技术领域
本发明涉及一种用于生产调节性T细胞的方法、通过所述方法获得的调节性T细胞、包含所述调节性T细胞的药物组合物等。本发明还涉及其中被引入表达构建体的iPS细胞。
背景技术
近年来,由于调节性T细胞(Treg)的免疫反应抑制功能,使用调节性T细胞的细胞疗法(Treg疗法)的研究和开发已经在世界范围内得到推广,以用于治疗各种免疫病症的目的。因此,调节性T细胞预期用于移植物抗宿主病(GvHD)和自身免疫性疾病的治疗中,以及用于炎性疾病和过敏性疾病的治疗和预防中。
调节性T细胞宜广泛地分为天然存在的调节性T细胞和诱导型调节性T细胞。天然存在的调节性T细胞被定义为在胸腺中天然产生的。诱导型调节性T细胞被定义为是在人工耐受诱导模型中响应于外部刺激而从外周幼稚T细胞分化诱导而来的。这些调节性T细胞根据细胞中表达的标志物类型而进一步细分。
当CD4+CD25+调节性T细胞(天然存在的调节性T细胞群之一)从活体中去除时,各种器官特异性自身免疫性疾病会自发发生。此时,移植CD4+CD25+调节性T细胞预防自身免疫性疾病的发生。因此,天然存在的CD4+CD25+调节性T细胞被认为在维持外周免疫自身耐受中发挥重要作用。从那时起,人们已经清楚CD4+CD25+调节性T细胞不仅可抑制自身免疫,还可抑制大多数免疫反应,诸如外来抗原引起的炎症、移植引起的排斥、感染免疫、过敏和肿瘤免疫。目前,转录因子FOXP3已被明确为CD4+CD25+调节性T细胞的主调节因子。
NPL 1公开了对于由FOXP3突变引起的免疫失调、多内分泌病、肠病、X连锁(IPEX)综合征,使用慢病毒载体将基因引入自体造血干细胞中以恢复FOXP3表达,并且将细胞施用于患有IPEX综合征的模型小鼠。据报告,造血干细胞因此分化为功能性调节性T细胞,从而从自身免疫表型中恢复。此外,PTL 1公开了包含编码人FOXP3蛋白的核苷酸序列的类似重组慢病毒载体。
PTL 2公开了Mcl-1与Foxp3一起表达对于提高Treg的存活很重要。另外,PTL 3公开了一种基因工程化哺乳动物细胞,其包含编码促进分化为CD4+Treg的谱系定向因子(lineage commitment factor)的转基因。PTL 4公开了一种用于通过培养三维细胞聚集体来制备来自干细胞或祖细胞的T细胞组合物的方法,所述三维细胞聚集体包含选定的表达Notch配体的基质细胞群体和选定的干细胞或祖细胞群体。
然而,尽管期望调节性T细胞具有高生产效率(产率)的工业生产方法,但目前还没有用于高效生产调节性T细胞的方法的报道。
FOXP3是Treg的主调节因子。已知FOXP3在Treg中组成型地表达,并且在不具有抑制功能的常规T细胞(Tconv,也称为效应T细胞或炎性T细胞)中未观察到组成型表达。因此,已经进行了关于在原代Treg中维持FOXP3表达以及在原代Tconv或大量CD4单阳性(SP)T细胞中诱导FOXP3表达的研究,并且已经报道了具有维持和诱导FOXP3表达的活性的化合物、细胞因子和其组合。此类化合物、细胞因子和其组合包括作为CDK8/19抑制剂的AS2863619(NPL 2)、作为mTOR抑制剂的雷帕霉素(rapamycin)(NPL 3、NPL 4、NPL 5和NPL 6)、TNFR2激动剂抗体(NPL 5、NPL 7和NPL 8)、细胞因子TGF-β(NPL 2和NPL 6)等。

技术问题
本发明的一个目的是提供一种用于生产调节性T细胞的方法,所述方法可高效地生产调节性T细胞。
问题的解决方案
作为实现上述目的的广泛研究的结果,本发明人发现可通过将包含(a)Foxp3基因的CNS1、CNS2和CNS3、(b)启动子和(c)FOXP3的cDNA的表达构建体引入能够分化为调节性T细胞的细胞(特别是iPS细胞)中并且诱导所获得的细胞分化诱导为调节性T细胞来高效地生产调节性T细胞。
基于此发现进行进一步检查后完成了本发明。本发明提供了下述的一种用于生产调节性T细胞的方法、iPS细胞、调节性T细胞、药物组合物等。
[1]一种用于生产调节性T细胞的方法,所述方法包括:
步骤(1)将其中被引入表达构建体的能够分化为调节性T细胞的细胞分化为调节性T细胞,所述表达构建体包含:
(a)Foxp3基因的保守非编码序列(CNS)1、CNS2和CNS3;
(b)启动子;以及
(c)编码FOXP3的核酸。
[2]根据[1]所述的方法,其中能够分化为调节性T细胞的所述细胞是多能干细胞、造血干细胞、造血内皮细胞、祖T细胞或中胚层祖细胞。

此类细胞表面标志物可使用对细胞表面标志物具有特异性的抗体通过免疫学测定诸如ELISA、免疫染色和流式细胞术来检测。
在本说明书中,术语“表达”被定义为由细胞内启动子驱动的指定核苷酸序列的转录和/或翻译。
在本说明书中,“多能干细胞”是指胚胎干细胞(ES细胞)和具有类似多能性的细胞,即具有分化为活体的各种组织(内胚层、中胚层和外胚层)的潜力的细胞。具有与ES细胞相同的多能性的细胞的实例包括诱导性多能干细胞(在本说明书中也称为“iPS细胞”)。
在本说明书中,“造血干细胞(HSC)”是能够分化为血细胞的多能干细胞。造血干细胞主要存在于人类活体的骨髓中,并且分化为白细胞(中性粒细胞、嗜酸性粒细胞、嗜碱性粒细胞、淋巴细胞、单核细胞和巨噬细胞)、红细胞、血小板、肥大细胞、树突状细胞等。在本说明书中,造血干细胞(HSC)可以是CD34阳性的并且是CD7阴性的(CD34+/CD7-)。在本说明书中,短语“CD34+/CD7-”等中使用的“/”意指“和”。
在本说明书中,“造血内皮细胞(HEC)”是表达CD34但不表达CD43、CD184和CD73的细胞(CD34+/CD43-/CD184-/CD73-)(CD34+/CD43-/CD184-/CD73-细胞不表达CD7,因此也被称为“CD34+/CD7-/CD43-/CD184-/CD73-细胞”)。
