Abstract
为克服现有可溶性微针的机械强度不足,影响给药质量的问题,本发明提供了一种微针制剂,其特征在于,包括以下A组分:聚乙烯醇和聚乙烯吡咯烷酮以及羟乙基纤维素;其中,聚乙烯醇中包括第一聚乙烯醇和第二聚乙烯醇,所述第一聚乙烯醇的粘度为2.4~12.3mpas,所述第二聚乙烯醇的粘度为17.2~42.6mpas;所述聚乙烯吡咯烷酮包括第一聚乙烯吡咯烷酮和第二聚乙烯吡咯烷酮,所述第一聚乙烯吡咯烷酮的分子量为3500~150000,所述第二聚乙烯吡咯烷酮的分子量为210000~1500000。同时,本发明还公开了一种微针贴片的制备方法。本发明提供的微针制剂能够有效提高制备得到的微针的结构强度。
Description
技术领域
本发明属于药物制剂技术领域,具体涉及一种微针制剂及微针贴片的制备方法。
背景技术
可溶性微针由于其较低的生产成本、较好生物稳定性,同时使用方便还能有效控制药物释放曲线,受到了微针市场广泛的关注。不同于实体针和中空针,可溶性微针只需要一次刺入皮肤即可留在皮肤内部并控制药物的释放速度,该速度主要视微针的结构而定,因此,可以通过改变微针的制剂处方来轻松的控制药物释放的速度。
然而,可溶性微针还是具有一定的缺陷,比如可溶性微针采用水溶性分子作为其赋形材料,在使用过程中的机械强度和在储存过程中的物理稳定性,相比其他类型的微针具有本质的差距。现有的可溶性微针制剂存在机械强度不足的问题,导致其在皮肤上施加时的穿刺深度不足,容易在未穿刺至目标深度时发生断裂,进而影响给药量,也不利于维持给药量的稳定性。另外目前可溶解微针生产效率,尤其是脱水效率也存在效率较低,且脱水过程中因为温度过高和脱水时间过长,导致可溶解微针内含的API容易丧失物理化学和生物学活性。因此,可溶解微针的机械强度受脱水的彻底程度深刻影响,脱水条件严苛还是温和,又对可溶解微针的产品质量产生深刻影响。
因此,找到合适可溶解微针的制剂处方,同时找到温和且高效的可溶解微针的脱水方式,对可溶解微针的生产效率和质量控制至关重要。
发明内容
针对现有可溶性微针的机械强度不足,生产效率低,影响给药质量的问题,本发明提供了一种微针制剂及微针贴片的制备方法。
本发明解决上述技术问题所采用的技术方案如下:
一方面,本发明提供了一种微针制剂,包括以下A组分:
聚乙烯醇、聚乙烯吡咯烷酮以及羟乙基纤维素;
其中,所述羟乙基纤维素的分子量为80000~780000,聚乙烯醇中包括第一聚乙烯醇和第二聚乙烯醇,所述第一聚乙烯醇的粘度为2.4~12.3mpas,所述第二聚乙烯醇的粘度为17.2~42.6mpas;
所述聚乙烯吡咯烷酮包括第一聚乙烯吡咯烷酮和第二聚乙烯吡咯烷酮,所述第一聚乙烯吡咯烷酮的分子量为3500~150000,所述第二聚乙烯吡咯烷酮的分子量为210000~1500000。
可选的,所述第一聚乙烯吡咯烷酮的粘度为0.8-10.6mpas,所述第二聚乙烯吡咯烷酮的粘度为16.5-88.4mpas。
可选的,所述A组分包括以下重量组分:
聚乙烯醇2~6份、聚乙烯吡咯烷酮9.5~15份、羟乙基纤维素5~7份;
其中,第一聚乙烯醇0.5~2.5份,第二聚乙烯醇1.5~3.5份;
第一聚乙烯吡咯烷酮2.5~5份,第二聚乙烯吡咯烷酮7~10份。
