The experimental procedure of the present invention, in which no specific conditions are noted in the following examples, is generally followed by routine conditions, such as, for example, sambrook et al, molecular cloning: conditions described in the laboratory Manual (New York: cold Spring Harbor Laboratory Press, 1989) or as recommended by the manufacturer. Example 1
Raw materials: 30 g of kudzuvine root, 15 g of hovenia dulcis thunb, 8g of bupleurum, and 10 g of red paeony root.
The steps are as follows: pulverizing the medicinal materials, placing into a proper decoction container, adding 10 times of water to submerge the medicinal materials, soaking for proper time, heating to boil, decocting for 30min for the first time, decocting for 20min for the second time, separating decoction, decocting medicinal residues for 2-3 times, collecting decoction, mixing decoctions, settling, filtering, concentrating at low temperature, and drying.
Mixing the dry extract powder with sugar powder, dextrin, and hot air drying at 80deg.C to remove small amount of water, making into soft material with 90% ethanol, making into wet granule with extrusion granulator, and drying to obtain granule.
Example 2
Raw materials: 40 g of kudzuvine root, 20 g of hovenia dulcis thunb, 10 g of bupleurum, and 15 g of red paeony root.
The steps are as follows: pulverizing the medicinal materials, placing into a proper decoction container, adding 10 times of water to submerge the medicinal materials, soaking for proper time, heating to boil, decocting for 30 min for the first time, decocting for 20 min for the second time, separating decoction, decocting medicinal residues for 2-3 times, collecting decoction, mixing decoctions, adding appropriate amount of 95% ethanol, monosaccharide slurry and sodium benzoate, filtering, and concentrating to obtain oral liquid.
Example 3
Raw materials: radix Puerariae 20 g, semen Hoveniae 10 g, bupleuri radix 5 g, radix Paeoniae Rubra 5 g.
The steps are as follows: pulverizing the medicinal materials, placing into a proper decoction container, adding 10 times of water to submerge the medicinal materials, soaking for proper time, heating to boil, decocting for 30min for the first time, decocting for 20min for the second time, separating decoction, decocting medicinal residues for 2-3 times, collecting decoction, mixing decoctions, settling, filtering, concentrating at low temperature, and drying.
Pulverizing the dry extract into the finest powder, making into micropill with ethanol as wetting agent, sieving, and drying at 60deg.C to obtain pill.
Example 4
In vitro efficacy test
1 Material
1.1 Animals: a total of 36C 57 mice.
1.2 Drugs and reagents: the Chinese medicinal composition granule (hereinafter referred to as honeydew melon soup) in example 1 has silymarin as positive medicinal component.
2 Method
2.1 The C57 mice were randomly divided into four groups, control group, model group, positive drug group, melon soup group. The three groups, except the control group, established a mouse model of alcoholic liver disease. The alcohol concentration was gradually increased from 1% to 5% for the first week of feeding, 2% every two days, 5% was maintained and feeding continued for 4 weeks. After 4 weeks, the model group, the positive medicine group and the melon soup group are respectively treated as follows: the model group irrigates the stomach with physiological saline at a dose of 60mg/kg per day; the positive medicine group is subjected to gastric lavage according to 50mg/kg per day; the honeydew melon soup group is subjected to stomach irrigation according to 60mg/kg per day. While the stomach was being irrigated, each group was fed with either alcohol feed or control feed.
2.2 Detection: detecting the serum ASL, ALT, TG, TC content of the mice after 4 weeks of gastric lavage; mouse liver oil red O staining, HE staining, sirius red staining.
3 Results
1 Triglyceride (TG) and Total Cholesterol (TC) content
As shown in FIG. 1, the positive drug group and the melon soup group have significantly reduced TG and TC contents compared with the model group, and the difference has statistical significance (P < 0.05).
2 Glutamic pyruvic transaminase (ALT) and glutamic oxaloacetic transaminase (AST) content
As shown in FIG. 2, the positive drug group and the melon soup group showed significantly lower ALT and AST contents than the model group, and the difference was statistically significant (P < 0.05).
3 Liver HE staining
Liver HE staining suggests that the liver tissue of the mice in the model group has remarkable fatty degeneration and inflammation degree, and the melon soup intervention can obviously reduce the fatty degeneration and inflammation degree of the liver tissue and improve the morphology and structure of liver cells (figure 3).
4 Liver visceral oil red O staining
Liver oil red O staining suggests that the liver tissue lipid deposition of mice in the model group is remarkable, and the liver tissue lipid deposition degree is reduced after the intervention of the honeydew melon soup is adopted (figure 4).
5 Liver Tilletia red dyeing
Liver sirius red staining suggests that the liver tissue fibrosis degree of mice in the model group is obviously higher than that of mice in the control group, the positive medicine group and the honeydew group, and the liver fibrosis degree of the mice is effectively relieved after the intervention of the honeydew (figure 5).
In conclusion, the composition provided by the invention has an obvious liver protection effect under the condition of only 4 raw materials, and can effectively reduce liver lipid deposition and liver fibrosis.