在本说明书中,“祖T细胞(proT)”是指在人类活体内由造血干细胞分化为CD3阳性T细胞的过程中产生的造血细胞,并且是CD34和CD7(CD34+/CD7+细胞)阳性的。此外,在本说明书中,proT可以是CD43、CD1a和/或CD116阴性的。
在本说明书中,“中胚层祖细胞”是指例如表达选自由T(与Brachyury同义)、KDR、FOXF1、FLK1、BMP4、MOX1和SDF1组成的组的至少一种标记基因的细胞。中胚层祖细胞与中胚层细胞没有区别。上述标记基因的表达较弱的细胞可称为“中胚层祖细胞”。
在本说明书中,“调节性T细胞”是指当经由T细胞受体刺激时具有抑制效应T细胞激活的能力并且负责抑制免疫反应(免疫耐受)的T细胞。调节性T细胞通常是CD25+/FOXP3+细胞,并且其中存在CD4+或CD8+细胞。在本发明中,调节性T细胞可以是CD4+细胞(即,CD4+/CD25+/FOXP3+)或CD8+细胞(即,CD8+/CD25+/FOXP3+),并且更优选是CD4+细胞。此外,已知转录因子FOXP3是CD4+/CD25+调节性T细胞的主调节因子。
在本发明中,造血干细胞、造血内皮细胞、祖T细胞和中胚层祖细胞可以是从生物组织(诸如骨髓、脐带血和血液)中分离的细胞,或者可以是衍生自多能干细胞(诸如ES细胞和iPS细胞)的细胞。
在本说明书中,“CD4CD8双阳性T细胞”是指T细胞中表面抗原CD4和CD8均呈阳性(CD8+/CD4+)的细胞。由于T细胞可通过表面抗原CD3和CD45呈阳性来鉴定,因此CD4CD8双阳性T细胞可鉴定为CD4、CD8、CD3和CD45阳性的细胞。
在本说明书中,“CD4CD8双阴性T细胞”是指T细胞中表面抗原CD4和CD8均呈阴性(CD8-/CD4-)的细胞。由于T细胞可通过表面抗原CD3和CD45呈阳性来鉴定,因此CD4CD8双阴性T细胞可鉴定为CD4和CD8阳性的并且CD3和CD45阳性的细胞。
就治疗应用而言,本发明中使用的各种细胞优选是通过良好生产规范(GMP)认证的细胞。
多能干细胞的实例包括胚胎干细胞(ES细胞)、诱导性多能干细胞(iPS细胞)、胚胎癌细胞(EC细胞)、胚胎生殖干细胞(EG细胞)和Muse细胞;并且优选iPS细胞(更优选是人iPS细胞)。当多能干细胞是ES细胞或衍生自人胚胎的任何细胞时,可通过破坏胚胎或不破坏胚胎来产生细胞,并且优选不破坏胚胎而产生的细胞。
关于“ES细胞”,可使用Ingenious Targeting Laboratory、RIKEN等建立的各种小鼠ES细胞株作为小鼠ES细胞,并且可使用威斯康星大学(the University of Wisconsin)、NIH、RIKEN、京都大学(Kyoto University)、国家儿童健康与发展中心(National Centerfor Child Health and Development)、Cellartis等建立的各种人ES细胞株作为人ES细胞。人ES细胞株的可用实例包括ESI Bio提供的CHB-1至CHB-12株、RUES1株、RUES2株、HUES1至HUES28株等;WiCell Research提供的H1株、H9株等;以及RIKEN提供的KhES-1株、KhES-2株、KhES-3株、KhES-4株、KhES-5株、SSES1株、SSES2株、SSES3株等。

作为诱导性多能干细胞株,可使用由NIH、RIKEN、京都大学等建立的各种iPS细胞株。人iPS细胞株的实例包括HiPS-RIKEN-1A株、HiPS-RIKEN-2A株、HiPS-RIKEN-12A株和Nips-B2株(RIKEN);Ff-I01s04株、QHJI株、RWMH株、DRXT株、RJWI株、YZWJ株、ILCL株、GLKV株、253G1株、201B7株、409B2株、454E2株、606A1株、610B1株和648A1株(京都大学)等。
“核酸”可以是通过聚合核苷酸和与核苷酸具有相同功能的分子而获得的任何分子。实例包括作为核糖核苷酸聚合物的RNA、作为脱氧核糖核苷酸聚合物的DNA、作为核糖核苷酸和脱氧核糖核苷酸的混合物的聚合物以及含有核苷酸类似物的核苷酸聚合物。核酸还可以是含有核酸衍生物的核苷酸聚合物。核酸可以是单链核酸或双链核酸。双链核酸包括其中一条链在严格条件下与另一条链杂交的双链核酸。
核苷酸类似物可以是任何分子,只要它是通过修饰核糖核苷酸、脱氧核糖核苷酸、RNA或DNA而获得的与RNA或DNA相比改进核酸酶抗性、稳定化、增加与互补链核酸的亲和力、增强细胞渗透性或可视化的分子即可。核苷酸类似物可以是天然存在的分子或非天然分子。实例包括糖修饰的核苷酸类似物(例如,2'-O-甲基核糖取代的核苷酸类似物、2'-O-丙基核糖取代的核苷酸类似物、2'-甲氧基乙氧基核糖取代的核苷酸类似物、2'-O-甲氧基乙基核糖取代的核苷酸类似物、2'-O-[2-(胍鎓)乙基]核糖取代的核苷酸类似物、2'-氟核糖取代的核苷酸类似物、桥接核酸(BNA)、锁核酸(LNA)、乙烯桥接核酸(ENA)、肽核酸(PNA)、氧肽核酸(OPNA)和肽核糖核酸(PRNA))、磷酸二酯键修饰的核苷酸类似物(例如,硫代磷酸酯键取代的核苷酸类似物和N3'-P5'氨基磷酸酯键取代的核苷酸类似物)等。
核酸衍生物可以是任何分子,只要它是通过向核酸添加另一种化学物质而获得的与核酸相比改进核酸酶抗性、稳定化、增加与互补链核酸的亲和力、增强细胞渗透性或可视化的分子即可。具体实例包括5'-多胺-加合物衍生物、胆固醇-加合物衍生物、类固醇-加合物衍生物、胆汁酸-加合物衍生物、维生素-加合物衍生物、Cy5-加合物衍生物、Cy3-加合物衍生物、6-FAM-加合物衍生物、生物素-加合物衍生物等。
本发明的用于生产调节性T细胞的方法(在本说明书中也简称为“本发明的生产方法”)特征性地包括以下步骤:
