实验模型组、对照模型组、实验对照组小鼠取器官心、肺、肝、脾、肾制备病理切片,HE染色,以及取血做血液检查,分析局部注射金前驱体对C57小鼠的安全性。提取总RNA和蛋白质分别进行RT-PCR与Western Blot实验,检测NFKB信号通路表达情况。
分别提取第1天,4天,和8天注射部位全层皮肤组织中的蛋白,BCA法测总蛋白浓度,酶联免实验ELISA检测TNFa的表达。
以上表征结果如下:注射3小时后实验模型组(DAu组)、对照模型组(DPBS组)、实验对照组(HAu组)小鼠背皮均无荧光;8小时后仅实验模型组(DAu组)小鼠背皮注射部位有荧光,平均荧光强度(Average radiant efficiency)高达1.04ⅹ108[p/s/cm2/sr]/[μW/cm2],并且荧光一直持续到24h,其平均荧光强度为1.03ⅹ108[p/s/cm2/sr]/[μW/cm2],而对照模型组(DPBS组)和实验对照组(HAu组)均没有荧光,表明金前驱体溶液只有在拔毛损伤皮肤中才能够原位生成得到金纳米簇,而在正常皮肤中并不能原位生成得到金纳米簇。两天后在主要的组织器官(liv,肝脏;kid,肾脏;spl,脾脏;lun,肺;hea,心脏)中无荧光纳米材料的累积(见图2),各组之间相对荧光强度(Relative fluorescent intensity)无变化。
在拔毛后的第9天实验模型组(DAu组)小鼠背皮注射部位出现抑毛现象,而对照模型组(DPBS组)小鼠背皮注射部位皮肤开始变黑长毛,实验对照组(HAu组)小鼠背皮无明显变化且毛囊仍处于静止期(见图3)。
图4为从DAu组皮肤中提取的荧光金纳米簇(FGNCs)的表征结果,材料表征分析发现在拔毛损伤皮肤中原位合成了金纳米簇(见图4),高价的金前驱体被还原成了低价的金簇(0价和1价),直径小于2nm的金簇由于具有很强的量子约束效应(对Au原子的数量甚至价电子的数量都很敏感),表现出与更大直径的金纳米颗粒不同的物理和化学性质,因此被称为纳米团簇。图4a是荧光金纳米簇的透射电镜成像,工作电压(Voltage)为200KV,左上角插入的蓝色直方图显示了荧光金纳米簇的粒径(Diameter)分布百分比(Percentage),TEM图像显示,FGNCs的平均尺寸为1.99nm,图4b是荧光金纳米簇的高分辨率透射电镜成像,其面间距为0.23nm,与Au的(111)面相对应。图4c是从皮肤中提取的FGNCs进行荧光光谱分析其荧光强度(Intensity),最大激发波长(Ex max wavelength)为462nm,最大发射波长(Emmax wavelength)为525nm;图4d是XPS分析和(e)是EDS分析,一价金和零价金4f轨道的电子结合能(Binding energy)所对应相对光电子流强度(intensity)的拟合曲线分析表明两种价态的Au(0)和Au(I)共存,样品中Au(0)和Au(I)共存的原子比分别为83.7%和16.3%。因此,它属于金纳米团簇的范围。
图5a是三组小鼠在首次给药后12天主要脏器的HE染色,图5b是各组小鼠12d内体重变化情况,从图看出,局部药物注射对实验模型组(DAu组)、对照模型组(DPBS组)、实验对照组(HAu组)小鼠心(Heart)、肝(Liver)、脾(Spleen)、肺(Lung)、肾(Kidney)等主要器官无影响,12天(day)内体重(Body weight)平稳增加,因此,FGNCs安全性好(见图5和表1与表2)。表1为各组小鼠首次给药后12天的血常规分析,表2为各组小鼠首次给药后12天的血生化分析。

从表1和表2的结果表明各指标均在正常范围之内,因此本发明的原位合成荧光金纳米簇是安全的。
图6是RT-PCR和Western Blot检测小鼠皮肤中NFKB信号通路关键分子的表达。第一次给药后8天,从DAu组、DPBS组和HAu组小鼠皮肤中提取总RNA和蛋白分别进行RT-PCR与Western Blot实验,结果表明原位合成的金纳米簇抑制NFKB信号通路(见图6)。
图7是ELISA实验显示在拔毛后1天到4天、8天,小鼠皮肤中TNF-a的表达水平,实验模型组(DAu组)小鼠皮肤中TNF-a的表达水平逐渐下降,而在对照模型组(DPBS组)小鼠的皮肤中TNF-a的表达水平保持不变,表明原位合成的金纳米簇抑制TNF-a表达。
为观察毛囊细胞的增殖情况,第一次给药后第9天皮肤组织石蜡包埋切片。免疫荧光染色观察细胞增殖标志物Ki67的表达(见图8)。DPBS组和DAu-NA组(DAu组皮肤的非给药部位,the non-dosed site,DAu-NA)组中的毛基质完全包裹真皮乳头,而DAu-A(DAu组给药部位的皮肤,the dosed site,DAu-A)中的毛基质未包裹或部分包裹真皮乳头。由箭头所示可知,Ki67在DPBS和DAu-NA的毛基质和根外鞘中表达强烈,但在DAu-A的次级毛芽和根外鞘中表达弱,表明DAu-A的毛囊细胞增殖弱。图9是各组小鼠的毛囊长度统计。与DPBS组皮肤的正常生长期毛囊和DAu组皮肤的非给药部位(the non-dosed site,DAu-NA)毛囊相比,DAu组给药部位皮肤(the dosed site,DAu-A)的毛囊体积更小,长度更短(见图9)。DAu-A毛囊长度(the length of hair follicle)显著短于DPBS组生长期毛囊,而且DAu-A毛囊长度跟HAu组静止期的毛囊长度无明显差异(No significance,NS),说明DAu-A组抑制效果具备显著性。因此,上述结果表明原位合成的金纳米簇通过负性调控NFKB/TNF-a信号轴抑制毛囊再生。