可选的,所述A组分还包括35~49重量份的第一溶剂。
可选的,所述可溶性微针还包括以下B组分:
糖类、缓冲剂和盐。
可选的,所述B组分包括以下重量组分:
糖类11~16份、缓冲剂0.3~0.8份、盐0.5~2份。
可选的,所述糖类包括1~3重量份的蔗糖和10~13重量份的海藻糖,所述B组分还包括0.002~0.01重量份的氢氧化钠和14~17重量份的第二溶剂。
可选的,所述A组分和所述B组分的重量比为(16.5~77):(11.8~35.81)。
可选的,所述微针制剂还包括有药物活性成分,所述A组分和所述药物活性成分的重量比为(16.5~77):(0.6~22.46)。
另一方面,本发明提供了一种微针贴片的制备方法,包括以下操作步骤:
获取药物活性成分和如权利要求1~8任意一项所述的微针制剂,制备用于微针成型的成型液;
将成型液施加于微针成型模具上,脱水至含水量低于15%,脱模得到微针贴片;
将微针贴片装入保护件中,并将其置于加有脱水剂的密封袋中,使微针贴片的含水量降低于3%。优选的,微针贴片的含水量低至1%以下。
可选的,所述密封袋的水汽透过率低于0.8g/㎡·24h。
可选的,所述密封袋为双向拉伸聚丙烯薄膜、聚对苯二甲酸乙二酯、氯化聚丙烯树脂、铝箔中的一种或多种混合膜。
可选的,所述泡罩和/或贴板的材质为水汽透过率为1~20g/㎡·24h的阻隔性材料。
可选的,所述泡罩的材质为聚氯乙烯或聚对苯二甲酸乙二醇酯。
可选的,所述贴板的材质为纺粘型烯烃或铝塑膜。
根据本发明提供的微针制剂,发明人通过大量试验发现,通过将特定分子量的羟乙基纤维素、特定粘度的第一聚乙烯醇和第二聚乙烯醇以及特定分子量的第一聚乙烯吡咯烷酮和第二聚乙烯吡咯烷酮进行组合作为微针制剂中起主要强度支撑作用的骨架材料,能够有效提高该微针制剂制备得到的微针的结构强度,进而在受到较大压力的作用下不发生断裂,进而能够刺入皮肤深处,提高给药质量;上述粘度范围内的第一聚乙烯醇、第二聚乙烯醇和上述分子量范围的第一聚乙烯吡咯烷酮、第二聚乙烯吡咯烷酮之间的配合,推测是由于聚乙烯醇和聚乙烯吡咯烷酮之间相互交缠,且其分子链的长度差异起到互补作用,进而使第一聚乙烯醇、第二聚乙烯醇、第一聚乙烯吡咯烷酮和第二聚乙烯吡咯烷酮之间相互交缠以形成更为稳固的支撑骨架,提高了微针的整体强度。

为了使本发明所解决的技术问题、技术方案及有益效果更加清楚明白,以下结合附图及实施例,对本发明进行进一步详细说明。应当理解,此处所描述的具体实施例仅仅用以解释本发明,并不用于限定本发明。
本发明的描述中,术语“分子量”指的是重均分子量。
术语“阻隔性材料”中的“阻隔性”指的是一定厚度的塑料制品,在一定的压力(1mPa)、一定的温度(23℃)、一定的湿度,单位时间(24h)、单位面积(1㎡)内透过小分子物质的体积或重量。
本发明一实施例提供了一种微针制剂,包括以下A组分:
聚乙烯醇和聚乙烯吡咯烷酮;
其中,其中,聚乙烯醇中包括第一聚乙烯醇和第二聚乙烯醇,所述第一聚乙烯醇的粘度为2.4~12.3mpas,所述第二聚乙烯醇的粘度为17.2~42.6mpas;