(1)将其中被引入表达构建体的能够分化为调节性T细胞的细胞分化为调节性T细胞,所述表达构建体(在本说明书中也称为“本发明的表达构建体”、“表达构建体”、“CNS-Foxp3构建体”或“构建体”)包含:
(a)Foxp3基因的保守非编码序列(CNS)1、CNS2和CNS3;
(b)启动子;以及
(c)编码FOXP3的核酸。
本发明的表达构建体用于表达FOXP3,并且包含(a)Foxp3基因的保守非编码序列1(CNS1)、保守非编码序列2(CNS2)和保守非编码序列3(CNS3),(b)启动子,以及(c)编码FOXP3的核酸。通过将这种表达构建体引入能够分化为调节性T细胞的细胞中,可高效地生产调节性T细胞。表达构建体没有特别限制,只要其可表达FOXP3即可。除了上文提及的(a)、(b)和(c)之外,表达构建体优选含有终止子、聚腺苷酸化信号、Foxp3 3’UTR等,并且更优选是在细胞中发挥作用的被引入这些细胞中的表达构建体。
人Foxp3基因的碱基序列注册为RefSeq登录号NM_001114377(SEQ ID No.1)和NM_014009(SEQ ID No.2),并且其氨基酸序列也注册为RefSeq登录号NP_001107849(SEQ IDNo.3)和NP_054728(SEQ ID No.4)。这些RefSeq ID在NCBI网站上注册。在本发明中,上述基因还包括具有上文提及的数据库中注册的碱基序列的基因以外的基因的简并产物和变体。期望的变体是编码具有与由上述氨基酸序列组成的蛋白质等同的生物活性的蛋白质的那些变体。变体的实例包括FOXP3变体,其具有与天然氨基酸序列具有80%或更多、85%或更多、90%或更多、91%或更多、92%或更多、93%或更多、94%或更多、95%或更多、96%或更多、97%或更多、98%或更多、或99%或更多的同一性的氨基酸序列。在本说明书中,FOXP3是指蛋白质,并且Foxp3是指基因;然而,当这种解释适当时,FOXP3和Foxp3分别是指基因和蛋白质。
编码FOXP3的核酸没有特别限制,只要其是编码FOXP3蛋白的核酸即可,并且优选是FOXP3的cDNA。
本发明中使用的CNS1、CNS2和CNS3均衍生自Foxp3基因。CNS1、CNS2和CNS3是Foxp3增强子元件。本发明中使用的启动子没有特别限制,并且实例包括CAG启动子、泛素基因启动子、Foxp3基因启动子、EF1α启动子、SRα启动子、SV40启动子、LTR启动子、巨细胞病毒(CMV)启动子、劳斯肉瘤病毒(RSV)启动子、莫洛尼鼠白血病病毒(MoMuLV)LTR、单纯疱疹病毒胸苷激酶(HSV-TK)启动子等;并且优选是Foxp3基因启动子。人Foxp3基因的CNS1、CNS2、CNS3和启动子的碱基序列的优选实例包括分别由SEQ ID No.5至8表示的碱基序列。即使启动子、CNS1、CNS2和CNS3不具有上述碱基序列,启动子、CNS1、CNS2和CNS3也包括变体。期望的变体是具有与由上述碱基序列组成的启动子、CNS1、CNS2和CNS3等同的生物活性的那些变体。变体的实例包括具有碱基序列的那些变体,所述碱基序列与天然碱基序列具有80%或更多、85%或更多、90%或更多、91%或更多、92%或更多、93%或更多、94%或更多、95%或更多、96%或更多、97%或更多、98%或更多、或99%或更多的同一性。
表达构建体中的启动子、CNS1、CNS2、CNS3和编码FOXP3的核酸的排列(顺序)没有特别限制。优选地,CNS1、CNS2和CNS3位于启动子的上游;并且更优选地从5'末端侧为CNS1、CNS2、CNS3、启动子和编码FOXP3的核酸的顺序。
其中被引入上述表达构建体的能够分化为调节性T细胞的细胞没有特别限制,只要其能够分化为调节性T细胞即可。实例包括多能干细胞、造血干细胞、造血内皮细胞、祖T细胞、中胚层祖细胞等;优选多能干细胞;并且更优选iPS细胞。
能够分化为调节性T细胞的细胞可衍生自人或人以外的哺乳动物(非人哺乳动物),并且优选人衍生的细胞。非人哺乳动物的实例包括小鼠、大鼠、仓鼠、豚鼠、兔、狗、猫、猪、牛、马、羊和猴。
用于将表达构建体引入能够分化为调节性T细胞的细胞中的手段没有特别限制,并且可使用各种已知或通用的手段。通常,使用表达载体将表达构建体引入能够分化为调节性T细胞的细胞中,并进行表达。表达载体可以是线性的或环状的,并且可以是非病毒载体(诸如质粒)、病毒载体或转座子载体。
用于将表达载体引入能够分化为调节性T细胞的细胞中的手段可根据实施方案进行适当的调整。例如,可通过已知的方法,诸如病毒感染法、磷酸钙法、脂转染法、显微注射法或电穿孔法,将表达载体引入能够分化为调节性T细胞的细胞中。表达载体可通过已知的手段或适当地使用可商购获得的试剂盒(根据其说明书)制备成适合在每个方法中使用的形式。
可通过病毒感染法将表达载体引入能够分化为调节性T细胞的细胞中。病毒载体的实例包括逆转录病毒载体、慢病毒载体、腺病毒载体和腺相关病毒载体。当使用这些病毒载体时,可使用对应的可商购获得的试剂盒将含有每种病毒的表达构建体和包装载体(质粒)的载体转染到宿主细胞中以产生重组病毒,然后可用所获得的重组病毒感染能够分化为调节性T细胞的细胞。
除了表达构建体之外,如果需要的话,表达载体还可含有诸如核定位信号(NLS)和多克隆位点(MCS)的序列。表达载体还可含有编码“功能基因”的核酸(碱基序列),所述基因诸如报告基因(例如,编码各种颜色荧光蛋白的基因)、药物选择基因(例如,卡那霉素(kanamycin)抗性基因、氨苄青霉素(ampicillin)抗性基因和嘌呤霉素(puromycin)抗性基因)以及自杀基因(例如,编码以下的基因:白喉A毒素、单纯疱疹病毒胸苷激酶(HSV-TK)、羧肽酶G2(CPG2)、羧酸酯酶(CA)、胞嘧啶脱氨酶(CD)、细胞色素P450(cyt-450)、脱氧胞苷激酶(dCK)、硝基还原酶(NR)、嘌呤核苷磷酸化酶(PNP)、胸苷磷酸化酶(TP)、水痘带状疱疹病毒(varicella zoster virus)胸苷激酶(VZV-TK)、黄嘌呤-鸟嘌呤磷酸核糖转移酶(XGPRT)、诱导型半胱天冬酶9等)。