实施例2
考察不同浓度的金前驱体对试验结果的影响,具体方法同实施例1,不同之处在于考察金前驱体的不同浓度对于结果的影响,所考察的金前驱体(氯金酸)的浓度分别是:1mmol/L、2mmol/L、2.5mmol/L。
结果表明,当浓度低于1.5mmol/L(比如0.5mmol/L和1mmol/L)时荧光强度低且抑毛效果不明显(见图10、11和12)。图12中拔毛损伤DAu组与正常皮肤HAu组荧光强度值(ROI)两者无者明显差异,其值均在104-105之间的水平(与未给药的皮肤荧光强度值无异),而且远低于108。如实施例1中所示,当浓度为1.5mmol/L时,DAu组荧光明显且其荧光强度(Radiant efficiency)显著高于另外两组,且其在24h时的荧光强度为1.03ⅹ108[p/s/cm2/sr]/[μW/cm2](见图1);当浓度为2mmol/L时,DAu组荧光明显且其荧光强度(Radiantefficiency)显著高于另外两组,且其荧光强度高达1.3ⅹ108[p/s/cm2/sr]/[μW/cm2](见图13);当浓度高于2mmol/L时(比如2.5mmol/L)反而会加重皮肤损伤,不利损伤愈合。因此,浓度对金前驱体发挥作用有一定的影响,并且在1.5mmol/L~2mmol/L内能原位合成荧光金纳米簇并表现出明显的抑毛效果。考虑到,有明显抑毛效果的同时用药浓度低安全性更好,所以后续采用1.5mmol/L为最佳浓度。
实施例3
考察不同金前驱体对试验结果的影响,试验方法同实施例1,不同之处在于考察不同金前驱体对于结果的影响,所考察的金前驱体是(浓度是1.5mmol/L):三氯化金。结果如下:DAu组荧光明显且强度显著高于另外两组,其荧光强度高达1.23ⅹ108[p/s/cm2/sr]/[μW/cm2](见图14);而且表现出明显的抑毛效果。可见,在损伤皮肤中,三氯化金作为前驱体,原位合成了荧光金纳米簇。
实施例4
不同损伤模型下的抑毛效果,试验方法同实施例1,不同之处在于考察不同损伤模型下对于抑毛效果的影响,所构建的损伤模型为全层皮肤缺失损伤模型。
1、全层皮肤缺失模型:选取处于毛囊静止期的C57小鼠(7-8周龄),背部皮肤呈粉红色,无皮肤病且无抓痕损伤。用造模器在小鼠背部皮肤形成一个直径大约3mm的圆形全层皮肤缺损构建全层皮肤缺失模型。
2、实验过程:将金前驱体从全层皮损小鼠的尾静脉注射或病灶部位原位给药作为实验组,同时设置给予PBS对照组小鼠;用小动物活体成像仪对各组小鼠进行实时原位成像观察并记录病灶的变化情况;与对照组相比,成像信号能快速识别实验组小鼠的病变部位并富集在该区域,皮肤组织切片进行HE染色以及Ki67增殖标记物免疫荧光染色等实验观察可见实验组小鼠毛囊再生受抑制。
Claims (10)
1.一种在损伤模型中原位生成的金纳米簇材料的制备方法,其特征在于,包括以下步骤:将金前驱体溶液处理损伤模型即得原位生成的金纳米簇材料,所述金前驱体溶液包括氯金酸溶液、氯金酸钠溶液、八氯化四金溶液、三氯化金溶液或一氯化金溶液中的一种或几种。
2.根据权利要求1所述的在损伤模型中原位生成的金纳米簇材料的制备方法,其特征在于,所述金前驱体溶液是将金前驱体溶于PH缓冲液中制备得到。
3.根据权利要求2所述的在损伤模型中原位生成的金纳米簇材料的制备方法,其特征在于,所述PH缓冲液包括磷酸盐缓冲液。
4.根据权利要求2所述的在损伤模型中原位生成的金纳米簇材料的制备方法,其特征在于,所述金前驱体溶液的浓度为1.5~2.0 mmol/L。
5.根据权利要求1所述的在损伤模型中原位生成的金纳米簇材料的制备方法,其特征在于,所述损伤模型包括小鼠拔毛损伤模型或小鼠全层皮肤缺失模型。
6.根据权利要求1所述的在损伤模型中原位生成的金纳米簇材料的制备方法,其特征在于,所述小鼠拔毛损伤模型的构建方法包括:选取处于毛囊静止期的小鼠,无皮肤病且无抓痕损伤,采用松蜡合剂拔去背毛,形成拔毛损伤模型。
7.根据权利要求1所述的在损伤模型中原位生成的金纳米簇材料的制备方法,其特征在于,所述小鼠全层皮肤缺失模型的构建方法如下:选取处于毛囊静止期的小鼠,无皮肤病且无抓痕损伤,造模器在小鼠背部皮肤形成直径3~5mm的圆形全层皮肤缺损。
8.权利要求1~7任一项所述的制备方法制备得到的在损伤模型中原位生成的金纳米簇材料。
9.权利要求8所述的在损伤模型中原位生成的金纳米簇材料在损伤皮肤成像鉴别中的应用,所述应用不涉及疾病的治疗和诊断目的。
10.权利要求9所述的在损伤模型中原位生成的金纳米簇材料或金纳米簇材料在制备针对皮肤损伤相关的多毛症的抑毛药物中的应用。