所述聚乙烯吡咯烷酮包括第一聚乙烯吡咯烷酮和第二聚乙烯吡咯烷酮,所述第一聚乙烯吡咯烷酮的分子量为3500~150000,所述第二聚乙烯吡咯烷酮的分子量为210000~1500000。
发明人通过大量试验发现,通过将特定分子量的羟乙基纤维素、特定粘度的第一聚乙烯醇和第二聚乙烯醇以及特定分子量的第一聚乙烯吡咯烷酮和第二聚乙烯吡咯烷酮进行组合作为微针制剂中起主要强度支撑作用的骨架材料,能够有效提高该微针制剂制备得到的微针的结构强度,进而在受到较大压力的作用下不发生断裂,进而能够刺入皮肤深处,提高给药质量;上述粘度范围内的第一聚乙烯醇、第二聚乙烯醇和上述分子量范围的第一聚乙烯吡咯烷酮、第二聚乙烯吡咯烷酮之间的配合,推测是由于聚乙烯醇和聚乙烯吡咯烷酮之间相互交缠,且其分子链的长度差异起到互补作用,进而使第一聚乙烯醇、第二聚乙烯醇、第一聚乙烯吡咯烷酮和第二聚乙烯吡咯烷酮之间相互交缠以形成更为稳固的支撑骨架,提高了微针的整体强度。
在优选的实施例中,所述第一聚乙烯吡咯烷酮的粘度为0.8-10.6mpas,所述第二聚乙烯吡咯烷酮的粘度为16.5-88.4mpas。
在一些实施例中,所述A组分包括以下重量组分:
聚乙烯醇2~6份、聚乙烯吡咯烷酮9.5~15份、羟乙基纤维素5~7份;
其中,第一聚乙烯醇0.5~2.5份,第二聚乙烯醇1.5~3.5份;
第一聚乙烯吡咯烷酮2.5~5份,第二聚乙烯吡咯烷酮7~10份。
通过对不同粘度的第一聚乙烯醇、第二聚乙烯醇和不同分子量的第一聚乙烯吡咯烷酮和第二聚乙烯吡咯烷酮的添加量进行限制,有利于维持所述微针制剂得到的微针的整体强度,当第一聚乙烯醇、第二聚乙烯醇、第一聚乙烯吡咯烷酮和第二聚乙烯吡咯烷酮的添加量不在上述范围中时,会对所得到的微针的结构强度产生劣化作用。
在一些实施例中,所述A组分还包括35~49重量份的第一溶剂。
所述第一溶剂用于分散所述A组分中的第一聚乙烯醇、第二聚乙烯醇、第一聚乙烯吡咯烷酮和第二聚乙烯吡咯烷酮等。
由于第一聚乙烯醇、第二聚乙烯醇、第一聚乙烯吡咯烷酮和第二聚乙烯吡咯烷酮均为水溶性分子,在优选的实施例中,所述第一溶剂选自水,更优选地,所述第一溶剂选自注射用水。
由于所述A组分中含有较多高分子量的物质,因此,需要在加热条件下,优选在65℃~95℃的条件下对A组分进行溶解分散,为避免对A组分的加热导致一些温敏性物质的变性,在一些实施例中,所述可溶性微针还包括B组分,可将温敏性物质加入B组分中,待A组分降温后再进行混合。
需要说明的是,在其他实施例中,若所述微针制剂中没有温敏性物质,或是不对A组分进行加热处理,也可不特别进行组分区分,将本发明中所提及的A组分和B组分作为同一组分进行混合。
在一些实施例中,所述B组分包括糖类、缓冲剂和盐。
在一些实施例中,所述B组分包括以下重量组分:
糖类11~16份、缓冲剂0.3~0.8份、盐0.5~2份。
所述糖类可以是蔗糖和/或海藻糖,在微针贴剂中,主要用作稳定剂。是一种生物相容性佳、稳定性高、成本低、安全性好的碳水化合物。制备的微针机械强度佳,给药时能在皮肤中快速释放有效成分,当皮肤吸收蔗糖时,其在体内可生物降解的,肾脏会逐渐将其清除。
所述盐选自氯化钠。
在一些实施例中,所述缓冲剂选自枸橼酸钠,枸橼酸钠在微针贴剂中,主要与氯化钠配伍组成缓冲剂,防止制剂配制过程中的PH值出现较大的波动。