本发明的生产方法还可包括(2)将表达构建体引入能够分化为调节性T细胞的所述细胞中,所述表达构建体包含:
(a)Foxp3基因的CNS1、CNS2和CNS3;
(b)启动子;以及
(c)编码FOXP3的核酸。
用于将表达构建体引入能够分化为调节性T细胞的细胞中的方法的实例包括上文提及的那些方法。
可根据已知的方法诱导其中被引入表达构建体的能够分化为调节性T细胞的细胞分化为调节性T细胞。此类方法的实例包括人工胸腺类器官(ATO)法(参见WO2017/075389等)。ATO法包括培养三维细胞聚集体,所述三维细胞聚集体含有能够分化为调节性T细胞的细胞或由其分化诱导的细胞以及表达Notch配体的基质细胞,由此可高效地诱导分化为调节性T细胞。ATO法是一种已知方法,并且可参考WO2017/075389等的公开内容进行。其他方法包括WO2021/092581中公开的分化诱导法(例如与MS5-DLL1/4基质细胞、OP9或OP9-DLL1基质细胞、或EpCAM-CD56+基质细胞共培养)。表达Notch配体的基质细胞优选是其中强制表达人DLL4蛋白的小鼠衍生的基质细胞株MS5。
能够分化为调节性T细胞的细胞或由其分化诱导的细胞是指“其中被引入表达构建体的能够分化为调节性T细胞的细胞”或“由其中被引入表达构建体的能够分化为调节性T细胞的细胞分化诱导而来的细胞”。可对其中被引入表达构建体的能够分化为调节性T细胞的细胞(即,待引入表达构建体的细胞)进行,或对由其中被引入表达构建体的能够分化为调节性T细胞的细胞分化诱导而来的细胞(即,待引入表达构建体的细胞)进行ATO法。
进行ATO方法的能够分化为调节性T细胞的细胞或由其分化诱导的细胞没有特别限制。实例包括CD34+细胞、中胚层祖细胞、CD4CD8双阴性T细胞、CD4CD8双阳性T细胞等;优选CD34+细胞或中胚层祖细胞;更优选CD34+/CD7-细胞或中胚层祖细胞;甚至更优选造血内皮细胞、造血干细胞和中胚层祖细胞;并且特别优选造血内皮细胞。经受ATO法的细胞更期望地是由能够分化为调节性T细胞的细胞分化诱导而来的细胞。CD34+细胞是表达CD34的细胞(CD34+),并且特别是表达CD34但不表达CD7的细胞(CD34+/CD7-)。CD34+细胞的实例包括造血内皮细胞和造血干细胞。
特别地,期望使用多能干细胞(特别是iPS细胞)作为能够分化为调节性T细胞、诱导细胞分化为CD34+细胞(特别是造血内皮细胞)的细胞,并且对CD34+细胞(特别是造血内皮细胞)进行ATO法以分化为调节性T细胞。
多能干细胞分化为CD34+细胞可根据已知的方法来诱导。当多能干细胞是iPS细胞时,可例如通过WO2017/221975、WO2018/135646和Cell Reports 2(2012)1722-1735中公开的方法诱导造血祖细胞的分化,从而产生CD34+细胞。当对造血内皮细胞进行ATO法时,可对含有造血内皮细胞的CD34+细胞进行ATO法(即在进行ATO法之前不分离造血内皮细胞),或也可对根据已知方法(例如,流式细胞术或磁性细胞分离法)分离的造血内皮细胞进行ATO法。
Notch配体没有特别限制,并且包括WO2017/075389中公开的规范Notch配体和非规范Notch配体。规范Notch配体的实例包括DLL4(δ样配体4)、DLL1(δ样配体1)、JAG1(Jagged 1)、JAG2(Jagged 2)等。他们可单独使用或以两种或更多种的组合使用。非规范Notch配体的实例包括接触蛋白-1(Contactin-1)、NOV/CCN3、接触蛋白-6、骨膜蛋白/OSF-2、DLK2/EGFL9、Pref-1/DLK1/FA1、DNER、血小板反应蛋白-2(Thrombospondin-2)、MAGP-1/MFAP2、血小板反应蛋白-3、MAGP-2/MFAP5、血小板反应蛋白-4和纺锤蛋白-1(Netrin-1)。优选使用人DLL4作为Notch配体。
三维细胞聚集体可例如通过对能够分化为调节性T细胞的细胞或由其分化诱导的细胞以及表达Notch配体的基质细胞进行离心来形成。基质细胞的实例包括小鼠基质细胞株、人基质细胞株等。优选将用于表达Notch配体的核酸引入基质细胞中。基质细胞与能够分化为调节性T细胞的细胞或由其分化诱导的细胞的比为例如100:1至1:100、20:1至1:20或10:1至4:1,并且优选为10:1至4:1。
用于培养三维细胞聚集体的培养基没有特别限制,并且实例包括无血清培养基,并且特别是含有胰岛素(还含有生物素、转铁蛋白和白蛋白)的无血清培养基。
用于培养三维细胞聚集体的基础培养基的实例包括AIMV、X-VIVO-15、NeuroBasal、EGM2、TeSR,BME、BGJb、CMRL1066、Glasgow MEM、经改良MEM锌选择(ImprovedMEM Zinc Option)、IMDM、199培养基、Eagle MEM、αMEM、DMEM、Ham、RPMI-1640、Fischer培养基等。这些培养基可单独使用或者作为两种或更多种的混合物使用。另外,例如,可合适地添加WO2017/075389中公开的培养基组分。
作为用于培养三维细胞聚集体的培养条件,例如,培养温度为约20℃至40℃,CO2浓度为约2%至10%,氧浓度约1%至20%,并且培养时间例如为约1至100天。培养开始时的细胞密度可以是例如1.0×104至1.0×1010个细胞/mL。
本发明的生产方法中使用的培养基、培养条件等可以是适合待培养的细胞类型的那些。
用作这种培养基的基础培养基没有特别限制,只要其可用于培养动物细胞即可。实例包括上文提及的那些。
培养基可含有血清,或可不含血清。培养基还可含有血清替代物(例如,白蛋白、转铁蛋白、敲除血清替代物(Knockout Serum Replacement,KSR)、脂肪酸、胰岛素、胶原前体、微量元素、2-巯基乙醇、3'-硫代甘油和ITS-补充剂)。血清替代物可单独使用或以两种或更多种的组合使用。
培养基还可含有以下物质中的一种或多种物质:脂类、氨基酸(非必需氨基酸等)、L-谷氨酰胺、维生素、生长因子、细胞因子、抗生素、抗氧化剂、丙酮酸、缓冲剂、无机盐等。作为培养基,期望使用不含有具有未知组分的物质(诸如血清)的化学成分确定的培养基,因为可减少批次间的培养基差异,并且可制备质量稳定的细胞。
培养基的pH通常为7.0至7.8,并且优选为7.2至7.6。为了防止使用前的污染,优选通过诸如过滤、UV照射、加热灭菌或辐射的方法对培养基进行灭菌。培养是在饲养细胞存在或不存在的情况下进行。培养条件没有特别限制,并且可使用通常用于细胞培养的条件。在培养中,可根据需要执行多次传代以获得期望量的细胞,或可执行培养基的添加和替换。培养容器没有特别限制,并且可从板、皿、培养皿、烧瓶、袋、瓶、槽(培养槽)、生物反应器等中适当地选择。
本发明的生产方法还可包括分离获得的调节性T细胞以浓缩调节性T细胞。调节性T细胞可通过已知方法诸如使用流式细胞术的方法或磁性细胞分离方法分离。
本发明的生产方法还可包括:
步骤(3A)在选自由CDK8和/或CDK19抑制剂、TNFR2激动剂、mTOR抑制剂和TGF-βR激动剂组成的组的至少一种物质的存在下培养调节性T细胞。

肿瘤坏死因子受体2(TNFR2)激动剂被定义为可与TNFR2结合以激活TNFR2信号传导的物质。实例包括抗体(例如,抗TNFR2抗体)、肽、低分子量化合物、蛋白质等。TNFR2激动剂抗体的实例包括与TNFR2结合的单克隆抗体,诸如克隆MR2-1(Hycult Biotech)和克隆MAB2261(R&D Systems)。TNFR2激动剂可以是作为激动剂的仅与TNFR2结合的TNF-α突变蛋白。TNFR2激动剂可单独使用或以两种或更多种的组合使用。
TNFR2激动剂优选为TNFR2激动剂抗体。抗体可以是其功能片段。功能片段的实例包括Fd、Fv、Fab、F(ab’)、F(ab)2、F(ab’)2、单链Fv(scFv)、双抗体、三抗体、四抗体和微型抗体。抗体的实例包括衍生自动物(诸如小鼠、大鼠、牛、兔、山羊、绵羊和豚鼠)的那些抗体。抗体的同种型没有特别限制。同种型的实例包括IgG(IgG1、IgG2、IgG3和IgG4)、IgA、IgD、IgE和IgM。抗体可以是单克隆抗体或多克隆抗体,并且优选是单克隆抗体。此外,抗体可以是人源化抗体、嵌合抗体、多特异性抗体(例如,双特异性抗体)等。抗体可通过已知的方法来产生。例如,可通过以下方式来产生抗体:构建含有编码所述抗体的核酸的表达载体,以及培养其中被引入所述核酸的转化体,或者培养产生所述抗体的杂交瘤。
转化生长因子-β受体(TGF-βR)激动剂被定义为可与TGF-βR结合以激活TGF-βR信号传导的物质。TGF-βR激动剂的实例包括属于TGF-β超家族的因子。TGF-β超家族包括TGF-β家族、激活素家族和骨形态发生蛋白(BMP)家族。属于TGF-β超家族的因子的实例包括TGF-β(TGF-β1、TGF-β2和TGF-β3)、激活素A、激活素B、GDF-8、GDF-11等。TGF-βR激动剂优选是TGF-β。TGF-βR激动剂可单独使用或以两种或更多种的组合使用。
此外,CDK8和/或CDK19抑制剂、TNFR2激动剂、mTOR抑制剂和TGF-βR激动剂可以游离形式或以盐形式使用。盐的实例包括与无机碱形成的盐,诸如钠盐、镁盐、钾盐、钙盐和铝盐;与有机碱形成的盐,诸如甲胺盐、乙胺盐和乙醇胺盐;与碱性氨基酸(诸如赖氨酸、鸟氨酸和精氨酸)形成的盐;以及铵盐。这些盐可以是酸加成盐。此类盐的具体实例包括与以下酸形成的酸加成盐:矿物酸,诸如盐酸、氢溴酸、氢碘酸、硫酸、硝酸和磷酸;有机酸,诸如甲酸、乙酸、丙酸、草酸、丙二酸、苹果酸、酒石酸、富马酸、琥珀酸、乳酸、马来酸、柠檬酸、甲磺酸和乙磺酸;以及酸性氨基酸,诸如天冬氨酸和谷氨酸。CDK8和/或CDK19抑制剂、TNFR2激动剂、mTOR抑制剂和TGF-βR激动剂还包括水合物、溶剂化物、多晶型物等。
当CDK8和/或CDK19抑制剂、TNFR2激动剂、mTOR抑制剂和TGF-βR激动剂含有不对称碳时,这些包括R-异构体、S-异构体以及含有任意比的两种异构体的混合物(例如,外消旋体)。
培养基中的CDK8和/或CDK19抑制剂的浓度没有特别限制,只要可广泛培养调节性T细胞即可,并且根据所使用的CDK8和/或CDK19抑制剂的类型等合适地调整。CDK8和/或CDK19抑制剂的浓度例如为0.1至300nM,并且优选为0.5至150nM。
培养基中的mTOR抑制剂的浓度没有特别限制,只要可广泛培养调节性T细胞即可,并且根据所使用的mTOR抑制剂的类型等合适地调整。mTOR抑制剂的浓度例如为1至3000nM,优选5至1000nM,并且更优选10至100nM。
培养基中的TNFR2激动剂的浓度没有特别限制,只要可广泛培养调节性T细胞即可,并且根据所使用的TNFR2激动剂的类型等合适地调整。TNFR2激动剂的浓度例如为0.0001至100μg/mL,优选0.01至10μg/mL,并且更优选1至5μg/mL。
培养基中的TGF-βR激动剂的浓度没有特别限制,只要可广泛培养调节性T细胞即可,并且根据所使用的TGF-βR激动剂的类型等合适地调整。TGF-βR激动剂的浓度例如为0.1至100ng/mL,并且优选1至50ng/mL。