在一些实施例中,所述糖类包括1~3重量份的蔗糖和10~13重量份的海藻糖,所述B组分还包括0.002~0.01重量份的氢氧化钠和14~17重量份的第二溶剂。
其中,所述蔗糖也可采用其他单糖或二糖替代,如麦芽糖、乳糖、果糖等。
所述氢氧化钠用于调节所述微针制剂的pH值。
需要说明的是,在微针成型的过程中,需要通过脱水去除其中的溶剂,所述第一溶剂和第二溶剂在此时被大部分去除,同时脱水后残留有小部分的溶剂是允许的,但过量的溶剂残留会影响微针的强度。
在一些实施例中,所述A组分和所述B组分的重量比为(16.5~77):(11.8~35.81)。
在一些实施例中,所述微针制剂还包括有药物活性成分,所述A组分和所述药物活性成分的重量比为(16.5~77):(0.6~22.46)。
在一些实施例中,所述药物活性成分为小分子药或生物药,其中,生物药包括DNA、RNA等核酸药、蛋白质、多肽等。
所述药物活性成分可以选自天然存在、合成或重组产生的合适的蛋白质、肽及其片段。用于输送的API类型的代表性实例包括抗生素、抗病毒药、止痛药、麻醉药、抗组胺药、抗炎药、抗凝血剂、过敏原、维生素、抗肿瘤药;生物活性成分可以包括各类激素、用于传染病的疫苗、用于癌症、神经系统疾病、过敏、戒烟或其他成瘾的治疗性疫苗。列举流感抗原(流感疫苗)、HBs抗原(B型肝炎病毒表面抗原)、HBe抗原(乙型肝炎Be抗原)、BCG(Bacillede Calmette et Guerin)抗原、麻疹抗原、风疹抗原、水痘抗原、黄热病抗原、带状疱疹抗原、轮状病毒抗原、Hib(流感杆菌b型)抗原、狂犬病抗原、霍乱抗原、白喉抗原、百日咳抗原、破伤风抗原、灭活脊髓灰质炎抗原、日本脑炎抗原、人乳头瘤病毒抗原或上述病毒混合抗原等。
在一些实施例中,所述微针制剂用于制备一体式微针或分层式微针。
如图9所示,当制备的微针为一体式微针时,所述A组分、所述B组分和所述药物活性成分直接混合,得到用于制备微针的微针制剂。
如图10所示,当制备的微针为分层微针时,可先将所述A组分和所述B组分混合得到基膜液,在部分基膜液的基础上进一步加入药物活性成分得到针尖液,针尖液用于形成微针的针尖部分,基膜液用于形成微针的针体部分和基膜部分。
本发明的另一实施例提供了如上所述的微针制剂的制备方法,包括以下操作步骤:
将羟乙基纤维素、第一聚乙烯醇、第二聚乙烯醇、第一聚乙烯吡咯烷酮和第二聚乙烯吡咯烷酮加入第一溶剂中,通过加热分散,得到A组分。
在一些实施例中,加热分散的温度为65℃~95℃。
在一些实施例中,还包括以下操作:
将糖类、缓冲剂、氢氧化钠和盐溶解于第二溶剂中,得到B组分;
将A组分和B组分混合,得到微针制剂。
在一些实施例中,在所述微针制剂中还加入有药物活性成分。
在可溶性微针制备过程中的脱水阶段,为了防止微针受热融化或是药物活性成分等物质的受热失效,往往都是采用常温以下温度进行风干脱水,同时不能使用过快的风速,避免微针受风力形变。然而采用这样的脱水条件导致脱水速率过于缓慢,尤其是在微针含水量处于较低的水平时,水分子脱除速度下降,其脱水速度相比于前期更加缓慢,不能满足规模生产的需要,同时,制备得到的微针其含水量过低也不利于其结构强度的提升。
为解决上述问题,本发明的另一实施例提供了一种微针贴片的制备方法,包括以下操作步骤:
获取药物活性成分和如上所述的微针制剂,制备用于微针成型的成型液;
将成型液施加于微针成型模具上,脱水至含水量为3~15%,脱模得到微针贴片;
将微针贴片装入保护件中,并将其置于加有脱水剂的密封袋中,使微针贴片的含水量降低于3%。
所述密封袋水汽透过率低于0.8g/㎡·24h,可以为双向拉伸聚丙烯薄膜、聚对苯二甲酸乙二酯、氯化聚丙烯树脂、铝箔中的一种或多种混合膜。
通过将微针贴片预先在-40~35℃的温度下脱水至含水量为3~15%,能够有效降低初步脱水所用的时间,加快了制备速率,降低了生产成本,如需要进一步脱水所需要的时间边际递增,故以3-15%的湿度作为初步脱水的临界点,更重要的是,在该含水量条件下,得到的微针贴片未完全固化,具有一定的韧性,此时有利于微针贴片的脱模操作,避免微针贴片在脱模是由于自身刚性过高而断针于微针成型模具中,提高微针形态完整性。
经过初步脱水后再将微针贴片装入保护件中,并将其置于加有脱水剂的密封袋中,脱水剂透过保护件吸收微针贴片的水分,使微针贴片的含水量降低于3%,所述脱水剂为分子筛或硅胶等吸水性材料,保护件可以作为微针贴片的包装件,只需在保护件中继续脱水3~5天即可将微针贴片的含水量降至低于3%。微针贴片在此含水量下具有最优的结构强度,且不需要付出多余的时间和成本。
在一些实施例中,所述脱水操作为冷风脱水,具体的,在-40-35℃的温度下风干3-4h。同时,将风速控制在范围风速为0.5-5m/s内,这样既不会将成型液吹出微针成型模具,也不会因风速太低导致固化速率降低,影响生产效率。
在一些实施例中,所述保护件包括泡罩和贴板,所述泡罩设置于所述贴板上,所述微针贴片置于所述泡罩中。
所述泡罩与所述贴板贴合形成良好的保护结构,能够防止所述微针贴片受外力作用而损坏,同时,也起到了良好的阻隔效果,能将外界细菌、病毒等污染源隔离在外,满足GMP法规的无菌要求。
泡罩和/或贴板的材质为水汽透过率为1~20g/㎡·24h的阻隔性材料,在此范围的水汽透过率能够保证微针贴片相对较快的脱水速率,同时也能保障外界的细菌不易透过保护件造成污染。在一些实施例中,所述泡罩的材质为聚氯乙烯或聚对苯二甲酸乙二醇酯。在一些实施例中,所述泡罩的材质还可以为聚酯镀铝膜、聚乙烯、双向拉伸聚丙烯、铝箔、聚偏二氯乙烯涂布聚丙烯、镀铝未拉伸聚丙烯膜。
在一些实施例中,所述贴板的材质为纺粘型烯烃或铝塑膜。在一些实施例中,所述贴板的材质还可以为无纺布型高密度聚乙烯膜、聚酯镀铝膜、聚乙烯、双向拉伸聚丙烯、铝箔、聚偏二氯乙烯涂布聚丙烯、镀铝未拉伸聚丙烯膜。
在一些实施例中,制备的微针贴片为分层微针,所述微针贴片的制备过程包括:
所述成型液包括基膜液和针尖液,获取如上所述的微针制剂;取一部分所述微针制剂作为基膜液,取另一部分所述微针制剂加入药物活性成分作为针尖液;
将针尖液施加于微针成型模具的微针腔中,脱水收缩得到微针的针尖部分,继续施加基膜液于微针成型模具上,脱水得到连接所述针尖部分的针体部分以及连接所述针体部分的基膜,脱水至含水量为3~15%,脱模得到微针贴片;
将微针贴片装入保护件中,并将其置于加有脱水剂的密封袋中,使微针贴片的含水量降低于3%。