步骤(3)或(3A)的培养中使用的培养基使用用于培养动物细胞的培养基作为基础培养基,并且含有上述CDK8和/或CDK19抑制剂、TNFR2激动剂、mTOR抑制剂和TGF-βR激动剂。培养基和培养条件包括上文提及的那些。培养时间也没有特别限制,并且可由本领域技术人员在监测调节性T细胞的数量的同时合适地确定。培养时间例如为10天或更久,优选1周或更久,并且更优选2周或更久。培养时间的上限没有特别限制,并且例如为35天或更短,优选28天或更短,并且更优选为21天或更短。
可将外来基因引入通过本发明的生产方法获得的调节性T细胞中,以进一步生产其中被引入外来基因的调节性T细胞。
“外来基因”是从外部引入以在调节性T细胞中表达期望蛋白质的基因,并且可根据调节性T细胞的用途合适地选择。
外来基因可以是例如用于表达嵌合抗原受体(CAR)的基因,并且还可含有用于表达细胞因子和/或趋化因子的基因。与一般或已知的CAR一样,由调节性T细胞表达的CAR基本上被配置成使得以下位点处的肽根据需要经由间隔子连接:(i)识别癌细胞的细胞表面抗原的抗原识别位点(例如,单链抗体),(ii)跨膜区,和(iii)诱导T细胞激活的信号转导区。外来基因还可以是例如用于表达外源T细胞受体(TCR)的基因。外源TCR意指对于其中待引入编码外源TCR的核酸的T细胞来说是外源的。外源TCR的氨基酸序列可与T细胞的内源TCR的氨基酸序列相同或不同。可引入一种或两种或更多种外来基因(例如,CAR和外源TCR)。
用于将外来基因引入调节性T细胞中的手段的实例包括上文提及的方法。
通过本发明的生产方法生产的调节性T细胞可用于治疗具有免疫反应异常增强的动物(特别是人)。例如,调节性T细胞可用于治疗和预防免疫失调、多内分泌病、肠病、X连锁(IPEX)综合征、移植物抗宿主病(GVHD)、器官移植排斥、自身免疫性疾病、炎性疾病、过敏性疾病(花粉热、哮喘、特应性皮炎、湿疹、食物过敏、食物超敏、荨麻疹、过敏性鼻炎、过敏性结膜炎和药物过敏)等,但不限于此。通过本发明的生产方法生产的调节性T细胞可用于自体移植或同种异体移植。此外,调节性T细胞可与其他药物组合使用。
当进行这种细胞疗法时,从不发生排斥的观点来看,从中分离细胞以产生调节性T细胞的对象优选具有与待施用调节性T细胞的对象相同的HLA类型,并且更优选与待施用调节性T细胞的对象相同。
本发明可生产包含调节性T细胞的药物组合物(在下文中也称为“本发明的药物组合物”)。本发明的药物组合物优选根据已知手段(例如,日本药典(JapanesePharmacopoeia)中描述的方法)通过将有效量的调节性T细胞与药学上可接受的载剂混合而生产为肠胃外制剂。本发明的药物组合物优选生产为肠胃外制剂,诸如注射剂、混悬剂或输注剂。肠胃外施用方法的实例包括静脉内、动脉内、肌肉内、腹膜内或皮下施用。药学上可接受的载剂的实例包括溶剂、基质、稀释剂、赋形剂、舒缓剂、缓冲剂、防腐剂、稳定剂、混悬剂、等渗剂、表面活性剂、增溶剂等。
本发明的药物组合物的剂量可根据各种条件(诸如患者的体重、年龄、性别和症状)合适地确定。一般来说,施用本发明的药物组合物使得对于体重为60kg的对象每次施用的细胞数通常为1×106至1×1010个细胞,优选1×107至1×109个细胞,并且更优选5×107至5×108个细胞。本发明的药物组合物可施用一次或多次。本发明的药物组合物可具有适合于肠胃外施用的已知形式,例如注射剂或输注剂。此外,本发明的药物组合物可含有生理盐水、磷酸盐缓冲盐水(PBS)、培养基等以便稳定地维持细胞。培养基的实例包括RPMI、AIM-V、X-VIVO10和其他培养基,但不限于此。另外,出于稳定化的目的,可将药学上可接受的载剂(例如,人血清白蛋白)、防腐剂等添加到药物组合物中。本发明的药物组合物适用于哺乳动物,包括人。
如本说明书和权利要求书中所用,除非上下文另有要求,否则单数术语应包括复数并且复数术语应包括单数。因此,除非另外明确指出,否则单数冠词(例如,在英语的情况下,“一个/种(a/an)”、“所述(the)”等)也应当被理解为涵盖其复数形式的概念。
实施例
下文提供实施例以便更详细地解释本发明。然而,本发明不限于这些实施例。
(1)CNS-Foxp3构建体的产生
通过将GenBank注册的序列(MK012431)中的mStrawberry蛋白质序列变更为dTomato序列而获得的DNA序列引入用于产生第三代慢病毒载体的转移质粒中来设计并合成质粒序列。将收到的质粒与包装质粒和包膜质粒一起转染到HEK293谱系细胞中以产生慢病毒载体。收集含有所产生的慢病毒载体的上清液,然后通过高速离心法浓缩以获得待引入iPS细胞中的慢病毒载体。
(2)构建体向iPS细胞中的引入和引入构建体的iPS细胞向HEC的分化
所使用的含有造血内皮细胞的细胞群体是一种浮游细胞群体,其是从由京都大学iPS细胞研究和应用中心(Center for iPS Cell Research and Application,KyotoUniversity)提供的iPS细胞(Ff-I01s04株:衍生自健康外周血单核细胞)通过已知方法(例如,Cell Reports2(2012)1722-1735和WO2017/221975中公开的方法)分化而来。
具体而言,将Ff-I01s04株以1×104个细胞/孔接种在24孔板中,并且以10μg/mL的最终浓度添加鱼精蛋白。然后,直接添加被引入CNS-Foxp3基因的慢病毒溶液以产生病毒感染的iPS细胞。将病毒感染的Ff-I01s04株以1×106个细胞/孔接种在已进行超低粘附处理的6孔板(Corning)中(第0天)。将50ng/ml BMP4(R&D systems)、50ng/ml bFGF(FUJIFILMWako Pure Chemical Corporation)、50ng/ml VEGF(R&D systems)和2μM SB431542(FUJIFILM Wako Pure Chemical Corporation)添加到EB培养基(StemPro34(Gibco)中,所述培养基补充有10μg/ml人胰岛素、5.5μg/ml人转铁蛋白、5ng/ml亚硒酸钠(ITS,Gibco)、2mM L-谷氨酰胺(Sigma-Aldrich)、45mMα-单硫代甘油(Nacalai Tesque,Inc.)和50μg/mlL-抗坏血酸2-磷酸(Sigma-Aldrich)),并且将细胞在低氧条件(5%O2)下培养4天(第4天)。随后,将细胞在补充有50ng/ml bFGF、50ng/ml VEGF和50ng/ml SCF(R&D systems)的培养基中进一步培养4天(第8天),从而获得含有HEC的细胞群体。使用下表1中的抗体组对含有HEC的浮游细胞群体进行染色。

通过流式细胞术方法确认,获得的细胞群体含有约15%的HEC(CD34+/CD43-/CD73-/CD184-细胞)。作为对照,以与上述相同的方式获得含有HEC的细胞群体(未转导),不同之处在于使用其中未引入(1)中产生的表达构建体的iPS细胞(Ff-I01s04株)。
(3)从HEC向Treg的分化诱导
参考ATO法(WO2017/075389等中公开的方法),使上述(2)中获得的含有HEC的细胞群体分化为Treg(CD25+/FOXP3+)。
具体而言,使用其中强制表达人DLL 4蛋白的小鼠衍生的基质细胞株MS5作为支撑物,并与上述(2)中产生的引入CNS-Foxp3的人iPS细胞衍生的HEC以4:1的细胞比共培养。使用的培养基是RPMI-1640(FUJIFILM Wako Pure Chemical Corporation),其最终浓度含有2x B27补充剂(Invitrogen)、1x PSG(Sigma-Aldrich)、1x Glutamax(Invitrogen)、5ng/mLIL-7(PeproTech)、5ng/mL FlT3L(PeproTech)和50μg/mL抗坏血酸(Sigma-Aldrich)。使在30mm Millicell(亲水性PTFE,孔径:0.4μm,高度:5mm,Merck Millipore)上共培养的细胞静置在6孔板(TPP)中,并且培养9周,同时每三或四天更换一次培养基。
培养后,收集获得的细胞,并且使用与上述(1)中所示的Foxp3基因的下游组合的dTomato为指标,使用FACS Aria获得靶细胞级分。根据WO2020/032179中公开的方法对其进行扩增,以确保足够数量的细胞,然后使用下表2中的抗体组进行细胞内染色,从而评估人iPS细胞衍生的Treg的诱导。使用流式细胞仪(FACS Aria Fusion,BD Bioscience)确认诱导了CD4+Treg(CD4+/CD25+/FOXP3+)。还确认了与对照(未转导)相比,CNS-Foxp3构建体转导组中以更高的比率诱导Treg(Foxp3转导的iPS细胞中的比率为56.4%,而未转导细胞中为11.2%)(图1)。

(4)iPS-Treg的扩增培养
将上述(3)中获得的从iPS细胞分化为Treg后的细胞在含有10nM雷帕霉素(Merck)和3μg/mL的抗TNFR2抗体(Hycult Biotech,克隆MR2-1)的以下培养基中进行扩增培养。
具体而言,将上述(3)中获得的引入CNS-Foxp3的人iPS细胞衍生的Treg在结合抗CD3抗体(eBioscience)的48孔细胞培养板中培养3天,然后重新接种在24孔G-Rex细胞培养板中,并且继续培养。使用的培养基是α-MEM(Invitrogen),其最终浓度含有FBS(15%,Corning)、L-谷氨酰胺-青霉素-链霉素溶液(1/100,Invitrogen,Sigma-Aldrich)、胰岛素-转铁蛋白-硒补充剂(1/100,Invitrogen)、抗坏血酸2-磷酸(50μg/mL,Sigma-Aldrich)、IL-2(10ng/mL,PeproTech)、IL-7(10ng/mL,PeproTech)、IL-12(50ng/mL,Merck)、IL-15(10ng/mL,PeproTech)、IL-18(50ng/mL,MBL)、IL-21(20ng/mL,PeproTech)和TL-1A(50ng/mL,PeproTech)。前三天,向上述配制物中进一步添加抗CD28抗体(1.5μg/mL,BioLegend)、抗CD30抗体(300ng/mL,R&D systems)和半胱天冬酶抑制剂(10μM,R&D systems)。培养进行9或10天,同时在培养开始后第3天替换培养基,并且此后每三或四天替换一次培养基。每个细胞群体用以下抗体组(Zombie NIR、BV510 CD3、BV421 CD4、PE/Cy7 CD8、APC CD25、FITCFoxp3)染色。在检查TSDR去甲基化量(5)和评价Treg抑制功能(6)时,使用通过在上述扩增培养之后使用FACSAria(Becton Dickinson)纯化CD4+Foxp3+细胞组而获得的细胞群体进行评价。在纯化中,使用以下抗体组进行染色(BV421 CD4、PE/Cy7 CD8、APC CD25)。
图2示出了在含有雷帕霉素和抗TNFR2抗体的培养基中扩增培养后的流式细胞术分析结果。已经确认了在通过在含有雷帕霉素和抗TNFR2抗体的培养基中扩增培养而获得的细胞群体中,引入CNS-Foxp3构建体的iPS细胞衍生的细胞维持Treg的比率高于对照(未转导)(CNS-Foxp3构建体转导组中的比率为84.8%,而未转导组中为10.4%)。此外,在通过在不含有雷帕霉素和TNFR2Ab的培养基中扩增培养而获得的细胞群体中,引入CNS-Foxp3构建体的iPS细胞衍生的细胞维持Treg的比率高于对照(未转导)(CNS-Foxp3构建体转导组中的比率为56.4%,而未转导组中为10.1%)。