在一些实施例中,制备的微针贴片为一体式微针,所述微针贴片的制备过程包括:
获取如上所述的微针制剂;在所述微针制剂中加入药物活性成分作为成型液;
将成型液施加于微针成型模具上,脱水得到微针以及连接所述微针的基膜,脱水至含水量为3~15%,脱模得到微针贴片;
将微针贴片装入保护件中,并将其置于加有脱水剂的密封袋中,使微针贴片的含水量降低于3%。
在一些实施例中,所述微针贴片上微针为锥形结构,如圆尖锥、椭圆形尖锥、规则多边形尖锥、不规则多边形尖锥等。所述微针的高度为0.001μm-1000μm,最大直径为0.005-3000μm。同时,为了保证微针阵列具有一定密度,所述微针之间的距离为4μm-1000μm。
在一些实施例中,所述微针成型模具包括支撑板和多个微针成型区,所述微针成型区的形状与所需制备的微针贴片的基膜形状一致,多个所述微针成型区间隔嵌入于所述支撑板上,且所述支撑板为刚性材料,所述微针成型区为柔性透气材料。
在一些实施例中,所述刚性材料选自单晶硅、不锈钢、铝板、钛板、硅酸盐玻璃、石英玻璃、陶瓷、聚四氟乙烯、聚醚醚酮(PEEK)、丙烷磺酸吡啶嗡盐等,所述柔性材料选自硅氧烷。
本实施例采用柔性材料和刚性材料结合制备微针成型模具,通过将柔性材料的微针成型区固定于刚性材料的支撑板上,使得所述支撑板对于所述微针成型区具有支撑和固定作用,用于保持所述微针成型区形态的稳定性;所述微针成型区上设置的微针腔用于微针的固化成型,采用柔性材料有利于减小微针脱模过程中所受应力的作用,提高微针脱模后的完整性,同时,通过刚性材料的支撑板对微针成型区进行形态固定,可避免所述微针成型区在成型液固化收缩的过程中发生形变,进而有效改善微针的成型效果和脱模完整性。
在一些实施例中,将成型液施加于微针成型模具时,对所述微针成型模具进行抽真空处理,以促进填充也对于微针成型模具的填充速率,同时避免在成型的微针中残留气泡。
在不同实施例中,施加成型液时,可采用加压喷射、雾化喷射、辊涂、刷涂、注射、丝网印刷、刮涂的方式将成型液导入到所述微针成型模具中。
在优选的实施例中,采用刮涂的方式在所述微针成型模具中施加成型液。以下通过实施例对本发明进行进一步的说明。

实施例1
本实施例用于说明本发明公开的微针贴片及其制备方法,包括以下操作:
按重量份称取0.5份第一聚乙烯醇(粘度2.4mpas)、1.5份第二聚乙烯醇(粘度17.2mpas)、5份羟乙基纤维素、2.5份第一聚乙烯吡咯烷酮(分子量3500)和7份第二聚乙烯吡咯烷酮(分子量210000)加入15份水中,通过90℃下加热分散,得到A组分,并降温冷却。
按重量份称取1份蔗糖、10份海藻糖、0.3份枸橼酸钠、0.5份氯化钠和0.002份氢氧化钠溶解于12份水中,得到B组分;
将A组分和B组分混合,得到微针制剂。
获取如上所述的微针制剂;取一部分所述微针制剂作为基膜液,取另一部分所述微针制剂加入药物活性成分作为针尖液;
将针尖液刮涂于微针成型模具的微针腔中,脱水收缩得到微针的针尖部分,继续刮涂基膜液于微针成型模具上,脱水得到连接所述针尖部分的针体部分以及连接所述针体部分的基膜,初步脱水至含水量为3%,脱模得到微针贴片;