(5)Treg特异性去甲基化区域(TSDR)的去甲基化量检查
使用从iPS细胞分化为Treg后的细胞,检查TSDR去甲基化。使用的细胞如下。原代Treg:从人PBMC分离的Treg(CD3+/CD4+/CD8-/CD25+/CD127-)细胞群体;原代非Treg:从人PBMC分离的非Treg(CD3+/CD4+/CD8-/CD25-)细胞群体;iPSC-Treg:从iPS细胞分化诱导而来的Treg(CD3+/CD4+/CD8-/CD25+/Foxp3+)细胞群体;HEC:由iPS细胞诱导的未分化的造血细胞(CD34+/CD43-/CD73-/CD184-)。作为iPSC-Treg,使用通过上述方法(1)至(3)从由CNS-Foxp3构建体转导的iPS细胞分化诱导而来的细胞,并且作为HEC,使用通过方法(1)和(2)分化而未转导CNS-Foxp3构建体的细胞。
具体而言,使用PureLink Genomic DNA迷你试剂盒(Thermo Fisher Scientific)从上述每一种细胞中提取DNA。对于DNA,使用PureQuant Treg测定(Thermo FisherScientific),通过qRCR量化基因组DNA的去甲基化。方案遵循试剂盒的推荐方案。图3示出了结果。在iPSC-Treg中,TSDR去甲基化的比率高于分化前的HEC中的比率(iPSC-Treg中为80%,而HEC中为0%),并且所述比率与原代Treg中的比率一样高(原代Treg中为88%)。
(6)Treg抑制功能评价
使用由上述(3)中获得的CNS-Foxp3构建体转导的iPS细胞分化诱导而来的Treg,在与人PBMC衍生的同种异体T细胞共培养后通过流式细胞术进行分析。
具体而言,通过用CellTrace Violet(Thermo Fisher Scientific)对未处理的PBMC或用衍生自与Treg供体不同的供体的抗CD3抗体处理过的PBMC进行染色来制备靶细胞,并与Treg共培养。使用的培养基是α-MEM(Invitrogen),其最终浓度含有FBS(15%,Corning)、L-谷氨酰胺-青霉素-链霉素溶液(1/100,Invitrogen,Sigma-Aldrich)、胰岛素-转铁蛋白-硒补充剂(1/100,Invitrogen)和抗坏血酸2-磷酸(50μg/mL,Sigma-Aldrich),并且将细胞培养4天。每个细胞群体用以下抗体组(Zombie NIR、PE/Cy7 CD3、FITC HLA-A24)染色。
图4示出了与人PBMC衍生的同种异体T细胞共培养后的流式细胞术分析结果。在与iPS-Treg共培养的组中,与单独培养靶细胞相比,靶细胞的细胞分裂被强烈抑制(在Treg:靶标=2:1的条件下,23.5%的靶细胞被诱导细胞分裂,而仅培养靶细胞时为74.9%)。此外,其抑制功能与原代Treg类似或更强(与原代Treg共培养时,47.5%的靶细胞被诱导细胞分裂)。

1.一种用于生产调节性T细胞的方法,所述方法包括:
步骤(1)将其中被引入表达构建体的能够分化为调节性T细胞的细胞分化为调节性T细胞,所述表达构建体包含:
(a)Foxp3基因的保守非编码序列(CNS)1、CNS2和CNS3;
(b)启动子;以及
(c)编码FOXP3的核酸。
2.根据权利要求1所述的方法,其中能够分化为调节性T细胞的所述细胞是多能干细胞、造血干细胞、造血内皮细胞、祖T细胞或中胚层祖细胞。
3.根据权利要求1所述的方法,其中能够分化为调节性T细胞的所述细胞是iPS细胞。
4.根据权利要求1所述的方法,其中CNS1、CNS2和CNS3位于所述启动子的上游。
5.根据权利要求1所述的方法,其中所述调节性T细胞是CD25+/FOXP3+细胞。
6.根据权利要求1所述的方法,其还包括:
步骤(2)将表达构建体引入能够分化为调节性T细胞的所述细胞中,所述表达构建体包含:
(a)Foxp3基因的CNS1、CNS2和CNS3;
(b)启动子;以及
(c)编码FOXP3的核酸。
7.根据权利要求1所述的方法,其中在步骤(1)中培养三维细胞聚集体,并且所述三维细胞聚集体包含能够分化为调节性T细胞的所述细胞或由其分化诱导的细胞,以及表达Notch配体的基质细胞。
8.根据权利要求7所述的方法,其中能够分化为调节性T细胞的所述细胞或由其分化诱导的细胞是CD34+细胞。
9.根据权利要求7所述的方法,其中能够分化为调节性T细胞的所述细胞或由其分化诱导的细胞是造血内皮细胞。
10.根据权利要求7所述的方法,其中所述Notch配体是选自由DLL4、DLL1、JAG1和JAG2组成的组中的至少一种。
11.一种iPS细胞,其中被引入表达构建体,所述表达构建体包含:
(a)Foxp3基因的CNS1、CNS2和CNS3;
(b)启动子;以及
(c)编码FOXP3的核酸。
12.根据权利要求11所述的iPS细胞,其中CNS1、CNS2和CNS3位于所述启动子的上游。
13.一种调节性T细胞,其通过根据权利要求1所述的方法获得。
14.一种药物组合物,其包含根据权利要求13所述的调节性T细胞。
15.根据权利要求14所述的药物组合物,其用于预防和/或治疗异常增强的免疫反应。
16.一种用于预防和/或治疗异常增强的免疫反应的方法,其包括向有需要的对象施用根据权利要求13所述的调节性T细胞。
17.根据权利要求13所述的调节性T细胞,其用于预防和/或治疗异常增强的免疫反应。
18.根据权利要求13所述的调节性T细胞在生产用于预防和/或治疗异常增强的免疫反应的药物中的用途。
19.根据权利要求1所述的方法,其还包括:
步骤(3)在mTOR抑制剂和TNFR2激动剂的存在下培养所述调节性T细胞。