将微针贴片装于保护件中,并置于加有脱水剂的铝塑膜中继续脱水,静置3天后,检测得到微针贴片的含水量降低至1%。
实施例2~7
实施例2~7用于说明本发明公开的微针贴片及其制备方法,包括实施例1中大部分操作,其不同之处在于:
采用表1中实施例2~7所示的物质含量组分,且实施例2-7的首次脱水含水量和继续脱水后的含水量如表2所示。
对比例1~4
对比例1~4用于说明本发明公开的微针贴片及其制备方法,包括实施例1中大部分操作,其不同之处在于:
采用表1中对比例1~4所示的物质含量组分,且对比例1~4的首次脱水含水量和继续脱水后的含水量如表2所示。
对比例5
对比例5用于说明发明公开的微针贴片及其制备方法,包括实施例1中大部分操作,其不同之处在于:
对比例5只采用了初步脱水,没有使用具有透水性的保护件和脱水剂进行后续干燥操作,并直接将含水量降至1%,但消耗了不对等的时间。
性能测试
1、万能材料试验机直压以测试针强度:将上述实施例2~7和对比例1~4制备得到的微针贴片置于万能材料实验压盘上,启动仪器,压力传感器向下位移,当压力传感器检测到微针断裂时记录断裂点压力。压力传感器上压头直径8mm,约共压到194根针,得到如下实验结果:
可以看出,通过本发明提供方法制备得到的微针贴片明显具有更高的微针结构强度,而缺少第一聚乙烯醇、第二聚乙烯醇、第一聚乙烯吡咯烷酮和第二聚乙烯吡咯烷酮中任一种的对比例1~4均会导致微针强度的劣化,说明第一聚乙烯醇、第二聚乙烯醇、第一聚乙烯吡咯烷酮和第二聚乙烯吡咯烷酮在提高微针强度方面起到了较好的协同作用。
2、parafilm封口膜穿刺实验:选取实施例1和对比例1~4的微针贴片,将多层parafilm膜层叠,将微针贴针尖面朝向parafilm膜,置于万能材料试验机下,采用147N(约等于成人手指按压力度)力按压微针,使微针刺入parafilm内。实验后取出parafilm膜,采用红墨水浸透parafilm膜,通过考察红墨水是否能透过parafilm膜,判断微针穿刺至parfilm膜层数。
实施例1得到的结果如图1所示,对比例1~4得到的结果如图2所示。
由图1和图2可以看出,实施例1的微针贴片经过parafilm封口膜穿刺后,红墨水都能浸染至第四层,即可以判断微针在147N下穿刺至第四层parafilm膜。
对比例1~4都仅能穿透第三层,没有浸染第四层。
3、树叶穿刺:选取实施例1和对比例1~4的微针贴片,将微针贴片穿刺贴敷树叶(金钱树),完成后取下微针贴,置显微镜下观察树叶表面微针穿刺情况。
实施例1的微针贴片树叶穿刺结果如图3所示,对比例1~4的微针贴片树叶穿刺结果如图4所示,可以看出实施例1提供的微针贴片能够完全穿刺树叶,且刺入深度较深,而对比例1~2刺入深度较浅,3~4更浅一些。
实施例1的微针贴片在穿刺前的形态如图5所示,穿刺后的形态如图6所示;对比例1~4的微针贴片在穿刺前的形态如图7所示,穿刺后的形态如图8所示。
可以看出,实施例1的微针贴片在穿刺后仍看到部分完整的针形,而对比例1~4的微针贴片在穿刺后则基本没有完整的针形。
以上所述仅为本发明的较佳实施例而已,并不用以限制本发明,凡在本发明的精神和原则之内所作的任何修改、等同替换和改进等,均应包含在本发明的保护范围之内。
Claims (16)
1.一种微针制剂,其特征在于,包括以下A组分:
聚乙烯醇、聚乙烯吡咯烷酮以及羟乙基纤维素;
其中,聚乙烯醇中包括第一聚乙烯醇和第二聚乙烯醇,所述第一聚乙烯醇的粘度为2.4~12.3mpas,所述第二聚乙烯醇的粘度为17.2~42.6mpas;
所述聚乙烯吡咯烷酮包括第一聚乙烯吡咯烷酮和第二聚乙烯吡咯烷酮,所述第一聚乙烯吡咯烷酮的分子量为3500~150000,所述第二聚乙烯吡咯烷酮的分子量为210000~1500000。
2.根据权利要求1所述的微针制剂,其特征在于,所述第一聚乙烯吡咯烷酮的粘度为0.8-10.6mpas,所述第二聚乙烯吡咯烷酮的粘度为16.5-88.4mpas。
3.根据权利要求1所述的微针制剂,其特征在于,所述A组分包括以下重量组分:
聚乙烯醇2~6份、聚乙烯吡咯烷酮9.5~15份、羟乙基纤维素5~7份;
其中,第一聚乙烯醇0.5~2.5份,第二聚乙烯醇1.5~3.5份;
第一聚乙烯吡咯烷酮2.5~5份,第二聚乙烯吡咯烷酮7~10份。
4.根据权利要求1所述的微针制剂,其特征在于,所述A组分还包括35~49重量份的第一溶剂。
5.根据权利要求1所述的微针制剂,其特征在于,所述可溶性微针还包括以下B组分:
糖类、缓冲剂和盐。
6.根据权利要求5所述的微针制剂,其特征在于,所述B组分包括以下重量组分:
糖类11~16份、缓冲剂0.3~0.8份、盐0.5~2份。
7.根据权利要求6所述的微针制剂,其特征在于,所述糖类包括1~3重量份的蔗糖和10~13重量份的海藻糖,所述B组分还包括0.002~0.01重量份的氢氧化钠和14~17重量份的第二溶剂。
8.根据权利要求5所述的微针制剂,其特征在于,所述A组分和所述B组分的重量比为(16.5~77):(11.8~35.81)。
9.根据权利要求1所述的微针制剂,其特征在于,所述微针制剂还包括有药物活性成分,所述A组分和所述药物活性成分的重量比为(16.5~77):(0.6~22.46)。
10.一种微针贴片的制备方法,其特征在于,包括以下操作步骤:
获取药物活性成分和如权利要求1~8任意一项所述的微针制剂,制备用于微针成型的成型液;
将成型液施加于微针成型模具上,在-40~35℃的温度下脱水至含水量为3~15%,脱模得到微针贴片;
将微针贴片装入保护件中,并将其置于加有脱水剂的密封袋中,使微针贴片的含水量降低于3%。
11.根据权利要求10所述的制备方法,其特征在于,所述保护件包括泡罩和贴板,所述泡罩设置于所述贴板上,所述微针贴片置于所述泡罩中。
12.根据权利要求10所述的制备方法,其特征在于,所述密封袋的水汽透过率低于0.8g/㎡·24h。
13.根据权利要求12所述的制备方法,其特征在于,所述密封袋为双向拉伸聚丙烯薄膜、聚对苯二甲酸乙二酯、氯化聚丙烯树脂、铝箔中的一种或多种混合膜。
14.根据权利要求11所述的制备方法,其特征在于,所述泡罩和/或贴板的材质为水汽透过率为1~20g/㎡·24h的阻隔性材料。
15.根据权利要求14所述的制备方法,其特征在于,所述泡罩的材质为聚氯乙烯或聚对苯二甲酸乙二醇酯。
16.根据权利要求12所述的制备方法,其特征在于,所述贴板的材质为纺粘型烯烃或铝塑膜。
